Comparison of Relative Antioxidant Capacity and Antiperoxidation Activity of Traditional Medicines in Vitro

  • To investigate the antioxidant capacity of traditional Korean medicines, water extracts from 57 species were tested on their antioxidant activity using radical scavenging effects against ABTSㆍ+. Of which, Rhus javanica, Caesalpinia sappan, Rosa rugosa, Spatholobus suberectus, and Magnolia obovata showed strong antioxidant capacities at 10㎍ concentration. Therefore, antioxidant capacities of 5 traditional medicine extracts in the different concentration (1㎍, 5㎍, and 10㎍) were determined. The 5 traditional medicine extracts was detected in antioxidant capacity dose dependently. R. javanica was showed the highest antioxidant capacity, the antioxidant activity at 1 ㎍ of herbal extract being 0.85 mM TE. At the same time, the antiperoxidation effects of these 5 medicines were determined. Lipid peroxidation in brain homogenates induced by NADPH and ADP-Fe/sup 2+/ was strong inhibited by S, suberectus, R. rugosa, and M. obovata. Extract of C. sappan was showed the highest inhibition against lipid peroxidation compared with 4 herbal extracts in vitro. These traditional medicines are a potent antioxidant capacity and antiperoxidation activity, further investigation into the in vivo antioxidant therapeutic potential for treatment of human disorders such as Alzheimer's disease.

Cabbage (Brassica oleracea L. var. capitata) Phytochemicals with Antioxidant and Anti-inflammatory Potential

  • Background: The objective of this study was to investigate antioxidant and anti-inflammatory activity of cabbage phytochemicals. Materials and Methods: Color coordinates were evaluated by colorimetry, and the antioxidant and anti-inflammatory activities were analyzed by spectrophotometer for some common cabbage varieties. Results: Red heads had the highest total antioxidant contents followed by Savoy, Chinese and green heads. The Chinese variety had the highest ABTS (2,2-azino-di-(3-ethylbenzthiazoline-sulfonic acid) antioxidant activity, was 5.72 ${\mu}mol$ TE/g fw (Trolox equivalent). The green variety had the highest DPPH (free radical scavenging activity) antioxidant activity, which was 91.2 ${\mu}mol$ TE/g fw. The red variety had the highest FRAP (ferric reducing antioxidant power) antioxidant activity, which was 80.8 ${\mu}mol$ TE/g fw. The total phenol amounts were 17.2-32.6 mM trolox equivalent antioxidant capacity (TEAC) and the total flavonoid amounts were 40.0-74.2 mg quercetin per gram. Methanolic extracts of different cabbage heads showed different anti-inflammatory activity values. Chinese, Savoy and green heads had the highest anti-inflammatory activity, while red heads had the lowest. Conclusions: The results suggest that these varieties of cabbage heads could contribute as sources of important antioxidant and anti-inflammatory related to the prevention of chronic diseases associated to oxidative stress, such as in cancer and coronary artery disease.

Antioxidant Effects of 2,3,6-tribromo-4,5-dihydroxybenzyl Methyl Ether (TDB) from the Red Alga, Symphyocladia latiuscula

  • 2,3,6-Tribromo-4,5-dihydroxybenzyl methyl ether (TDB) from the methanolic extract of the red alga Symphyocladia latiuscula exhibits major antioxidant activity. In this study, the activity of TDB against oxidative damage in deoxyribose and DNA was investigated in vitro for potential applications in preventing mutagenesis caused by DNA damage. TDB inhibited the oxidation of deoxyribose at concentrations of up to $1{\mu}g$/mL in the presence of $Fe^{+3}$-EDTA/$H_2O_2$. Furthermore, TDB showed no pro oxidant activity as determined by absence of the reduction of bleomycin-$Fe^{+3}$ to bleomycin-$Fe^{+2}$, which leads to DNA damage. Based on these results, TDB demonstrated considerable antioxidant activity without prooxidant properties.

인진쑥 추출물의 항산화 및 항암 활성 (Antioxidant and Anticancer Activities of Extract from Artemisia capillaries)

  • Artemisia capillaries is a major important food and medicinal resource in Korea. In order to confirm the biological activities of Artemisia capillaries, we investigated antioxidant and anticancer activities from in vitro assays. The Artemisia capillaries methanol (MeOH) extracts was used for the evaluation of DPPH scavenging, total phenolic content, total flavonoid content, hydroxyl radical (${\bullet}OH$) scavenging, reducing power assay as antioxidant activity, as well as anticancer activities as MTT assay. As a result, the Artemisia capillaries MeOH extracts showed potent antioxidative activity and anticancer activity in vitro. These results suggest that the Artemisia capillaries MeOH extracts have a potential alleviated oxidation process, cell motility activity, and tumorigenesis.

Antioxidative Activity on Human Low Density Lipoprotein(LDL) Oxidation by 2,6-Dimethoxyphenol Purified from Bacillus sp. KS-96

  • This study was designed to develope the antioxidative activity on oxidation of human low density lipoprotein(LDL) from marine microbials. Bacillus KS-96 producign antioxidant have been isolated and identified from seawater, Bacillus sp. KS-96. The optimal medium pH was 7.0 and incubation temperature was 30$^{\circ}C$. The antiosidant of potential substance produced extracellularly in the culture broth by Bacillus sp. KS-96 was obtained by elution of silica gel culumn chromatography with hexane, ethylacetate and water. The ethylacetate faction are shown at highest level of antioxidant activity using thiocyanate method among them. By IR, NMR, and GC/MS, antioxidant purified from ehtylacetate fraction was identified and named as 2,6-dimethoxyphenol. 2,6-dimethoxyphenol inhibited the metal mediated oxidation of human LDL at concentration of 50∼100 ${\mu}$g/mL in the presence of 5uM CuSO4 with macrophage or J774 cells.

The antioxidant and cytotoxic activities of Sonchus oleraceus L. extracts

  • This study investigated in vitro antioxidant activity of Sonchus oleraceus L. by extraction solvent, which were examined by reducing power, hydroxyl radical-scavenging activity(HRSA) and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assays. 70% MeOH extract had the greatest reducing power while EtOH extract had the greatest HRSA. The antioxidant activity of S. oleraceus extracts was concentration dependent and its $IC_{50}$ values ranged from 47.1 to $210.5\;{\mu}g/ml$ and $IC_{50}$ of 70% MeOH, boiling water and 70% EtOH extracts were 47.1, 52.7 and $56.5\;{\mu}g/ml$, respectively. 70% MeOH extract of S. oleraceus contained the greatest amount of both phenolic and flavonoid contents. The extracts tested had greater nitrite scavenging effects at lower pH conditions. The cytotoxic activity showed that EtOH extract had the best activity against the growth of stomach cancer cell. These results suggest that S. oleraceus extract could be used as a potential source of natural antioxidants.

Antioxidant Constituents from Leonurus japonicus

  • Two phenolic acids, gallic acid (1) and syringic acid (2), and five flavonoids, apigenin (3), luteolin (4), kaempferol (5), quercetin (6), and myricetin (7), were isolated from the aerial parts of Leonurus japonicus. Their structures were elucidated by chemical and spectral analysis. The antioxidant activities of the crude extracts, partitioned fractions and isolated compounds were evaluated by DDPH free radical-scavenging assay. Results suggested that the EtOAc partitioned fraction and compounds 1, 4, 5, 6, and 7 showed significantly high antioxidant potential compared with $\alpha-tocopherol$ and BHT, which were used as controls.

In vitro anti oxidant activity of methanol extract of Clerodendrum infortunatum Linn

  • The antioxidant potency of methanolic extract Clerodendrum infortunatum Linn. (MECI), which are widely used in the Indian indigenous system of medicine for different purposes, was studied. The antioxidant potential was evaluated using different established in vitro antioxidant tests viz. determination of total amount of polyphenolics compounds, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging, nitric oxide scavenging, superoxide anion radical scavenging, hydroxyl radical scavenging and reductive power assay. It was found that MECI contain a high amount of polyphenolics and possesses significant free radical scavenging activity in all the assay. The higher activity was may be due to presence of richest amount of polyphenolics and flavonoids in it.

Isolation of Luteolin 7-O-rutinoside and Esculetin with Potential Antioxidant Activity from the Aerial Parts of Artemisia montana

  • The antioxidant activity of Artemisia montana was determined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and inhibitory activity against free radical generation of hepatocytes ($AC_2$F). The methanol extract of A. montana showed strong radical scavenging activity at a concentration of $10.1{\mu}g/m{\ell}$, and thus fractionated by solvent extraction. Esculetin and luteolin 7-O-rutinoside (scolymoside) were isolated as the active principles from the EtOAc and interphase fractions, respectively. The antioxidant activity of these compounds were comparable to that of L-ascorbic acid.

Antioxidant activity of 3,5-dicaffeoyl-epi-quinic acid (DEQA) from the halophyte Atriplex gmelinii

  • In this study, the antioxidizing effect of 2,3-dicaffeoyl-epi-quinic acid (DEQA) was investigated. The antioxidant activity was evaluated by measuring the scavenging effect on DPPH radical and peroxynitrite and the reducing power on ferric ion. DEQA showed a scavenging effect and reducing power comparable to vitamin C used as a positive control. Also, DEQA effectively inhibited production of intracellular reactive oxygen species (ROS) in HT-1080 cells, showing the scavenging ratio of 43.8% even at 10 µM concentration of DEQA after 2 hours in HT-1080 treated with H2O2. In addition to this, DEQA inhibited the production of nitric oxide (NO) very effectively in Raw 264.7 cells. The above results suggest that DEQA has the potential to be developed as a natural antioxidant.
