Marssonina coronaria associated with apple blotch disease causes severe premature defoliation, and is widely distributed in Korea. Thirteen isolates were collected from orchards located in Gyeongbuk Province from 2005~2007. All isolates displayed over 99.6% and 99.2% sequence similarity to each other in internal transcribed spacer regions and partial sequences of 28S rDNA, respectively. The isolates were phylogenetically closely related to Chinese isolates. Selected isolates did not differ in their pathogenicity. The optimum conditions for fungal growth were $20^{\circ}C$ and pH 6 on peptone potato dextrose agar (PPDA). Peptone and mannose were the best nitrogen and carbon source, respectively. Fungal growth was better on PPDA than on common potato dextrose agar. This study provides valuable information for integrated disease management program and facilitates the routine culturing of M. coronaria.

This study was carried out to establish a spatial decision support system for evaluating climatic aspects of a given geographic location in complex terrains with respect to the quality apple production. Monthly climate data from S6 synoptic stations across South Korea were collected for 1971-2000. A digital elevation model (DEM) with a 10-m cell spacing was used to spatially interpolate daily maximum and minimum temperatures based on relevant topoclimatological models applied to Jangsoo county in Korea. For daily minimum temperature, a spatial interpolation scheme accommodating the potential influences of cold air accumulation and the temperature inversion was used. For daily maximum temperature estimation, a spatial interpolation model loaded with the overheating index was used. Freezing risk in January was estimated under the recurrence intervals of 30 years. Frost risk at bud-burst and blossom was also estimated. Fruit quality was evaluated for soluble solids, anthocyanin content, Hunter L and A values, and LID ratio, which were expressed as empirical functions of temperature based on long-term field observations. AU themes were prepared as ArcGlS Grids with a 10-m cell spacing. Analysis showed that 11 percent of the whole land area of Jangsoo county might be suitable for quality 'Fuji' apple production. A computer program (MAPLE) was written to help utilize the results in decision-making for site-selection of new orchards in this region.

This study quantified the storage and annual uptake of carbon by apple trees in orchards as a production-type greenspace, and computed the annual carbon emissions from apple cultivation. Tree individuals in the study orchards were sampled to include the range of stem diameter sizes. The study measured biomass for each part including the roots of sample trees through a direct harvesting method to compute total carbon storage per tree. Annual carbon uptake per tree was quantified by analyzing the radial growth rates of stem samples at ground level. Annual carbon emissions from management practices such as pruning, mowing, irrigation, fertilization, and use of pesticides and fungicides were estimated based on maintenance data, interviews with managers, and actual measurements. Regression models were developed using stem diameter at ground level (D) as an independent variable to easily estimate storage and annual uptake of the carbon. Storage and annual uptake of carbon per tree increased as D sizes got larger. Apple trees with D sizes of 10 and 15 cm stored 9.1 and 21.0 kg of carbon and annually sequestered 1.0 and 1.6 kg, respectively. Storage and annual uptake of carbon per unit area in study orchards were 3.81 t/ha and 0.42 t/ha/yr, respectively, and annual carbon emissions were 1.30 t/ha/yr. Thus, the carbon emissions were about 3 times greater than the annual carbon uptake. The study identified management practices to reduce the carbon footprint of production-type greenspace, including efficient uses of water, pesticides, fungicides, and fertilizers. It breaks new ground by including measured biomass of roots and a detailed inventory of carbon emissions.

Spring phonology of the oriental fruit moth, Grapholita molesta, was monitored using sex pheromone traps in apple cultivating areas. Their occurrence was earlier in southern areas and their population sizes were significantly different among orchards even in a local cultivating zone. The overwintering populations appeared to move between local orchards, based on the fact that monitoring data obtained at the sites between orchards were similar to those of nearby orchards. However, within orchards, these adult movements appeared to decrease and showed skewed occurrences at the side of upwind direction or close to neighboring orchards. At initial occurrence peak(April 20-25), the ovenvintering populations of the different localities were collected and analyzed in their genetic distances. PCR-RAPD analysis indicated that there were significant genetic differences among the overwintering populations of G. molesta. This genetic differentiation of overwinterin populations may be due to genetic bottleneck following differential selection pressures against the subpopulations of G. molesta during winter on the basis of the RAPD analysis that each early spring population was significantly different to its previous fall population in the same locality.

Most apple trees in South Korea are grafted on M.9 and M.26 rootstocks; however, these rootstocks are susceptible to fire blight. Although M.7 rootstocks are moderately resistant to fire blight, they tend to exhibit excessive vigor, which is unsuitable for high-density planting, unless weak cultivars are used. This study investigated the vegetative growth, yield, and fruit quality of apple trees grafted onto M.7, M.9, or M.26 rootstocks to assess the feasibility of establishing high-density apple orchards domestically using the M.7 rootstock a period of seven years (1-7 years after planting). Rootstocks were tested using three cultivars with contrasting induced vigor and harvesting times: vigorous and late-maturing 'Fuji,' moderate vigor and middle-maturing 'Hongro,' and low vigor and early-maturing 'Sansa.' The planting density was maintained constant, with 190 trees per 10 a. Primary thinning (leaving only the king fruit on clusters) was performed, whereas secondary thinning (controlling crop load) was not. Vegetative growth, accumulated yield per 10 a, and yield efficiency varied depending on cultivars and rootstocks; however, the cultivars had a more notable effect on fruit quality than the rootstocks. Biennial bearing often occurred in the M.26 rootstock. 'Fuji'/M.7 was overly vigorous for high-density planting. The fruit quality and accumulated yield per 10 a of M.7 were similar to those of M.9 with the 'Hongro' and 'Sansa' cultivars. In particular, 'Hongro'/M.7 did not show tree vigor reduction due to heavy crop load, and the degree of biennial bearing in 'Sansa'/M.7 was not particularly high. These results indicated that high-density apple planting using the M.7 rootstock was achievable using the 'Hongro' and 'Sansa' cultivars.
Peach (Prunus persica L.) is one of the major fruit crops in South Korea, along with apple, persimmon, and Asian pears. Peach anthracnose is a continuing threat to growers and is accountable for enormous economic loss. In July 2018, anthracnose of peach appeared at different peach orchards in Gyeongsangbuk-do region, Korea. The typical anthracnose symptoms (brown, circular, and necrotic lesions) were observed on the fruits. Anthracnose of peach was surveyed in different peach orchards of Gyeongsangbuk-do, and 20 fungal isolates from 19 diseased fruits were collected. Multigene phylogenetic analyses coupled with morphological characteristic analysis approaches were used for identifying the fungal species isolated from diseased fruits. This study confirmed three Colletotrichum species. Based on the results, Colletotrichum siamense are reported for the first time as causal agents of peach anthracnose alongside C. fructicola and C. fioriniae, which has been reported previously. Pathogenicity assays were performed for the three isolates representing all the species identified, and Koch's postulates on detached healthy peach fruits were verified. All the identified species were pathogenic on peach fruits as the typical anthracnose symptoms were reproduced. Significant variations in the virulence were observed among fungal species on peach fruit.
This study was carried out to evaluate the effect of pre-harvest factors on fruit quality and the incidence of storage disorders in 'Fuji' apple during CA storage. Incidence of storage disorders varied, depending on the growing regions, field conditions, and altitude of the orchards. Results indicated that fruit maturity may play a crucial role in the incidence and severity of flesh browning and watercore. The incidence of these storage disorders increased with fruit maturity until 180 days after full bloom (DAFB). In addition, occurrence of watercore was correlated with the incidence of flesh browning during CA storage. The incidence of flesh browning was positively correlated with the sum of the diurnal temperature range from September through October and amount of precipitation from August to October in 1996-1999 growing years. These results suggested that 'Fuji' apple should be harvested no later than 180 DAFB because the late harvested fruits were prone to development of watercore which was correlated with the incidence of flesh browing during CA storage in Korea.

BACKGROUND: Since 2015, fire blight disease caused by Erwinia amylovora has been devastating apple and pear orchards every year. To quickly block the disease spreading, infected apple and pear trees have been buried in soil. However, concern on the possibility of the pathogen survival urgently requires informative data on the buried host plants. Therefore, this study was conducted to investigate the survival of the pathogen from the buried host plants. METHODS AND RESULTS: Apple trees buried in 42 months ago in a Jecheon site and pear trees buried in 30 months ago in an Anseong site were excavated using an excavator. Plant samples were taken from stems and twigs of the excavated trees. The collected 120 samples were checked for rotting and used for bacterial isolation, using TSA, R2A, and E. amylovora selection media. The purely isolated bacteria were identified based on colony morphology and 16S rDNA sequences. Wood rotting and decay with off smells and discoloring were observed from the samples. A total of 17 genera and 48 species of bacteria were identified but E. amylovora was not detected. CONCLUSION: Our investigation suggests that the survival of E. amylovora doesn't seem possible in the infected hosts which have been buried in soil for at least 30 months. Therefore, the burial control can be considered as a safe method for fire blight disease.
A destructive collar rot of apple caused by a species of Phytophthora has widely occurred in Kyungbuk and less extended in Chungbuk, Chungnam and Chunbuk provinces of Korea. Significantly higher incidence of the disease was observed on cv. Fuji when M26 or M9 was used as dwarfing stocks. Incidence of the disease at several orchards in Uisung, Kunwi, Yesan and Muju ranged from 45 to 80%. Twenty-five isolates of the causal fungus were collected and all isolates were identified as P. cactorum on the basis of their cultural and morphological characters. The fungus produced markedly papillate and broadly ovoid deciduous sporangia both on agar and in water, and a short pedicel was attached to each sporangium. Oospores were readily formed on clarified V8 agar by single isolates and all the antheridia were paragynous. The fungus neither grew nor produced oospores under 5 and over 33$^{\circ}C$. The destructive collar rot of apple caused by P. cactorum has not been reported in Korea previously.
Fast and accurate diagnosis is needed to eradicate and manage economically important and invasive diseases like fire blight. Loop-mediated isothermal amplification (LAMP) is known as the best on-site diagnostic, because it is fast, highly specific to a target, and less sensitive to inhibitors in samples. In this study, LAMP assay that gives more consistent results for on-site diagnosis of fire blight than the previous developed LAMP assays was developed. Primers for new LAMP assay (named as DS-LAMP) were designed from a histidine-tRNA ligase gene (EAMY_RS32025) of E. amylovora CFBP1430 genome. The DS-LAMP amplified DNA (positive detection) only from genomic DNA of E. amylovora strains, not from either E. pyrifoliae (causing black shoot blight) or from Pseudomonas syringae pv. syringae (causing shoot blight on apple trees). The detection limit of DS-LAMP was 10 cells per LAMP reaction, equivalent to $10^4$ cells per ml of the sample extract. DS-LAMP successfully diagnosed the pathogens on four fire-blight infected apple and pear orchards. In addition, it could distinguish black shoot blight from fire blight. The $B{\ddot{u}}hlmann$-LAMP, developed previously for on-site diagnosis of fire blight, did not give consistent results for specificity to E. amylovora and on-site diagnosis; it gave positive reactions to three strains of E. pyrifoliae and two strains of P. syringae pv. syringae. It also, gave positive reactions to some healthy sample extracts. DS-LAMP, developed in this study, would give more accurate on-site diagnosis of fire blight, especially in the Republic of Korea, where fire blight and black shoot blight coexist.
