천해역 먹이망 생태계에 대한 무기부유입자와 카드뮴의 영향 II. Tigriopus japonicus에 대한 카드뮴의 급성 및 만성독성 (Effects of Suspended Solid and Cadmium on the Shallow-sea Ecosystem II. Acute and Chronic Toxicity of Cadmium to a Herbivorous Copepod, Tigriopus japonicus)

  • 초식성 요각류인 Tigriopus japonicus를 대상으로 사망률과 반수치사농도$(LC_{50})$ 및 호흡대사에 미치는 카드뮴의 급성 및 만성독성영향을 조사하였던바 다음과 같은 결과를 얻었다. 염분 32, 26 및 $18\%_{\circ}$에서 T. japonicus에 대한 카드뮴의 96시간 반수치사농도$(96hr.-LC_{50})$$20^{\circ}C$에서 각각 15.1, 13.7 및 9.9 ppm이었고, $10^{\circ}C$에서는 각각 21.6, 15.0 및 11.0ppm으로 나타나 수온의 감소와 염분의 증가에 따라 반수치사농도는 증가되었다. 96시간동안 카드뮴에 노출시킨 후 산소소비율은 $10^{\circ}C$에 비해 $20^{\circ}C$에서 높게 나타났으며, 카드뮴농도의 증가에 따라 보다 크게 감소되었다. 염분 $32\%_{\circ}$ 및 Cd농도 4ppm에서 T. japonicus의 생존률과 25일동안의 반수치수농도$(25days-LC_{50})$$10^{\circ}C$의 경우 각각 $5\%$와 0.05ppm이었고, $20^{\circ}C$에서는 $0\%$와 0.28ppm을 나타내었다.

  • 우리나라산 주요 패류에 대한 독의 분포특성 등에 대한 연구의 일환으로 우선 1986년 3월 부산시 사하구 구평동 지선에 소재하는 모회사(폐선을 해체하여 고철로 활용)의 작업인부들이 먹고 식중독 사고를 일으킨 원인식품인 진주담치를 시료로 하여 biassay를 통한 마비성 패류독의 독성을 조사하고 독소를 분리하여 electrophoresis, TLC, HPLC로 분석한 결과를 요약하면 다음과 같다. 1. 식중독사고의 원인이 된 진주담치의 마비성 패류독 함량은 $132\~295MU/g$, 또는 $26.4\~58.9{\mu}g/g$ 였다. 2. 독화된 진주담치의 중장선부위에는 독성이 $439\~979MU/g$나 되어 육질부위의 약 9배에 달하였으며 전체 독성의 $70\%$ 정도가 중장선부위에 축적되어 있었다. 3. 마비성 패류독을 분리하여 전기영동, TLC, HPLC로 분석한 결과 주성분은 $Gonyautoxin_{1\~4}$ 였으며, Saxitofin 군은 아주 미량검출되었다. 이상의 결과로서 본 마비성 패류독에 의한 식중독 사고의 원인물질은 Saxitoxin이 아니고 Gonyautoxin 임을 알 수 있었다.

  • 연안 및 천해에 서식하는 반부유성 갑각류인 곤쟁이, Neomysis awatschensis를 대상으로 수온별 PCB의 시험농도에 따른 성장, 호흡, 질소배설, 섭이 및 에너지 함량을 측정하여 에너지 수지를 분석하였다. 성장, 탈피 및 호흡에 사용한 에너지를 파악하기 위해 곤쟁이의 몸체 및 탈피체의 에너지 함량을 구한 결과 각각 5.52cal와 2.17ca1였다. 곤쟁이 치하가 성체로 성장하는 동안 $10^{\circ}C$의 경우, 섭이에너지는 대조구에서 3.755ca1, 2.0ppb에서는 3.420cal 였으며, 이 가운데 각각 $70.19\%$$67.53\%$를 동화하였다. 그리고 $20^{\circ}C$에서는 섭이에너지가 대조구에서 5.998ca1, 2.0ppb에서는 4.166cal를 섭이하였으며, 동화효율은 각각 $76.50\%$$70.50\%$로 2.0ppb 농도구에서 낮았다. 그리고 $20^{\circ}C$의 경우 총동화에너지의 각 부분별 사용에너지 중 호흡에 사용한 에너지가 대조구에서 $45.78\%$, 2.0 ppb농도에서 $62.27\%$으로 대조구에 비해 2.0ppb 농도구 에서 대사에 사용하는 에너지가 많았다.

Paralytic shellfish toxins (PSTs) and tetrodotoxin (TTX) of Korean pufferfish

  • Paralytic shellfish toxins (PSTs) and tetrodotoxin (TTX) are neurotoxins that display pharmacological activity that is similar to that of specific sodium channel blockers; they are the principle toxins involved in shellfish and puffer fish poisoning. In Korea, puffer fish is a very popular seafood, and several cases of accidental poisoning by TTX have been reported. Therefore, it is necessary to determine whether puffer fish poisoning incidents are caused by PSTs or by TTX. In this study, we used mouse bioassay (MBA) and liquid chromatograph-tandem mass spectrometry (LC-MS/MS) to determine the presence of PSTs and TTX in puffer fish from an area near Mireuk-do, Tong-Yeong on the southern coast of Korea from January through March, 2014. The toxicity of PSTs and TTX extracts prepared from three organs of each specimen was analyzed by MBA. Most of the extracts killed mice with typical signs of TTX and PSTs. The LC-MS/MS analysis of seven specimens of Takifugu pardalis and Takifugu niphobles, each divided into muscles, intestines, and liver, were examined for TTX. In T. pardalis, the TTX levels were within the range of 1.3-1.6 ㎍/g in the muscles, 18.8-49.8 ㎍/g in the intestines, and 23.3-96.8 ㎍/g in the liver. In T. niphobles, the TTX levels were within the range of 2.0-4.5 ㎍/g in the muscles, 23.9-71.5 ㎍/g in the intestines, and 28.1-114.8 ㎍/g in the liver. Additionally, the toxicity profile of the detected PSTs revealed that dcGTX3 was the major component in T. pardalis and T. niphobles. When PSTs were calculated as saxitoxin equivalents the levels were all less than 0.5 ㎍/g, which is below the permitted maximum standard of 0.8 ㎍/g. These findings indicate that the toxicity of T. pardalis and T. niphobles from the southern coast of Korea is due mainly to TTX and that PSTs do not exert an effect.

  • The life cycle assessment method for environmental impact assessment was used, in this study, to assess the production process of wheat flour which is the most important material in the food industry. Environmental impact assessments were compared between that of the Ministry of Environment, Republic of Korea (method I) with that of the Ministry of Commerce, Industry and Energy (method II). Life cycle inventories (LCI) was performed using internal and external databases and the production statistics database of company S. The procedure of life cycle impact assessment (LCIA) was followed in terms of classification, characterization, normalization and weighting to identify the key issues. The impact categories of method I were divided into 8 categories with consideration of : abiotic resources depletion, global warming, ozone depletion, photochemical oxidant creation, acidification and eutrophication. The impact categories of method II were divided into 10 categories with consideration of: abiotic resources depletion, global warming, ozone depletion, photochemical oxidant creation, acidification, eutrophication, human toxicity, freshwater aquatic ecotoxicity, marine aquatic ecotoxicity and terrestrial ecotoxicity.

지중해담치, Mytilus galloprovincialis의 배 발생에 미치는 다환방향족탄화수소류 (2-methylnaphthalene, fluorene, dibenzothiophene, phenanthrene, pyrene) 의 영향 (Effect of five PAHs (2-methylnaphthalene, fluorene, dibenzothiophene, phenanthrene, and pyrene) on the embryonic development in the mussel, Mytilus galloprovincialis)

  • Mussels have been commonly used in bioassay for quality assessments of environment. Moreover, several standard protocols for the developmental bioassay of bivalves have been proposed. In this study, the EC50 of polycyclic aromatic hydrocarbons (PAHs) was determined using mussel, Mytilus galloprovincialis embryonic developmental bioassay. To determine the sensitivity of M. galloprovincialis, their fertilized eggs were exposed to five PAHs (2-metylnaphthalene, fluorene, dibenzothiophene, phenanthrene, pyrene). The EC50 of 2-metylnaphthalene, fluorene, dibenzothiophene, phenanthrene, and pyrene were 232, 273, 67.9, 43.2, and $33.1{\mu}g/L$, respectively. The overall sensitivity of M. galloprovincialis in the present developmental bioassay was similar to or more sensitive than that of other marine organisms commonly used in aquatic bioassays. The results of this study could be provide with fundamental data of setting standard for protection of marine life and or can use prediction the aquatic toxicity of PAHs.

Nutritional Components and Safety of Purified Pufferfish (Lagocephalus gloveri) Liver Oil

  • The safety of pufferfish (Lagocephalus gloveri) liver oil and the contents of some nutritional components were examined to obtain important information on their use as high valued functional foods. Pufferfish liver oil was extracted by the hot-water method using $1\%$ NaOH solution to remove toxic compounds, and then purified using a general purifying method of fish oil. Any extraordinary clinical symptoms were not observed from all groups administrated with pufferfish liver oil throughout the test period. None of the rats died when administrated the highest concentration of 10 mL/kg of the pufferfish liver oil. Vitamin A content was 114.2 ppm, as a retinal equivalent in the oil extracted using hot-water, but the content was higher (169.3 ppm) in oil extracted using n-hexane. Vitamin D and E were not detected in ppm in oil extracted using hot-water. Vitamin D in the pufferfish liver oil extracted using n-hexane was also undetected, but vitamin E was at 32.5 ppm. Among the 18 minerals detected, the sodium content was the highest at 253.5 ppm, followed by 13.9 ppm ofpotassium, 1.5 ppm of calcium, 0.2 ppm of magnesium, and other trace minerals. The contents of EPA and DHA were $0.8\%\;and\;14.8\%$, respectively, in the pufferfish liver oil extracted using hot-water. Considering these results, there is potential that pufferfish liver oil could be used as a functional food.

냉동과 해동에 따른 국매리복 근육의 독화 (Muscle Intoxication of 'Gukmeri-bok' Puffer (Takifugu vermicularis radiatus) by Freezing and Thawing)

  • We here report that the muscle of 'gukmeri-bok' puffer fish (Takifugu vermicularis radiatus) is intoxicated after freezing and thawing processes even though it is not toxic when it is fresh. This study was carried out to investigate the effect of different freezing and thawing conditions as well as the effect of the presence of skin or viscera on the intoxication of muscle of 'gukmeri-bok' puffer during freezing and thawing process. As a result of this investigation, thawing condition, but not freezing conditions or periods played an important role in muscle intoxication. Tetrodotoxin secretory grands exist in the skin of toxic puffer fish, the skin was more responsible for the muscles intoxication than the viscera during the freezing-thawing process. In other words, no toxicity was measured in the muscle of skinned-frozen specimens even when thawed. According to this result, it is recommended that the skin and viscera must be removed before being frozen for edible purpose. Otherwise, when a whole fish should be frozen, alternative half-thawing and removal of skin from frozen specimens is recommended.

Aluminum Inhibits Vitellogenin Production via Toxic Effects on Hepatocytes in the Rockfish Sebastes schlegelii

  • Effects of aluminum (Al) on plasma vitellogenin (VTG), alkaline-labile phosphorus (ALPP), calcium (Ca), glutamate pyruvate transaminase (GPT), the hepatosomatic index (HSI), and hepatic Al concentration were examined in estradiol-$17{\beta}$ ($E_2$)-administered immature rockfish Sebastes schlegeli. Fish were injected intraperitoneally with $E_2$ (5 mg/kg body weight [BW]) and/or Al (0, 0.1, 1, 5, and 10 mg/kg BW) and plasma and liver samples were extracted 7 days later. After sodium dodecyl sulfate polyacrylamide gel electrophoresis, the relative amount of VTG was determined by integrated optical density. VTG accounted for 23.6% of the total proteins in the control group, but this value decreased with increasing Al administration. Al reduced the concentrations of ALPP and Ca in a concentration-dependent manner and significant reduction occurred at Al concentrations greater than 5 mg/kg. The concentration of GPT increased in a concentration-dependent manner in all Al-administered rockfish. The concentrations of Al in the liver also increased, and HSI was decreased, in a concentration-dependent manner. These results suggest that Al inhibits $E_2$-induced VTG production by being toxic to hepatocytes in marine fish.

Anti-inflammatory Activities of Undaria pinnatifida and Laminaria japonica (Phaeophyta)

  • The anti-inflammatory activities of dichloromethane, ethanol, and boiling water extracts of the brown seaweeds Undaria pinnatifida (Harvey) Suringar and Laminaria japonica Areschoug were examined. Ethanol extracts (0.4 mg/ear) of U. pinnatifida inhibited inflammatory symptoms in mouse ear edema by 95.3%, and dichloromethane extract inhibited erythema by 65.5%. Dichloromethane and ethanol extracts (4 g/kg bw) of L. japonica demonstrated potent antipyretic activity. Activities of the seaweed extracts were similar to those of the commonly used drugs indomethacin and acetyl salicylic acid. No acute toxicity was observed after p.o. administration of each extract (5 g/kg bw). These results were in agreement with the claims of the health care industry and indigenous medicine that the above seaweeds can be used as an effective remedy for inflammation-related symptoms.
