P53 Expression in Adenoid Cystic Carcinoma of the Head and Neck (두경부 영역에 발생한 선양낭포암에서의 p53 단백 발현)

  • Overexpression of the mutant p53, derived from mutation of the p53 gene which is located on the short arm of chromosome 17 and plays a role in suppression of the tumor, was reported in some human malignancy such as breast or colon carcinoma and suggested to be a prognostic factor. The authors investigated expression of p53 protein by immunohistochemical staining using anti-p53 monoclonal antibody in the paraffin embedded blocks of the 23 patients who were diagnosed at Seoul National University Hospital as adenoid cystic carcinoma of the head and neck between 1982 and 1991, and could be followed-up. 7 cases(34%) out of the 23 cases showed p53 expression, and there was no significant association between p53 positivity and local recurrence(p=0.31) or distant metastasis(p=0.16)

A Study on the Association between Sasang Constitutions and Body Composition (사상체질분류검사에 의한 체질 진단과 체성분분석과의 상관성에 관한 연구)

  • Objectives : The purpose of this study was to evaluate the body shape of Sasang Constitutions by body composition and body measurement. Methods : This study was carried out 98 cases(68 males and 30 females)of Oriental Medical College at Kyunghee University. After determining the Sasang Constitutions(Soeumin, Soyangin, Taeumin) by QSCCII, the body shape of each group were analysed by body composition(INBODY 2.0, Biospace, seoul, korea) and body measurement. Results : 1) The result of Sasang Constitution analysis of 98 cases were classified as Soumin(52%), Soyang(24.5%), Taeumin(23.5%). 2) In the body composition and body measurement, there were significant difference from intercellular fluid, protein, mineral mass, weight, percent body fat, body water of left arm and BMI between Taeumin, Soyangin and Soumin. 3) In the body composition of male, Taeumin group showed that the arms were more developed than the legs and the upper trunk were more developed than the lower trunk. Soumin group showed the asymmetry of the right and left limbs. Soyang group showed the asymmetry of the right and left arms. In the body composition of female, Taeumin group showed the asymmetry of the right and left upper limbs. Conclusion : There are significant relationships between Sasang Constitutions, Body composition and BMI. The findings indicated here Body composition could be useful mean to make a diagnosis of the body shape, body type, physical constitution of Sasang constitutions.

A Study of Nutrient Intakes, Blood Lipids and Bone Mineral Density according to Obesity Degree by Percentage of Body Fat and Age between Male and Female Teacher in Jeonbuk Province, Korea (전북지역 일부 남녀교사의 체지방률에 의한 비만도와 연령에 따른 영양소 섭취와 혈중지질 및 골밀도에 관한 연구)

  • The purpose of this study was to compare nutrient intakes, blood lipids and bone mineral density of male (n = 59) and female (n = 172) teachers according to the obesity index by percentage of body fat and age. The energy intakes of obesity group were higher than normal group in male (p < 0.05), but were not significant in female. The protein intake ratio among three energy nutrients for male was higher than female (p < 0.001), and lipid intake ratio of obesity group in female was a little higher than male that was not significant. TC, LDL, TC/HDL, risk of coronary heart disease, blood glucose and blood pressure of obesity group were higher than normal group in female (p < 0.01 ~ p < 0.001), but were little significance in male. Risk of coronary heart disease was affected by gender (p < 0.001), obesity degree (p < 0.01), age (p < 0.001), and interaction of gender and age (p < 0.001). Blood glucose was affected by obesity degree (p < 0.05), but was not affected by age. T-scores of forearm for female (= -1.42) were lower than that of male = -0.95), and T-scores of obesity group in male (= 0.12) were higher than that of normal group (= -0.33) but were not significant in female. The T-scores of forearm for female were affected by age (p < 0.05) and gender (p < 0.01), but calcaneus was not affected by gender. These results suggest lipid intake ratio should be balanced for obesity group in female. Nutritional education for treatment obesity to prevent hyperlipidemia and arteriosclerosis is necessary for obesity group and older age groups. T-scores of forearm were lower than calcaneus, so arm exercise would be especially required to prevent osteoporosis for older age women groups.

Expression analysis and characterization of rice oligopeptide transport gene (OsOPT10) that contributes to salt stress tolerance

  • Knock-out of a gene by insertional mutagenesis is a direct way to address its function through the mutant phenotype. Among ca. 15,000 gene-trapped Ds insertion lines of rice, we identified one line from selected sensitive lines in highly salt stress. We conducted gene tagging by TAIL-PCR, and DNA gel blot analysis from salt sensitive mutant. A gene encoding an oligopeptide transporter (OPT family) homologue was disrupted by the insertion of a Ds transposon into the OsOPT10 gene that was located shot arm of chromosome 8. The OsOPT10 gene (NP_001062118.) has 6 exons and encodes a protein (752 aa) containing the OPT family domain. RT-PCR analysis showed that the expression of OsOPT10 gene was rapidly and strongly induced by stresses such as high-salinity (250 mM), osmotic, drought, $100\;{\mu}M$ ABA. The subcellular localization assay indicated that OsOPT10 was localized specifically in the plasma membrane. Overexpression of OsOPT10 in Arabidopsis thaliana and rice conferred tolerance of transgenic plants to salt stress. Further we found expression levels of some stress related genes were inhibited in OsOPT10 transgenic plants. These results suggested that OsOPT10 might play crucial but differential roles in plant responses to various abiotic stresses.

A plant-based multivitamin, multimineral, and phytonutrient supplementation enhances the DNA repair response to metabolic challenges

  • Purpose: DNA damage and repair responses are induced by metabolic diseases and environmental stress. The balance of DNA repair response and the antioxidant system play a role in modulating the entire body's health. This study uses a high-fat and high-calorie (HFC) drink to examine the new roles of a plant-based multivitamin/mineral supplement with phytonutrients (PMP) for regulating the antioxidant system and cellular DNA repair signaling in the body resulting from metabolic stress. Methods: In a double-blind, randomized, parallel-arm, and placebo-controlled trial, healthy adults received a capsule containing either a PMP supplement (n = 12) or a placebo control (n = 12) for 8 weeks. Fasting blood samples were collected at 0, 1, and 3 hours after consuming a HFC drink (900 kcal). The blood samples were analyzed for the following oxidative stress makers: areas under the curve reactive oxygen species (ROS) levels, plasma malondialdehyde (MDA), erythrocytes MDA, urinary MDA, oxidized low-density lipoprotein, and the glutathione:oxidized glutathione ratio at the time points. We further examined the related protein levels of DNA repair signaling (pCHK1 (Serine 345), p-P53 (Serine 15), and 𝛄H2AX expression) in the plasma of subjects to evaluate the time-dependent effects of a HFC drink. Results: In a previous study, we showed that PMP supplementation for eight weeks reduces the ROS and endogenous DNA damage in human blood plasma. Results of the current study further show that PMP supplementation is significantly correlated with antioxidant defense. Compared to the placebo samples, the blood plasma obtained after PMP supplementation showed enhanced DNA damage response genes such as pCHK1(Serine 345) (a transducer of DNA response) and 𝛄H2AX (a hallmark of DNA damage) during the 8 weeks trial on metabolic challenges. Conclusion: Our results indicate that PMP supplementation for 8 weeks enhances the antioxidant system against oxidative stress and prevents DNA damage signaling in humans.

Effects of Omega-3 Fatty Acid Supplementation on Skeletal Muscle Mass and Strength in Adults: A Systematic Review

  • Previous studies have suggested that omega-3 polyunsaturated fatty acids, predominantly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), have several health benefits. However, their effect on changes in skeletal muscle mass and strength has not been established, owing to differences in study designs. This systematic review aimed to investigate the recent evidence regarding the role of dietary EPA and DHA in muscle mass changes and their association with muscle strength. Databases including PubMed and Google Scholar were searched for randomized controlled trials and single-arm interventions that investigated the effects of omega-3 fatty acids on skeletal muscle mass, strength, and body composition in adults aged 18 years and older. A total of 18,521 studies were retrieved from the databases and manual searches; 21 studies were quality assessed, and the findings were summarized. Studies were categorized into 3 main categories according to the type of omega-3 fatty acid supplementation: pure compounds such as oil tablets, formulated forms with protein, leucine, and vitamin D, and ingredients added to enteral nutrition support products. Overall, the majority of the study results appeared to indicate that omega-3 fatty acids are beneficial for muscle health. However, meta-analysis was not conducted because of the heterogeneity of the study participants, evaluation method of muscle indices, and intervention periods among the studies. High-quality studies are required to validate our conclusions. However, this systematic review of the effects of EPA and DHA on skeletal muscle and body composition provides evidence that can be applied in both clinical and industrial settings.

Status of Early Childhood and Maternal Nutrition in South Korea and North Korea (남북한 영유아 및 가임기 여성의 영양상태 비교)

  • The purpose of this study was to compare the nutritional status of children aged 5 or under and women aged 20 to 34 years between the Republic of Korea (South Korea) and the Democratic Peoples' Republic of Korea (DPRK: North Korea). For the source of nutritional status of North Koreans, the DPRK 2004 Nutrition Assessment-Report of Survey Results was used. As the comparable data of South Koreans, the anthropometric data for children and women were obtained from the reports of the Korean Pediatric Society and the Korean Agency for Technology and Standards, respectively. The blood hemoglobin data of South Korean women were obtained from the data file of the 2001 National Health and Nutrition Survey and analyzed. In regard to the North Korea, the Prevalence of underweight (weight for age Z-score < -2.0) in children under 12 months was about $10\sim15%$, and thereafter progressively increased until 30 to 35 months reaching 30%. In South Korea, the prevalence of underweight was less than 3% in most age groups both in boys and girls. In North Korea, the prevalence of stunting (height for age Z-score < -2.0) reached 20% in children under 12 months and increased with age over the level of 50% in children aged 54 to 59 months. In South Korea, the prevalence of stunting was less than 3% in children under 12 months and was less than 10% throughout the age groups. Maternal protein-energy malnutrition and anemia were assessed for the women aged 20 to 34 years using mid-upper arm circumference (< 22.5 cm) and blood hemoglobin level (< 12g/DL), respectively. The prevalence of protein-energy malnutrition was 39.6%, 30.7%, 31.7% in North Korea and 12.5%, 5.0%, 1.5% in South Korea fir the women in $20\sim24,\;25\sim29,\;30\sim34$ years, respectively. The prevalence of anemia in the North Korean women was about $34\sim36%$ while that in the South Korean women was $15\sim18%$. In conclusion, the disparity of nutritional status in early childhood and maternity between South Korea and North Korea is so huge that active and well-planned nutrition support policy and programs for women and children in North Korea is imperative to prepare for the future unified nation.

The Dietary Status of Kindergarten Child from a High Socioeconomic Apartment Compound in Seoul (일부(一部) 고소득(高所得) 아파트 단지내(團地內) 유치원(幼稚園) 어린이의 성장발육(成長發育) 및 영양(營養)에 관(關)한 연구(硏究))

  • A dietary survey of 100 children, aged 5 to 6, of Y.C. Kindergarten located in Yowido apartment compound of Seoul, was conducted July 18 to August 21, 1979. The results are summarized as follows: General family environment: Ninety four percent of the subjects were from families having two to three children, without grandparents in the home. One hundred percent of fathers were educated at or above the college level, while 83.9% of mothers had completed college. Mothers' ages ranged from 30 to 40 years, with 58% in the $31{\sim}35$ age bracket. Nearly 60% of the fathers were office and government employees, 10% were professors and teachers, 30% were engaged in businesses and other work, while 91.3% of mothers were unemployed. Anthropometric measurements: Mean values of standing height, body weight, girth of head and girth of chest, ranged from the 75th to the 90th percentile of Korean standards. None of the subjects had anthropometric indicators below the 5th percentile, representing the malnutrition borderline, of the Korean standards; while 12%, 24%, 17% and 14% of subjects supposed the 97th percentile in standing height, body weight, girth of head, and girth of chest, respectively. Furthermore, 58% of subjects exceeded Jelliffe's standard in arm circumferences; 60%, in triceps skinfold thickness; and 14.5% of subjects exceeded 120% of the latter standard. From this data, a certain number of these children were judged to he rather overweight. Dietary intake: Mean intake per day of energy and of nutrients, excluding iron, reached or exceeded Korean recommended dietary allowances. The energy input ratio of carbohydrate: protein: fat was 61.4 : 13.2 : 25.4, showing lower dependance on carbohydrates and higher dependance on fat for energy needs, compared with the average for Korean children of the same age group. Mean FAO-protein score of each subject's diet was shown to be as high as 90.6. Energy intake was divided among breakfast, lunch, and evening meal, on the average, in a ratio of 19.6 : 24.1 : 23.0. Approximately 33.3% of the daily caloric intake came from the snacks. Nutrition Knowledge of mothers: Nutrition knowledge of mothers was highly scored. Mothers' self-evaluations of their knowledge of nutrition was highly correlated with attitudes toward nutrition behavior.

P53 Expression in Squamous Cell Carcinomas of Tongue and Tonsil (설과 편도 편평 상피세포암에서 P53의 표현양상에 관한 연구)

  • The nuclear phosphoprotein p53 is expressed in all normal cells and appears to function in cell cycle regulation. Abnormally high levels of the protein are found in many different types of cancer. In human cancer overexpression of p53 is associated with point mutations within highly conserved regions of p53 gene. These altered genes encode stable p53 proteins that can detected by standard immunocytochemical techniques unable to detect rapidly degraded wild-type protein. Using of a monoclonal antibody to p53 antigen, immunocytochemical analysis of 29 squamous cell carcinomas of tongue(n= 19) and tonsil(n= 10) was performed. Non-tumor nuclei showed all negative reactivity. Positive reactivity was found in 4/29(13.8%)of SCCs of tongue and tonsil. In sizes of primary tumor, the cases over 4cm showed more positive reactivity than the cases under 4cm(p < 0.05). There was no stastical correlation between the reactivity and histopathologic grades, the primary sites of tumor or the presence of cervical metastasis.

Effect of Maturity Scores and Number of Extractions on the Chemical Properties of Water Extract from Hanwoo Shank Bones (한우 성숙도와 추출횟수가 사골용출액의 화학적 특성에 미치는 영향)

  • This study was conducted to investigate the effect of maturity scores [2 (bull), 2 (steer), 3-9 (cow)] and the number of extractions (up to 4 times) on the chemical properties of water extract from Hanwoo shank bones (arm, fore shank, round and hind shank). The turbidity, meat color (CIE L value), collagen, protein, caloric and chondroitin sulfate contents of samples were observed. The turbidity and lightness were higher for water extract of Hanwoo shank bones with a maturity score of 2 (bull and steer) than maturity scores of 3-9 (cow) (p<0.05). The turbidity and lightness of water extract from shank bones of all Hanwoo maturity scores significantly increased with the 1st and 2nd extractions, but significantly decreased with 3rd and 4th extractions (p<0.05). The collagen and protein contents were highest for water extract from Hanwoo shank bones of maturity score 2 (bull and steer) (p<0.05). The caloric and chondroitin sulfate contents were higher for water extract from Hanwoo shank bones of maturity score 2 (bull and steer) than maturity scores of 3-9 (cow) (p<0.05). As the number of extractions increased, the chondroitin sulfate content significantly decreased (p<0.05). Based on these results, differences correlating with maturity scores were found only with collagen and protein contents. Therefore, further studies should be considered to address whether different maturity scores affect the price of shank bones in the meat industry.
