고콜레스테롤혈증 성인에서 울금과 명일엽 복합 추출물의 복용에 따른 혈중지질, 항산화 및 염증 관련 지표의 변화 (The Effects of Angelica keiskei Koidzumi and Turmeric Extract Supplementation on the Blood Lipids, and Antioxidant and Inflammatory Markers in Hypercholesterolemic Adults in Korea)

  • This study investigated the effects of Angelica keiskei Koidzumi and turmeric extract supplementation(ATE) on blood lipids, antioxidant and inflammatory markers in 35 hypercholesterolemic Korean adults with high blood cholesterol levels (serum total cholesterol$\geq200mg/d{\ell}$ or LDL-cholesterol$\geq130mg/d{\ell}$). They received ATE(n=21, 14 females and 7 males) or placebo(control group, n=14, 11 females and 3 males) for 4 weeks. There was no significant change in serum total cholesterol, LDL-cholesterol and HDL-cholesterol levels after ATE supplementation in the both groups. However, the LDLcholesterol: HDL-cholesterol ratio(LPH) was significantly decreased and both serum prostagrandin E2(PGE2) levels were significantly decreased in those receiving ATE. No significant changes were evident in interleukin(IL)-$1\beta$, IL-6, IL-8, 8-isoprostane, malondialehyde, total antioxidant capacity and oxidized-LDL. These results suggest that complex extract of Angelica keiske and turmeric has the potential to decrease cardiovascular risk by reducing LPH and inflammatory mediator $PGE_2$ in hypercholesterolemic adults.

Reproductive Performance of Dairy Buffaloes Supplemented with Varying Levels of Vitamin E

  • The effect of vitamin E supplementation on plasma ${\alpha}$-tocopherol level, total antioxidant level and reproductive performance in Murrah buffaloes was studied during periparturient period. Twenty-four advance pregnant buffaloes were randomly divided into four equal groups as $T_1$, $T_2$, $T_3$ and $T_4$ and were supplemented with 0, 1,000, 1,500 and 2,000 IU of ${\alpha}$-tocopheryl acetate (Merck) from 60 days prepartum to 30 days postpartum and 0, 500, 750 and 1,000 IU from 30 to 60 days postpartum, respectively. Blood samples were collected at -60, -45, -30, -15, -7, 0, 7, 15, 30 and 60 days of parturition and were analyzed for plasma ${\alpha}$-tocopherol and total antioxidant activity (TAA). The intake of DM, CP and TDN did not vary among different groups. Plasma ${\alpha}$-tocopherol and TAA around parturition (-7 to 15 day) in $T_3$ and $T_4$ were significantly higher than the control group. There was 17% reduction in retention of fetal membranes (RFM) and metritis in $T_4$ than control. The post partum estrus interval averaged 58.00, 55.33, 51.83 and 43.00 days in $T_1$, $T_2$, $T_3$ and $T_4$ respectively. There was significant reduction in days open in both $T_3$ and $T_4$ in comparison to $T_1$ group (127,130 Vs.146). All the vitamin E supplemented groups showed reduction in days open than their previous lactation performance. Supplementation of vitamin E at $1,500IU\;d^{-1}$ from 60 day prepartum to 30 day post partum to buffaloes exhibited beneficial effect on plasma ${\alpha}$-tocopherol level and TAA around parturition and continuation of its supplementation at $1,000IU\;d^{-1}$ from 30 to 60 days of lactation improved post partum reproductive performance of buffaloes.

The Effects of Green Tea Supplementation on Behavioral Changes, Striatal Dopamine Level, and Hepatic Antioxidant Parameters of Parkinson's Disease Model Rats

  • Green tea has attracted attention with respect to its potential for preventing and treating neurodegenerative disease. The neurotoxin, 6-hydroxydopamine (6-OHDA), was used to produce experimental Parkinson's disease (PD) model. The purpose of this study was to investigate the effects of green tea diet on behavioral changes, striatal dopamine content, and hepatic antioxidant parameters of PD model rats. In this study, we used male Sprague-Dawley rats weighing $200\sim220g$ and injected 6-OHDA into the right substantia nigra and medial forebrain bundle of the brain. The supply of green tea diet was started at 2 weeks before 6-OHDA lesion and continually supplied during 0, 2, and 4 weeks after 6-OHDA lesion (GT-0, GT-2, GT-4). Behavioral disturbance was measured by the stepping and d-amphetamine drug-induced rotation tests. Then, we assayed the striatal dopamine content and the hepatic malondialdehyde (MDA), hydrogen peroxide $(H_2O_2)$, and superoxide dismutase (SOD) activity. The percentage of lesioned forepaw to non-lesioned forepaw step scores was the highest in GT-4 group among all groups at both 3 and 4 weeks after 6-OHDA lesion. At 4 weeks after 6-OHDA lesion, the rotation score was the lowest in GT-2 group (p<0.05). However, increasing rate of the rotation score from 2 to 4 weeks after 6-OHDA lesion was the lowest in GT-4 group. The striatal dopamine content was not significantly different among four groups by green tea diet. The hepatic MDA level was the lowest in GT-4 group among four groups. The hepatic SOD activity was increased with the prolongation of green tea diet period These results suggest that green tea diet affects behavioral changes in rats of PD model. It seems that continuous green tea supplementation has an influence on the reduction of behavioral disturbance and the hepatic MDA level. Accordingly, continuous green tea supplementation was recommended for the prevention and treatment of PD. However, further studies are needed to investigate the mechanisms and efficacy of green tea in PD.

흰다리새우(Litopenaeus vannamei) 사료 내 괭생이모자반(Sargassum horneri) 분말의 이용가능성 평가 (Evaluation of Dietary Supplementation of Sargassum horneri for Pacific White Shrimp Litopenaeus vannamei)

  • This study was conducted to evaluate dietary supplementation of Sargassum horneri (SH) meal on growth performance, feed utilization, innate immunity, antioxidant capacity and disease resistance of Pacific white shrimp. The diets were added with graded levels of SH meal by 0, 0.5, 1, 2, 4 and 8% (designated as Con, SH0.5, SH1, SH2, SH4 and SH8, respectively). Quadruplicate groups of shrimp were hand-fed with one of the diets five times daily for 39 days. The innate immunity and antioxidant capacity of shrimp were significantly improved by the dietary SH supplementation. The cumulative survival of shrimp exposed to Vibrio parahaemolyticus in a challenge was higher in shrimp group fed SH diets, except for SH8, than that of shrimp fed the control diet. Growth performance and feed utilization of the shrimp were significantly decreased with the dietary SH inclusions except for 0.5%. Results indicated that dietary SH meal could be used as a functional supplement for improvements in innate immunity, antioxidant capacity and disease resistance in Pacific white shrimp. The suggested dietary inclusion level of dried SH is approximately 0.5% for the shrimp.

The relationship between reactive oxygen species, DNA fragmentation, and sperm parameters in human sperm using simplified sucrose vitrification with or without triple antioxidant supplementation

  • Objective: This study examined whether the addition of triple antioxidants (3A)-10 µM acetyl-L-carnitine, 10 µM N-acetyl-L-cysteine, and 5 µM α-lipoic acid-in freezing-thawing medium during human sperm cryopreservation using the sucrose vitrification (SuV) and liquid nitrogen vapor (Vapor) techniques could improve post-thaw survival of spermatozoa. Methods: We analyzed 30 samples from healthy human sperm donors. Each sample was allocated into one of five groups: fresh control, SuV, SuV+3A, Vapor, and Vapor+3A. The sperm motility, morphology, viability, intracellular and extracellular reactive oxygen species (ROS) levels, and sperm DNA fragmentation (SDF) were evaluated. Results: The cryopreserved spermatozoa had significantly reduced percentages of motility (p<0.05) and viability (p<0.05). Antioxidant supplementation non-significantly improved these parameters (p>0.05). No significant differences were found in sperm morphology between the fresh and frozen-thawed groups (p>0.05). After freezing, the extracellular ROS levels in the frozen-thawed groups were significantly higher (p<0.05) than in the fresh group. However, we did not find any differences in intracellular ROS parameters among these groups (p>0.05). The SDF was higher in the SuV and Vapor groups than in the fresh group, but without statistical significance (p=0.075 and p=0.077, respectively). Conclusion: Cryopreservation had detrimental effects on sperm motility, viability, and extracellular ROS levels, without changing the morphology or intracellular ROS levels. Antioxidant supplementation was slightly effective in preventing SDF in frozen-thawed spermatozoa.

  • This study investigated the effects of dietary supplementation with Sargassum horneri celluclast hydrolysate (SHCH) on the growth performance, feed utilization, non-specific immunity and antioxidant capacity of the Pacific white shrimp Penaeus vannamei. Six diets were prepared by SHCH supplemention levels of 0, 5, 10, 20, 40 and 80 g/kg (designated as Con, S5, S10, S20, S40 and S80, respectively). A total of 540 shrimp were randomly distributed in triplicate across 18 tanks and fed one of the diets for five weeks. Weight gain did not differ significantly among the groups except for S80. The feed conversion ratio was significantly lower in S10 than in S80. Phenoloxidase and nitro-blue tetrazolium activities were significantly higher in S5 than in Con. The prophenoloxidase gene was significantly upregulated in S10 compared to that in Con. The gene expression of crustin, glutathione peroxidase, lipopolysaccharide and β-1,3-glucan-binding protein were significantly upregulated in S5 compared to Con. The superoxide dismutase gene was significantly upregulated in S20 and S80 compared to Con. Supplementing shrimp diets with up to 8% of SHCH did not impair non-specific immunity or antioxidant capacity. However, based on growth results, supplementation up to 4% was considered appropriate.

Effects of Vitamin C or E on the Pro-inflammatory Cytokines, Heat Shock Protein 70 and Antioxidant Status in Broiler Chicks under Summer Conditions

  • The present study was carried out to investigate the effects of dietary antioxidants on pro-inflammatory cytokines, heat shock protein (HSP) and antioxidant status in broiler chicks under summer conditions. A total of 162, 3-d-old broiler chicks were randomly assigned to a basal diet (CON) and the basal diet supplemented with vitamin C (200 mg/kg diet, VCD) or vitamin E (100 mg/kg, VED) until 35 day of age. All birds were exposed to summer diurnal heat stress at average daily fluctuations of temperature between $32^{\circ}C$ to $34^{\circ}C$ at day to $27^{\circ}C$ to $29^{\circ}C$ at night for the entire feeding periods. There was no significant difference in body weight, feed to gain ratio and the relative organ weight except the thymus in response to dietary vitamin C or E supplementation. However, the mRNA expression of interleukin (IL)-$1{\beta}$, IL-6, interferon (IFN)-${\gamma}$, Toll like receptor (TLR)-4 and HSP70 in the liver of birds fed diet containing vitamin C significantly (p<0.05) decreased compared with those in birds fed basal diet. Dietary vitamin E also showed a significant (p<0.05) decrease in the mRNA expression of IL-6 and HSP70 compared with a basal diet. Total antioxidant status (TAS) in serum of birds fed vitamin C supplemented diet was significantly (p<0.05) higher with than that in birds a basal diet. Lipid peroxidation in serum and liver resulted in a significant (p<0.05) decrease in response to dietary vitamin C or E supplementation. In conclusion, dietary supplementation with antioxidant vitamins, especially vitamin C resulted in a significant decrease in the mRNA expression of pro-inflammatory cytokines and HSP70, and higher antioxidant parameters than that of birds on the basal diet under summer conditions.

Effect of Copper on Plasma Ceruloplasmin and Antioxidant Ability in Broiler Chickens Challenged by Lipopolysaccharide

  • The effects of dietary copper (Cu) supplementation in broiler chickens challenged with a single injection of Salmonella typhymurium lipopolysaccharide (LPS) on the antioxidant capacity and plasma levels of ceruloplasmin (Cp) were evaluated. The broiler chickens were provided with a basal diet or diets supplemented with 8 and 50 mg/kg Cu from 1d of age. At 25d of age, 48 chickens with similar body weight were selected from each diet. Half of the chickens in each dietary treatment were injected intraperitoneally (i.p.) with LPS (1 mg/kg body weight). The other half was injected with saline, serving as the control. Body weight gain and feed consumption were significantly suppressed by LPS challenge during the first 12-h after injection, regardless of dietary Cu levels. Feed efficiency was reduced by LPS injection during the 72-h experimental period. Dietary Cu levels had no significant effect (p>0.05) on the plasma ceruloplasmin concentrations in chickens injected with saline. In contrast, high dietary level of Cu elevated plasma Cp levels in chickens with LPS challenge. Short-term LPS challenge had no significant effect on the antioxidant ability of broilers, as indicated by superoxide dismutase, ferric reducing/antioxidant power and the thiobarbituric acid reacting substances in the plasma. The result suggests that high dietary Cu level (as much as 50 mg/kg supplementation) is favorable for coping with short-term LPS challenge through upregulating plasma Cp levels.
