The methanol extract of Aralia elata root bark was found to decrease the blood glucose level in normal and alloxan-treated rats. The fractionation process confirmed positive activity in ethylacetate fraction. It is suggested that this fraction may show potent antihyperglyceimic activity through stimulation of insulin secretion.
In the previous report, it was described that the butanol fraction of the root bark extract of Aralia elata had potent antigastritic and antiulcerative effects in rats, In this pharmacological profile, the fraction revealed low acute toxicities in mice and rats, no central nervous system activities, moderate analgesic action, weak inhibitory action in carrageenin edema of rats and no direct effects on isolated smooth muscles of rats and guinea-pigs. Slight lowering of blood pressure as given i.v. in rabbit was observed.
It is well known that lipidperoxide, formed in vivo, induced the denaturation of enzyme and destruction of cell membrane to acute injury of tissue. Aralia elata have physiological activates, the improvement of lipid metabolism, antidiabetic activity etc., which was thought to have the relationship to lipid peroxidation. The anti-lipidperoxidative effect of Aralia elata have not yet established. In this study, we examined the anti-lipidperoxidative effects of Aralia elata (Butanol fraction) on CCI$_{4}$ induced lipidperoxidation in rats, and elucidated the anti-lipidperoxidative mechanism. In rat liver homogenate intoxicated with CCI$_{4}$ (0.5 ml/100g), BuOH fraction of Aralia elata (80 mg/Kg/day) exhibited 85.41% anti-lipidperoxidative effect but in serum 69.63% inhibitory effects, respectively. In mitochondrial and microsomal fraction showed inhibition of 55.85% and 69.30%, respectively. In order to elucidate the mechanism of anti-lipidperoxidation effects of Aralia elata, enzymatic (NADPH dependent) and non-enzymatic (Ascorbic acid catalyzed) reaction, in vitro, were performed. In enzymatic reation, Aralia elata exhibited 59.43% anti-lipidperoxidation effects, but in non-enzymatic reaction exhibited 43.27% inhibition. Therefore, it is noteworthy that antioxidative power of them may mainly results from the inhibition by enzymatic reaction.
In order to investigate the safety of Aralia elata extract causing the reduction in the blood glucose level and oxidative stress in diabetes animals, these genotoxicity studies in bacterial and mammalian cell assay system such as Ames bacterial reverse mutation test and chromosomal aberration assay were performed. As results, in Ames bacterial reversion assay the extract in the range of 5,000-625 ug/plate did not induce mutagenicity in Salmonella typhimurium TA 98, TA 100, TA 1535 and TA 1537 strains with and without metabolic activation of S-9 mixture. For chromosomal aberration assay, $IC_{50}$ (50% inhibition concentration of cell growth) of the extract were determined; 792 $\mu\textrm{g}$/$m\ell$ without and 524 $\mu\textrm{g}$/$m\ell$ with S-9 mixture in Chinese hamster lung (CHL) fibroblast cell culture. Any significant chromosomal aberration was not observed in CHL cells treated with the extract at the concentrations of 792, 396 and 198 $\mu\textrm{g}$/$m\ell$ or 524, 262 and 131 $\mu\textrm{g}$/$m\ell$ in the absence or presence of S-9 metabolic activation, respectively. From these results, Aralia elata extract did not induce any harmful effects on the gene in bacteria and mammalian cell system used in these experiments.
Aralia elata seeds were successively extracted with water, methanol, ethanol, acetone and chloroform. The crude extracts were investigated for antioxidant and antidiabetic activities. The antioxidant properties of various extracts were evaluated by antioxidant tests, such as DPPH free radical-scavenging activity, hydroxyl radical-scavenging assay, metal-chelating activity, lipid peroxidation inhibition activity and reducing power assay. The 70% methanol extract exhibited the highest activity in the in vitro models of DPPH free radical-scavenging activity, metal-chelating activity, and reducing power assay. Acetone extract showed good effects on lipid peroxidation inhibition and hydroxyl radical-scavenging assay at a low concentration. In addition, the $\alpha$-glucosidase inhibition assay showed that 70% methanol extract had the highest activity. These results indicate the high possibility of using A. elata seeds for medical application due to their efficient antioxidant properties.
This study has examined the effects of Aralia elata Seemann ethanol extract on antioxidant enzyme systems inrats along with benzo($\alpha$) pyrene(B(a)P) administration . The ethanol extract of Aralia elata Seemann (50mg/kg body wt.) was fed to rats for 4 weeks by stomach tubing. The extract administration increased antioxidant activities of glutathione sulfur transferase(GST) comparing to the control. also total superoxide dismutase(SOD) and Cu, Zn-SOD activities were stimulated. Catalase activities were increased by 50% with the extract feeding compared to the control . Combined administration of B($\alpha$)P and the extract increased GST activity in B($\alpha$) P group. Although total SOD acitivity was decreased , Cu, Zn-SOD was greately increased from 0.10unit to 0.18 unit and catalase activity also was increased compared to the group of B($\alpha$) P. GST activity in CLE group was 1.32 unit, increased by 33% comparing to the group CL of 0.99unit. Cu, Zn-SOD and catalase activities in thegroup fed high fat and ethanol extracts were increased by 25% and 39%, respectivley comparing to the group of high fat. In addition , total SOD was decreased but, Cu, Zn-SOD acitivity was increased from 0.09 unit to 0.18unit. Catalase activity was 76.05 unit in the group of B($\alpha$) P and extract comparing to 65.26 units in B($\alpha$)P group. Serum $\alpha$-tocopherol of rat was markedly increased by theextract. Administration of B9$\alpha$)P reduced $\alpha$-tocopherol levels in the serum, on the other hand, lard in the diet increased $\alpha$-tocopherol levels in the serum. The above results indicate that Aralia bud exerts antioxidant functions in vivo against B($\alpha$)P. Further research may be necessary for the identification fo the biologically active material.
The purpose of this study was to investigate effect of methanolic extracts of 10 kinds of wild vegetables cultivated in Gangwon province on antioxidant activity, acetylcholinesterase, and β-secretase inhibitory activities. Results showed that among the wild vegetables, Aralia elata(Miq.) Seem shoot extract exhibited the highest total phenol content (84.65±1.08 mg GAE/g) and total flavonoids content (70.77±0.55 mg RE/g), respectively. The antioxidant activity of wild vegetables extracts was measured by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay. Aralia elata(Miq.) Seem shoot extracts had the highest DPPH and ABTS scavenging activity (90.16%, 40.18% at 2 mg/mL). As a result, Aralia elata(Miq.) Seem shoot extract was the most effective in terms of acetylcholinesterase inhibitory activity (35.94% at 1 mg/mL). In the β-secretase activity assay, all 10 kinds wild vegetables extracts showed low inhibitory activity, and Aralia elata(Miq.) Seem shoot extract had highest inhibitory activity among the 10 wild vegetables extracts was 14.99%. Taken together, these results showed that Aralia elata(Miq.) Seem shoot extract has potential cognition improvement impact, suggesting that it may provide an effective strategy for improving cognition.
Effect of Aralia elata extract on experimentally alloxan-induced diabetes ill rabbits and toxicity of mouse were studied; $LD_50$ of Aralia elata extract (A. E. E.) was 5.5g/kg intraperitoneally ill mice. A. E. E. 800mg/kg exhibited more excellent hypoglycemic effect after 6 days and 14 days in 200mg/kg, compared to the control group. Blood urea nitrogen level was significantly decreased after 6 days in 800mg/kg. and blood levels of total cholesterol and GPT activity were showed more significant decreasing effect ill 800mg/kg than 200mg/kg.
Objectives : This study was performed to investigate the antioxidant activities of isolated compounds from the shoot of Aralia elata. Methods : The methanol extract from the shoot of Aralia elata was fractionated into ethyl acetate, n-BuOH and $H_2O$ layers through solvent fractionation. Repeated silica gel, ODS column chromatography of n-BuOH layer afforded four flavonol glycosides. Their antioxidant activity was determined by measuring free radical scavenging activity by DPPH, ABTS and superoxide dismutase (SOD) like activity assay. Results : They were identified as quercetin 3,7-di-O-$\alpha$-rhamnopyranoside (1), quercetin 3-O-$\beta$-D-galactoside-7-O-$\alpha$-L-rhamnoside (2), kaempferol 3-O-$\beta$-glucosyl($1{\rightarrow}2$)-$\alpha$-rhamnoside-7-O-$\alpha$-rhamnoside (3) and quercetin 3-O-$\beta$-glucosyl($1{\rightarrow}2$)-$\alpha$-rhamnoside-7-O-$\beta$-rhamnoside (4) on the basis of spectroscopic data. The result showed that 1 is the most active compound in the DPPH and ABTS radical scavenging test. Conclusions : Isolated Compounds from the shoot of Aralia elata showed anti-oxidative effect.
Aralia elata, a common medicinal and culinary herb, has beer consumed for centuries without any reported adverse effects. However, due to its limited safety information, we decided to investigate the repeated-dose toxicity of ethanolic extract of Aralia elata. The test was administered once daily by the gavage to male and female rats at doses of 0, 250, 500 and 1,000 mg/kg/day for four weeks. Throughout the study, no treatment-related deaths or clinical signs were observed. Also, no apparent changes were detected in ophthalmoscopy, urinalysis, serum biochemistry, hematology and gross necropsy. The test result showed a significant decrease in body and heart weight of males treated with 250 mg/kg of extract of Aralia elata compared to normal control, a significant increase in relative brain weight and adrenal weight in females treated with 250 mg/kg of extract compared to normal control. However, all these changes were not considered toxicologically important due to irrelevant dose-response relationship to gross and microscopic findings. Histopathologically, abnormal changes were not observed in any target organs. On the basis of these results, the NOAEL of extract of Aralia elata was estimated to be more than 1,000 mg/kg/day under the tested conditions.
