  • This study was examined the characteristics on the pollinating activities of Apis mellifera, Bombus ignitus and B. terrestris as pollinators on the watermelon flowers(Citrullus vulgaris Schrad), and the effects of the pollination by pollinators compared to the artificial pollination in the watermelon green houses in Nonsan City on summer season in 2005. As a result, B. ignitus and B. terrestris could not be used for summer season, because the bumblebees were not pollinated effectively indeed even two days after hives were released at watermelon green houses during the summer cultivating period. It could not be controlled a poor watermelon house conditions; high temperature and low humidity on summer season which were the bad environment for pollinating activities of bumblebees. But this study was showed that A. mellifera could be used comparatively good pollinator instead of artificial pollination in spite of the bad environment for bee within the watermelon green houses on summer season.

  • Hygienic behavior of Honey bees, Apis mellifera, was evaluated by uncapping and removing ability of dead broods from the nest. Hygienic behavior is originated from quantitative traits, which are expected to express key roles in colony defense against mite parasites and bacterial and fungal diseases. It is regarded as one of important characteristics of honey bee's resistance to parasites and pathogens. In this study, five inbreed and two hybrid lines of A. mellifera, the former five inbreed lines, which have been reared for over eight years at the National Academy of Agricultural Science in Korea, and the latter two hybrid lines, which have been bred by crossing between the inbreed lines, were investigated on their hygienic behavior by a pin-killed brood assay at 12hrs and 24hrs after treatment. The results indicated that after 12hrs one inbred line was proved to be hygienic (removal rate of dead brood >90%), three inbred and two hybrid lines showed intermediate behavior, and one inbred line belonged to non-hygienic (removal rate of dead brood <70%). However, after 24hrs, only one line was considered to be intermediate as removal rate was below 90%, thus all except this line had shown hygienic behavior.

High Level of Sequence-Variation in Sacbrood Virus (SBV) from Apis mellifera

  • Sacbrood virus (SBV) is one of the main pathogenic RNA viruses of honeybee. SBV is found worldwide and many local strains have been reported, such as kSBV, cSBV, and wSBV. In this study, SBV-specific DNA fragments were cloned and sequenced by reverse-transcription PCR from 4 populations of A. mellifera, 4 sequences from 1 population belonged to the 2134D51 genotype (349 nucleotides, nt) and 12 sequences from 3 populations belonged to the 2100D0 genotype (400 nt) among the 16 determined sequences. A total of 87 points of mismatches were found by comparison with the most similar sequences in GenBank. Seventeen single-nucleotide polymorphisms (SNP) were detected, and 6 SNP-patterns in the 2100D0 genotype and 2 SNP-patterns in the 2134D51 genotype were identified based on SNP positions. In SNP-pattern 2, 10 SNPs were detected, but only 2 SNPs were found in SNP-pattern7. Meanwhile, one SNP-pattern was found from one RNA-sample, multi SNP-patterns were detected from other RNA-samples. Large numbers of SNP variants indicate that vast numbers of point-mutations on SBV have occurred since SBV invaded Korea and that SNP smay have been introduced individually over time. Thorough analysis of SNP variants will not only define the local infection-route, but also the relationships between SNP-pattern and SBV-pathogenic abilities.

Identification of Mariner-Like Element(MLE) Gene from Nombyx mori. (누에에서의 Mariner 유사 전이인자유전자의 동정)

  • We have cloned an internal fragment of the putative transoisase gene of MLE in the silkworm, Bombyx mori, using PCR method with degenerative oligonucleotide primers designed to represent regions of amino acids encoding transposase. The resulting PCR clone, designed as BmoMAR, cords a partial ORF(152 a.a.) of MLE in which interrupted by five stop codons, and the sequence of its deduced amino acids showed 37% homology with Mos1, an active mariner, from Drosophila mauritiana. Furthermore, the BmoMAR exhibits nucleotide and amino acid homology with 59% and 37% from Apis mellifera and D. mauritiana 7.9 clone, respectively. This result strongly that a MLE is present in the genome of B. mori.

Therapeutic Effect of Bee Venom in Calves with Bacterial Diarrhea (송아지의 세균성 설사증에 관한 생봉독의 치료효과)

  • This study was designed to examine therapeutic effect of bee(Apis mellifera L.) venom in calves with bacterial diarrhea. Calves with bacterial diarrhea were administered with bee venom and therapeutic drug, respectively. In the bee venom-treated group, 69 calves were acupunctured once a day for 3 conseutive days. Two local acupoints of Jiao-chao(GV-1, at the indentation between the base of tail and the anus) and Bai-hui(GV -20, in the dorsal midline of the lumbo-sacral space) were stung by the bee. In the therapeutic drug-treated group, 55 claves were intramuscularly injected with a standard dosage of antibiotic (ciprofloxacin, 2.5 mg/kg of body weight) and antidiarrheal drug (berberine cholride, 10mg/head) once a day for 3 consecutive days. At post-treatment, 81.2% of the bee venom-treated calves and 76.4% of the therapeutic drugtreated calves were recovered from bacterial diarrhea. Bee acupuncture therapy showed in calves without side effects such as allergy hemorrhage, or infection. It might be concluded that apitherapy was affective in controlling of calves with bacterial diarrhea.

Therapeutic Effects of Natural Bee Venom in Sows with Mastitis, Metritis and Agalactia Syndrome (MMA 증후군 모돈에서 생봉독의 치료효과)

  • This study was designed to examine the clincotherapeutic effect of natural honeybee (Apis mellifera) venom in sows with mastitis, metritis and agalactia (MMA) syndrome. Sows with MMA syndrome after parturition were assigned to treated and nontreated control groups. In treated group, 22 sows were bee acupunctured once a day for 3 consecutive days. Acupuncture points of Jiao-chao (GV-1, at the indentation between the base of tail and the anus), Yang-ming (ST-18, outside at the base of teat) and Hai-men (ST-25, about 1 cm lateral to the umbilicus) were stung by the natural honeybees. In control group, 20 sows were intramuscularly injected with a standard dosage of penicillin G (400,000 IU/kg of body weight) once a day for 3 consecutive days. At post-treatment, 85.0% of control sows and 90.9% of sows in treated group recovered from MMA syndrome. Bee acupuncture therapy didn't show any side effects such as allergy, intoxication, hemorrhage, or infection. It might be concluded that apitherapy was effective in controlling of sows with MMA syndrome.

Effects of Flower Thinning Formulation on Activities of Digestive Enzymes and Acetylcholine Esterase in Honey bee Apis mellifera

  • The effects of a newly developed flower thinning formulation (FTF) on the vitality of the honey bee Apis mellifera were examined by measuring the activities of various digestive enzymes in adult worker bees. First, direct spraying of the FTF solution did not cause any behavioral changes or lethal effects for the honey bees based on 24 h observation. Second, oral ingestion of a sugar solution containing the FTF did not produce any significant change in the activities of amylase, proteinases, lipase, or acetylcholine esterase (AChE) in the worker bees 6 h or 24 h after treatment. Meanwhile, a commercial formulation containing sulfur compounds showed slightly reduced activities for several digestive enzymes and AChE, although no behavioral disturbance. Thus, the results of the present study suggest that the FTF is not toxic for honey bees, in terms of contact and ingestion. Therefore, this newly developed FTF can be used for flower thinning without any detrimental effects on pollinating insects.

Effects of the blood chemistry of honeybee (Apis mellifera L.) venom on the Hanwoo calves (신생송아지의 봉독 처리가 혈액성상에 미치는 영향)

  • This study was conducted to investigate effects of honeybee venom (Apis mellifera L. BV) on the calving, the growth performance and blood chemistry occurrence of Hanwoo calves. A total of twenty, a 3 day old male experimental calves were allocated into four groups, BV treated (0.05 mg/kg, 0.1 mg/kg and 0.2 mg/kg BV, n=5, respectively) and non-treated (n=5, Control) of Hanwoo calves in Icheon, Kyonggi province. Treatment of BV in calves increased body weight and weight gain during 60 days compared with control group. The concentrations of IgG in blood were significantly increased in BV groups compared with control at 60 days after treatment with BV. Cholesterol and glucose concentrations in BV group were significantly lower with control at 60 days. There were no differences in plasma biochemical components including aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphate (AKLP), total protein, albumin, globuline, total bilirubin, creatinine, blood urea nitrogen (BUN) and $Ca^+$. In conclusion, the BV treatment had effect on growth, the score of the calf scours and blood biochemical profile.

Antimicrobial Property of honeybee (Apis mellifera L.) venom against Propionibacterium acnes and Aerobic Skin Flora (국내산 봉독의 여드름 유발균 및 피부 상재균 증식 억제 효과)

  • The in vitro antibacterial activities of honeybee(Apis mellifera. L) venom collected by a bee venom collector were investigated against several bacteria including antibiotic-susceptible and resistant Propionibacterium acnes. Honeybee venom was prepared with different concentrations and they showed strong antibacterial activites. Honeybee venom inhibited the growth of the tested antibiotic-resistant P. acnes at the concentration of 1 mg/ml. The inhibitory activities of the honeybee venom showed time-dependent manner. Honeybee venom did not influence the viability of human dermal fibroblast at the high concentration of less than 10 mg/ml. From these results, we expect that honeybee venom has strong antibacterial activities and has advantage for treating cure.

Chemical Composition and Bioactivity of Korean Green Tea (Camellia sinensis) Pollen collected by Honey Bee (한국 녹차 화분의 화학적 조성과 기능성 연구)

  • In this study, we investigated the nutritional composition including proximate, amino acid, vitamin, minerals, and the antioxidant activity of green tea (Camellia sinensis) pollen grains collected by Apis mellifera bees, for use as a health food. The crude protein and fat content was estimated at 26.14% and 3.49%, respectively. Eighteen amino acids were identified in green tea pollen, including 8 essential amino acids and 10 non-essential amino acids. The predominant amino acids were glutamic acid, proline and aspartic acid accounting for about 33.3% of total free amino acids. The concentration of vitamin C was the highest value of 35.7%, followed by $B_3$ and $B_2$ among the detected vitamins. The predominant minerals were potassium (790.32 mg/100g), followed by phosphorus (707.52 mg/100g) and sulfur (302.67 mg/100g), whereas copper, zinc and sodium were detected as minor elements. The antioxidant activity and phenolic content accounted for 33.8% at $500{\mu}g/mL$ extract and $2.55 {\mu}g/mg$, respectively.
