Biosynthesis of Cephalexin in PEG400-Ammonium Sulfate and PEG400-Magnesium Sulfate Aqueous Two-Phase Systems

  • The biosynthesis of cephalexin was carried out in the aqueous two-phase systems using penicillin acylase as a catalyst, and 7-aminodeacetoxicephalosporanic acid (7-ADCA) and phenylglycine methyl ester (PGME) as substrates. 20% PEG400-l7.5% ${(NH_4)}_2SO_4$ containing 0.5 M NaCl and 1.5 M methanol aqueous two-phase systems (ATPS) were selected as reaction medium, and 53% product yield was obtained using immobilized penicillin acylase as a catalyst. 20% PEG400-l5% $MgSO_4$ ATPS was also used for the synthesis of cephalexin, and 60-62% product yield was obtained by using free penicillin acylase as a catalyst. When batch reactions were repeated in the ATPS, the cephalexin yields decreased during the reactions due to deactivation, loss, and product inhibition of penicillin acylase. The effect of different ratio of phenylglycine methyl ester to 7-ADCA on the product yield was investigated, and high cephalexin yield was obtained at a high molar ratio.

수성 2상계를 이용한 알파-아밀라제의 생산 (Production of $\alpha$-Amylase using Aqueous Two-Phase System)

  • 세포외 효소를 효율적으로 생산하기 위한 새로운 발효시스템으로서 수성 2상계를 시험하였다. 실험대상으로는 Bacillus amyloliquefaciens 균주에 의하여 $\alpha$-amylase를 생산하는 시스템을 선정하였다. Polyethylene glycol 5%, dextran 7%, 그리고 발효액이 88% 포함된 수성 2상계 발효시스템에 의하여 $\alpha$-amylase의 생합성은 25% 정도 증가하였으며 효소의 비활성화는 관찰되지 않았다. 또한 Polyethylene glycol 이 효소의 생합성을 증진시키는 것으로 관찰되었다. 효소의 생합성을 증가시키며 동시에 효소를 발효액으로부터 분리하기 위하여는 polyethylene glycol을 성장도중에, dextran은 효소의 활성이 최고에 달한 후에 발효액에 가하는 발효방법이 바람직하다.

  • The alkylation of resorcinol with cyclic allylic alcohol in non-aqueous acid medium gives intermolecular Friedel-Craft compounds. The products are primary alkylation products. The likage always occurs between carbon 3 of cyclic allylic alcohol and the position 2, 4, 5 or 6 of the resorcinol.

Thermo-Physical Properties of Some Coumarin Complexes

  • A number of complexes of transition metal ions with some coumarin derivatives have been prepared and their structures were elucidated with the help of conductometric, photometric and infrared studies. The stability constants of various complexes were determined, in aqueous medium, at different temperatures potentiometrically. The thermodynamic characteristics, ΔG, ΔH and ΔS, were calculated. The electrical behaviour of prepared compounds was followed.

  • The complex of the composition LaL(N$O_3)_3\;(H_2O)_2$ is prepared by the reaction of La($NO_3)_3{\cdot}6H_2O$ with Nethylmorpholine in aqueous medium. The ligand is involved in the complex as a neutral species where the chelation occurs via the oxygen of the ligand moiety and the nitrate groups as bidentate ligand. The chemical structure of the studied complex is confirmed through IR, XRD, and thermal analysis data. The complexation equilibria of La(III) with N-ethylmopholine is studied in aqueous medium at ionic strength I = 0.1 mol${\cdot}dm^{-3}\;KNO_3$ and at 25, 35 and 45 ${^{\circ}C}$, respectively. The thermodynamic parameters $\Delta$G, $\Delta$H and $\Delta$S values were calculated to prove the association with the complex formation. It is clearly observed that the process is accompanied by absorption of heat, i.e. endothermic process, while the entropy does not change greatly attributed to the release of constant number of water molecules during chelate formation.

Micro-Chemical Structure of Polyaniline Synthesized by Self-Stabilized Dispersion Polymerization

  • A variety of NMR techniques were applied to the micro-chemical structural characterization of polyanilines prepared via an efficient synthetic method in a self-stabilized dispersion medium in which the polymerization was conducted in a heterogeneous organic/aqueous biphasic system without any stabilizers. Here, the monomer and growing polymer chain were shown to function simultaneously as a stabilizer, imparting compatibility for the dispersion of the organic phase, and as a form of flexible template in an aqueous reaction medium. Polymerizations predicated on this concept generated polyanilines with a low defect content: solution state $^{13}C-NMR$ and solid $^{13}CDD/CP/MAS$ spectroscopy indicated that the synthesized HCPANi and its soluble derivative, HCPANi-t-BOC, evidenced distinctly different NMR spectra with fewer side peaks, as compared to conventionally prepared PANis, and the complete structural assignments of the observed NMR peaks could be determined via the combination of both 1D and 2D techniques. Ortho-linked defects in HCPANi were estimated to be as low as 7%, as shown by a comparison of the integration of the carbonyl carbon resonance peaks.
