해삼내장(內臟)젓의 맛성분(成分) (The Taste Compounds in Fermented Entrails of Trepang, Stichopus Japonicus)

  • 해삼내장(內臟)젓은 그 향미(香味)가 독특(獨特)하여 기호식품(嗜好食品)으로서 널리 애용(愛用)되고 있으나 그의 맛성분(成分)에 대한 연구보고(硏究報告)는 없다. 그래서 내장(內臟)젓의 맛성분(成分)을 밝혀 식품영양학적(食品營養學的) 기초자료(基礎資料)를 얻고져 삼천포산(三千浦産) 해삼 내장(內臟)을 원료(原料)로 써 숙성(熟成) 중(中) 유리(遊離)아미노산(酸), 유리당(遊離糖), 핵산관련물질(核酸關聯物質), TMAO, TMA 및 betaine의 변화(變化)를 실험(實驗)하였다. 해삼내장(內臟)의 유리(遊離)아미노산(酸) 조성(組成)을 보면 함량(含量)이 많은 것은 glutamic acid, alanine, glycine 및 proline 이고 함량(含量)이 적은 것은 leucine, valine phenylalanine, isoleucine, methionine 및 tyrosine 등이었다. 함량(含量)이 많은 아미노산(酸)의 전유리(全遊離)아미노산(酸)에 대한 비율을 보면 glutamic acid가 32.3%, alanine 이 16.4%, glycine이 12.0%, proline 이 10.54%로서 이들 4종(種)의 아미노산(酸)이 전유리(全遊離)아미노산(酸)의 71.2%를 차지하였다. 젓갈 숙성중(熟成中) 유리(遊離)아미노산(酸) 조성(組成)에는 변화(變化)가 없으나 원료(原料)에 많았던 glutamic acid, alanine, glycine proline, lysine, arginine 및 leucine 등은 숙성기간(熟成期間)에 따라 양적(量的) 변화(變化)는 있었지만 대체(大體)로 보아 젓갈 제품(製品)에도 함량(含量)이 많았다. 해삼내장(內臟)젓의 유리당(遊離糖)으로서는 galactose 가 $933.7{\sim}938.0mg%$로서 월등히 많았고, arabinose가 78.7mg%, xylose가 $55.2{\sim}77.1mg%$였으며 도당(萄糖)은 흔적량이었다. 핵산관련물질(核酸關聯物質)은 젓갈 숙성(熟成) 중(中) 대체(大體)로 증가(增加)하는 경향(傾向)이었고 특(特)히 함량(含量)이 많은 것은 hypoxanthine으로서 $47.1{\sim}62.5{\mu}mole/g$였으며, 정미성(呈味性)이 강(强)한 lMP도 비교적(比較的) 함량(含量)이 많았다. 젓갈 숙성(熟成) 중(中) TMA는 증가(增加)하는 반면(反面) TMAO는 점차 감소(減少)하는 경향(傾向)이었으며, 해삼내장(內臟)젓에는 TMAO질소(窒素)가 30.0mg% 전후(前後)로서 비교적(比較的) 함량(含量)이 많았다. 젓갈 숙성(熟成) 중(中) betaine은 점차 증가(增加)하는 경향(傾向)이었고, 해삼내장(內臟)에 545.0mg%, 젓갈에는 $734.2{\sim}934.2mg%$로서 월등히 그 함량(含量)이 많았다. 해삼내장(內臟)젓의 정미성분(呈味成分)으로서는 좋은맛을 가진 glutamic acid, 단맛을 가진 alanine, glycine, lysine, proline, arginine, 쓴맛을 가진 leucine을 주체(主體)로 한 유리(遊離)아미노산(酸), 그리고 galactose, arabinose 및 xylose등의 유리당(遊離糖), 단맛을 가진 betaine TMAO 및 핵산관련물질(核酸關聯物質)로서는 lMP, hypoxanthine 등이 중요(重要)한 성분(成分)이고 이들 성분(成分)들이 해삼내장(內臟)의 독특(獨特)한 향기(香氣)와 texture등과 조합(組合)되어 해삼내장(內臟)젓의 풍미(風味)에 중요(重要)한 구실을 할 것이라는 결론(結論)을 얻었다.

Characterization of a New Acidophilic Acetobacter sp. Strain HA Isolated from Korean Traditional Fermented Vinegar

  • A new strain of acidophilic, acetogenic bacterium, Acetobacter sp. strain HA was isolated by selective enrichment from the traditionally fermented rice wine vinegar in Korea. It was a gram-negative, non-motile short rod and oxidized acetate and lactate. The optimal temperature and pH for growth were $28^{\circ}C$ and 4.0, respectively. The strain HA differed from other Acetobacter species by growing well on methanol, xylitol, inositol, dulcitol, D-xylose, L-arabinose, and D-mannose as sole sources of carbon and energy. The isolated strain HA did not produce $\gamma$-pyrones from glucose and did not produce ketone bodies from glycerol. The quinone system used in this study was an ubiquinone-9 isoprene unit. The guanine-plus-cytosine content of the DNA was 50.7 mol%, and the major cellular fatty acids were $C_{18:1} and C_{16:0}$.

지유로부터 분리한 다당류의 분석과 항응고작용 (Compositional Analysis of Polysaccharide from Sanguisorba officinalis and Its Anticoagulant Activity)

  • Polysaccharide from Sanguisorba officinalis was separated and fractionated using DEAE-Sephadex ion-exchange chromatography and Sephacry HR-200 gel filtration chromatography. One of the fraction(Fr. II) was sulfated and its anticoagulant activity was tested in vitro. Sulfation could increase the clotting time 50 times compared to unsulfated one. Fr. II was hydrolyzed and its composition was analyzed by conjugation with 7-amino-1, 3-naphthalene disulfonic acid using HPLC and electrophoresis. Arabinose and galactose were mainly composed at the ratio of 4 : 1. In addition, xylose and rhamnose were also found.

세포외 Cytosine Deaminase 생산균의 분리 및 동정 (Isolation and Identification of Bacterium Producing Extracellular Cytosine deaminase)

  • A bacterium, strain YK 391 producing extracellular cytosine deaminase, has been isolated from soil sample collected near Taegu City and identified. The strain YK 391 was observed to be a motile Gram-negative rod, and did not produced capsule nor spore. The bacterium produced acid from glucose and trehalose, not from arabinose. Esculine was nto hydrolyzed. The isolate could grow anaerobically at 37$\circ $C, but not at 4$\circ $C. Palmitoleic and palmitic acids comprised over 80% of the fatty acid composition of the strain. The strain. The strain YK 391 was identified as Chromobacterium violaceum YK 391 based on its morphological and physiolohical characteristics, and on the fatty acid composition. The extracellular cytosine deaminase produced by Chromobacterium violaceum YK 391 is believed to be unique because it was active not only on cytosine and 5-fluorocytosine but also on cytidine.

  • 본 연구는 고수율의 아라비녹실란을 얻기 위해 필수요소인 hemicellulose B의 회수율을 검토하였다. 검토결과 10% acetic solution으로 pH 4.2로 적정 후 반응시키는 단계에서의 온도를 $65^{\circ}C$, 에탄올 침전단계에서 3 volume일 때 가장 좋은 수율을 얻었다.

$\alpha$-1,3-glucanase를 생산하는 미생물의 분리, 동정 및 배양조건

  • A strain producing $\alpha$-1, 3-glucanase was isolated from soil samples. The strain has a grey aerial mycelium and showed brown color from the other side. The temperature and pH range for growth were from 10$\circ$C to 42$\circ$C and from 4 to 10, respectively. Arabinose, dextrin and mannose were utilized for growth. Analysis of cell wall components revealed that the strain was classified as type I. From the results, the strain was identified as Streptomyces sp. By using this strain, the maximum production of 0.65 units/ml for $\alpha$-1, 3-glucanase was achieved at 37$\circ$C for 48 hrs.

Direct Fermentation of D-Xylose to Ethanol by Candida sp. BT001

  • A yeast strain, BT001, which can directly ferment D-xylose to ethanol was isolated from forest soils, and then identified as Candida sp. Cultural conditions for the optimum ethanol production, along with the effects of aeration on cell growth and ethanol production were investigated. Aeration stimulated the cell growth and the volumetric rate of ethanol production, but decreased the ethanol yield. Optimum temperature and initial pH for the ethanol production were $33{\circ}^C$ and 6.0, respectively. In a shake flask culture, this strain produced 52.3 g ethanol per liter from 12%(w/v) D-xylose after incubation for 96 hours. Ethanol yield was 0.436 g per g D-xylose consumed. This corresponds to 85.8% of theoretical yield. Also, this yeast strain produced ethanol from D-galactose, D-glucose and D-mannose, but not from L-arabinose and L-rhamnose. Among these sugars, D-glucose was the fastest in being converted to ethanol sugars.

Enterobacter agglomerans TY-25 에 의한 D-Galactose로부터 D-Tagatose의 생산

  • A variety of microbial strains isolated from soil were tested for their ability to produce D-tagatose from D-galactose. An organism that can convert D-galactose into D-tagatose was selected and was identified as Enterobacter agglomerans. The cells grown on the induction medium containing 20 g/l arabinose were found to the best conversion potential among different carbohydrates and the conversion yield was about 15% when 20 gll galactose was used. The isolated crystals were obtained from the culture broth after the purification process such as treatment of ion resins, crystallization, and drying. The recovery yield was 70% after the purification. The crystals were identified as D-tagatose by the infrared spectroscopy, HPLC, specific optical rotation, and melting point.

Production of a Platelet Aggregation Inhibitor, Salmosin, by High Cell Density Fermentation of Recombinant Escherichia coli

  • Optimal conditions for a high cell density fermentation were investigated in a recombinant Escherichia coli producing salmosin, a platelet aggregation inhibitor. The optimized carbon and nitrogen sources were glycerol 10 g/l, yeast extract 30 g/l, and bacto-tryptone 10 g/l, yielding the dry cell weight (DCW) of 10.61 g/l in a 500 ml flask culture. The late-stage induction with 1% L-arabinose in a 5 l jar fermentor showed the highest DCW of 65.70 g/l after 27 h of the fed-batch fermentation. Around 2,200 mg/l of the protein was expressed as an inclusion body that was then refolded to obtain the active salmosin of 96 mg/l. We also confirmed the inhibitory activity against platelet aggregation of the active salmosin from the high cell density fermentation.

Structure, Origin, Development and Senescence of Collecters in Nerium indicum Mill. (N. odorum Soland., Apocynaceae)

  • Nerium indicum has adaxial colleters on each petiole, bract, bracteole, calyx and corolla. The colleters are finger-shaped sometimes lobed structures with tall heads on short stalks. The petiolar colleters are more abundant than those on the bract, bracteole, calyx and corolla but their structure is similar. Meristematic activity in a small group of protodermal and hypodermal cells at the base of the developing leaf gives rise to the colleter that further differentiates by frequent anticlinal and periclinal divisions. Many of the colleters are vascullarized, sometimes a layer of redially elongated cells is present beneath the epithelial cells. Senescence of a colleter begins when the leaves have either completed their growth or are still expanding. In senescent colleters, the central cell cytoplasm and nuclei are considerably reduced. Thin layer chromatography of the freshly harvested exudate of colleter shows the presence of rhamnose, glucose and arabinose sugars.
