Mouse Single Oral Dose Toxicity Test of Red Koji Fermented Scutellariae Radix Aqueous Extracts (홍국 발효 황금의 마우스 단회 경구투여 독성시험)

  • Objectives : The objectives of this study was to obtain acute information (single oral dose toxicity) of Red-Koji (Monascus purpureus 12002) Fermented Scutellariae Radix Aqueous Extracts (fSR), has been traditionally used in Korean medicine for treating various diseases including inflammatory diseases. Methods : In order to observe the 50% lethal dose (LD50), approximate lethal dosage (ALD) and target organs, fSR powders were once orally administered to female and male ICR mice at dose levels of 2,000, 1,000, 500 and 0 (control) mg/kg (body weight.). The mortality and changes on body weight, clinical signs and gross observation were monitored during 14days after single oral treatment of fSR with organ weights and histopathological observations of 12 types of principle organs. Results : After single oral treatment of fSR, we could not find any mortality and toxicological evidences up to 2,000 mg/kg treated group, the limited dosages in rodents, on the body and organ weights, clinical signs, gross and histopathological observations, except for some accidental findings. Conclusions : The results obtained in this study suggest that the LD50 and ALD of fSR in both female and male mice after single oral treatment were considered as over 2,000 mg/kg because no mortalities were detected up to 2,000 mg/kg and can be safety used in clinics.

Single Oral Dose Toxicity Test of Taeumjowi-tang in ICR Mice (태음조위탕(太陰調胃湯)의 마우스 단회투여 독성시험)

  • The object of this study was to evaluate the single dose toxicity of Taeumjowui-tang(TUJWT), a polyherbal formula have been traditionally used as prevention or treatment agent for obesity, in male and female mice. Aqueous extracts of TUJWT (Yield = 10.5%) was administered to female and male ICR mice as an oral dose of 2000, 1000 and 500 mg/kg (body wt.) according to the recommendation of KFDA Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy, organ weight and histopathology of 12 principle organs were examined. As results, we could not find any mortality, clinical signs, and changes in the body and organ weight except for diarrhea restricted in TUJWT 2000 mg/kg treated one male mouse at 1 day after administration, and increases of popliteal lymph node weights in all TUJWT administered groups. In addition, no TUJWT-treatment related abnormal gross findings and changes in histopathology of principle organs were detected except for increases of the frequency of hypertrophy and hyperplasia of lymphoid cells in the popliteal lymph nodes in all TUJWT treatment groups with some sporadic accidental findings. These increases of popliteal lymph node weights with hypertrophy and hyperplasia of lymphoid cells were considered that related to the immune modulate effect of TUJWT not toxicological signs. The results obtained in this study suggest that the TUJWT does not cause any toxicological signs. The 50% lethal dose and approximate lethal dose of TUJWT aqueous extracts in both female and male mice were considered as over 2000 mg/kg.

In order to observe the 50% lethal dose, approximate lethal dosage, maximum tolerance dosage and target organs, test articles were once orally administered to ICR mice at dose levels of 2000, 1000, 50 mg/kg according to the recommendation of KFDA Guidelines. The mortality and changes on body weight, clinical signs and gross observation were monitored during 14 days after dosing according to KFDA Guidelines with organ weights of 12 types of principle organs. In addition, after twice oral treatment of Mahwangbujaseshin-tang extracts 2000, 1000 and 500 mg/kg, we checked the changes on the number of MNPCE. We could not find any mortality, clinical signs, changes in the body weight and gross findings upto 2000 mg/kg treated group. The limited dosages in rodents except for increases of lymphoid organ weights and hypertrophy encounted as results from pharmacological effects of Mahwangbujaseshin-tang extracts, immune modulator effects with some sporadic accidental findings not toxicological signs. No evidence of increases of MNPCE numbers were also detected in all three different dosages of Mahwangbujaseshin-tang extracts treated mice. The results obtained in this study suggest that the LD50 and ALD of Mahwangbujaseshin-tang extracts in mice were considered as over 2000 mg/kg because no mortalities were detected upto 2000 mg/kg that was the highest dose recommended by KFDA and OECD. And the results of mouse bone marrow micronucleus test of Mahwangbujaseshin-tang extracts is negative results.

Results : After single oral treatment of SR, we could not find any mortality and toxicological evidences up to 2,000 mg/kg treated group, the limited dosages in rodents, on the body and organ weights, clinical signs, gross and histopathological observations, except for some accidental findings. Conclusions : The results obtained in this study suggest that the $LD_{50}$ and ALD of SR in both female and male mice after single oral treatment be considered as over 2,000 mg/kg because no mortalities were detected up to 2,000 mg/kg that was the highest dose recommended by KFDA and OECD, and can be safely used in clinics.

Prediction of Wind Power by Chaos and BP Artificial Neural Networks Approach Based on Genetic Algorithm

  • It is very important to make accurate forecast of wind power because of its indispensable requirement for power system stable operation. The research is to predict wind power by chaos and BP artificial neural networks (CBPANNs) method based on genetic algorithm, and to evaluate feasibility of the method of predicting wind power. A description of the method is performed. Firstly, a calculation of the largest Lyapunov exponent of the time series of wind power and a judgment of whether wind power has chaotic behavior are made. Secondly, phase space of the time series is reconstructed. Finally, the prediction model is constructed based on the best embedding dimension and best delay time to approximate the uncertain function by which the wind power is forecasted. And then an optimization of the weights and thresholds of the model is conducted by genetic algorithm (GA). And a simulation of the method and an evaluation of its effectiveness are performed. The results show that the proposed method has more accuracy than that of BP artificial neural networks (BP-ANNs).

  • RBFN has some problem that because the basis function isn't orthogonal to each others the number of used basis function goes to big. In this reason, the Wavelet Neural Network which uses the orthogonal basis function in the hidden node appears. In this paper, we propose the composition method of the actual function in hidden layer with the scaling function which can represent the region by which the several wavelet can be represented. In this method, we can decrease the size of the network with the pure several wavelet function. In addition to, when we determine the parameters of the scaling function we can process rough approximation and then the network becomes more stable. The other wavelets can be determined by the global solutions which is suitable for the suggested problem using the genetic algorithm and also, we use the back-propagation algorithm in the learning of the weights. In this step, we approximate the target function with fine tuning level. The complex neural network suggested In this paper is a new structure and important simultaneously in the point of handling the determination problem in the wavelet initialization.

  • We consider transmission beamforming techniques for frequency-division-duplex (FDD) wireless communication systems using adaptive arrays to improve the signal quality of the array transmission link. We develop a simple effective transmission beamforming technique based on an approximated frequency tranlsation (AFT) to derive the tranmsiion beamforming weights from the uplink channel vector. This technique exploits the invariance of the short-time averaged fast fading statistics to small frequency translations. A simple approximate relationship that relates the transmission channel vector to the reception channel vector is derived. We have developed its practical alternative in which the frequency translation of the channel vector is performed at the principal angle of arrival (AOA) of the u;link synthestic angular spectrum instead of the mean AOA. To analyze the performance of the proposed methods, we consider the power loss incurred by applying the estimated channel vector instead of the true downlink channel vector. The performance is analyzed as a function of the mean AOA, the angular spread, the number of elements, frequncy difference between the uplink and the downlink, and the angle distribution. Their performance is also compared with that of the direct weight reuse method and the AOA based methods.

  • This paper presents a neural net based nonlinear adaptive controller for an autonomous underwater vehicle (AUV). AUV's dynamics are highly nonlinear and their hydrodynamic coefficients vary with different operational conditions, so it is necessary for the high performance control system of an AUV to have the capacities of learning and adapting to the change of the AUV's dynamics. In this paper a linearly parameterized neural network is used to approximate the uncertainties of the AUV's dynamics, and a sliding mode control is introduced to attenuate the effects of the neural network's reconstruction errors and the disturbances of AUV's dynamics. The presented controller is consist of three parallel schemes; linear feedback control, sliding mode control and neural network. Lyapunov theory is used to guarantee the asymptotic convergence of trajectory tracking errors and the neural network's weights errors. Numerical simulations for motion control of an AUV are performed to illustrate to effectiveness of the proposed techniques.

Neural-based Blind Modeling of Mini-mill ASC Crown

  • Neural network can be trained to approximate an arbitrary nonlinear function of multivariate data like the mini-mill crown values in Automatic Shape Control. The trained weights of neural network can evaluate or generalize the process data outside the training vectors. Sometimes, the blind modeling of the process data is necessary to compare with the scattered analytical model of mini-mill process in isolated electro-mechanical forms. To come up with a viable model, we propose the blind neural-based range-division domain-clustering piecewise-linear modeling scheme. The basic ideas are: 1) dividing the range of target data, 2) clustering the corresponding input space vectors, 3)training the neural network with clustered prototypes to smooth out the convergence and 4) solving the resulting matrix equations with a pseudo-inverse to alleviate the ill-conditioning problem. The simulation results support the effectiveness of the proposed scheme and it opens a new way to the data analysis technique. By the comparison with the statistical regression, it is evident that the proposed scheme obtains better modeling error uniformity and reduces the magnitudes of errors considerably. Approximatly 10-fold better performance results.
