Physicochemical Profiles of Chub Mackerel Scomber japonicus Bones as a Food Resource

  • The present study was conducted to characterize physicochemical properties of chub mackerel bones (CMB) to evaluate its potential as a food resource. The proximate composition of CMB showed 40.4% moisture, 13.8% crude fat, 15.2% crude protein, and 28.7% ash. The major minerals in CMB were calcium (26.27 g/100 g) and phosphorus (15.88 g/100 g). The amino acids were rich in glycine, proline, glutamic acid, and alanine. The contents of total and neutral lipids, glycolipid and phospholipid were shown to be 16.05%, 95%, 2.32%, and 3.15%, respectively. The major fatty acids were C22:6, C16:0, C18:1, C20:5, C18:0, C17:0, C14:0, C20:1 in order. The fatty acid contents of total and neutral lipid were in a range of 39.25% and 44.54% for saturated and 33.61% and 34.05% for polyunsaturated, respectively. The breaking strength and hardness of intact CMB were 10.01 and $50.03kgf/cm^2$, whereas those of CMB heated for 45 min at $121^{\circ}C$ were 0.40 and $1.94kgf/cm^2$, respectively.

Characterization of Biosurfactant Produced by Pseudomonas fluorescens PD101 (Pseudomonos fluorescens PD101이 생산하는 생물유화제 특성)

  • Biosurfactant-producing bacteria, showing strong crude oil degrading activity, were isolated from the caverns of National Oil Storage Basement. From the results of biochemical and molecular biological tests, the isolate was identified as Pseudomonas fluorescens PD101. It grows well on liquid media at temperature range from $20^{\circ}C\;to\;37^{\circ}C,$ but it does not produce biosurfactant when grown at $37^{\circ}C$ or at higher temperature. The biosurfactant was stable at broad pH range from 5 to 11 and under heat treatment condition of $100^{\circ}C$ for 30 min. The biosurfactant produced dark blue halo around the colony when grown on SW agar plates, which could confirm the biosurfactant as one of rhamnolipid group. The 700 bp of PCR product could be amplified from DNA of P. flurorescens PD101 by using PCR primers designed from rh1A gene of P. aeruginosa, and it showed $99\%$ of sequence homology with rh1A gene of P. aeruginosa encoding rhamnosyltransferase 1.

Evaluation of the Quality of Canned Seafood with Added Spice-oil Extract

  • The influence of spice (cinnamon, allspice, black pepper)-oil extract on canned seafood quality was studied. During the processing of canned seafood, the substitution of spice-oil extract for vegetable oil (refined sunflower, corn, soybean and olive oil) resulted in a decrease in the heat resistance of spore microorganisms, making it possible to reduce the duration of sterilization for canned food to 5-10 min at $115^{\circ}C$. This reduction in the sterilization duration of canned seafood with spice-oil extract inhibited residual microflora in the product, thus reducing the deleterious effect of heating on the main food compounds while preserving protein digestibility.

  • A synthesis method for a chitosan-coated magnetic drug-delivery system of cisplatin is proposed. Here, cisplatin was conjugated to the surface of Magnetite ($Fe_3O_4$) nanoparticles via a (3-Aminopropyl)-trimethoxysilane (APTS) coupling agent. To reduce the cytotoxic effect of cisplatin, the magnetic drug was then encapsulated in chitosan (CS-cisplatin-$Fe_3O_4$) through the water/oil (W/O) emulsion method. The CS-cisplatin-$Fe_3O_4$ nanoparticles were synthesized in a spherical shape with a diameter of 190 nm. The cytotoxicity assay was performed using HeLa cells. The cisplatin uptake of the cells was determined using High Performance Liquid Chromatography (HPLC) to calculate the drug content. The controlled release of cisplatin was demonstrated by regulating the dissolution and diffusion of the drug through the chitosan matrix.

  • The sustainable development is a key issue in the whole field of economy, culture and society, which can be accomplished by the improvement of environment. Recently, life cycle assessment(LCA) has been applied to reduce environmental impacts preliminarily by evaluating the environmental performance of a product through its life cycle. In this study, life cycle assessment was performed to analyze quantitatively the environmental impact on the interior panel of electric motor unit(EMU). As a result, the interior panel with aluminum showed the most global warming(GW), while that with phenol and plastic showed high fresh water aquatic ecotoxicity(FAET) and marine water aquatic ecotoxicity(MAET), respectively. Global warming was occurred mainly due to the emission of $CO_2$ by energy consumption. FAET and MAET were caused by the pollutants released from acid-washing and paints coating process. Therefore, an environmental-friendly EMU can be designed considering the environmental impacts of interior panel.

  • Environmental problem is one of important global issues. Transportations are main pollutant emission sources. Although railroad is stilt an environmental-friendly transportation, its environmental impact has been increased continuously. Especially, because a large amount of environmental impact is released from vehicles and facilities, it is necessary to assess and to reduce their environmental impact. Life cycle assessment (LCA) is a representative method which can evaluate environment impact through the whole life cycle of a product or a process. In this study, the environmental impact of seat in the electric motor unit (EMU) was analyzed quantitatively with its material using lift cycle assessment (LCA). As a result, the characteristics of environmental impact were investigated differently with the material of seat. Among ten impact categories, the seat with aluminum and FRP showed the highest ozone depletion (OD). On the other hand, in the seat with stainless steel and plastic, fresh water aquatic ecotoxicity (FAET) and marine water aquatic ecotoxicity (MAET) were high relatively. Therefore, the parts of EMU must be selected considering the characteristics of environmental impact in future.

  • In order to isolate and clarify the antioxygenic substances from the browning reaction products, the antioxidant activity of various amino acids and their browning reaction products were measured when they were reacted with xylose. Among nonpolar amino acids Met and Trp appeared to have stronger antioxidant effect than others. Most of polar and basic amino acids, however, did not have antioxidant activity. Ser and Cys showed a rather slight prooxidant effect. The browning reaction products of Trp and His had a higher level of antioxidant activity than that they were reacted as free amino acids. But the browning product of Met did not show the antioxidant activity. When all amino acids were divided on their polar characteristics, the higher optical density of the browning reaction products showed, the stronger antioxidant activity revealed.

Distillers Dried Grain from Makgeolli By-product Is Useful as a Dietary Ingredient for Growth of Juvenile Sea Cucumber Apostichopus japonicus

  • We tested the effects of various distillers dried grains (DDGs) in a formulated diet on growth and body composition of juvenile sea cucumber Apostichopus japonicus. DDG is a solid residue obtained by filtering an aqueous mixture of fermented rice with Aspergillus oryzae and yeasts. Six isonitrogenous and isocaloric diets (DDG0, DDG-G, DDG-C, DDG-K, DDG-W, and DDG-P) were formulated to contain 20% DDG obtained from different Makgeolli factories. Juvenile sea cucumber (average weight, $2.1{\pm}0.02$ g) were randomly distributed in eighteen 50-L rectangular plastic tanks (40 L of water each) in a seawater flow-through system at a density of 50 juveniles/tank. Three replicate groups of sea cucumber were fed each of the six experimental diets at a feeding rate of 5% body weight per day for 22 weeks. At the end of the feeding experiment, survival and weight gain of the juvenile sea cucumber were not affected by the type of dietary DDG (P > 0.05), and the proximate and amino acid compositions of the whole body were not affected by dietary DDG diets (P > 0.05). These results indicate that rice-based DDG is a potential dietary ingredient that could be used at dietary concentrations of up to 20% for growth of juvenile sea cucumber.

Studies in Browning Reaction in Dried Fish Lipid Oxidative Browning in Dried Conger eel and Properties of Browning Products (수산건제품의 갈변에 관한 연구 붕장어육 및 유의 산화, 갈변 물질의 성상)

  • This studies was carried out in order to investigate the browning reaction of lipid originated compound with nitrogenous compound in dried conger eel. The major fatty acids were $C_{16:0},\;C_{16:1},\;C_{18:1},\;C_{20:5}\;and\;C_{22:6}$. The nonpolar lipid contained the highest percentage of $C_{16:0}$, while the polar lipid contained the highest percentage of $C_{22:6}$. The browning reaction there was a rapidly developed with the beginning of the decline in carbonyl value and remarkable decrease in polyunsaturated fatty acids such as $C_{20:5},\;C_{22:5},\;C_{22:6}$ compared with the other fatty acid, in the water soluble fraction of the browning product obtained from tile fish was detected some antioxidation activity but in the lipid soluble fraction which covers most of the browning reactions in the fish meat antioxidation activity was not detected. In the test of conger eel oil, the phosphatidylcholine was largest in quantity and browning products provided in this experiment showed very low reducing activity.

  • Yellow sea bream contains comparatively larger amount of fat among white muscle fishes, so that rancidity might easily occur during drying and storage. For the purpose of the protection of rancidity, the effect of some antioxidants was studied when yellow sea bream was sun-dried after dipping in the solutions and packed in PVC film$0.3mm\times12cm\times30cm $ for storage at room temperature. The inhibitory effect of additives was in order of Tenox-II, BHA, Sustane and NDGA, while EDTA, potassium sorbate, CTC and $\alpha-naphthylamine$ were ineffective. The results suggest that the treatment of $0.1\%$ Tenox-II solution and packing in PVC film is better condition to improve the quality of product and during drying and storage.
