Studies on the antitumor components of Korean Basidiomycetes $(IV)^*$ Antitumor Components of Namatoloma Fasciculare (Fr.) Karst.

  • The carpophores of a mushroom, Naematoloma fasciculare (Fr.) Karst, were extracted with 0.2 N NaOH and the extract was dialyzed through visking tube. It was found to contain an antitumor activity against sarcoma 180 implanted in mice. The components of this aqueous extract were found to be a polysaccharide and a protein by color reactions including Anthrone and Lowery-Folin tests. The hydrolysis of the polysaccharide with 3% HCL-Me-OH and trimethylsily lation of the hydrolysate yielded five monosaccharides, i. e. glucose, frutose, mannose, galactose and xylose, which were detected and analyzed by GLC. After hydrolysis of the protein with 6N HCl, 15 amino acids, including aspartic acid and glutamic acid, were detected and analyzed by an auto amino acid analyzer.

야생 비늘 버섯의 항암 성분에 관한 연구 (Studies on Antitumor Components of Wild Pholiota squarrosa (Fr.) Quel.)

  • In order to investigate the antitumor components of Korean wild higher fungi, the carpo-phores of Pholiota squarrosa collected in Kaung Nung area were extracted with water by refluxing for eight hours at 80-90.deg. C. The extracts were dialized for six days against distilled water and lyophilized to produce crude polysaccharide powders. They were found to have the antitumor activity against sarcoma 180 implanted in mice. Especially, the inhibition ratio of the extract of Pholiota squarrosa was 78.7% in the dose of 20mg/kg/day for the period of ten days. The tumor in two of the ten mice was completely regressed. The components of these aqueous extracts were found to be polysaccharide and protein. The total polysaccharide was 42% by Anthrone method and the protein was 55% by Lowry-Folin method in the antitumor component of Pholiota squarrosa.

  • To find antitumor components in the shake-cultured mycelia of Volvariella bombycina, the mycelia were extracted with hot water. After the extract was dialyzed and freeze-dried, a protein-polysaccharide fraction was obtained and examined for antitumor activity against the solid form of sarcoma 180 in ICR mice. It showed 60.3% inhibition ratio at a dose of 20mg/kg/day for 10 days. It was found to consist of a polysaccharide moiety and a protein moiety. After gel filtration on Sepharose 4B, Fraction B was obtained and showed the highest inhibition ratio of 71.1%. When the antitumor component was examined for immunopotentiating activity, it was found to increase the macrophage accumulation in the peritoneal cavity as well as the antibody production of the spleen cells of the mice.

  • To investigate a possibility of producing the antitumor component by shake culture method, the mycelia of Flammulina velutipes were cultured in flasks on a shaker at $26{\sim}28^{\circ}C$ at 180 rpm for seven days. The extract of the mycelia was concentrated under vacuum. The precipitate obtained by adding a three-fold volume of ethanol was centrifugated and freeze-dried after dialysis. The fraction was tested against sarcoma 180 in the mice. The inhibition ratio of the fraction against the tumor was 68.0% at the dose of 20 mg/kg/day for the period of ten days and the tumors in three of the mice were completely regressed. The results showed, therefore, that the antitumor component was produced by the shake culture method.

Immunostimulation Effects of Cell Wall Components Isolated from Lactobacillus plantarum

  • Immunostimulation effects of the cell wall components isolated from Lactobacillus plantarum were investigated by studying the macrophage s tumorcidal activity, splenocyte proliferation, anticomplementary activity and the inhibition of peritoneal tumor cell growth measured with ICR mice inoculated with sarcoma 180. The immunopotentiating cell wall components were a complex of peptidoglycan and exopolysaccharides. The tumorcidal activity of macrophage against Yacl and B16 tumor cells was enhanced when the cell wall components were added into the macrophage s culture medium. They also stimulated splenocytes to proliferate up to the same level as when the concanavalin A was added into the splenocyte's culture medium. The complementary activity was inhibited by 50% when the cell wall components were incubated with the sheep red blood cells treated with hemolysin and guinea pig complement. This result confirmed that the cell wall components had an antitumor effect, because the anticomplementary activity is usually accompanied by an antitumor activity at the same time. This fact was confirmed again by the inhibition of the growth of sarcoma 180 when the cell wall components were injected intraperitoneally into ICR mice inoculated with sarcoma 180. As a result, it is concluded that the cell wall components isolated from Lactobacillus plantarum had multifunctional immunostimulation effects in vitro and in vivo.

  • To investigate antitumor components of Korean higher fungi, the carpophores of Schizophyllum commune and Auricularia auricula-judae collected in Kyeong Buk Province were extracted with hot water or 0.1N-NaOH solution. The concentrated extracts were precipitated by addition of ethanol, and the precipitates were purified by dialyzing through visking tube and polysaccharide fractions were obtained. They were found to show antitumor activity against sarcoma 180 implanted in mice. Especially, the inhibition ratio of the extract of Auricularia adicula-judae was 90.8% in the doses of 100mg/kg/day for the period of ten days. The tumor in five of the eight mice was completely regressed. The components of these aqueous extracts were found to be polysaccharide and protein. The hydrolysis of the respective polysaccharide yielded four monosaccharides. After hydrolysis of the protein fraction, 15 amino acids were identified in the respective fraction of S. commune and A. auricula-judae.

갓버섯의 항균 분석 (Antitumor Components of the Cultured Mycelia of Lepiota procera)

  • 한국산 담자균류인 갓버섯 Lepiota procera의 균사를 액내 배양하여 항암성분인 단백성 다당체를 분리하였다. 이 성분은 DEAE-Sephadex A-50 이온교환수지와 Sepharose-4B gel Filtration을 이용하여 정제하여 Fraction C1을 얻었으며 이 Fr, Cr은 단백질과 다당체로 구성되어 있으며 항암 효과는 10mg/kg/day 투여군에서 64%의 저지유을 나타내었다. 이러한 항암작용의 기전을 밝히기 위한 연구의 일환으로 면역에 미치는 영향을 실험함 결과 이 단백다당체는 용혈반형성 세포수를 증가시켰으며, 저하된 지연성 과민반응을 회복시켰을 뿐만 아니라, carrageenan 투여에 의해 억제된 면역능을 다시 증강시켰음을 알 수 있었다. 이러한 결과들은 이 버섯의 항암작용이 세포독성에 의한 것이 아니라 종양에 대한 면역능을 강화시켜 발휘됨을 제시하고 있다.

  • To investigate antitumor components in Korean higher fungi, the carpophores of Pholiota squarrosa belonging to the family Strophariaceae were collected and extracted with hot water. A protein-bound polysaccharide fraction was obtained by adding ethanol to the extract and by dialyzing through Visking tube. The fraction was examined for antitumor activity against sarcoma 180 implanted in mice. It showed an inhibition ratio of 78.7% at the dose of 20mg/kg/day. The tumor in two of the ten mice was completely regressed. The chemical analysis of the antitumor fraction by Anthrone and Lowry-Folin methods showed that it consisted of 42% polysaccharide and 55% protein. The enzyme fraction of the carpophores showed no proteolytic activity on casein.

  • To find antitumor components with low toxicity in natural products of Korean Basidiomycete, the carpophores of Flammulina velutipes (Fr.) Sing. were extracted with hot water for eight hours. The extract was purified by dialyzing through Visking tube and a protein-bound polysaccharide fraction was obtained as pale brownish amorphous powder after it was freeze-dried. The fraction was examined for antitumor activity against sarcoma 180 implanted subcutaneously in the left groins of I.C.R. mice. The inhibition ratio of this fraction of against the tumor was 62.3% at the dose of 10 mg/kg/day for the period of ten days. The tumors in three of the ten treated mice were completely regressed.

Studies on the antitumor activity of duchesnea indicae herba

  • As a part of finding the biologically active components from Duchesnea indica (Andr.) Focke (Rosaceae), antitumor activities of its water soluble fractions have been studied. The fractions were examined for antitumor activity against sarcoma 180 implanted in mice. The anitumor inhibition ratios of the water soluble fractions from Duchesnea indica were 17.9, 37.1, 62.1, 62.4%, repectively.
