김치 발효에 관여하는 젖산균에서의 Bacteriocin의 검색 (Dectection of the Bacteriocin from Lactic Acid Bacteria Involved in Kimchi Fermentation)

  • Lactic acid bacteria in Kimchi fermentation were tested for inhibitory activity against Gram positive bacteria and Gram negative bacteria. The Lactobacillus brevis (KCCM 35464) was found to produce a antimicrobial substance. It showed relatively wide range of inhibition spectrum against gram positive and gram negative bacteria and maintained the inhibitory activity between pH 4.0 and pH 9.0. The antimicrobial substance was obtained in the stationary growth phase and was purified by gel chromatography. The inhibitory effect of the antimicrobial substance on sensitive bacterial strains was determined by filter paper test. The activity of antimicrobial substa- nce was stable at 75$\circ$C. On the basis of its electrophoretic pattern is SDS-PAGE, antimicrobial substance appeared as a single band of 59 KDalton.

오매의 에틸아세테이트 추출물이 Staphylococcus aureus의 증식에 미치는 영향 (Effect of Ethyl Acetate Extract of the Dried Prunus mume on the Growth of Staphylococcus aureus)

  • The dried Prunus mume, an alkaline food abundant in organic acids (citric acid, malic acid and tartaric acid), has been largely used in both folklore remedies and Chinese herbal medicine for a long time. This study was performed to investigate the antimicrobial activity of the dried Prunus mume. The fractionation of the methanol extracts from Prunus mume was conducted using petroleum ether, chloroform, ethyl acetate and butanol, respectively. The antimicrobial activity of the Prunus mume extracts was then determined against food-borne pathogens using a paper disc method. The ethyl acetate extract showed the strongest antimicrobial activity against the eigth food-born pathogens used in this present study. Diaion HP 20 column chromatography was performed to remove some sugars that might inhibit the antimicrobial activity of Prunus mume. The strongest antimicrobial activity of ethyl acetate fraction of Prunus mume was shown against Staphylococus aureus. The growth inhibition curve was determined using ethyl acetate extracts of Prunus mume against Staphylococus aureus, which showed the growth inhibition up to 72 hours at 1,000 ppm concentration.

초피추출물의 항균특성 (Antimicrobial Activities of Chopi(Zanthoxylum piperitum DC.) Extract)

  • In order to evaluate the antimicrobial function of natural herb extracts as antimicrobial agent or packaging material for the preservation of foods and greenhouse produce, the water extract of chopi (Zanthoxylum piperitum DC.) was prepared and its antimicobial activity was determined. In the paper disk test its antimicrobial activity was increased in proportion to its concentraion. The growth of microorganisms was completely inhibited above 500ppm of its concentration. It showed wide spectrum of thermal(40 to 180oC) and pH(4 to 10) stabilities. In the electronic microscopic observation(TEM and SEM) of microbial morphological change it showed to decrease the activation of physiological enzymes and to lose the function of cell membranes. Even in the activation test of galactosidase, it seemed to weaken the osmotic function of cell membranes remarkably in comparison with chloroform and its activation corresponded to 40~50% of toluene. Zanthoxylum piperitum DC. extract seemed to be an excellent antimicrobial for the inhibition of food borne microorganisms as well as the pre servation of greenhouse produces.

Antimicrobial Effect of Portulaca oleracea Extracts on Food-Borne Pathogens

  • This study was performed to investigate the antimicrobial effects of Portulaca oleracea extracts against food-borne pathogens. First, the Portulaca oleracea was extracted with methanol at room temperature, and then further fractionated by using petroleum ether, chloroform, ethyl acetate and methanol, respectively. The antimicrobial activity of the Portulaca oleracea extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of Portulaca oleracea showed the highest antimicrobial activity against Staphylococcus aureus and Shigella dysenteriae. There was also a synergistic effect of the combined extracts of Portulaca oleracea and Indigofera kirilowii as compared to each extract alone. Finally, the growth inhibition curve of ethyl acetate extracts of Portulaca oleracea against Staph­ylococcus aureus and Shigella dysenteriae was determined The ethyl acetate extract of Portulaca oleracea showed strong antimicrobial activity against Staphylococcus aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate extract from Portulaca oleracea, retarded the growth of S. aureus by more than 24 hand Shigella dysenteriae up to 12 h at $37^{\circ}C$.

Antimicrobial Effect of Sophora angustifolia Extracts on Food-Borne Pathogens

  • This study was performed to investigate the antimicrobial effect of Sophora angustifolia extracts against food-borne pathogens. First, Sophora angustifolia was extracted with methanol at room temperature, and the methanol extracts from Sophora angustifolia were fractionated using petroleum ether, chloroform, ethyl acetate, and methanol. The antimicrobial activity of the Sophora angustifolia extracts was determined using the paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Sophora angustifolia showed the highest antimicrobial activity against Staphylococcus aureus and Salmonella typhimurium. A synergistic effect was found in the combined extracts of Sophora angustifolia and Portulaca oleracea, compared to the activity of each extract alone. Finally, the growth inhibition curve was determined using the methanol extracts of Sophora angustifolia against Staphylococcus aureus and Salmonella typhimurium. The methanol extract of Sophora angustifolia showed strong antimicrobial activity against Staphylococcus aureus at a concentration of 5,000 ppm. The 5,000 ppm methanol extract from Sophora angustifolia retarded the growth of S. aureus for more than 24 hours and of Salmonella typhimurium for up to 12 hours.

  • 다양한 미생물에 대하여 높은 불활성화 성능을 지니고 있는 은(Ag)은 최근 환경 기술 분야, 나노 기술 분야 등에서 응용 가능성이 높아 큰 주목을 받고 있으며, 새로운 적용 제품들이 활발하게 연구, 개발되고 있다. 하지만 다양한 응용 연구에도 불구하고 정확한 항균 성능 및 기작에 대한 연구 결과와 이해가 부족하여 관련 연구자와 소비자들에게 논쟁과 혼동을 야기시키고 있다. 본 글에서는 기존 연구를 중심으로 은의 미생물 불활성화 성능과 기작, 다른 항균 물질과의 시너지 효과, 응용 분야 등에 대해서 정리, 검토하여 이에 대한 연구 및 개발에 도움이 되고자 한다.

  • The antimicrobial effects of silver (Ag) ion or salts are well known. Recently, silver nanoparticle is attracting an interest in a wide variety of fields since it has been known to be safe and effective as an antimicrobial agent against a broad spectrum of microorganisms. Although silver nanoparticle has been applied to various kinds of products owing to its potent antimicrobial activity, the effects of silver nanoparticle on microorganisms and antimicrobial mechanism have not been revealed clearly. In this paper, we summarized the characteristics, antimicrobial activities and mechanisms, cytotoxicity and applicability of silver nanoparticle.

목초액의 향균 및 DPPH 라디칼 소거 활성에 관한 연구 (Study of Antimicrobial and DPPH Radical Scavenger Activity of Wood Vinegar)

  • The antimicrobial and antioxidative activities were investigated to confirm the utility of wood vinegar. Antimicrobial activity was performed by paper disc method and liquid culture. The growth inhibition was observed in all microbial species at a dose of as low as 25 $\mu$m of wood vinegar by paper disc method. Also, in liquid culture, S. cerevisiae and P. aeruginosa were more inhibited the growth than others in the concentration of $2\%$ (v/v). For measuring of antioxidative activity, wood vinegar was fractionated with acidic, phenolic, basic and neutral fraction; and their antioxidant activities were measured by the radical scavenging effect on DPPH radical. In four fractions, phenolic fraction showed high antioxidative activity.

천연 물질의 항균 효과 (The effect of Natural Antimicrobial Agent)

  • 천연 물질로 지용성인 뉴질랜드산 차나무 정유(Manuka oil), 그 구성 성분인 $\alpha$-pinene, Oxy’less clear, 수용성인 자몽 종자 추출물(Citrex) 및 그 구성성분인 R-limonene의 항균력 을 측정 평가하여 항균력을 보다 향상시키고 광범위하게 사용할 수 있는 방부제의 처방을 제안하였다. 항균력 측정에는 disk diffusion법이 사용되었으며, 균주로는 그람 음성균인 Escherichia coli와 그람 양성균인 Staphylococcus aureus을 사용하였다. Manuka oil과 $\alpha$-pinene은 항균력이 10${\mu}\ell$/paper disk일 때에는 큰 차이가 없었으나 2${\mu}\ell$/paper disk이하의 소량의 시료에서는 Manuka oil이 $\alpha$-pinene에 비해 월등하게 우수하였다. Citrex가 R-limonene보다 두 균주에 대하여 항균력이 모든 농도에서 우수한 성능을 보였다 Oxy’less clear은 두 균주에 대하여 모든 농도에서 파라벤류 propyl para hydroxybenzoate(PPHB)나 R-limonene보다는 항균효과가 우수하며 $\alpha$-pinene과는 비슷하게 나타났고, Manuka oil과 Citrex보다 약간 낮게 나타났다. 이상을 종합해 본 결과 Manuka oil과 Citrex가 다른 물질들에 비해 항균력이 가장 우수한 것으로 나타났다. 항균력이 우수한 지용성 물질 Manuka oil과 확산도가 큰 수용성 물질 Citrex를 적절히 혼합하여 항균력을 향상시키고 광범위하게 사용할 수 있는 이상적인 혼합비(Manuka oil : Citrex)를 구하였는데 Escherichia coli에 대해서는 75 : 25이었고 Staphylococcusureus에 대해서는 25 : 75 이었다.

택사 추출물의 항균 및 항산화 효과 (Antimicrobial and Antioxidative Effects of Alismatis Rhizoma Extract)

  • 택사(Alismatis Rhizoma)는 중국에서 많이 생산되고, 우리나라에서도 그 생산량의 50%이상이 전남 순천지방에서 생산되고 있는데, 옛부터 생약으로 많이 사용되어 왔으며, 최근 택사분말은 무좀에 도포약으로 쓰이고 있어 택사에는 다양한 생리활성이 있는 것으로 생각된다. 따라서 본 연구에서는 추출용매에 따른 택사 추출물에 대한 항균 및 항산화성에 대하여 조사하였다. Paper disc법에 의한 택사 추출물의 항균 test결과 물추출물에서는 거의 모든 시험균주에서 항균활성이 나타났으나, 메탄올 및 에탄올 추출에서는 그 활성이 작게 나타났으며, 시험균주중 Vibrio parahaemolyticus균에 가장 강한 항균활성을 나타내었다. Linoleic acid에 대한 항산화력의 조사에서 택사 물추출물 100$\mu{l}$첨가시 메탄올 및 에탄올 추출물은 항균효과 나타나지 않았고, 물추출물은 효과가 나타났으나 0.1% BHT첨가시 보다는 그 효과가 적었다.
