천잠견단백질 가수분해 분말의 생리활성 (Biological Activities of Antheraea yamamai Silk Fibroin Hydrolysates)

  • 천잠 견단백질 가수분해 분말의 생리활성을 검토하여 비의류용 소재개발을 위한 기초 자료로 삼기 위하여 우선 항산화 및 항당뇨 활성을 탐색하였다. 1. 천잠 고치를 가수분해하여 제조한 분말은 항산화제인 비타민 C 대비 75% 내외의 항산화 활성을 나타내었다. 2. 천잠 견단백질 가수분해 분말은 혈당강하 효과도 나타내어 기능성 식품소재 적용 가능 물질로 사료된다.

삼나무와 편백나무 정유의 미백 및 항산화 효능 평가 (Whitening and Antioxidant Activities of Essential Oils from Cryptomeria japonica and Chamaecyparis obtusa)

  • 본 연구는 삼나무와 편백나무 정유의 tyrosinase 저해 활성과 DPPH (1,1-diphenyl-2- picryl-hydrazyl) radical 소거 활성, SOD (superoxide dismutase) 유사활성을 구명하여 정유의 미백 및 항산화 효능을 평가하였다. 본 연구를 통해 삼나무와 편백나무 정유의 미백 및 항산화 기능성 소재로 개발하기 위한 기초자료로 이용하고, 건조 중 발생하는 nVOCs의 부가적 가치를 확인하고자 하였다. 삼나무 및 편백나무 잎의 정유는 수증기 증류법으로 24시간 추출하였고, 목부는 고온 열처리기를 이용하여 80, 100, $120^{\circ}C$의 온도조건에서 열기 건조하여 건조 중 목부로부터 발생하는 부산물인 천연 휘발성 유기화합물(nVOCs, Natural Volatile Organic Compounds)을 응축액의 형태로 추출하였다. 삼나무 잎의 정유가 편백나무 잎의 정유 보다 미백 및 항산화 활성에서 모두 높은 효과를 나타내었고 건조 시 발생한 nVOCs는 온도가 올라갈수록 미백 활성이 높아지는 것을 확인할 수 있었으며 항산화 활성은 $120^{\circ}C$에서 발생한 삼나무 nVOCs에서만 나타났다.

번행초 추출물의 항산화 활성 (Antioxidant Activities of the Solvent Extracts from Tetragonia tetragonioides)

  • 본 연구에서는 예로부터 위장병의 예방 및 치료 효과가 뛰어난 작물로 알려진 번행초(Tetragonia tetragonioides)의 각종 용매 추출 분획물의 항산화 효과를 명확히 하기 위해 총 폴리페놀과 플라보노이드 함량, 비타민 E함량, DPPH free radical 소거활성, Superoxide 음이온 소거활성, 과산화물생성 억제 활성, 환원능력 그리고 금속 킬레이트 효과를 측정하였다. 먼저 총 폴리페놀 화합물의 함량은 DCM 분획, EA 분획에서 높게 나타났고, 플라보노이드 함량은 HX 분획, EA 분획 순으로 나타났다. 비타민 E의 함량은 HX 분획, DCM 분획 순으로 높게 나타났다. DPPH radical 소거활성 효과는 DCM과 EA 추출 분획에서 $IC_{50}$ 값이 각각 $554.25{\mu}g/ml,\;394.96{\mu}g/ml$으로 산화방지제로 널리 이용되고 있는 BHT의 $784.7{\mu}g/ml$보다 우수하게 측정되었다. 과산화물 생성 억제 활성에서 번행초 분획은 지질단백질을 산화로부터 보호한다고 알려진 ${\alpha}$-tocopherol과 유사한 능력을 보였으나, 페놀계 합성 항산화제인 BHT, BHA에 비해서는 억제능이 다소 낮음이 확인되었다. 환원능력을 통한 항산화 활성 측정에서는 1.5 mg/ml농도에서 EA 분획의 활성이 ${\alpha}$-tocopherol보다 유의하게 높게 나타나 좋은 항산화 소재임이 확인되었다.

정전가매이진탕(正傳加味二陳湯)이 과산화지질 급여 비만 쥐의 지질강하, 항산화효과 및 염증매개물질의 생산에 미치는 영향 (Effects of Jengjengamiyijin-tang (zhengzhuanjiaweierchentang) on Lowering Lipid, Antioxidation and Production of Inflammatory Mediators Being Used Rats Fed on High Oxidized Fat)

  • Objectives The purpose of this study was investigating effects of Jengjengamiyijin-tang (zhengzhuanjiaweierchentang) (JGYT) on lowering lipid, antioxidation and production of inflammatory mediators being used rats fed on high oxidized fat. Methods We divided fat Sprague-Dawley rats fed on high oxidized into 4 groups. Each of 8 rats was divided into a control group and experimental groups. We fed a control group of rats a basal diet and administered normal saline (100 mg/kg, 1 time/1 day) for 4 weeks. And We fed each experimental group of rats basal diet and administered an extract of JGYT extracts (100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of proinflammatory cytokines, antioxidative activity and plasma tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), plasma interleukin-6 (IL-6), Apo-B, Apo-E and Leptin gene expression. Results 1. Concentration of plasma FFA, LDL-cholesterol, plasma and liver total cholesterol showed a significant decrement in JGYT groups. However, concentration of plasma HDL-cholesterol showed a significant increment in JGYT groups. 2. Concentration of plasma and liver TG, TBARS showed a significant decrement in JGYT groups. However, concentration of liver GSH-Px, SOD and CAT showed a significant increment in JGYT groups. 3. Plasma GPT activity and concentration of plasma IL-6, TNF-${\alpha}$, NO, Ceruloplasmin, ${\alpha}1$-acid glycoprotein showed a significant decrement in JGYT groups. 4. In the analysis of RT-PCR, gene expression of Apo-B and Apo-E in the JGYT groups showed a low expression than that of control group. However, the gene expression of leptin showed no difference in all the treatment groups. 5. The ratio of leptin expression per ${\beta}$-actin expression showed no significant difference among all treatment groups. However, The ratio of Apo-B and Apo-E expression per ${\beta}$-actin expression showed a significant decrement in JGYT groups. Conclusions According to this study, extract of JGYT showed a positive effect in lowering lipid, antioxidation and control of inflammatory mediators production.

  • This study was carried out in order to investigate the browning reactions of fish oil-amino acid model system and the properties of browning products. The lysine was added to the hair tail fish and mackerel oil, and incubated at $40^{\circ}C$ for 178 hours. Browning was rapidly developed with the begining of decline in PoV and CoV and a remarkable decrease in such polyunsaturated fatty acids as $C_{20:5},\;C_{22:5},\;C_{22:6}$ compared with the other fatty acids, was resulted. And among the polyenoic acids the role of $C_{22:6}$ was considered to come first as appeared in the test with authentic samples. In the water soluble fraction of the browning products obtained from the fish meat or the lysine added fish oil, some antioxidation activity was detected. But in the liposoluble fraction which covers most of the browning reactions in the fish meat, little antioxidation activity was detected. And all of the browning products provided in this experiment shelved very low reducing activity.

생맥산(生脈散)과 구성 약물의 성분 분석 및 항산화(抗酸化) 효능 비교 (An Analysis of Saengmaegsan's Ingredients and a Comparison Study on Anti-Oxidation Effects According to Kinds of Extract)

  • Objectives: The experiment was to estimate the value of Saengmaegsan as a therapeutic anti-aging agent with an analysis of ingredients and their bio-activating effects by enzymologic methods. Methods: We analyzed anti-aging effects of the ingredients of Saengmaegsan being Liriope Platyphylla, Schizandrae Chinensis, and Schizandrae Chinensis. Results: In Liriope Platyphylla and Schizandrae Chinensis, reducing sugar concentration was shown at the highest level. Of mineral contents, K+ and Na+ were the highest in Saengmaegsan and each ingredient also, and then Ca++ and Al. The amount of polyphenol showed was in order Schizandrae Chinensis > Saengmaegsan > Panax Ginseng > Liriope Platyphylla. The electron donating ability of Schizandrae Chinensis doubled that of Saengmaegsan and Panax Ginseng, at the same time that one per 1 ml in Saengmaegsan was significantly found the greatest level as compared to each single ingredient. SOD-like activity was high in Saengmaegsan and Schizandrae Chinensis. SOD-like activity of Saengmaegsan was higher than in single components. Nitric oxide inhibition in pH 1.2 was high in Saengmaegsan and Schizandrae Chinensis, and for per 1 ml it showed the same pattern as above. In pH 3.0 the result was not different from in pH 1.2. Xanthine oxidase inhibition was high in Schizandrae Chinensis and for per 1 ml showed the greatest activity in Liriope Platyphylla. Tyrosinase inhibition in Omija was the most noticeable, and for per 1 ml was Liriope Platyphylla more than any others. Conclusions: With this analysis of ingredients, it is proven that Saengmaegsan and each component fosters antioxidation. On the whole, the composite prescription Saengmaegsan was superior to each individual component.

사염화탄소에 의한 간손상에 대한 황백의 간보호 효과 연구 (Hepatoprotective Effect of Bark of Phellodendron amurense RUPR. on Liver Damage Induced by Carbon Tetrachloride)

  • We evaluated the hepatoprotective activity of crude hot-water extracts of the traditional Korean medicinal herb, Phellodendri Cortex (PC; Bark of Phellodendron amurense RUPR.), in an experimental model of hepatic damage induced by carbon tetrachloride ($CCl_4$). The serum marker of liver damage, sGOT, sGPT and sALP, were significantly decreased in the liver of the PC treated rats, compared with that of $CCl_4$ treated group. The histological observation of liver section of rats, showed the same protective effect of PC treatment. And the protective activity of PC was more significant in the post-treated group than pre-treated group. The significant decrease of malodialdehyde and increase of glutathion, catalase activity were observed in the liver homogenate of PC treated rats. Based on these findings, it is suggested that PC has potent hepatoprotective effects and the mechanism of the protection may be related to antioxidation pathways.

병포장 갓김치의 항산화 효과에 대한 연구 (Antioxidation Activities of Bottled Mustard Leaf Kimchi during Fermentation)

  • Mustard Leaf Kimchi (MLK) is a traditional fermented Korean vegetable food. This study was conducted to investigate the effects of part vacuum treatment on MLK packed in a glass bottle during fermentation. There have been a few previous studies that examined the chemical and microbial changes during MLK fermentation. However, the major object of this study was to investigate the antioxidative activities of vacuum treated MLK. In this study, the antioxidative activity of vacuum treated mustard leaf Kimchi (VM) and control mustard leaf Kimchi (CM) were examined. VM and CM were fermented at $5^{\circ}C$ for 8 weeks. A model system was designed to evaluate the antioxidative activity of crude chlorophylls and carotenoids (CCC) extracts from Mustard leaf Kimchi. The oxidative reaction of the linoleic acid mixture system at $30^{\circ}C$ in the dark was quantified determining the peroxide value and conjugated dienoic acid content. The effect of the CCC extracts on lipid peroxidation in a rat liver homogenate was examined. Formation of lipid peroxides was estimated by the TBA value, and the CCC extracts were found to inhibit the TBA value. Chlorophyll a and b, and carotenoids, Which are the major components in the CCC extracts of Kimchi were isolated on a DEAE-sepharose CL-6B and Sepharose CL-6B column and TLC. The effects of chlorophyll a and b, caroteins on linoleic acid autoxidation were measured by determining the peroxide value. In addition, their effects on free radical scavenging were investigated by DPPH. In this assay, chlorophyll a showed the greatest antioxidative activity followed by chlorophyll b, and carotenoids. MLK contains a sufficient content of chlorophyll a and b, and carotenoid which have strong antioxidative activities.

감마선 조사가 오미자의 생리 활성과 색상 변화에 미치는 영향 (Effects of Gamma-Irradiation on Biological Activities and Color Changes of Extracts of Schizandrae fructus)

  • 오미자 추출물을 감마선 조사하여 색택 개선, 항산화성, 항균성에 대하여 실험하였다. 오미자의 추출 방법은 열수추출, ethanol, methanol 그리고 acetone으로 추출하였으며, 감마선 조사는 10, 20 그리고 30 kGy로 하였다. 감마선 조사 선량이 증가할수록 명도(Hunter color L-value)의 경우 비조사구보다 조사구가 월등히 밝은 색을 보였으며, 적색도(a값)와 황색도(b값)는 감소하는 결과를 보여줬다. TBA가 분석 결과는 ethanol로 추출한 경우 가장 높은 항산화력을 확인하였다. 전자 공여능의 측정 결과는 열수 추출한 경우에 비조사구와 조사구 모두 가장 활성이 높았으며 ethanol과 methanol의 경우에는 비조사구와 비슷한 활성을 보여줬다. 오미자 추출 용매별 및 감마선 조사 선량에 따른 항균효과는 B. subtilis, B. natto, B. megaterium, S. aureus, Sal, typhymurium과 E. coli 등 모든 균주에 대해 아주 높은 항균 활성을 나타내었다. 또한 오미자 추출물은 30 kGy까지 감마선을 조사하여도 추출물의 생리 활성에는 영향을 미치지 않았다 따라서 감마선 조사 기술은 천연 식품 보존제 및 화장품 원료로서 사용하기 위한 오미자 추출물의 색택 개선에 활용 가능성이 높을 것으로 기대된다.
