Streptavidin이 융합된 DR4 항원에 특이적인 single-chain Fv 항체의 개발 (The development of anti-DR4 single-chain Fv (ScFv) antibody fused to Streptavidin)

  • Streptavidin (STR)과 Biotin system은 Biotin의 Streptavidin에 대한 높은 비공유 친화력(non-covalent affinity; $K_D=10^{-14}M$)과 4 Biotin 결합부위를 갖는 Streptavidin의 tetramer 구조로 인해 복수의 항원결합부위 및 복수의 항원특이성을 갖는 항체를 제조할 수 있기 때문에 가장 활발하게 연구되고 있다. 이 system을 활용하기 위해 우리는 Streptomyces avidinii 염색체 DNA로부터 PCR을 통해 Streptavidin (STR) 유전자를 증폭하고 이를 TRAIL (tumor necrosis factor ${\alpha}$ related apoptosis induced ligand) receptor인 death receptor 4 (DR4)에 특이적으로 결합하는 hAY4 single-chain Fv 항체유전자에 융합시켰다. 대장균에서 발현시킨 STR에 융합된 hAY4 ScFv (hAY4-STR) 항체는 가열시킨 SDS-PAGE에서 43 kDa monomer를 나타내었다. 그러나 가열하지 않은 SDS-PAGE와 Size-exclusion chromatography에서는 tetramer인 172 kDa을 나타내었는데 이는 hAY4 ScFv-STR 항체가 STR의 자연적인 비공유결합에 의해 유도된 tetramer를 형성하고 있음을 나타내고 있다. 본 융합 단백질은 Ouchterlony assay와 ELISA에서 보여주는 것처럼 자연 Streptavidin과 유사한 Biotin 결합력을 유지하고 있었다. ELISA와 Westernblot을 이용하여 정제된 hAY4-STR 융합항체의 DR4 항원결합력 또한 확인하였다. 게다가 표면 플라즈몬 공명(surface plasmon resonance) 분석에서 hAY4 ScFv-STR tetramer는 tetramerization에 의해 hAY4 ScFv monomer보다 60배 더 높은 항원결합력을 나타내었다. 요약하면 hAY4 ScFv-STR 융합단백질은 E. coli에서 soluble tetramer로 성공적으로 발현 및 정제되었으며 Biotin과 DR4 항원에 동시에 결합함을 보여 주었다. 이는 bifunctional and tetrameric ScFv 항체를 제조 할 수 있음을 제시해 주고 있다.

배양신경세포의 저산소증모델에서 대황 물추출물에 의한 유전자 표현 변화의 microarray 분석 (Microarray Analysis of Gene Expression by Rhei Rhizoma Water Extracts in a Hypoxia Model of Cultured Neurons)

  • 대황(Rhei Rhizoma; RR, 대황(大黃))은 Rheum officinale Baill.와 Rheum palmatum L.(polygonaceae)의 땅속부분으로 남아시아의 민속의학에서 간 및 신장의 손상을 치료하는데 널리 이용되고 있다. 본 연구에서는 배양한 흰쥐 해마신경세포의 저산소증모델을 이용하여 대황의 물추출물이 유전자 표현에 미치는 영향을 microarray 방법을 이용하여 조사하였다. 배양 후 10일 (DIV10)에 추출물을 배지에 $2.5{\mu}g/ml$ 농도로 첨가하고, DIV13에 저산소증(2% $O_2$/5% $CO_2$, $37^{\circ}C$, 3 h)을 유발한 후 24 시간 후에 total RNA를 분리하여 microarray에 사용하였다. MA-plot에 의하면 표현이 연화된 대부분의 유전자는 ${\pm}2$배 이내로 증감되었다. 이 가운데 Global M 값이 0.2(즉, 15%)보다 더 증가한 유전자는 472종, Global M 값이 -0.2(즉, -15%)보다 더 감소한 유전자는 725종이였다. 세포의 생존과 관련된 유전자 가운데 세포자연사 억제유전자인 Tegt(2.4배), Nfkb1 (2.4배), Veg (1.8배), Ngfr (1.6배) 등이 크게 증가하였으며, 반면에 자연사 촉진유전자인 Bad (-64%), Cstb (-66%)는 감소하였다. 스트레스를 극복하는데 필요한 유전자인 Defb3 (2.7배), Cygb (2.2배), Ahsg (2.18배), Alox5 (2배) 등도 크게 증가하였다. 그리고 세포 성장을 촉진하는 유전자인 Erbb2 (1.84배), Mapk12 (1.8배)도 크게 증가하였다. 따라서 대황의 물추출물은 세포생존에 필요한 유전자를 증가시키고, 세포사를 유도하는 유전자는 감소시킴으로서 저산소층 스트레스에서 신경세포의 사망을 억제하는 것으로 해석된다.

Dysregulation of MicroRNA-196b-5p and MicroRNA-375 in Gastric Cancer

  • Purpose: Dysregulated microRNAs (miRNAs) can contribute to cancer development by leading to abnormal proliferation of cells, apoptosis, and differentiation. Although several miRNAs that are related to gastric cancer have been identified, the reported results have been inconsistent. The aim of this study was to determine miRNA expression profiles and validate miRNAs up- and down-regulated in gastric cancer. Materials and Methods: We evaluated 34 primary gastric cancer tissues and paired adjacent nontumorous gastric tissues. Total RNA was extracted, and low-molecular-weight RNAs (<200 nucleotides) were isolated for further analysis. Two pairs of tissues were processed for GeneChip microarray analysis, and the identified up- and down-regulated miRNAs were validated by real-time quantitative polymerase chain reaction (qPCR). Results: In the set of differentially expressed miRNAs, 5 were overexpressed by more than 2 fold, and 5 were reduced by 2 fold or less in gastric cancer tissues compared with normal gastric tissues. Four of these miRNAs (miR-196b-5p, miR-375, miR-483-5p, and miR-486-5p) were then validated by qPCR, and the relative expression levels of 2 miRNAs (miR-196b-5p and miR-375) were significantly different between cancer and normal tissues. Conclusions: Our results revealed that the expression of miR-196b-5p and miR-375 significantly correlates with gastric cancer. These miRNAs could therefore serve as diagnostic biomarkers of gastric cancer.

Expression profiles of circular RNAs in sheep skeletal muscle

  • Objective: Circular RNAs (circRNAs) are a newfound class of non-coding RNA in animals and plants. Recent studies have revealed that circRNAs play important roles in cell proliferation, differentiation, autophagy and apoptosis during development. However, there are few reports about muscle development-related circRNAs in livestock. Methods: RNA sequencing analysis was employed to identify and annotate circRNAs from longissimus dorsi of sheep. Reverse transcription followed by real-time quantitative (q) polymerase chain reaction (PCR) analysis verified the presence of these circRNAs. Targetscan7.0 and miRanda were used to analyse the interaction of circRNA-microRNA (miRNA). To investigate the function of circRNAs, an experiment was conducted to perform enrichment analysis hosting genes of circRNAs using gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) pathways. Results: About 75.5 million sequences were obtained from RNA libraries of sheep skeletal muscle. These sequences were mapped to 729 genes in the sheep reference genome. We identified 886 circRNAs, including numerous circular intronic RNAs and exonic circRNAs. Reverse transcription PCR (RT-PCR) and DNA sequencing analysis confirmed the presence of several circRNAs. Real-Time RT-PCR analysis exhibited resistance of sheep circRNAs to RNase R digestion. We found that many circRNAs interacted with muscle-specific miRNAs involved in growth and development of muscle, especially circ776. The GO and KEGG enrichment analysis showed that hosting genes of circRNAs was involved in muscle cell development and signaling pathway. Conclusion: The study provides comprehensive expression profiles of circRNAs in sheep skeletal muscle. Our study offers a large number of circRNAs to facilitate a better understanding of their roles in muscle growth. Meanwhile, we suggested that circ776 could be analyzed in future study.

MPTP-induced vulnerability of dopamine neurons in A53T α-synuclein overexpressed mice with the potential involvement of DJ-1 downregulation

  • Familial Parkinson's disease (PD) has been linked to point mutations and duplication of the ${\alpha}$-synuclein (${\alpha}$-syn) gene. Mutant ${\alpha}$-syn expression increases the vulnerability of neurons to exogenous insults. In this study, we developed a new PD model in the transgenic mice expressing mutant hemizygous (hemi) or homozygous (homo) A53T ${\alpha}$-synuclein (${\alpha}$-syn Tg) and their wildtype (WT) littermates by treatment with sub-toxic (10 mg/kg, i.p., daily for 5 days) or toxic (30 mg/kg, i.p., daily for 5 days) dose of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). Tyrosine hydroxylase and Bcl-2 levels were reduced in the ${\alpha}$-syn Tg but not WT mice by sub-toxic MPTP injection. In the adhesive removal test, time to remove paper was significantly increased only in the homo ${\alpha}$-syn Tg mice. In the challenging beam test, the hemi and homo ${\alpha}$-syn Tg mice spent significantly longer time to traverse as compared to that of WT group. In order to find out responsible proteins related with vulnerability of mutant ${\alpha}$-syn expressed neurons, DJ-1 and ubiquitin enzyme expressions were examined. In the SN, DJ-1 and ubiquitin conjugating enzyme, UBE2N, levels were significantly decreased in the ${\alpha}$-syn Tg mice. Moreover, A53T ${\alpha}$-syn overexpression decreased DJ-1 expression in SH-SY5Y cells. These findings suggest that the vulnerability to oxidative injury such as MPTP of A53T ${\alpha}$-syn mice can be explained by downregulation of DJ-1.

인체폐암세포 NCI-H460 및 A549의 증식에 미치는 삼기보폐탕의 영향 비교 (Induction of Apoptosis by Samgibopae-tang in Human Non-small-cell Lung Cancer Cells)

  • In the present study, we investigated the antiproliferative activity of the water extract of Samgibopae-tang (SGBPT) in NCI-H460 and A549 non-small-cell lung cancer cell lines. We found that exposure of A549 cells to SGBPT resulted in the growth inhibition in a dose-dependent manner as measured by MTT assay, however SGBPT did not affect the growth of NCI-H460 cells. The antiproliferative effect by SGBPT treatment in A549 cells was associated with morphological changes such as membrane shrinking and cell rounding up. SGBPT treatment did not induce the cell cycle arrest in both cell lines, however the frequency of sub-G1 population was concentration-dependently increased by SGBPT treatment in A549 cells. SGBPT treatment partially induced the expression of tumor suppressor p53 in A549 cells and the expression of cyclin-dependent kinase inhibitor p21(WAF1/CIP1) was markedly increased in both transcriptional and translational levels in A549 cells. The up-regulation of p21 by SGBPT occurred in a similar a concentration dependent manner to that observed with the inhibition of cell viability and induction of sub-G1 population of the cell cycle. However SGBPT treatment did not affect other growth regulation-related genes such as early growth response-1 (Egr-1), nonsteroidal anti-inflammatory drug-activated gene-1 (NAG-1), inducible nitric oxide synthease (iNOS), cyclooxygenases (COXs), telomere-regulatory factors in A549 as well as NCI-H460 cells. Taken together, these findings suggested that SGBPT-induced inhibition of human lung carcinoma A549 cell growth was aoosciated with the induction of p21 and the results provided important new insights into the possible molecular mechanisms of the anti-cancer activity of SGBPT.

암종양유전자 SETDB1과 FosB 발현에 대한 p53의 음성 조절기작 (Negative Regulation of Tumor Suppressor p53 at the Promoter Regions of Oncogenic SETDB1 and FosB Genes)

  • 암세포에 항암제를 처리하게 되면, 세포증식, 이동성 또는 약물 내성과 관련된 많은 유전자들의 발현 변화가 발생하며, 유전자 발현 변화는 상호간의 조절 네트워크에 의해 밀접하게 연결될 수도 있다고 추측된다. 본 연구에서 p53 유전자 유무가 다른 A549와 H1299 인간 폐암세포에 독소루비신을 처리하면, 원종양유전자인 FosB의 발현은 증가하지만, 원종양유전자인 SETDB1의 발현은 감소하지만, 단백질 발현의 양적인 차이가 발생한다는 사실을 알 수 있었다. TF motif binding 분석 프로그램을 이용하여 SETDB1과 FosB 프로모터지역에서의 p53단백질의 결합가능성을 분석한 결과, SETDB1의 경우 18부위, FosB의 경우 21 부위의 p53 결합부위를 예측할 수 있었다. SETDB1과 FosB 프로모터의 subcloning하여 luciferase 분석을 수행한 결과, p53은 SETDB1과 FosB을 음성적으로 조절한다는 사실을 알 수 있었다. 또한, H1299 세포에 p53의 과발현은 SETDB1 과 FosB의 발현을 감소시킬 수 있음을 RT-PCR, western blot, qPCR, 면역염색 실험을 통해 확인하였다. 이러한 결과를 종합하여 본다면, p53에 의한 SETDB1과 FosB 유전자 발현 조절은 항암제 처리과정에서 나타나는 암세포의 사멸과 생존에 대한 기능적 조절 네트워크로 사료된다.

Cigarette Smoke Extract-Treated Mouse Airway Epithelial Cells-Derived Exosomal LncRNA MEG3 Promotes M1 Macrophage Polarization and Pyroptosis in Chronic Obstructive Pulmonary Disease by Upregulating TREM-1 via m6A Methylation

  • Cigarette smoke extract (CSE)-treated mouse airway epithelial cells (MAECs)-derived exosomes accelerate the progression of chronic obstructive pulmonary disease (COPD) by upregulating triggering receptor expressed on myeloid cells 1 (TREM-1); however, the specific mechanism remains unclear. We aimed to explore the potential mechanisms of CSE-treated MAECs-derived exosomes on M1 macrophage polarization and pyroptosis in COPD. In vitro, exosomes were extracted from CSE-treated MAECs, followed by co-culture with macrophages. In vivo, mice exposed to cigarette smoke (CS) to induce COPD, followed by injection or/and intranasal instillation with oe-TREM-1 lentivirus. Lung function and pathological changes were evaluated. CD68+ cell number and the levels of iNOS, TNF-α, IL-1β (M1 macrophage marker), and pyroptosis-related proteins (NOD-like receptor family pyrin domain containing 3, apoptosis-associated speck-like protein containing a caspase-1 recruitment domain, caspase-1, cleaved-caspase-1, gasdermin D [GSDMD], and GSDMD-N) were examined. The expression of maternally expressed gene 3 (MEG3), spleen focus forming virus proviral integration oncogene (SPI1), methyltransferase 3 (METTL3), and TREM-1 was detected and the binding relationships among them were verified. MEG3 increased N6-methyladenosine methylation of TREM-1 by recruiting SPI1 to activate METTL3. Overexpression of TREM-1 or METTL3 negated the alleviative effects of MEG3 inhibition on M1 polarization and pyroptosis. In mice exposed to CS, EXO-CSE further aggravated lung injury, M1 polarization, and pyroptosis, which were reversed by MEG3 inhibition. TREM-1 overexpression negated the palliative effects of MEG3 inhibition on COPD mouse lung injury. Collectively, CSE-treated MAECs-derived exosomal long non-coding RNA MEG3 may expedite M1 macrophage polarization and pyroptosis in COPD via the SPI1/METTL3/TREM-1 axis.

Rosmarinic Acid Inhibits Ultraviolet B-Mediated Oxidative Damage via the AKT/ERK-NRF2-GSH Pathway In Vitro and In Vivo

  • Rosmarinic acid (RA) is a phenolic ester that protects human keratinocytes against oxidative damage induced by ultraviolet B (UVB) exposure, however, the mechanisms underlying its effects remain unclear. This study aimed to elucidate the cell signaling mechanisms that regulate the antioxidant activity of RA and confirm its cyto-protective role. To explore the signaling mechanisms, we used the human keratinocyte cell line HaCaT and SKH1 hairless mouse skin. RA enhanced glutamate-cysteine ligase catalytic subunit (GCLC) and glutathione synthetase (GSS) expression in HaCaT cells in a dose- and time-dependent manner. Moreover, RA induced nuclear factor erythroid-2-related factor 2 (NRF2) nuclear translocation and activated the signaling kinases protein kinase B (AKT) and extracellular signal-regulated kinase (ERK). Treatment with the phosphatidylinositol 3-kinase (PI3K) inhibitor LY294002, the ERK inhibitor U0126, and small interfering RNA (siRNA) gene silencing suppressed RA-enhanced GCLC, GSS, and NRF2 expression, respectively. Cell viability tests showed that RA significantly prevented UVB-induced cell viability decrease, whereas the glutathione (GSH) inhibitors buthionine sulfoximine, LY294002, and U0126 significantly reduced this effect. Moreover, RA protected against DNA damage and protein carbonylation, lipid peroxidation, and apoptosis caused by UVB-induced oxidative stress in a concentration-dependent manner in SKH1 hairless mouse skin tissues. These results suggest that RA protects against UVB-induced oxidative damage by activating AKT and ERK signaling to regulate NRF2 signaling and enhance GSH biosynthesis. Thus, RA treatment may be a promising approach to protect the skin from UVB-induced oxidative damage.
