Establishment of the Successive Rearing System of Brush-footed Butterflies (Lepidoptera: Nymphalidae) (네발나비과 나비류의 계대사육법 체계확립)

  • In order to establish the successive rearing system brush-footed butterflies (Lepidoptera : Nymphalidae) were reared in a room. Artificial diets were developed for a year-round rearing. Bu-diet was best to rear these butterflies among 3 kinds of diet used. The freeze-dried host plant leaf powder in diet was better than heat-dried one $(60^{\circ}C)$ in the growth of larvae. The rearing results were best in the diet C/N ratio was 1:1. The 24-hrs old eggs could be stored for 5 days at $15^{\circ}C$ or for 3 days at $5^{\circ}C$ and showed 75% of hatchability. On the other hand, pupae could be stored for maximum 15 days at $15^{\circ}C$ because the emergence of abnormal adults appeared much more as the cold storage period got longer. And the adult was able to be stored until 60 days at refrigerator without relation of nectar-sucking period before cold-storage and storage temperature. Also a simple artificial ovipositing kit was devised by ${\Phi}9$ cm of petri-dish and a female oviposited $278{\pm}27$ of eggs with adding the ether extract of host plant to this kit. The systematic successive rearing method of brush-footed butterflies in a room was completed.

Species Identification and Developmental Biology of a Red Bean Pest in Ostrinia sp. (Lepidoptera: Crambidae) (Ostrinia속(나비목: 포충나방과) 팥 해충의 종 동정과 발육 특성)

  • Ostrinia larvae feed the pods and stem of red bean and seriously damage the bean production from farmers. In this study we investigated biological and developmental characteristics including field collection, host feeding preference, artificial rearing diet, morphological and molecular taxonomical identification, and pheromone analysis for an Ostrinia sp. in Korea. The male adults have massive tibia in the middle legs and 3-lobed uncus in the genitalia. The partial nucleotide sequences of mitochondrial cytochrome oxidase I (COI) and II (COII) were not corresponded to those DNA sequences from other Ostrinia species reported previously in Japan and China. Host plants for this species are also different from the previous species reported. In the gas chromatography (GC) analyses, (Z)-9-tetradecenyl acetate was not detected from the pheromone gland of our species while the component as a sex pheromone was found in O. zaguliaevi and O. zealis, With taken results, we conclude this Ostrinia species in Korea is Ostrinia scapulalis or closely related species. When larvae collected in a fall were incubated in the outdoor condition, they emerged to adult between June and July in the next year. The result indicates that the winter diapause could be started in late larval stage. In addition, we developed a semi-synthetic artificial diet adopted for mass rearing of the O. scapulalis in laboratory.

Root Rot of Moth Orchid Caused by Fusarium spp.

  • Moth orchid plants with yellowing blight and root rot symptoms were collected, and a total of 54 isolates of Fusarium spp. was obtained from roots and leaf bases of the diseased plants. The isolates were identified based on their morphological characteristics. Out of the 54 isolates of Fusarium spp., 42 isolates were identified as F. solani, 5 isolates as F. oxysporum, and 7 as F. proliferatum. Isolates of the three Fusarium spp. were tested for pathogenicity to moth orchid plants by artificial inoculation. All the Fusarium spp. induced root rot of the host plants. The symptoms progressed up to the basal part of the leaves, which later caused yellowing blight. The symptoms induced on the plants by artificial inoculation with the Fusarium spp. isolates were similar to those observed in greenhouses. The present study reveals that F. oxysporum, F. proliferatum, and F. solani cause root rot of moth orchid, and that F. solani is the main pathogen of the disease.

  • The purpose of this study was designed to compare the storage stage of spermatozoa in the utero-vaginal (U-V) glands and the infundibular glands of high-fecundity hens. These laying hens were assigned to four groups by date intervals after last artificial inseminations. The U-V glands and the infundibular glands in the tissue preparation of the each hen uterus were observed microscopically, and also the appearance rates of spermatozoa-contained U-V glands were compared with those of the infundibular glands. 1. The appearance rates of spermatozoa-contained U-V glands were found to be 27.8, 28.8, 19.9 and 11.1% respectively at the hen groups of 1, 3, 7 and 10 days after artificial insemination. 2. The appearance rates of spermatozoa-contained infundibular glands were found to be 0.5, 1. 1, 0.6 and 0.4% of 1, 3, 7 and 10 days after AI and number of spermatozoa contained in a infundibular glands tended predominantly to be 1 to 2. So this study concluded as follows: The appearance rates of spermatozoa-contained glands were found to be higher in the U-V glands than in the infundibular glands and also spermatozoa number per gland were more numerous in U-V glands than in infundibular glands.

  • Being recently increased interests of our healthcare, a host of symptoms-diagnostic sites has been introduced on the World Wide Web. But conventional healthcare sites provide users with only a very restricted functions. In this paper, we propose the use of Artificial Neural Networks (ANNs) as a flexible symptoms-diagnostic tool that enables learning effects of ANNs (not expert's knowledge) to be incorporated into the diagnostic process. We develop a novel algorithm for predicting patient\`s disease that satisfy user (or expert)-specified symptoms on WWW. Our algorithm provides two important benefits : 1) enables users (patients) to be taken early diagnostic, and 2) enables experts to perform confidently diagnostic by referencing the predicted diseases-list with its respective possibility.

  • Gas piping systems in power plants and factories are always influenced by the mechanical vibrations of rotational machines such as pumps, blowers, and compressors. Unusual vibrations in a gas piping system influence possible leakages of liquids or gases, which can lead to large explosive accidents. Real-time measurements of unusual vibrations in piping systems in situ prohibit them from being possible leakages owing to the repeated fatigue of vibrations. In this paper, a non-contact 3-dimensional measurement system that can detect the vibrations of a solid body and monitor its vibrational modes is introduced. To detect the displacements of a body, a stereoscopic camera system is used, through which the major vibration types of solid bodies (such as X-axis-major, Y-axis-major, and Z-axis-major vibrations) can be monitored. In order to judge the vibration types, an artificial neural network is used. The measurement system consists of a host computer, stereoscopic camera system (two-camera system, high-speed high-resolution camera), and a measurement target. Through practical application on a flat plate, the measured data from the non-contact measurement system showed good agreement with those from the original vibration mode produced by an accelerator.

Occurrence of Dry Rot on Cymbidium Orchids Caused by Fusarium spry. in Korea

  • Cymbidium orchids with blight and rot symptoms were collected, and a total of 63 isolates of Fusarium app. was obtained from pseudobulbs, roots, and leaves of the diseased plants. The isolates were identified based on their morphological characteristics. Out of the 63 isolates of Fusatium sup., 51 isolates were identified as F. oxysporum, 10 isolates as F. solani, and the rest as F. proliferatum. F. oxysporum was isolated from all the Cymbidium spp., while F. solani and F. proliferatum were isolated only from Cymbidium ensifolium and C. ginatum, respectively. Isolates of the three Fusarium spp. were tested for pathogenicity to their hosts by artificial inoculation. The strongly pathogenic isolates of Fusarium spp. induced severe dry rot of pseudobulbs and roots of the host plants. The symptoms progressed up to the basal part of the leaves, which later caused blight of the entire plant. The dry root symptoms induced on the plants by artificial inoculation with the isolates of Fusarium app. were similar to those observed in the growers'greenhouses. This is the first report of dry rot of Cymbidium spp. caused by F. oxysporum, F. solani, and F. proliferatum in Korea.

Foot Rot of Bok Choy and Kale Caused by Rhizoctonia solani AG-2-1 in Korea

  • Foot rot symptoms were occasionally observed on young bok choy (Brassica rapa subsp. chinensis) and kale (B. oleracea var. viridis) plants grown in vinyl greenhouses located in Icheon and Yangpyeong, Gyeonggi Province, Korea. These observations were made during disease surveys in April 2020. The incidence of diseased plants in the vinyl greenhouses investigated was 0.5-1.0% in bok choy and 0.5-5.0% in kale. Five isolates of Rhizoctonia sp. were obtained from diseased roots of bok choy and three isolates of Rhizoctonia sp. were taken from diseased stems of kale. All the Rhizoctonia sp. isolates were identified as Rhizoctonia solani AG-2-1 based on the morphological characteristics and anastomosis test. Three isolates each of R. solani AG-2-1 from bok choy and kale were tested for pathogenicity in their host plants by artificial inoculation. The tested isolates induced foot rot symptoms on the inoculated bok choy and kale plants. The symptoms on the bok choy and kale, induced by the artificial inoculation, were similar to those observed on plants from the vinyl greenhouses that were investigated. This is the first report of R. solani AG-2-1 causing foot rot in bok choy and kale in Korea.

  • Integrated pest management is essential for controlling plant diseases that reduce crop yields. Rapid diagnosis is crucial for effective management in the event of an outbreak to identify the cause and minimize damage. Diagnosis methods range from indirect visual observation, which can be subjective and inaccurate, to machine learning and deep learning predictions that may suffer from biased data. Direct molecular-based methods, while accurate, are complex and time-consuming. However, the development of large multimodal models, like GPT-4, combines image recognition with natural language processing for more accurate diagnostic information. This study introduces GPT-4-based system for diagnosing plant diseases utilizing a detailed knowledge base with 1,420 host plants, 2,462 pathogens, and 37,467 pesticide instances from the official plant disease and pesticide registries of Korea. The AI plant doctor offers interactive advice on diagnosis, control methods, and pesticide use for diseases in Korea and is accessible at https://pdoc.scnu.ac.kr/.

  • Due to the inconsistent fluctuation of blood supply for transfusion, much attention has been paid to the development of artificial blood using other animals. Although mini-pigs are candidate animals, contamination of mini-pig T cells in artificial blood may cause a major safety concern. Therefore, it is important to analyze the cross-reactivity of IL-7, the major survival factor for T lymphocytes, between human, mouse, and mini-pig. Thus, we compared the protein sequences of IL-7 and found that porcine IL-7 was evolutionarily different from human IL-7. We also observed that when porcine T cells were cultured with either human or mouse IL-7, these cells did not increase the survival or proliferation compared to negative controls. These results suggest that porcine T cells do not recognize human or mouse IL-7 as their survival factor.
