Acute Toxicity of Heavy Metals, Tributyltin, Ammonia and Polycyclic Aromatic Hydrocarbons to Benthic Amphipod Grandidierella japonica

  • Benthic amphipod, Grandidierella japonica widely inhabits the Korean coastal waters and is developed as a standard test species for sediment toxicity tests. We exposed G. japonica to various pollutants including 4 kinds of inorganic metals (Ag, Cd, Cu and Hg), tributyltin [TBT], ammonia and 7 polycyclic aromatic hydrocarbon (PAH) compounds (acenaphthene, chrysene, fluoranthene, fluorene, naphthalene, phenanthrene and pyrene) to estimate the no observed effect concentration (NOEC) and the median lethal concentration (LC50) of each pollutant during the 96-hour acute exposure. Among all tested pollutants, TBT was most toxic to G. japonica, and Rg was most toxic among inorganic metals. The toxicity of pyrene to G. japonica was greatest among PAH compounds, followed by fluoranthene, phenanathrene, acenaphthene, fluorene and naphthalene. The toxicity of PAH compounds was closely related to their physico-chemical characteristics such as $K_ow$ and water solubility. G. japonica responded adequately to pollutant concentrations and exposure durations, and the sensitivity of G. japonica to various inorganic and organic pollutants was generally comparable to other amphipods used as standard test species in ecotoxicological studies, indicating this species can be applied in the assessment of environments polluted by various harmful substances.

  • White rot fungi have been useful source of enzymes for the degradation of environmental pollutants including polycyclic aromatic hydrocarbons (PAHs) and synthetic dyes. PAHs are widespread organic compounds present in fossil fuels and are routinely generated by incomplete fuel combustion. PAHs are some of the major toxic pollutants of water and soil environments. Synthetic dyes are major water-pollutants, which are toxic to organisms in water environments and interfere photosynthesis of water plants. Removal of PAHs and synthetic dyes has been of interests in the environmental science especially in the environmental microbiology. Mushrooms are fungal groups that function as primary degraders of wood polyphenolic lignin. The ligninolytic enzymes produced by mushroom, including manganese peroxidase, lignin peroxidase, and laccase, mediate the oxidative degradation of lignin. The catalytic power of these enzymes in the degradation of aromatic ring compounds has been sought for the degradation of various organic compounds. In this project, we have screened 60 wild mushroom strains for their degradation activity against two representative PAHs, naphthalene and anthracene, and five aromatic dyes, including alizarin red S, crystal violet, malachite green, methylene blue, rose bengal. The degradation of PAHs was measured by GC while the decolorization of dyes was measured by both UV spectrophotometer and HPLC. As results, 9 wild mushroom strains showed high activity in degradation of PAHs and textile dyes. We also describe the secretive enzyme activities, the transcription levels, and cloning of target genes. In conjunction with this, activities of degradative enzymes, including laccase, lignin peroxidase, and Mn peroxidase, were measured in the liquid medium in the presence of PAHs and dyes. Our results showed that the laccase activity was directed correlated with the degradation, indicating that the main enzyme acts on PAHs and dyes is the laccase. The laccase activity was further simulated by the addition of $Cu^{2+}$ ion. Detailed studies of the enzyme system should be sought for future applications.

Simultaneous determination of ultra-trace phenols, polycyclic aromatic hydrocarbons and pesticides in surface water by gas chromatography-mass spectrometry (지표수에서 GC-MS에 의한 극미량 페놀류, 다환방향족탄화수소류와 농약류의 동시 분석법)

  • A gas chromatography-mass spectrometric (GC-MS) method was developed for determining 17 hazard compounds containing phenols, polycyclic aromatic hydrocarbons and pesticides in surface water. A 1.0 L surface water sample was placed in a separatory funnel and saturated with NaCl, and the solution was extracted with 40 mL of methylene chloride. Under the established condition, the lowest quantification limit was 1.0-10 ng/L and the relative standard deviations were less than 22%. The method was used to analyze 70 surface water samples collected from 35 regions in Gum-River. The samples revealed the compounds concentrations in the range of 1.1-26,604 ng/L. Maximum concentrations of compounds detected were not exceeded guidelines established in other countries. The developed method may be valuable for monitoring hazards in water.

A Study on Reduction Effects of Air Pollutant Emissions by Automotive Fuel Standard Reinforcement (자동차연료 기준강화에 따른 대기오염물질 배출량 저감효과)

  • The air pollutants from vehicle exhaust gas are affected by many factors including fuel qualities, engine and vehicle technologies, driving patterns. In particular, fuel qualities and after-treatment devices could directly affect the emission level of pollutants. The pollutant reduction characteristics that caused by enforced fuel quality standard were analyzed. Three types of test fuel were selected in accordance with Korean automotive fuel standard in 2006, 2009, 2012 and used for vehicle emission test in chassis dynamometer. European COPERT correction equation of fuel impact was considered as reference information to quantify the vehicle emission test results. The contribution rates of exhaust emission by COPERT correction equation showed that aromatic compounds and oxygen contents in gasoline fuel was most important. In case of diesel fuel, cetane index and polycyclic aromatic compounds accounted for the greater part. The exhaust emission effects by COPERT correction equation revealed that CO and VOC was increased 0.86%, 1.57% respectively in after 2009 gasoline when compared to before 2009 gasoline fuel. In case of light-duty diesel vehicle CO, VOC and PM were decreased in range of 3~7%. The result from this study could be provided for developing future fuel standards and be used to fundamental information for Korean clean air act.

A Novel Bioassay System for Screening of Compounds Affecting Anthocyanin Biosynthesis Pathway in White Corn Leaf Segment (백화옥수수 잎절편을 이용한 안토시아닌 생합성 조절제 탐색용 신규 검정법 확립)

  • This study was carried out to establish a novel bioassay system for screening of compounds affecting aromatic amino acid or anthocyanin biosynthesis through investigating a degree of sucrose-induced anthocyanin formation such as size of plant material, buffer conditions, light intensity and irradiated duration, incubation temp., etc were determined and standard procedure (suitable experimental condition) was set up as follows. The second leaf blade of white corn seeding induced by fluridone treatment were segmented into a size of 5${\times}$5 min. The segments were floated on the solution of 1% sucrose in 1.0mM MES buffer (pH6.0∼6.5) and incubated at 26$^{\circ}C$ for 2days under the continuous light condition(70∼100$\mu$mol m$^{-2}$ s$^{-1}$ ). Anthocyanin in the purpled tissues was extracted with methanol containing 1% HCl and the optical density of the clear supematants was determined at 528mm. Influences of some chemicals were tested using this system. Glyphosate, 5-enolpyruvylshikimate 3-phosphate synthase inhibitor, showed most sensitive response with I$_{50}$ value at 3.3$\mu$M. Dicyclohexylcarbodiimide(DCCD) and parachloromercuribenzenesulfonic acid(PCMBS) had a relatively strong ingibition with I50 value at 7.1$\mu$M and 10.2$\mu$M, respectively. These results show that sucrose-induced anthocyanin formation in white com leaf segment provide a very simple and rapid system for searching new compounds affecting aromatic amino acid or anteocyanin biosynthesis by screening at less than 10$\mu$M.

  • In Korea, the Air Quality Conservation Act and the Petroleum and Petroleum Substitute Fuel Business Act stipulate certain quality standards for fuels distributed in Korea, thereby striving to reduce vehicle performance and emissions. Domestic petroleum products import and produce all the crude oil from each oil refiner so that the quality of the petroleum product is different according to the characteristics of the crude oil. As a result, vehicles have been improved by using the physical properties calculated through the physical property measurement that has tried to improve the accuracy of the measurement of the energy consumption efficiency of the automobile by using standard fuel from abroad. In this study, the same test procedure and method as the test method of domestic gasoline vehicle emission are applied using four samples of gasoline and the latest gasoline vehicle which are actually distributed, and the performance evaluation is performed. The purpose of this study is to contribute to improvement of vehicle technology and fuel quality by collecting necessary basic data and obtaining data on the effect of differences in gasoline property on vehicle emissions. The results of the test showed that the emission of gases (NMOG, CO) from gasoline vehicles was the most influenced by the sulfur content, unlike the previous studies that the vehicles emission had the greatest influence on the distillation characteristics and the specific gravity of aromatic compounds. The catalytic reaction such as the poisoning action of the three-way catalyst which is the abatement device was interfered and the emission was increased. The distillation characteristics and specific gravity of aromatic compounds were found to affect the emission of vehicles. According to the physical properties of the fuel, the emission difference was 28.0% in the urban mode and 17.6 % in the highway mode.

  • Polyaniline/$SiO_2$ catalyzed one-pot mannich reaction of acetophenone, aromatic aldehydes and aromatic amines are carried out in ethanol to afford various ${\beta}$-amino ketones. The various wt% of polyaniline were supported on pure silica synthesized by using chemical oxidative method. The catalyst prepared has been characterized by means of thermal analysis (TG-DTA), X-ray diffraction (XRD), scanning electron microscopy (SEM) energy dispersive spectroscopy (EDS), and Fourier transform infrared spectroscopy (FT-IR). Solvent stability of catalyst was tested using UV-Visible spectroscopy. This protocol has several advantages such as high yield, simple work up procedure, non-toxic, clean, easy recovery and reusability of the catalyst.

Phylogenetic characteristics of bacterial populations and isolation of aromatic compounds utilizing bacteria from humus layer of oak forest (상수리림 부식층으로부터 방향족 화합물 분해세균의 분리 및 세균군집의 계통학적 특성)

  • In this study, we isolated aromatic compounds (lignin polymers) utilizing bacteria in humus layer of oak forest and investigated phylogenetic characteristics and correlation with major bacterial populations in the humus layer by pyrosequencing. Forty-two isolates using aromatic compounds such as p-anisic acid, benzoic acid, ferulic acid and p-coumaric acid were isolated and phylogentic analyses based on 16S rRNA gene sequences showed that the isolates belonged to the genus Rhizobium, Sphingomonas, Burkhorlderia, and Pseudomonas. Among these, Burkhorlderia species which belong to Betaproteobacteria class occupied 83% among the isolates. The bacterial populations in humus layer of oak forest were characterized by next generation pyrosequencing based on 16S rRNA gene sequences. The humus sample produced 7,862 reads, 1,821 OTUs and 6.76 variability index with 97% of significance level, respectively. Bacterial populations consist of 22 phyla and Betaproteobacteria were the major phylum consisting of 15 genera including Burkholderia, Polaromonas, Ralstoria, Zoogloea, and Variovorax. Approximately fifty percentage of them was Burkholderia. Burkholderia as the majority of population in the humus was considered to play a role in degrading lignin in humus layer of oak forest.

TRIBOCHEMICAL ACTIVITY OF NASCENT METAL SURFACES

  • Chemical nature of nascent metal surfaces which is one of the important active sources for tribochemical reactions was investigated using a newly developed method. Some enhanced activities were observed. For example, organic compounds chemisorbed on nascent gold surfaces and aromatic compounds decomposed on nascent nickel surfaces resulting in hydrogen evolution. Non-polar compounds such as organic sulfides had a higher chemisorption activity on nascent steel surfaces than polar compounds such as fatty acids and phosphates. Organic sulfides reacted directly with nascent steel surfaces and the surface was covered with metal sulfides. The activity for the chemisorption of organic compounds was closely dependent on the electronic structure of metals. Although benzene chemisorbed very easily on nascent surfaces of transition metals, it did not chemisorb ell nascent surfaces of simple metals. Boundary lubricating behaviors of extreme pressure additives were explained on the bases of the chemical activities of nascent surfaces obtained in this investigation. Under mild conditions, polar compounds such as fatty acids and phosphates were effective for boundary lubrication, because surfaces are covered with oxide layers. On the other hand, sulfides were more effective under severe conditions where the oxide layers were removed and the nascent surfaces were formed.

Biodegradation of Polycyclic Aromatic Hydrocarbons by White Rot Fungi (백색부후균을 이용한 다환방향족 탄화수소(PAHs) 의 분해)

  • The white rot fungi Phanerochaete chrysosporium(IFO 31249) Trametes sp and Pleurotus sp. were studied for their ability to degrade Polycyclic Aromatic Hydrocarbons(PAHs) using anthracene and pyrene as model compounds. The disapperarance anthracene and pyrene of from cultures of wild type strains. P chrysosporium Trametes sp. and Pleurotus sp was observed However the activities of ligninolytic enzymes were not detected in P chrysosporium cultures during degradation while ligninolytic enzymes were detected in both culture of Trametes sp. and Pleurotus sp. Therefore our results showed that PAHs was degraded under ligninolytic as well as nonligninolytic conditions. The results also indicate that lignin peroxidase(LiP) mananese peroxidase(MnP) and laccase are not essential for the biodegradation of PAHs by white rot fungi.
