인진(茵蔯)의 에탄올 추출물이 담도암 세포주 SNU-1196의 apoptosis에 미치는 효과에 관한 연구 (Study on the Pro-apoptotic Effect of Artemisiae Capillaris Herba Extracted with Ethanol on Biliary Tract Cancer Cell Line, SNU-1196)

  • Objectives : This study was performed to elucidate the pro-apoptotic effect of Artemisiae capillaris herba extracted with ethanol on biliary tract cancer cells. Materials and Methods : The biliary tract cancer cell line SNU-1196 was used in this study. Cells were treated with different concentrations of Artemisiae capillaris herba for 24, 48 and 72 hours. After the treatment, cell viability, apoptosis, caspase activities and the mRNA expressions of the Bcl-2, Bax, P53, and P21 were measured by using MTT assay, cell cycle analysis, apoptosis assay, and RT-PCR. The cell cycle analysis was done by flow cytometry and apoptosis assay by cell death detection ELISA kit. Results : Artemisiae capillaris herba inhibited proliferation of SNU-1196 in long-time culture group with dose-dependent manner. All cells treated with Artemisiae capillaris herba showed increased apoptosis with dose- and time-dependent manner. Exposure of SNU-1196 to Artemisiae capillaris herba induced caspase-3 activation. However, apeoptosis was blocked when SNU-1196 was treated together with the pan-caspase inhibitor Z-VAD-FMK and the caspase-3 inhibitor Z-DEVE-FMK. After the treatment of Artemisiae capillaris herba, the mRNA expressions of caspase -3, -8, -9, p53, and p21 was increased in all cells. Artemisiae capillaris herba resulted in a significant decrease in Bcl-2 and an increase in Bax mRNA levels. Conclusions : These results suggest that Artemisiae capillaris herba would be beneficial in the treatment of biliary tract cancer.

인진(茵蔯)이 간성상세포의 섬유화 억제에 미치는 영향 (Inhibitory Effect of Artemisiae Capillaris Herba on Fibrogenesis in Rat Hepatic Stellate Cells)

  • Objectives : This study was performed to investigate the anti-fibrogenic effect of Artemisiae Capillaris Herba on cultured rat hepatic stellate cells. Materials and Methods : Hepatic stellate cells(HSC-T6) were treated with various concentrations of Artemisiae Capillaris Herba extract for 24 hours. The extraction was done either with distilled water or 50% EtOH. After the treatment, cell viability, proliferation, procollagen levels and the mRNA of the collagen type 1a2 and ASMA were measured by using MTT assay, BrdU assay, RT-PCR, and Procollagen Type I C-peptide EIA Kit. Results : The viability and proliferation of the hepatic stellate cells were decreased as the concentration increased. The mRNA expression decreased consistently with the volume of the secreted procollagen with the extraction made with distilled water, which indicates the herb has inhibitory effect on fibrogenesis of the liver by regulating one of the fibrosis associated genes in transcription. However, it increased in 50% EtOH extraction, which shows that a more stable reaction is expected of the extraction made with distilled water than the extraction made with 50% EtOH. The production of procollagen was decreased by a low-concentration treatment with Artemisiae Capillaris Herba, but increased by a high concentration. It seemed that the cells were responding to Artemisiae Capillaris Herba in low- concentrations, thus producing small amounts of collagen. When the drug was administered at high enough concentration to give direct toxicity to cells, the ability of cells to produce collagen was activated, and the overproduction of collagen was observed as an undesirable results. Conclusion : These results suggest that Artemisiae Capillaris Herba is beneficial in the treatment of cirrhotic patients as well as for the patients with chronic hepatitis when extracted with water in the proper concentrations.

인진(茵蔯) 에탄올추출물이 ${\alpha}$-MSH로 유도된 과색소 형성에 미치는 영향 (Effect of the Ethanol Extract of Artemisiae Capillaris Herba on the Hyperpigmentation Induced by ${\alpha}$-MSH)

  • Melanogenesis is induced mainly by ultraviolet radiation of sunlight and ${\alpha}$-Melanocyte stimulation hormone (${\alpha}$-MSH) which binds to a specific G protein coupled receptor. ${\alpha}$-MSH and cAMP-elevating agents are known to melanin syntheisis and dendrite outgrowth. The purpose of this study was to investigate the mechanism of melanogenesis inhibition in B16/F10 cells by ethanol extract of Artemisiae Capillaris Herba. In the present study, ${\alpha}$-MSH led to a stimulation of melanin synthesis that appeared to result from an increased tyrosinase activity and melanin content. However, the ethanol extract of Artemisiae Capillaris Herba inhibited the ${\alpha}$-MSH-induced tyrosinase activity and melanin content. In control conditions, B16/F10 cells displayed a fibroblastic appearance while ${\alpha}$-MSH treatment promoted the emergence of small and numerous dendrites from the plasma membrane. The ethanol extract of Artemisiae Capillaris Herba abolished the ${\alpha}$-MSH-induced dendricity. Regarding protein levels of the melanogenic enzymes, the amounts of tyrosinase were increased after incubation with ${\alpha}$-MSH. The treatment of Artemisiae Capillaris Herba ethanol extract decreased the ${\alpha}$-MSH expression levels of tyrosinase. Based on these findings, it is likely that the ethanol extract of Artemisiae Capillaris Herba exerts its depigmenting effects in B16/F10 cells through the suppression of tyrosinase expression, which are key enzymes for melanogenesis.

인진과 황련 추출물의 췌장암 세포주 MIA PaCa-2에 대한 세포사멸 효과 (Apoptotic Effect of ARTEMISIAE CAPILLARIS HERBA and COPTIDIS RHIZOMA Extracts on MIA PaCa-2 Cells)

  • Purpose : The purpose of this study is to investigate the effect of ARTEMISIAE CAPILLARIS HERBA and COPTIDIS RHIZOMA Extracts on cell death in pancreatic cancer cells. Method : Human-derived pancreatic cancer cell line, MIA PaCa-2 cells were treated by Prescription A with various concentrations and the cytotoxicity was determined by MTT assay. To investigate the effects of Prescription A on pancreatic cancer cells, the staining of Annexin V/PI, cell cycle arrest, nuclear chromatin condensation and the production of reactive oxygen species (ROS) were examined. The effect of Prescription A's effective components, ARTEMISIAE CAPILLARIS HERBA and COPTIDIS RHIZOMA Extracts on cell death were also observed. Results : The viability of MIA PaCa-2 cells treated with Prescription A were decreased in a dose dependent manner. Prescription A induced cell death in MIA PaCa-2 cells as shown by result of Annexin V/PI double staining, chromatin condensation and cell cycle arrest. In addition, production of ROS was increased by Prescription A treatment, suggesting that ROS induced by Prescription A mediated cell death. Furthermore, Prescription A's effective components, ARTEMISIAE CAPILLARIS HERBA and COPTIDIS RHIZOMA Extracts were also induced apoptosis of MIA PaCa-2 cells through ROS production. Conclusion : These results suggest that Prescription A's effective components, ARTEMISIAE CAPILLARIS HERBA and COPTIDIS RHIZOMA Extracts induced death of MIA PaCa-2 through ROS production.

Franz Diffusion Cell을 이용한 인진호 함유 경피제의 피부투과 특성 연구 (Permeation Characteristics of Transdermal Preparations Containing Artemisiae Capillaris Herba in Franz Diffusion Cell)

  • Artemisiae Capillaris Herba is a dried aerial part of Asteraceae capillaris Thunb.(Compositae), which has been used in Korean traditional medicine for the treatment of various diseases. It has a variety of pharmacological activities and has been evaluated for potential as an active ingredient in cosmeceutical products. In the cosmetics industry, animal experiments is besides the major concern of ethics, there are few more disadvantages of animal experimentation like demand of skilled manpower, time consuming protocols and high cost. Therefore, various alternatives to animal experiments have been proposed. The purpose of this study was to investigate the skin permeation characteristics of chlorogenic acid and dimethyleculetin, which are constituent of Artemisiae Capillaris Herba by using Franz diffusion cell. As a result, skin permeability was characterized by flux(penetration rates) and $K_p$(permeability coefficient) value, chlorogenic acid had lower flux and $K_p$ than dimethylesculetin. According to the definitions of Marzulli, chlorogenic acid and dimethylesculetin would be classified as 'Moderate' and 'Very fast' respectively. In conclusion, skin permeation characteristics of chlorogenic acid and dimethylesculetin were confirmed through Franz diffusion cell, and suggests the direction of alternative method for skin permeation of natural compounds.

인진(茵蔯)의 생식.발생 독성 시험 -배자(胚子)및 태자(胎子)발생을 중심으로- (Terato Genicity Test of Oral Administration of Artemisiae capillaris herba Extracts on New Zealand White Rabbit)

  • Objectives : The objective of this study was to access the effect of Artemisiae capillaris herba Extracts on the organogenetic reproductive toxicity of New Zealand White rabbits when administered by oral gavage. Methods : Gestational New Zealand White rabbits were dosed from 6 days of gestation to 18 days of gestation. It was conducted in accordance with the recommendations of the KFDA Guideline for Detection of Toxicity to Reproduction for Medicinal Products. In addition, serum AST, ALT, LDH and ALP levels were detected with their necropsy and histopathological observation Results: No significant changes of body weights, gains and food consumption were demonstrated in all dosed groups compared to those of vehicle control group except for 2,000mg/kg-dosing group, which showed significantly increase of body weight compared to that of vehicle control group. Normal gross findings were demonstrated in all tested groups. No significant changes of number of corpora lutea, implantation, implantation rate, number of fetal death, loss rate of post-implantation, number of live youngs at C-section, survival rate of post-implantation, number of male live youngs at C-section, number of female live youngs, sex ratio of live young, external anomalies of live youngs and body weight of live youngs were demonstrated in all dosed groups compared to those of vehicle control group. Although some visceral variations such as thymic reminant in the neck, dilation of renal pelvis, dilated ureter and malpositioned left common carotid artery, no significant changes were demonstrated in all dosed groups compared to that of vehicle control group. Some malformations and variation were demonstrated with retardations. However, no significant changes were demonstrated in all dosed groups compared to that of vehicle control group. In addition, similar ossification numbers were detected in sternebrae, metacarpals, metatarsals, phalanges, cervical vertebrae, and sacral and caudal vertebrae. No Artemisiae capillaris herba Extracts treatment-related changes of serum AST, ALT, LDH and ALP levels were demonstrated in all dosed levels in this study. Conclusions : We could conclude that oral administration of Artemisiae capillaris herba Extracts hasn't an influence on the organogenetic reproductive toxicity.

인진의 안정성에 대한 연구 - 주산$\cdot$수유기 생식 독성을 중심으로 - (Studies on the Safety of Artemisiae Capillaris Herba - With the Perinatal and Lactational Reproductive Toxicity -)

  • Objectives: To study the effect of Artemisiae Capillaris Herba extracts, that have been used as oriental medicine to treat liver disease, on the perinatal and lactational n;)productive toxicity of SD rats when administered by oral lavage. Methods: Female SD rats were dosed from 6 days of gestation to 3 weeks postpartum. This was conducted in accordance with the recommendations of the KFDA Guidelines for Detection of Toxicity to Reproduction for Medicinal Products. Results: No Artemisiae Capillaris Herba extracts treatment-related changes in clinical signs, mortalities, implantation number, dead fetus number, loss rate of fetus, number of live young, survival rate of fetus, sex ratio of live young, external anomalies, pregnancy periods, viability index, lactational index, survival rate of litters at 4 days after birth or delivery index were demonstrated in any dosed levels in this study. However, the body weight and gains, food consumption and absolute organ weights of brain, adrenal glands, liver, spleen, kidney, ovaries and heart were significantly increased in 2000 or 1000mg/kg-dosing groups and the relative organ Weights of adrenal glands were significantly increased in 2,000mg/kg-dosing groups. Therefore, it was concluded that this increase was natural according to growth. Also, no changes of gross findings, clinical signs, mortalities, body weight and gains, physical development results, necropsy findings, organ weight, faculty test, open filed test and water-filled simple T-maze test, copulation, fertility, pregnancy indices, body weight and gains during gestation periods, necropsy findings, corpora lutea number, implantation number, implantation rate, dead fetus number, post-implantation loss rate, live young, post-implantation survival rate, sex ratio of live young, external anomalies and individual body weights of live young were demonstrated in any dosed levels in this study. Conclusions: It is considered that the NOAEL (No-Observed-Adverse-Effect Level) for perinatal and lactational reproductive toxicity of Artemisiae Capillaris Herba extracts was up to 2000mg/kg/day because no changes of other perinatal and lactational reproductive indices were demonstrated.

주상에 활용되는 수종의 한약물이 알콜올대사 및 간장해에 미치는 영향 (The Effects of Some Oriental Herbs Which Have Been Used in the Treatment of Alcoholic Diseases on Alcoholic Metabolism and Alcoholic Liver Damages)

  • Objectives : This experiment was conducted to verify the effects of some oriental herbs(Alny Cortex et Ramulus, Artemisiae Capillaris Herba, Aurantii Nobilis Pericarpium, Giseng Radix, Hoveniae Semen, Puerariae Flos, Puerariae Radix, and Xanthii Fructus) which have been used in the treatment of alcoholic diseases, on alcoholic metabolism, and on alcoholic liver damage. Methods : The effects of the herbs on the activities of alcohol dehydrogenase(ADH), aldehyde dehydrogenase(ALDH) were evaluated and their protective effects of liver function and cells from alcoholic damage were analysed. For the evaluation of the protective effects, the levels of glucose, triglyceride, BUN, AST, and ALT in serum of rats were measured. Results and Conclusions : It is concluded that Puerariae Radix interferes with the ADH activity directly, thereby reducing the toxicity of alcohol, resulting in enhancing alcohol-tolerance and protecting liver functions. Also Artemisiae Capillaris Herba interferes both ADH and ALDH activities. Isolation of the biologically active compounds from Puerariae Radix and its detailed characterization are matters for future research.

인진이 간성상세포의 섬유화 억제에 미치는 영향에 대한 연구 (Inhibitory Effect of Artemisiae Capillaris Herba on Fibrogenesis in Primary Cultured Rat Hepatic Stellate Cells)

  • Object : This study was performed to investigate the anti-fibrogenic effect of Artemisiae Capillaris Herba(ACH) on cultured rat hepatic stellate cells. Methods : Hepatic Stellate Cells were obtained from a 350gm Sprague-Dawley rat by tissue perfusion system, and the cells for the study were selected after 3 passages of culture on non-coated plastic culture dishes which enable the cells to activate, thus producing collagens in the cell media. Cells were treated with various concentrations of Artemisia Capillaris Herba(ACH) extract powder for 24 or 48 hours. After the treatment, Soluble collagen, procollagen levels and the mRNA of the procollagen type I C were measured by using assay kit and RT-PCR method. Results : Procollagen production by the hepatic stellate cells decreased after the treatment in a time-dependent dose-dependent manner. The mRNA expression decreased consistently with the volume of the secreted procollagen which indicates the herb hat inhibitory effect on fibrogenesis of the liver by regulating one of the fibrosis associated genes in transcription. Conclusion : These results suggest that ACH is beneficial in the treatment of cirrhotic patients as well as for the patients with chronic hepatitis.

Nuclear Factor kappa B 억제를 통한 인진추출물의 inducible Nitric Oxide synthase 및 Cytokine 억제효과 (Anti-inflammatory effects of Herba Artemisiae Capillaris as a consequence of the inhibition of NF-kappa B-dependent iNOS and pro-inflammatory cytokines production.)

  • Herba Artemisiae Capillaris is the dried bud of Artemisia capillaris Thunb, which has been used for expelling heat to loosen the bowels and normalizing gallbladder function to cure jaundice in traditional oriental medicines. In the present study, we evaluated the anti-inflammatory effects of the aqueous extracts of Herba Artemisiae Capillaris (HAC) in LPS-activated Raw 264.7 cells. Cells were treated with $1\;{\mu}g/ml$ of LPS 1 h before adding HAC extract. Cell viability was determined by MTT assay, and the relative level of NO was measured with Griess reagent. TNF-$\alpha$, IL-$1{\beta}$, and IL-6 cytokines were detected by ELISA. During the entire experimental period, all three doses of HAC extract (0.03, 0.10 and 0.30 mg/ml) had no significant cytotoxicity. LPS-activated cells showed increased NO levels and iNOS expressions compared to control. However, these increases were dramatically attenuated by treatment with HAC extract. Moreover, the inhibitory effects of HAC extract occurred in a dose-dependent manner. In addition, HAC extract reduced the translocation of $NF{\kappa}B$ into nuclear. HAC reduced production of IL-$1{\beta}$ and IL-6 by LPS, although it had no effects on TNF-$\alpha$. These results demonstrate that liquiritigenin exerts anti-inflammatory effects, which results from the inhibition of $NF{\kappa}B$ activation in macrophages, thereby decreasing production of iNOS and proinflammatory cytokines. Taken together, these results indicate that the aqueous extracts of Herba Artemisiae Capillaris warrant further development as an anti-inflammatory agent for the treatment of gram-negative bacterial infections.
