Ursolic Acid Promotes Apoptosis of SGC-7901 Gastric Cancer Cells through ROCK/PTEN Mediated Mitochondrial Translocation of Cofilin-1

  • Ursolic acid, extracted from the traditional Chinese medicine bearberry, can induce apoptosis of gastric cancer cells. However, its pro-apoptotic mechanism still needs further investigation. More and more evidence demonstrates that mitochondrial translocation of cofilin-1 appears necessary for the regulation of apoptosis. Here, we report that ursolic acid (UA) potently induces the apoptosis of gastric cancer SGC-7901 cells. Further mechanistic studies revealed that the ROCK1/PTEN signaling pathway plays a critical role in UA-mediated mitochondrial translocation of cofilin-1 and apoptosis. These findings imply that induction of apoptosis by ursolic acid stems primarily from the activation of ROCK1 and PTEN, resulting in the translocation of cofilin-1 from cytoplasm to mitochondria, release of cytochrome c, activation of caspase-3 and caspase-9, and finally inducing apoptosis of gastric cancer SGC-7901 cells.

Induction of Apoptosis in the Testes of SD Rats After Exposure to 2-Bromopropane

  • Exposure to 2-Bromopropane has been known to cause degeneration of male germ cells. However the mechanism underlying this process is poorly understood. The objective of this study was to determine whether or not the exposure of male Sprague-Dawley rats to 2-BP induces apoptosis in male germ cells. Male rats(N=3 or 4 in each group) were orally administered either with the corn oil vehicle (10 ml/kg body weight) or with 2-BP (3,500 mg/kg) once a day for 3 days. The presence of apoptosis was determined by TUNEL detection in situ and by an increase in DNA fragmentation. A low spontaneous incidence of apoptosis was observed in vehicle control animals, especially in pre-meiotic germ cells of stages I-VI and stages XII-XIV the seminiferous tubules. In 2-BP exposure rats, the incidence of apoptosis markedly increased at 4 h, reached a peak at 8 h (about 7-fold over control), and then decreased rapidly to control level by 48 h after the last administration. Although apoptosis induced by 2-BP occurred in all stages of germ cells, it was most pronounced in spermatogonia and early spermatocytes in stages I-VI and stages XII-XIV. Taken together, our results suggest that apoptosis is involved in the toxicity of testicular germ cells resulting in oligospermia or azoospermia after exposure to 2-BP.

  • Induction of apoptosis is considered to be the underlying mechanism that accounts for the efficiency of chemotherapeutic drugs. It has recently been proposed that doxorubicin (DOX) can induce apoptosis in human leukemic cells via the Fas/Fas Ligand (FasL) system. Comparison of Fas and FasL mRNA expression between drug- and anti-Fas antibody(Fas-Ab)- induced apoptosis was analyzed for examining the role of Fas/FasL system in the mediation of drug-induced apoptosis. After HL-60 cells were routinely cultured, MTT assay was performed for cytotoxicity test. Giemsa staining was carried out to monitor the apoptosis morphologically. By semiquantitative RT-PCR analysis, the expression of Fas and FasL at 4, 10, 24 hours was determined after DOX and Fas-Ab treatment. Dose-dependent cytotoxicity was induced by DOX-treatment, while Fas-Ab treatment showed the similar dose-dependent pattern but the cytotoxicity is not reached at LD$_{50}$ at 100 ng/ml concentration of Fas-Ab. In the 10ng/m1 DOX and 10ng/m1 Fas-Ab treated group, typical apoptotic cell morphology was shown such as fragmented nuclei and cell membrane budding in the Giemsa-stained slide. Fas mRNA expression was not changed significantly in the both groups. But, FasL mRNA expression was induced significantly at initial period of apoptosis. In this study, Fas/FasL interaction assumed to be involved in drug-induced apoptosis.s.

Sasa quelpaertensis Nakai extract induces p53-independent apoptosis via the elevation of nitric oxide production in human HCT116 colon cancer cells

  • Induction of apoptosis in human cancer cells by Sasa quelpaertensis Nakai has been considered to be a potential therapeutic target for cancer treatment; however, the underlying mechanisms of action are not well understood. The present study investigated the role of nitric oxide (NO•) and inhibitors of apoptosis (IAPs) during apoptosis induced by Sasa quelpaertensis Nakai extracts (SQE) in p53-wild type (WT) and p53-null HCT116 colon carcinoma cells. Trypan blue exclusion and Annexin V/propidium iodide assays were used to test for antiproliferation, and apoptosis and cell cycle. Griess and reverse transcription-polymerase chain reaction and western blotting assays were carried out to assay NO• production, and to detect the mRNA and protein levels of Bcl-2, PARP and IAPs. A colorimetric assay was utilized to measure the time-dependent increase in caspase-3 activity. SQE inhibited cell growth and promoted apoptosis by the elevation of NO• in a dose- and time-dependent manner. In addition, both cell types underwent a reduction in mRNA and protein levels of IAPs (survivin, CIAP-1 and -2, and X-linked inhibitor of apoptosis) as well as anti-apoptotic Bcl-2, whereas an increase in protein expression of poly (ADP-ribose) polymerase 1 and caspase 3 activity was observed; however, an equivalent cytotoxic and apoptotic effect by SQE was observed in p53-WT and p53-null cells. Collectively, the results indicated that SQE-induced apoptosis was independent of p53 status and associated with modulation of endogenous NO• and IAP family gene expression.

Anti-tumor Activity and Apoptosis-regulation Mechanisms of Bufalin in Various Cancers: New Hope for Cancer Patients

  • The induction of apoptosis in target cells is a key mechanism for most anti-tumor therapies. Bufalin is a cardiotonic steroid that has the potential to induce differentiation and apoptosis of tumor cells. Research on bufalin has so far mainly involved leukemia, prostate cancer, gastric cancer and liver cancer, and has been confined to in vitro studies. The bufadienolides bufalin and cinobufagin have been shown to induce apoptosis in a wide spectrum of cancer cell. The present article reviews the anticancer effects of bufalin. It induces apoptosis of lung cancer cells via the PI3K/Akt pathway and also suppressed the proliferation of human non-small cell lung cancer A549 cell line in a time and dose dependent manner. Bufalin, bufotalin and gamabufotalin, key bufadienolides, significantly sensitize human breast cancer cells with differing ER-alpha status to apoptosis induction by the TNF-related apoptosis-inducing ligand (TRAIL). In addition, bufadienolides induce prostate cancer cell apoptosis more significantly than that in breast epithelial cell lines. Similar effects have been observed with hepatocellular carcinoma (HCC) but the detailed molecular mechanisms of inducing apoptosis in this case are still unclear. Bufalin exerts profound effects on leukemia therapy in vitro. Results of multiple studies indicate that bufalin has marked anti-tumor activities through its ability to induce apoptosis. Large-scale randomized, double-blind, placebo or positive drug parallel controlled studies are now required to confirm the efficacy and apoptosis-inducing potential of bufalin in various cancers in the cliniucal setting.

  • 본 연구에서는 인체전립선 상피세포인 267B1 세포에서 HDAC 저해제인 TSA에 의한 증식억제가 apoptosis 유도에 의한 것임을 제시하였다. 이러한 TSA에 의한 267B1 세포의 apoptosis에는 c-IAP-1 및 c-IAP-2와 같은 IAP family의 발현감소가 동반되었으나 Bax 및 Bcl-2와 같은 Bcl-2 family의 발현에는 큰 변화가 없었다. 그리고 TSA에 의한 267Bl 세포의 apoptosis는 caspase의 활성에 의한 표적 단백질들의 분해와 연관성이 있었다. 또한 TSA에 의한 apoptosis 유도에서 $NF-{\kappa}B$의 활성이 증가된다는 것을 세포질에서 $NF-{\kappa}B$의 핵 내로의 이동에 따른 전사활성의 증가 현상에 의한 것임을 다양한 방법으로 제시하였다. 본 연구의 결과는 TSA와 같은 HDAC 저해제에 의한 apoptosis 유도에는 $NF-{\kappa}B$의 활성 증가가 동반될 수 있음을 보여주는 결과로서 HDAC 저해제의 항암활성에 대한 $NF-{\kappa}B$의 새로운 역할 가능성을 제시하여 주는 것으로서 이에 관한 추가적인 연구의 필요성을 제시하였다.

Trichoplusia ni 세포의 apoptosis 메커니즘 규명을 위한 기초연구 (Basic Studies on the Apoptosis Mechanism of Trichoplusia ni Cell Line)

  • 본 연구에서는 Ttichoplusia ni 세포의 apoptosis 유도 및 억제 현상의 기초연구를 수행하였다. Apoptosis 유도제로 알려진 hygromycin B에 의한 세포 성장 저해는 $200\;{\mu}/ml$의 수준에서부터 나타났고, $400\;{\mu}/ml$ hygromycin B를 처리한 세포에서는 배양 후 2일부터 DNA가 분절되어지는 것을 확인할 수 있었다. 그러나 dexamethasone과 sodium butyrate를 첨가시 세포성장은 저해되었지만 DNA 분절현상이 보이지 않아 apoptosi의 유발여부를 확인할 수 없었다. 그리고 caspase 기능억제제의 apoptosis 지연효과를 보기 위해 $200\;{\mu}/ml$ hygromycin B로 apoptosis를 유발한 상태에서 Ac-DEVD-CHO를 첨가하여 세포성장을 비교해 본 결과 이 저해제에 의해 약 36%정도 apoptosis가 억제되었음을 확인하였다. N-acetylcysteine의 경우도 apoptosis지연 효과가 있었다. Bcl_계에 속하는 anti-apoptotic 유전자의 발현연구로서 apoptosis 저해 단백질인 bcl-2 유전자를 곤충세포에 형질전환시킨 후 이 단백질이 한시적으로 발현되는 것을 western blot분석법으로 확인하였으며 apoptosis가 지연된 곤충세포주의 개발이 가능하다는 결론을 보였다.

K562 백혈병 세포주에서 방사선에 의해 유도되는 Apoptosis에 미치는 PTK Inhibitors의 영향 (Radiation-induced Apoptosis is Differentially Modulated by PTK Inhibitors in K562 Cells)

  • 목적 : 방사선에 의해 유도되는 apoptosis에 내성을 가진 세포로 알려진 KS62 세포를 대상으로, PTK inhibitors인 herbimycin A와 genistein을 이용한 방사선에 의한 apoptosls의 내성 기전을 연구하고자 하였다. 대상 및 방법 : 6 MV 체외 X-선 방사선 치료기를 이용하여 200$\~$300 cGy/min의 선량률로 10 Gy의 X-선을 세포에 균일하게 조사하였다. Apoptosis의 관찰은 래arose gel electrophoresis를 이용하여 DNA frgmentation의 지표인 ladder를 관찰하였고, TUNEL 염색을 이용하여 정량 분석을 시행하였다. Western blot 방법으로 apoptosls 관련 유전 단백인 bel-2, bel-X$_L$ 및 bax들의 발현을 관찰하였다. 방사선 조사 및 약물 처치 후의 세포 주기 분석은 flow cytometry로 분석하였다. 결과 : Agarose gel electrophoresis 실험에서 방사선을 조사하지 않은 K562 세포와 방사선을 10 Gy 조사한 세포를 48시간에 걸쳐 12시간 간격으로 관찰하였을 때 DNA fragmentation를 관찰할 수 있었다. 이러한 현상은 genistein을 투여한 세포들에서도 동일한 현상을 관찰할 수 있었지만, 방사선 조사 후 herbimycin A를 투여한 세포들에서는 48 시간째 확연한 DNA fragmentation을 관찰할 수 있었다. 이를 TUNEL assay에서 정량적으로 확인하였다. 방사선만조사한 세포들과 방사선과 genistein 투여 후 48시간째 관찰한 세포들에서는 10$\%$, 미만의 apoptosis 양성 세포의 빈도를 관찰할 수 있었지만, 방사선 조사 후 herbimycin A를 투여한 세포들에서는 30$\~$35$\%$ 빈도로 apoptosis 양성 세포들이 관찰되었다. Western blot analysis에서 bcl-2의 경우 방사선을 조사하지 않았던 대조군에 비하여 전체적인 발현은 증가되었지만 방사선 및 약제간의 발현의 차이는 없었다. 그 외 bcl-X$_{L}$과 bax는 대조군에 비해 방사선 및 약제간의 발현의 차이를 관찰할 수 없었다. KS62 세포에 방사선을 10 Gy 조사하였을 때 나타나는 세포 주기의 변화는 시간이 경과함에 따라 전형적인 G2/M block의 소견을 보였다. 이러한 소견은 genistein을 투여했을 경우에는 특별한 변화를 보이지 않지만, herbimycin A를 투여했을 경우에는 12시간째부터 G2/M block이 소실되면서 세포가 세포 주기를 재 순환하는 양상을 보였고, 48시간째 관찰한 소견에서는 G2/M block이 거의 소실된 양상을 띠었다. 이러한 소견을 토대로 apoptosis 유도와의 상호 연관성을 유추할 수 있었다. 결론 : herbimycin A는 방사선에 의해 유도되는 apoptosis가 억제된 K562 세포에서 apoptosis를 유도할 수 있었다. 이러한 유도 기전에 apoptosis 관련 유전 단백들인 bcl-2, bcl-X$_{L}$ 및 bax와 관련된 영향은 관찰되지 않았다. 세포 주기의 분석에서 G2/M block의 해소와 apoptosis 유도와의 연관성을 유추할 때, 세포 주기 관련 인자들에 대한 연구가 방사선에 의한 apoptosis의 내성의 극복 및 방사선에 의한 세포의 감수성 조절 약제로서의 역할에 이바지할 것으로 생각한다.

NF-${\kappa}B$ Inhibitor Suppresses Hypoxia-induced Apoptosis of Mouse Pancreatic ${\beta}$-cell Line MIN6

  • Hypoxia is one of the main reasons for islet apoptosis after transplantation as well as during isolation. In this study, we attempted to determine the potential usefulness of NF-${\kappa}B$ inhibitor for suppression of hypoxia-induced ${\beta}$-cell apoptosis as well as the relationship between IP-10 induction and ${\beta}$-cell apoptosis in hypoxia. To accomplish this, we cultured the mouse pancreatic ${\beta}$-cell line MIN6 in hypoxia (1% $O_2$). Among several examined chemokines, only IP-10 mRNA expression was induced under hypoxia, and this induced IP-10 expression was due to NF-${\kappa}B$ activity. Since a previous study suggested that IP-10 mediates ${\beta}$-cell apoptosis, we measured hypoxia-induced IP-10 protein and examined the effect of anti-IP-10 neutralizing Ab on hypoxia-induced ${\beta}$-cell apoptosis. However, IP-10 protein was not detected, and anti-IP-10 neutralizing Ab did not rescue hypoxia-induced MIN6 apoptosis, indicating that there is no relationship between hypoxia-induced IP-10 mRNA expression and hypoxia-induced ${\beta}$-cell apoptosis. Since it was still not clear if NF-${\kappa}B$ functions as an apoptotic or anti-apoptotic mediator in hypoxia-induced ${\beta}$-cell apoptosis, we examined possible involvement of NF-${\kappa}B$ in hypoxia-induced ${\beta}$-cell apoptosis. Treatment with 1 ${\mu}M$ NF-${\kappa}B$ inhibitor suppressed hypoxiainduced apoptosis by more than 50%, while 10 ${\mu}M$ AP-1 or 4 ${\mu}M$ NF-AT inhibitor did not, indicating involvement of NF-${\kappa}B$ in hypoxia-induced ${\beta}$-cell apoptosis. Overall, these results suggest that IP-10 is not involved in hypoxia-induced ${\beta}$-cell apoptosis, and that NF-${\kappa}B$ inhibitor can be useful for ameliorating hypoxia-induced ${\beta}$-cell apoptosis.

Expression of bcl-2 and p53 in Induction of Esophageal Cancer Cell Apoptosis by ECRG2 in Combination with Cisplatin

  • Aim: To investigate the mechanisms of induction of apoptosis of esophageal cancer cells by esophageal cancer-related gene 2 (ECRG2) in combination with cisplatin (DDP). Methods: Hoechest staining was performed to analyze the effects of single ECRG2 and ECRG2 in combination with DDP on apoptosis of EC9706 cells. The expression levels of p53 and bcl-2 mRNA and protein were determined by RT-PCR and Western blotting, respectively. Results: The number of apoptotic cells after the treatment with ECRG2 in combination with DDP for 24 hours was more than that after the treatment with single ECRG2. RT-PCR and Western blotting showed that the expression levels of bcl-2 mRNA and protein were both down-regulated, while p53 mRNA and protein were both up-regulated in the cells treated with ECRG2 in combination with DDP compared with those given ECRG2 alone. Conclusion: ECRG2 in combination with DDP can enhance the apoptosis of EC9706 cells, possibly by down-regulating bcl-2 expression and up-regulating p53.
