Angiotensin I-Converting Enzyme (ACE) Inhibitory Activity of Elk (Cervus elaphus) Velvet Antler

  • Angiotensin I-converting enzyme (ACE) inhibitory activities of elk antler hydrolysates prepared with three kinds of proteases, pepsin, trypsin and $\alpha-chymotrypsin$, were investigated. The ACE inhibitory activity of the pepsinolytic hydrolysate was the highest with an $IC_{50}$ value of $9.3\mu g/mL.$ In addition, three kinds of pepsinolytic hydrolysates with relatively high molecular weights (over 10,000 Da), medium molecular weights (5,000 to 10,000 Da), and low molecular weights (below 5,000 Da) were fractionated using an ultrafiltration membrane system. The below 5,000 Da hydrolysate exhibited the highest ACE inhibitory activity. These results indicate that the pepsinolytic hydrolysates of elk velvet antler could be a good source of peptides with ACE inhibitory activity.

발효녹용 추출물에 의한 비장세포의 생존율 및 interleukin-12 생산 증진 (Fermented antler extract enhances the viability and interleukin-12 production of spleen cells)

  • The effects of antlers have long been known in traditional Asian medicine. However, few studies have investigated the effects of antlers on immunity. In this study, we investigated whether fermented antler extract (FAE) has immunomodulatory effects on spleen cells. FAE enhanced the activity of spleen cells in a concentration dependent manner compared to antler extract. Interestingly, FAE significantly increased the production of interleukin-12, a representative cytokine of cell-mediated immunity, while it marginally increased that of tumor necrosis factor-alpha. Flow cytometry analysis demonstrated that FAE can protect spleen cells from spontaneous cell death without a significant proportional change in subsets, mainly lymphocytes. Taken together, the results of the present study showed that FAE has beneficial effects on spleen cells, a major type of immune cell, indicating that it can function as an immunomodulator without significant cytotoxicity. These data may broaden the use of FAE in basic research and clinical areas.

녹용이 흰쥐의 비만억제에 미치는 영향 (Effect of Deer Antler on the Inhibitory Action of Obese Rats)

  • Objective: We investigated the effects of deer antler extract (DAE) on the expression of the weight, leptin, and serum levels (lipid metabolism) of the obese SD rats induced by a $2\%$ cholesterol diet in vivo. Methods: In vivo, the weight, leptin and serum levels were measured after a period of 7 weeks for obese SD rats induced by a $2\%$ cholesterol diet and treated with DAE of three concentrations (0.938 g/kg/5ml, 1.875 g/kg/5ml, 3.75 g/kg/5ml). Results: 1. Body weight didn't change significantly. 2. Leptin levels significantly decreased (p<0.05). 3. Total cholesterol, triglyceride, LDH, sGOT & sGPT levels significantly decreased in the sample (p<0.05). 4. The HDL cholesterol levels significantly increased in the sample (p<0.05). 5. The average size of the epididymal fat cells significantly decreased as the concentration of deer antler extract increased (p<0.05). Conclusion: Deer antler effects inhibition of obese rats.

Structure Characterization and Antihypertensive Effect of an Antioxidant Peptide Purified from Alcalase Hydrolysate of Velvet Antler

  • Recently, interest in food-derived bioactive peptides as promising ingredients for the prevention and improvement of hypertension is increasing. The purpose of this study was to determine the structure and antihypertensive effect of an antioxidant peptide purified from velvet antler in a previous study and evaluate its potential as a various bioactive peptide. Molecular weight (MW) and amino acid sequences of the purified peptide were determined by quadrupole time-of-flight electrospray ionization mass spectroscopy. The angiotensin I-converting enzyme (ACE) inhibition activity of the purified peptide was assessed by enzyme reaction methods and in silico molecular docking analysis to determine the interaction between the purified peptide and ACE. Also, antihypertensive effect of the purified peptide in spontaneously hypertensive rats (SHRs) was investigated. The purified antioxidant peptide was identified to be a pentapeptide Asp-Asn-Arg-Tyr-Tyr with a MW of 730.31 Da. This pentapeptide showed potent inhibition activity against ACE (IC50 value, 3.72 μM). Molecular docking studies revealed a good and stable binding affinity between purified peptide and ACE and indicated that the purified peptide could interact with HOH2570, ARG522, ARG124, GLU143, HIS387, TRP357, and GLU403 residues of ACE. Furthermore, oral administration of the pentapeptide significantly reduced blood pressure in SHRs. The pentapeptide derived from enzymatic hydrolysate of velvet antler is an excellent ACE inhibitor. It might be effectively applied as an animal-based functional food ingredient.

꽃사슴의 녹용 성장기간 중 혈액 내 미네랄 및 효소활성 변화에 관한 연구 (A Study on the Changes of Blood Mineral and Enzyme Activity During Growth Period of Velvet Antler in Korean Spotted Deer(Cervus Nippon))

  • 본 연구는 축사 내에서 집약적으로 사육되고 있는 꽃사슴의 녹용 성장기간 중의 혈액성분변화와 녹용성장과의 관련성 해석을 위한 기초자료를 제공하고자 수행하였다. 녹용성장기간 중 혈액성분의 변화를 측정하기 위하여 우리 내에서 사육 중인 4${\sim}$6년 생 꽃사슴 웅록 25두를 무작위로 선별하여, 낙각 직후부터 10일 간격으로 절각일(50일)까지 혈액을 채취하여 미네랄과 효소 활성치를 측정하였다. 칼슘과 인의 농도는 녹용성장기간 중 커다란 변동이 없었으며 나트륨, 칼륨, 염소의 농도는 낙각 직후와 녹용성장에 따른 경시적인 유의차가 나타났다. AST, ALT, amylase, CK, GGT 및 LDH의 농도는 낙각 직후와 녹용성장에 따른 유의차는 인정되지 않았으나, ALK-P의 농도는 녹용성장이 진행되면서 계속 증가하여 낙각 후 50일 경에는 최고치를 나타내어 낙각시의 농도와 유의차가 나타났으며(p〈0.01), CK와 LDH의 활성은 다른 동물들보다 높게 나타났다.

Composition of Biologically Active Substances and Antioxidant Activity of New Zealand Deer Velvet Antler Extracts

  • Deer velvet antler was subjected to the extraction process using boiling water at three different temperatures (100, 110 and $120^{\circ}C$) and 70% ethanol solution. Functional components such as uronic acid, sulfated-glycosaminoglycans (sulfated-GAGs) and sialic acid in the extracts were analyzed, and their antioxidant activities were investigated using several in vitro models. Uronic acid and sulfated-GAGs content of each extract significantly decreased with increasing extraction temperature (p<0.05), while the residues obtained from the upper and middle part of the antler had a higher uronic acid content than the residues obtained from the base section. Sialic acid contents were highest in compounds extracted at $110^{\circ}C$, followed by 120 and $100^{\circ}C$. The 70% ethanol extracts also had a high levels of uronic acid content, but not for sulfated-GAGs and sialic acid. All extracts showed good antioxidant ability in a dose-dependant manner, with the $100^{\circ}C$ residue exhibiting the strongest activity compared to the 110 and $120^{\circ}C$ extracts. In relation to the hydroxyl radical scavenging activity and reduction power, the 70% ethanol extract exhibited the strongest activity. Furthermore, the velvet antler extracts inhibited apoptosis in hydrogen peroxide-induced PC-12 cells.

노화 촉진 생쥐에서 녹용의 조혈 작용에 관한 연구 (A Study of Hematopoietic Action of Pilose Antler in Senescence Accelerated Mice)

  • In previous studies we reported that the levels of RBC, hemoglobulin (HGB) and hematocrit (HCT) in SAM P6 were increased significantly from 7 day after oral administration of the pilose antler extract, 5 g/kg/day, and were lasted during the study. Thus, this study was performed to elucidate mechanism of erythropoietic action by the extract administration. SAM R1 and SAM P6 were chosen as experimental animals. At age of 12 weeks, pilose antler extract were given 0.3 and 5 g/kg/day (p.o.) each for 0, 7 and 14 days in both animals. Complete blood cells such as RBC, HGB, and HCT were counted. And plasma concentration of erythropoietin (EPO) which is the major regulator of erythropoiesis was measured using $^{125}I-antierythropoietin$ IgG. Total iron concentration in plasma was also analyzed. The levels of RBC and HCT were increased significantly after administration at both doses of 0.3 and 5 g/kg/day in SAM P6, however, these were increased only at dose of 5 g/kg/day in SAM R1. The plasma EPO concentration was increased significantly after administration in SAM P6. The plasma concentrations of total iron were significantly decreased after administration of the extract in SAM P6. These results suggest that the changes in erythropoietic effects after the administration of pilose antler extract may be mediated, at least in part, through the change in the plasma EPO.

Antioxidant, Anti-acetylcholinesterase and Composition of Biochemical Components of Russian Deer Velvet Antler Extracts

  • Russian deer velvet antlers were divided into three parts and subjected to a extraction process using hot water at 100, 110, and $120^{\circ}C$ or an extraction with 70% ethanol. Each extract was analyzed for its biochemical components, including uronic acid, sulfated-glycosaminoglycans (sulfated-GAGs), and sialic acid, and the antioxidant and anti-acetylcholinesterase activities were investigated. Different levels of uronic acid and sulfated-GAGs were observed in the extracts according to the water temperature used for the extraction, and contents decreased with increasing extraction temperature. The upper layer of each extract showed high amounts of uronic acid and sulfated-GAGs, followed by the middle and base layers. Ethanol extraction was more effective for recovering uronic acid than sulfated-GAGs. Sialic acid content was the highest in the $110^{\circ}C$ extracts but was not observed in the ethanol extracts. Velvet antler extracts showed strong antioxidant activities against DPPH and hydrogen peroxide as well as strong reducing power in a dose-dependent manner. However, the antioxidant activities were different in each layer and according to the extraction method. Additionally, velvet antler extracts exhibited inhibitory activity against acetylcholinesterase, which is associated with Alzheimer's disease, in a dose-dependent manner. These results suggest that velvet antler extracts are useful as a functional food ingredient and/or a pharmaceutical.

Molecular cloning and expression analysis of annexin A2 gene in sika deer antler tip

  • Objective: Molecular cloning and bioinformatics analysis of annexin A2 (ANXA2) gene in sika deer antler tip were conducted. The role of ANXA2 gene in the growth and development of the antler were analyzed initially. Methods: The reverse transcriptase polymerase chain reaction (RT-PCR) was used to clone the cDNA sequence of the ANXA2 gene from antler tip of sika deer (Cervus Nippon hortulorum) and the bioinformatics methods were applied to analyze the amino acid sequence of Anxa2 protein. The mRNA expression levels of the ANXA2 gene in different growth stages were examined by real time reverse transcriptase polymerase chain reaction (real time RT-PCR). Results: The nucleotide sequence analysis revealed an open reading frame of 1,020 bp encoding 339 amino acids long protein of calculated molecular weight 38.6 kDa and isoelectric point 6.09. Homologous sequence alignment and phylogenetic analysis indicated that the Anxa2 mature protein of sika deer had the closest genetic distance with Cervus elaphus and Bos mutus. Real time RT-PCR results showed that the gene had differential expression levels in different growth stages, and the expression level of the ANXA2 gene was the highest at metaphase (rapid growing period). Conclusion: ANXA2 gene may promote the cell proliferation, and the finding suggested Anxa2 as an important candidate for regulating the growth and development of deer antler.

  • In order to investigate the reducing effect of velvet antler water extract (VAWE) on the toxicity of anti-cancer drug, cis-platin (CDDP) and mitomycin C (MMC), we examined effects of co-administration with VAWE and anti-cancer drugs on their toxicities. We recognized that $LD_{50}$ of CDDP/MMC were increased by co-administration with VAWE and them in mice. It was found that co-administration of VAWE and MMC increased the survival rate in mice treated by lethal dose of MMC. Also, co-administration of VAWE and CDDP/MMC inhibited decrease of the body weight and organ weight in mice intoxificated by CDDP/MMC. The increase of serum blood urea and serum creatinine levels in rats intoxicated by CDDP were significantly inhibited by the co-administrationin with VAWE and CDDP. The decrease of RBC and WBC in rats intoxificated by MMC were significantly inhibited by the co-administration with VAWE and MMC. These results suggest that the combined usage of VAWE and CDDP/MMC drugs may be a new method for prevented or minimized the toxicity of them.
