Antimicrobial Characteristics of Scutellariae Radix Extract (황금추출물의 항균특성)

  • It was carried out for research and development of natural antimicrobial on Scutellariae Radix extract against food-borne infection bacteria .Scutellariae Radix extract showed remarkable antimicrobial activites against Bacillus cereus, Listeria monocytogenes, Escherichia coli, and Vivro parahaemolyticus when examined by disk method, it was very stable on the wide rane of temperature and pH,.The growth rates of Bacillus cereus, Listeria monocytogenes, Escherichia coli, and Vivro parahaemolyticus were decreased at the concentration of more than 500 ppm Scutellariae Radix extract, Indicating that the minimum inhibitory concentration (MIC)of the Bacillus cereus, Listeria monocytogenes, Escherichia coli, and Vivro parahaemolyticus to Scutellariae Radix extract were around 500 ppm . The morphological changes were observed by transmission electron microscope and scanning electron microscope and the microbial cells membran was destroyed by Scutellariae Radix extract. It was identified that the membrane integrity of the sensitive cells was disrupted by exposure to Scutellariae Radix extract as the $\beta$-galactosidase test on experimental substrate ONPG(o-nitrophenyl-$\beta$-D-galacto-pyranoside)

Effects of Water Extract from Omija (Schizandra chinensis Baillon) on Nabak Kimchi Preservation (오미자 물추출물이 나박김치의 저장성에 미치는 영향)

  • In order to investigate the possible use of omija extract as natural preservatives for nabak kimchi omija extract was tested for antioxidation and electron-donating ability and further more antimicrobial activites against lactic acid bacteria of nabak kimchi. The concentration of the test sample used were 0.5, 1.0, 1.5, 2.0%. Antioxidative activity measured the TBA value, when omija extract concentration % is higher the antioxidation effect were more evident with the increased omija extract concentrations, and showed a high electron donating activity more than 1.0%. As the result of isolated lactic acid bacteria from nabak kimchi the primary separation tool 117 strains, of these 4 strains which had excellent growth and a strong acid formation capability was selected the second time and Lactobacillus brevis, Lactococcus faecalis, Leuconostoc mesenteriodes, Lactobacillus palantarum were identified. After measuring the antimicrobial activity of the four lactid acid bacteris, except Lactobacillus brevis the other three bacteria showed strong antimicrobial activities. The results suggest the possible use of the omija extract as natural preservative for nabak kimchi.

Antimicrobial Effects of Essential Oils for Multidrug-Resistant Acinetobacter baumanii (다제내성 아시네토박터 바우마니의 에센셜 오일에 대한 항균효과)

  • Acinetobacter baumannii is categorized as a red alert pathogen that is increasingly associated with a high mortality rate in infected patients because of its resistance to extensive antibiotics. This study evaluated the antibacterial activities of some essential oils (tee tree, rosemary, and lavender oils) against 18 clinical isolates of multidrug-resistant A. baumannii (MRAB). The carbapenemase screening Hodge test showed that all 20 strains of A. baumannii were resistant to imipenem. The identification of multidrug-resistant microbes was carried out using the VITEK system. The antimicrobial activity of essential oils was tested by a disk diffusion method against MRAB. In the disk diffusion method, tea tree showed the largest increase in inhibition size compared to lavender oil, and rosemary had no antibacterial effect. These results proved the antimicrobial effect of multidrug resistance A. baumannii. Tee tree oil would be a useful alternative natural product for the treatment and prevention of most common human pathogens and MRAB infections. This is expected to be used as an antimicrobial agent, such as hand disinfectant using natural essential oil in the future.

Isolation and Antimicrobial Susceptibility of Nontuberculous Mycobacteria in a Tertiary Hospital in Korea, 2016 to 2020

  • Background: There is a global increase in isolation of nontuberculous mycobacteria (NTM). The aim of the study was to analyze longitudinal trends of NTM identification and pattern of antimicrobial susceptibility testing. Methods: NTM recovery rates, distribution of NTM species identification, and antimicrobial susceptibility pattern of NTM at Pusan National University Yangsan Hospital between January 2016 and December 2020 were retrospectively analyzed. Results: A total of 52,456 specimens from 21,264 patients were submitted for mycobacterial culture, of which 2,521 from 1,410 patients were NTM positive over five years (January 2016 to December 2020). NTM isolation showed an increasing trend from 2016 to 2020 (p<0.001, test for trend) mainly caused by Mycobacterium avium complex. The vast majority of M. avium complex were susceptible to key agents clarithromycin and amikacin. For Mycobacterium kansasii, resistance to rifampin and clarithromycin is rare. Amikacin was the most effective drug against Mycobacterium abscessus subspecies abscessus and Mycobacterium subspecies massiliense. Most of M. subspecies massiliense were susceptible to clarithromycin, while the majority of M. abscessus subspecies abscessus were resistant to clarithromycin (p<0.001). Conclusion: There was an increasing trend of NTM isolation in our hospital. Resistance to key drugs was uncommon for most NTM species except for M. abscessus subspecies abscessus against clarithromycin.

Study on antimicrobial resistance and detection of MRSA of Staphylococcus aureus isolated from carcass in Daegu slaughterhouses (대구지역 도축장 출하가축의 도체에서 분리한 Staphylococcus aureus의 항생제 내성과 MRSA 검출에 관한 연구)

  • At the present study, it was aimed to explore the states of antimicrobial resistant Staphylococcus aureus isolates from 1,360 chickens, pigs and cattle carcass (400 chickens, 480 pigs and 480 cattle) in Daegu province from January 2022 to December 2022. Among 1,360 samples, 81 of S. aureus were isolated cattle (1.4%), pigs (7.7%) and chickens (9.2%). In antimicrobial susceptibility test, all of the isolates were demonstrated susceptibility to rifampin. But the isolates were showed resistance other antibiotics in order of tetracycline (62.9%), ciprofloxacin (62.9%), tobramycin (58.0%), gentamicin (51.8%), amikacin (40.7%), penicillin (39.5%), clindamycin (35.8%), enrofloxacin (33.3%), trimethoprim/sulfamethoxazole (30.8%), oxacillin (30.8%), minocycline (29.6%), erythromycin (25.9%), quinupristin/dalfopristin (20.9%), chloramphenicol (12.3%), cefoxitin (9.8%). Among the 81 S. aureus isolates, 25 (30.8%) methicillin-resistant staphylococcus aureus (MRSA) were observed. Seven (28.0%) of 25 MRSA harbored mecA gene. About 96% of MRSA were multidrug resistance to at least 3 more drugs. A continuous monitoring and surveillance program to prevent antimicrobial resistance in livestock products is demanded.

  • A series of 1,4-disubstituted octahydroquinoxaline-2,3-dione derivatives was prepared through two steps reaction. The latter involves the formation of N,N-disubstituted cyclohexane-1,2-diamine derivatives (la-j) through reductive alkylation of 1,2-cyclohexanediamine with different aldehydes in presence of sodium cyanoborohydride. Fusion of compounds (1a-j) with diethyl oxalate affording the target compounds (2a-j). Elucidation of structures of compounds (2a-j) was based upon different spectral data as well as the elemental methods of analyses. In addition, mass spectrometry and X-ray diffraction analyses were carried out. Moreover, the lipophilicity of the target compounds as expressed from the Clog P. Most of the test compounds (2a-j) showed weak to moderate antibacterial and antifungal activities against most of the used bacterial and fungal strains in comparison to chloramphenicol and clotrimazole as reference drugs respectively.

  • The antibacterial and antifungal effect of grapefruit seed extract(GFSE) was investigated for its purpose of application to a diverse spectrum of field as sanitizers, disinfectants and preservatives. GFSE showed coparatively high content of such flavoniods as naringin and hesperidin and ascorbic acid. GFSE containing a low level of organic acids is a heavy viscous and water-soluble liquid. As a result of the antimicrobial test of GFSE, Bacillus subtilis and Aspergillus oryzae did not survive at detectable levels when treated with more than 100 ppm of GFSE. The minimum inhibitory concentrations of GFSE for a wide variety of pathogenic and putrefactive bacteria, fungi and yeasts were 100 ppm and 250 ppm, respectively. In the comparable electron micrograph of microbial cells treated with GFSE or not, we could conclude that GFSE destroy microorganisms by disrupting the functions of the cell wall membrane and microbial spores.

  • The antimicrobial action of various dental cements evaluated against common micro-organisms most frequenty found within the components of the normal bacterial flora of oral cavity. They include Streptococcus mutans (2 strains), Lactobacillus acidophilus, Actinomyces viscosus, and Streptococcus sanguis. The test was done by the use of brain heart infusion (BHI) agar plates. A standard mix of each cement was made and placed on the plates which were seeded with a standard culture of microorganisms. After incubation, the halo of bacterial growth inhibition around the cement was identified and its size was measured. Some of the cements tested had obvious antibacterial effect. The cements listed in decreasing order of effectiveness are 1) zinc phosphate and oxyphosphate, 2) silicate, 3) zinc oxide-eugenol, 4) calcium hydroxide, and 5) carboxylate.

  • 1.2-bis(aminoacyl)hydrazine derivatives and dipeptides were synthesized by conventional peptide synthesis procedures. Their antioxidant activity were inverstigated by over-storage test using corn oil as substrte. 1.2-bis (aminoacyl) hydrazine derivatives and dipetides containing hydrophobic side chain amino acid showed higher antioxidant activity. A free N-terminal amino group was also found to be important for the appearance of antioxidant activity. 1.2-bis (aminoacyl) hydrazine derivatives showed higher antioxidant activity than dipeptides. Antimicrobial activites of dipeptides and 1.2-bis (aminoacyl) hydrazine derivatives were also examined by the paper disc method. All of these compounds had shown no antimicrobial activity.

Antimicrobial Resistance Patterns and Resistance genes assay of Shigella sonnei Isolated in Korea for Five Years (최근 5년 동안 국내에서 분리된 Shigella sonnei의 항균제 내성 유형과 내성유전자형 분석)

  • This study has been carried out for investigating the relatedness of representative 135 Shigella sonnei strains isolated from 2000 to 2004 by using biotyping and antimicrobial resistance. All strains showed typical biochemical characterisics of Shigella strain. Among 135 strains,79 (58.5%) strains were biotype "g",54 (40.0%) strains were biotype "a" and 2 (1.5%) strains were biotype "e". The results of susceptibility test against 16 antimicrobial agents were like this. Most of strains were susceptible to AN, CIP, C and GM. 129 (95.6%) strains were resistant to SXT, 126 (93.3%) strains were resistant to TE and 122 (90.4%) strains were resistant to SM. One hundred thirty two (97.8%) strains were resistant to more than two antimicrobial agents. R28 type (antimicrobial resistance patterns 28: resistant to AM, SAM, TE, TIC, SXT, K, SM and AmC) were 42 strains (31.1%). The other strains were showed 33 kinds of R patterns. The results of $bla_{TEM}$, sulII, tetA and strA gene detection were coincided with phenotype of antimicrobial resistance by disk diffusion method. But some strains which had sulII and strA genes were not showed the resistance against SXT and SM.
