Identification and Characterization of a New Alkaline Thermolysin-Like Protease, BtsTLP1, from Bacillus thuringiensis Serovar Sichuansis Strain MC28

  • Thermolysin and its homologs are a group of metalloproteases that have been widely used in both therapeutic and biotechnological applications. We here report the identification and characterization of a novel thermolysin-like protease, BtsTLP1, from insect pathogen Bacillus thuringiensis serovar Sichuansis strain MC28. BtsTLP1 is extracellularly produced in Bacillus subtilis, and the active protein was purified via successive chromatographic steps. The mature form of BtsTLP1 has a molecule mass of 35.6 kDa as determined by mass spectrometry analyses. The biochemical characterization indicates that BtsTLP1 has an apparent Km value of 1.57 mg/ml for azocasein and is active between 20℃ and 80℃. Unlike other reported neutral gram-positive thermolysin homologs with optimal pH around 7, BtsTLP1 exhibits an alkaline pH optimum around 10. The activity of BtsTLP1 is strongly inhibited by EDTA and a group of specific divalent ions, with Zn2+ and Cu2+ showing particular effects in promoting the enzyme autolysis. Furthermore, our data also indicate that BtsTLP1 has potential in cleaning applications.

  • 본 연구는 광합성 미생물 R. rubrum KS-301을 이용한 연속 반응기인 고정층 반응기와 연속 교반탱크 반응기에서 실험을 수행하여 도입 포도당 농도, 희석속도에 대한 수소 생산성과 고정화 담체에 의해서 형성되는 내부, 외부 및 총괄 물질전달 저항, 반응 속도식 변수 등을 평가한 후 비교하였다. 고정층 반응기에서의 겉보기 $K_m$은 희석속도의 증가에 따라 감소하며, 연속 교반탱크 반응기의 경우는 희석속도 $0.4h^{-1}$ 이상에서는 거의 일정한 값을 나타내었다. 고정층 반응기에서의 수소 생산성과 물질전달 실험결과는 연속 교반탱크 반응기에서와 비슷한 경향을 보이나 두 반응기를 비교하면 수소 생산성과 외부 효율인자는 교반탱크 반응기일 때가 크고, 내부 효율인자는 고정층 반응기일 때가 컸다. 그러나 총괄 효율인자는 두 반응기가 도입 포도당 농도에 따라서는 비슷한 수치를 나타내었다.

Estimation of the excavation damage zone in TBM tunnel using large deformation FE analysis

  • This paper aims to estimate the range of the excavation damaged zone (EDZ) formation caused by the tunnel boring machine (TBM) advancement through dynamic three-dimensional large deformation finite element analysis. Large deformation analysis based on Coupled Eulerian-Lagrangian (CEL) analysis is used to accurately simulate the behavior during TBM excavation. The analysis model is verified based on numerous test results reported in the literature. The range of the formed EDZ will be suggested as a boundary under various conditions - different tunnel diameter, tunnel depth, and rock type. Moreover, evaluation of the integrity of the tunnel structure during excavation has been carried out. Based on the numerical results, the apparent boundary of the EDZ is shown to within the range of 0.7D (D: tunnel diameter) around the excavation surface. Through series of numerical computation, it is clear that for the rock of with higher rock mass rating (RMR) grade (close to 1st grade), the EDZ around the tunnel tends to increase. The size of the EDZ is found to be direct proportional to the tunnel diameter, whereas the depth of the tunnel is inversely proportional to the magnitude of the EDZ. However, the relationship between the formation of the EDZ and the stability of the tunnel was not found to be consistent. In case where the TBM excavation is carried out in hard rock or rock under high confinement (excavation under greater depth), large range of the EDZ may be formed, but less strain occurs along the excavation surface during excavation and is found to be more stable.

KINEMATIC OSCILLATIONS OF POST-CME BLOBS DETECTED BY K-COR ON 2017 SEPTEMBER 10

  • We investigate 20 post-coronal mass ejection (CME) blobs formed in the post-CME current sheet (CS) that were observed by K-Cor on 2017 September 10. By visual inspection of the trajectories and projected speed variations of each blob, we find that all blobs except one show irregular "zigzag" trajectories resembling transverse oscillatory motions along the CS, and have at least one oscillatory pattern in their instantaneous radial speeds. Their oscillation periods are ranging from 30 to 91 s and their speed amplitudes from 128 to 902 km s-1. Among 19 blobs, 10 blobs have experienced at least two cycles of radial speed oscillations with different speed amplitudes and periods, while 9 blobs undergo one oscillation cycle. To examine whether or not the apparent speed oscillations can be explained by vortex shedding, we estimate the quantitative parameter of vortex shedding, the Strouhal number, by using the observed lateral widths, linear speeds, and oscillation periods of the blobs. We then compare our estimates with theoretical and experimental results from MHD simulations and fluid dynamic experiments. We find that the observed Strouhal numbers range from 0.2 to 2.1, consistent with those (0.15-3.0) from fluid dynamic experiments of bluff spheres, while they are higher than those (0.15-0.25) from MHD simulations of cylindrical shapes. We thus find that blobs formed in a post-CME CS undergo kinematic oscillations caused by fluid dynamic vortex shedding. The vortex shedding is driven by the interaction of the outward-moving blob having a bluff spherical shape with the background plasma in the post-CME CS.

An assessment of responses to egg production and liver health of Japanese quails subjected to different levels of metabolizable energy

  • Objective: Current quail production is configured as an economic activity in scale. Advancements in quail nutrition have been limited to areas such as breeding and, automation of facilities and ambience. The objective of this study was to evaluate the performance responses, liver and oviduct morphometry, and liver histology of Japanese laying quails subjected to different levels of nitrogen-corrected apparent metabolizable energy (MEn). Methods: A completely random design was used that consisted of nine levels of MEn, six replicates, and five hens per cage with a total of 270 quails. The experimental period lasted for 10 weeks. The variables of performance were subjected to analysis of variance and then regression analysis using the broken-line model. The morphometric and histological variables were subjected to multivariate exploratory techniques. Results: The MEn levels influenced the responses to zootechnical performance. The broken-line model estimated the maximum responses for feed intake, egg production, egg weight, and egg mass as 3,040, 2,820, 1,802, and 2,960 kcal of MEn per kg of diet, respectively. Multivariate analysis revealed that the occurrence of hepatic steatosis and increased levels of Kupffer cells were not related to MEn levels. Conclusion: The level of 2,960 kcal/kg of MEn meets performance variable requirements without compromising hepatic physiology.

Tethering Meat Goats Grazing Forage of High Nutritive Value and Low to Moderate Mass

  • Twenty-four yearling Boer$\times$Spanish goats were used in a crossover design experiment to determine effects of tethering on forage selection, intake and digestibility, grazing behavior and energy expenditure (EE) with forage high in nutritive value and low to moderate in mass. Objectives were to determine if tethered goats could be used as a model for study of unrestrained animals and to characterize tethering as a production practice. Four 0.72-ha pastures of wheat (Triticum aestivum) and berseem clover (Trifolium alexandrium) were grazed in December and January. Each pasture hosted six animals, three with free movement and three attached to a 4.11-m tether for access to a circular area of $53.1m^2$. Tethering areas were moved each day. One animal of each treatment and pasture was used to determine forage selection, fecal output or grazing behavior and EE; therefore, there were eight observations per treatment. Mass of forage DM before grazing in Tethered areas averaged 1,280 and 1,130 kg/ha in periods 1 and 2, respectively. The CP concentration in ingesta was greater ((p<0.05) 239 and 209 g/kg; SE = 8.0) and the NDF level was lower (p<0.05) for Free vs. Tethered animals (503 and 538 g/kg; SE = 12.0); in vitro true DM digestion was similar between treatments (0.808 and 0.807 for Free and Tethered, respectively; SE = 0.0096). Intakes of DM (1,013 and 968 g/d; SE = 78.6), NDF (511 and 521 g/d; SE = 39.9) and ME (10.9 and 10.7 MJ/d; SE = 0.90) were similar between treatments, but CP intake was greater (p<0.05) for Free vs. Tethered animals (241 and 203 g/d; SE = 17.2). There were small treatment differences in in vivo apparent digestibility of OM ((p<0.05) 0.780 and 0.814; SE = 0.0049), CP ((p<0.05) 0.800 and 0.817; SE = 0.0067) and NDF ((p<0.09) 0.777 and 0.760 for Free and Tethered, respectively; SE = 0.0078). There were no treatment effects on time spent ruminating or grazing (346 and 347 min/d for Free and Tethered, respectively; SE = 42.5), but EE was considerably greater (p<0.05) for Free vs. Tethered animals (571 and 489 kJ/kg $BW^{0.75}$; SE = 8.9). In conclusion, with forage of high nutritive value and low to moderate in mass, tethering can offer a production advantage over free grazing of less energy used for activity despite similar grazing time. With forage removal considerably less than that available for grazing, effects of tethering on chemical composition of selected forage were small and less than needed to markedly affect digestion. Tethering may offer a means of studying some aspects of grazing by ruminants, but would not seem suitable for energy metabolism.

하악에 발생된 Ewing 육종의 1증례 (A CASE REPORT OF EWING′S SARCOMA OF THE MANDIBLE)

  • The authors observed a 27-year-old male patient who came to the Infirmary of Kyungpook National University Hospital who had complained of dull pain in right mandibular angle area 1 month ago. As a result of careful analysis of clinical, radiological, and histopathological findings, the authors diagnosed it as Ewing's sarcoma and obtained the results as follows: 1. In clinical examination, main clinical symptoms were continuous dull pain and gingival swelling on the 2nd and 3rd molar area of the right mandible. 2. In radiographic examination, ill-defined radiolucent area was seen on mandibular right angle area, and apparent periosteal reaction of sun-ray spicule was emanated from the lingual cortex of mandibular angle area. And computed tomograph also revealed moderate-defined 3. soft tissue mass of the same area. In histopathological examination, small round-shaped, and closely packed cells with scanty cytoplasm were observed in this specimen, and positive reaction in PAS staining was also observed.

Effects of Magnetic Fields on the Gaseous Structures in Spiral Galaxies

  • Stellar spiral arms and magnetic fields in disk galaxies are important in the formation of gaseous structures such as spurs/feathers and wiggles as well as in angular momentum transport between stars and gas. We present our recent results of global magnetohydrodynamic simulations to study nonlinear responses of self-gravitating and magnetized gas to an imposed stellar spiral potential. We vary the arm strength, the arm pattern speed, and magnetic field strength to explore various galactic situations. Magnetic fields not only reduce the peak density of galactic spiral shocks but also make angular momentum transport more efficient via magnetic pressure and tension forces. The extent and shapes of gaseous arms as well as the radial mass drift rate depend rather sensitively on the magnetic field strength. The wiggle instability apparent in unmagnetized models is suppressed with increasing magnetic field strength, while magnetic fields promote the development of magneto-Jeans instability of the arms and magnetic islands in between arms. We quantify the angular momentum transport by spiral shocks, focusing on the effects of magnetic fields. We also present physical interpretations of our numerical results and discuss astronomical implications of our findings.

Chloride penetration in anchorage concrete of suspension bridge during construction stage

  • Steel corrosion in embedded steel causes a significant durability problems and this usually propagates to structural degradation. Large-scaled concrete structures, PSC (Pre-stressed Concrete) or RC (Reinforced Concrete) structures, are usually constructed with mass concrete and require quite a long construction period. When they are located near to sea shore, chloride ion penetrates into concrete through direct or indirect exposure to marine environment, and this leads durability problems. Even if the structures are sheltered from chloride ingress outside after construction, the chloride contents which have been penetrated into concrete during the long construction period are differently evaluated from the initially mixed chloride content. In the study, chloride profiles in cores extracted from anchorage concrete block in two large-scaled suspension bridge (K and P structure) are evaluated considering the exposure periods and conditions. Total 21 cores in tendon room and chamber room were obtained, and the acid-soluble chlorides and compressive strength were evaluated for the structures containing construction period around 3 years. The test results like diffusion coefficient and surface chloride content from the construction joint and cracked area were also discussed with the considerations for maintenance.

Vibration and mode shape analysis of sandwich panel with MWCNTs FG-reinforcement core

  • The goal of this study is to fill this apparent gap in the area about vibration analysis of multiwalled carbon nanotubes (MWCNTs) curved panels by providing 3-D vibration analysis results for functionally graded multiwalled carbon nanotubes (FG-MWCNTs) sandwich structure with power-law distribution of nanotube. The effective material properties of the FG-MWCNT structures are estimated using a modified Halpin-Tsai equation. Modified Halpin-Tsai equation was used to evaluate the Young's modulus of MWCNT/epoxy composite samples by the incorporation of an orientation as well as an exponential shape factor in the equation. The exponential shape factor modifies the Halpin-Tsai equation from expressing a straight line to a nonlinear one in the MWCNTs wt% range considered. Also, the mass density and Poisson's ratio of the MWCNT/phenolic composite are considered based on the rule of mixtures. Parametric studies are carried out to highlight the influence of MWCNT volume fraction in the thickness, different types of CNT distribution, boundary conditions and geometrical parameters on vibrational behavior of FG-MWCNT thick curved panels. Because of using two-dimensional generalized differential quadrature method, the present approach makes possible vibration analysis of cylindrical panels with two opposite axial edges simply supported and arbitrary boundary conditions including Free, Simply supported and Clamped at the curved edges. For an overall comprehension on 3-D vibration analysis of sandwich panel, some mode shape contour plots are reported in this research work.
