Antimicrobial, Antioxidant and Cytotoxic Activities of Dendropanax morbifera Léveille extract for mouthwash and denture cleaning solution

  • PURPOSE. The purpose of this study was to analyze the antimicrobial, antioxidant activity and cytotoxicity of Dendropanax morbifera $L{\acute{e}}veille$ extract for assessing whether Dendropanax morbifera $L{\acute{e}}veille$ can be used for the development of natural mouthwash and denture cleaning solution. MATERIALS AND METHODS. The extract was obtained from branches of Dendropanax morbifera $L{\acute{e}}veille$. The solvent fractions were acquired by fractionating Dendropanax morbifera $L{\acute{e}}veille$ extract using n-hexane, ethyl acetate, chloroform and butanol solvent. Paper disc test was used to evaluate the antimicrobial and antifungal activity of Dendropanax morbifera $L{\acute{e}}veille$ extract and solvent fractions against Streptococcus mutans and Candida albicans. The analysis of antioxidant activity was carried out through DPPH radical scavenging assay. The cytotoxicity of Dendropanax morbifera $L{\acute{e}}veille$ extract was analyzed through MTT assay using normal human oral keratinocytes. RESULTS. Dendropanax morbifera $L{\acute{e}}veille$ extract showed antimicrobial activity against Streptococcus mutans and especially Candida albicans. The solvent fractions of Dendropanax morbifera $L{\acute{e}}veille$ showed strong antimicrobial activity against Streptococcus mutans and Candida albicans in n-hexane and butanol solvent fraction, respectively. Dendropanax morbifera $L{\acute{e}}veille$ extract also showed outstanding antioxidant activity. Butanol, ethyl acetate, and chloroform solvent fraction of Dendropanax morbifera $L{\acute{e}}veille$ tended to have increased antioxidant activity as the concentration increased. Dendropanax morbifera $L{\acute{e}}veille$ extract showed high cell survival rate in cytotoxicity test. CONCLUSION. Dendropanax morbifera $L{\acute{e}}veille$ extract turned out to have antimicrobial, antioxidant activity and cytophilicity. Based on these results, it is expected that Dendropanax morbifera $L{\acute{e}}veille$ is applicable as an ingredient for natural mouthwash and denture cleanser.

Antimicrobial and Cytotoxic Activities of Some Malaysian Flowering Plants

  • A total of 43 extracts from 21 species of Malaysian flowering plants were screened for antimicrobial and cytotoxic activities. Antimicrobial activity was tested against fungi, including yeast and candida, as well as Gram-positive and Gram-negative bacteria, and cytotoxicity was assayed using the CEM-SS and HT-29 cell lines. The methanol extracts of the roots and stems of Plumeria acutifolia showed the highest antimicrobial activity, i.e. against Saccharomyces cerevisiae and Candida lipolytica, and mostly moderate activity against the other microbes such as Aspergillus ochraceous and Saccharomyces lipolytica. Phyllanthus emblica also showed moderate activity especially that of the methanol extracts. The methanol extract of the roots of Plumeria acutifolia showed the strongest cytotoxic activity $(CD_{50}=\;3\;{\mu}g/ml)$. Most of Zingiberaceae species gave negative results for antimicrobial activity and showed low cytotoxic activity.

Antimicrobial activity and cytotoxicity of Eclipta prostrata

  • The plant Eclipta prostrata, a member of the Compositae family, has folkloric reputation of being used as a medicinal agent in Bangladesh. In the present investigation, attempt was taken to explore the antimicrobial potency and cytotoxicity of its extractives and purified compounds. The methanolic extract of the whole plant, its n-hexane, carbon tetrachloride, chloroform, aqueous soluble fractions and two purified compounds, eclalbasaponin I (1) and II (2), obtained from Eclipta prostrata were subjected to screening for inhibition of microbial growth by the disc diffusion method at 300 and 100 ${\mu}g$/disc for extracts and pure compounds, respectively. In this case, the carbon tetrachloride and chloroform soluble fractions of the methanolic extract appeared very potent in terms of both zone of inhibition and spectrum of activity. However, all the extractives were also subjected to brine shrimp lethality bioassay for preliminary cytotoxicity evaluation. Here, the carbon tetrachloride soluble fraction of methanolic extract revealed the strongest cytotoxicity having $LC_{50}$ of 1.318 ${\mu}g$/ml.

국내산 주요허브 열수 추출물의 항산화, 항균활성 및 세포독성 (Antioxidant, Antimicrobial Acitivities and Cytotoxicity of Hot Water Extracts of Major Herbs in Korea)

  • 본 연구는 10종 허브 추출물에 대한 DPPH 자유라디칼 소거능, 항균효과 및 세포독성에 미치는 영향을 조사함으로써 항생제 대체제로서 활용 가능성을 위한 기초 자료로 제시하고자 수행하였다. 10종의 허브 추출물의 DPPH 자유라디칼 소거능은 로즈마리, 오레가노 및 캐모마일이 각 49.9, 48.9, 47.7%으로 유의적으로 가장 높았다. 허브 추출물의 E. coli K99와 S. typhimurium에 대한 항균활성은 베르가못, 오레가노, 타임과 로즈마리가 두 균주에 공통적으로 활성이 높게 측정되었으며, 활성이 나타나는 농도에서의 세포독성이 비교적 낮아 안전한 것으로 관찰되었다. 본 연구 결과 유기 축산을 위한 가축 내 천연 항생제 대체제로 항균 및 항산화 활성이 높고, 세포독성이 낮게 나타난 오레가노와 로즈마리가 천연물질로 활용 가능할 것으로 판단되며, 다양한 추출 조건으로 최적 추출 효율을 검토하여 추가적인 생리활성물질 검정과 해당 물질에 대한 연구가 필요하다.

Antimicrobial activity and toxicity of Quisqualis indica

  • The plant Quisqualis indica (Compositae) has ehnopharmcological reputation of being used as a healing agent in Bangladesh. In this study, preliminary screenings were conducted to look at the antimicrobial susceptibility and cytotoxicity of the plant extract. The extractives of the plant were subjected to screening for inhibition of microbial growth by the disc diffusion method. The zones of inhibition demonstrated by the n-hexane, carbon tetrachloride, chloroform and aqueous soluble partitionates of the methanolic extract ranged from 8 - 15 mm, 8 - 18 mm, 12 - 20 mm and 10 - 16 mm, respectively at a concentration of 400 ${\mu}g$/disc. All the extractives were also subjected to brine shrimp lethality bioassay for primary cytotoxicity evaluation. Here, the carbon tetrachloride soluble materials demonstrated the highest cytotoxicity with $LC_{50}$ of 0.826 ${\mu}g$/ml, while n-hexane, chloroform and aqueous soluble partitionates of the methanolic extract revealed the $LC_{50}$ of 1.254, 3.866 and 5.366 ${\mu}g$/ml, respectively. This is the first report of the antimicrobial activity and cytotoxicity Q. indica.

Magnolol과 Honokiol이 항균, 교원질 분해효소, 세포독성 및 Cytokine생산에 미치는 영향 (THE EFFECTS OF HONOKIOL AND MAGNOLOL ON THE ANTIMICROBIAL, BACTERIAL COLLAGENASE ACTIVITY, CYTOTOXICITY AND CYTOKINE PRODUCTION)

  • The oral microbiota such as P. gingivalis, P. intermedia and A. actinomycetemcomitans play a primary role in the initiation and progression of the periodontal disease. The purpose of this study was to evaluate the antimicrobial effects and inhibitory effects of honokiol and magnolol on the bacterial collagenase activity, cytotoxicity and cytokine production of periodontopathic microorganisms. The antimicrobial activities of honokiol and magnolol was evaluted with minimum inhibition concentration. Honokiol was more active than magnolol, but less than chlorhexidine on antimicrobial activity. The inhibitory effects of magnolol and honokiol on the collagenolytic activity and cytotoxicity were evaluated using a Collagenokit CLN-100 and rapid colorimetric assay (MTT method) for cellular growth and survival of gingival fibroblast and periodontalligament cell and $[^3H]-thymidine$ incorporation for the gingival epithelial cell. The inhibitory effects on the collagenolytic activity was the highest in chlorhexidine, and the lowest in magnolol. Magnolol had the lowest cytotoxic effect and chlorhexidine had the highest. The inhibitory effects on cytokine production was evaluated using $interleukin-1{\beta}$ ELISA kit (Cistron Biotech.), IL-6, $TNF-{\alpha}$ ELISA kit (Genzyme) and inhibitory effects were higher than bacterial LPS and there is no difference among the honokiol, magnolol and chlorhexidine. From these results, the antimicrobial and antienzymatic activities of honokiol and magnolol were seemed to inhibit bacterial growth and enzyme activities with lesser cytotoxic activities. Therefore, it was suggested that honokiol and magnolol are very effective antimicrobial agents on periodontal pathogens.

  • Capsicum frutescen is known in Sudan, is one of the most commonly used pepper species in cooking and in Sudanese folk medicine. The present study was conducted to investigate antimicrobial (bacteria and fungi) and cytotoxicity (Brine Shrimp Lethality Test) of methanolic extract of Capsicum frutescen (fruits). The extract have been tested in the present study to investigate the in vitro potential effects against Gram positive, Gram negative bacteria and fungi. The selected organisms were Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumonia and Candida albicans using the cup plate agar diffusion method. The methanol extract of Capsicum frutescen (fruits) exhibited inhibitory effects against Escherichia coli with zone of inhibition (23 mm) and Klebsiella pneumonia with zone of inhibition (17 mm). The phytochemical screening revealed the presence of Tannins, Saponin, Alkaloids, Anthroquinoles and Terpenoids. The Cytotoxicity of methanolic extract of Capsicum frutescens was $LD_{50}$ $64.68{\mu}g/ml$. The activity and presence of compounds known to be biologically active are a validation for the use of Capsicum as a food ingredient and as a therapeutic element of traditional medicine.

Antibacterial Activity and Synergism of the Hybrid Antimicrobial Peptide, CAMA-syn

  • A 20-residue hybrid peptide CA(1-8)-MA(1-12) (CAMA) incorporating residues 1-8 of cecropin A (CA) and residues 1-12 of magainin 2 (MA) has high antimicrobial activity without toxicity. To investigate the effects of the total positive charges of CAMA on the antibacterial activity and toxicity, a hybrid peptide analogue (CAMA-syn) was designed with substitutions of $Ile^{10}\;and\;Ser^{16}$ with Lys. According to CD spectra, structure of CAMA-syn with increase of cationicity was very similar to that of CAMA in DPC micelle. CAMA-syn showed antimicrobial activity similar with CAMA while CAMA-syn has no hemolytic activity and much lower cytotoxicity against RAW 264.7 macrophage cells than CAMA. Also, CAMA and CAMA-syn significantly inhibited NO production by LPSstimulated RAW264.7 macrophage at 10.0∼20.0 $\mu$M. CAMA-syn displayed salt resistance on antimicrobial activity against Escherichia coli at the physiological concentrations of $CaCl_2\;and\;MgCl_2$. The combination studies of peptides and antibiotics showed that CAMA-syn has synergistic effects with synthetic compound and flavonoid against Enterococcus faecalis and VREF. CAMA-syn can be a good candidate for the development of new antibiotics with potent antibacterial and synergistic activity but without cytotoxicity.

메탄올로 추출한 여주 분획성분의 항균 및 항발암 효과 (The Effects on Antimicrobial and Anticarcinogenic Activity of Momordica Charantia L.)

  • 중국에서 약용 및 음용식물로 사용되고 있는 여주 (Mo-mordica charantia L.)를 이용하여 추출, 분획한 후 항균, 항암 및 암예방 효과를 살펴보았다 여주의 MCU층을 제외한 모든 층에서 항균력을 보였으며 특히 Staphylococru aureus의 MCMEE층에서 가장 높은 항균활성을 나타내었다 실험에 사용한 각균에 대한 여주 분획물의 암세포 증식 억제 효과를 MTT assay로 실험한 결과, 3종의 인체 암세포 HepG2, HeLa 및 MCF극에서 모두 여주의 ethyleth-er 분획층인 MCMEE와 ethylacetate 분획층인 MCMEA 층에서 아주 높은 암세포 증식억제 효과를 나타내었다. HepG2세포를 이용하여 암예방 QR 유도 활성을 측정한 결과, 다른 분회층에 비해 비극성 용매층인 hexane 분획층 MCMH에서 아주 높은 QR 유도를 활성시키는 것으로 나타났다. 본 실험 결과, 여주에는 식품 보존과 항균제로서의 개발가능성이 보이며. 항발암 효과를 보기위한 암세포 증식 억제 효과는 ethylether층 MCMEE과 ethylacetate층 MCMEA층에서 월등하였고, 비극성 용매층인 hexane 분획층에는 암발생을 예방하는 quinone reductase inducer가 있는 것으로 사료되며. 나아가 단계적인 생리활성 연구의 분리 동정이 계속 이루어져야 할 것으로 사료된다.

적채 분획물의 항균, 암세포 증식 억제효과 (The Effects on Antimicrobial and Cytotoxicity of Brassica oleracea L. Fractions.)

  • 노화억제, 항균, cholesterol 저하작용 등 다양한 생리활성을 가지고, 기관지염, 천식 등 호흡기 질환에도 옛부터 효과가 있는 것으로 알려져 있으며 식품으로 애용되고 있는 적채를 metanol (BOM)로 먼저 추출하고 이를 hexane 분획물(BOMH), ethyl ether 분획물(BOMEE), ethyl acetate 분획물(BOMEA), butanol 분획물(BOMB) 및 물 분획물(BOMA) 등 다섯가지의 각 용매별로 분획하여 적채의 항균, 암세포 증식 억제 및 QR유도 효과를 연구하였다. 먼저 적채의 각 분획물을 Streptococcus mutans, Bacillus subtilis, Escherichia coli. Pseudomonas aeruginosa 및 Aspergil-lus oryzae의 5가지 균주에 첨가시료농도를 증가시키면서 첨가하였다. 즉, 각 분획 별 시료를 500, 1000, 1500 및 2000 $\mu{g}$/ml의 농도로 사용균주에 각각 처리하였을때 BOMEA에서 비교적 높은 항균 활성 효과를 나타내었고, 그 다음으로는 BOMEE이었다. 적채의 암세포 증식억제 효과(cytotoxicity)를 MTT assay 로 실험한 결과, 3종의 암세포주 HepG2, HeLa 및 MCF-7은 모두 BOMEE에서 높은 암세포 증식억제 효과를 보였으며, HepG2 세포주를 이용한 암예방 QR 유도 활성은 다른 분획층에 비해 비극성 용매층인 BOMH과 BOMEE에서 유의적으로 QR유도 활성을 증가시키는 것으로 나타났다. 이 결과를 기초로 독특한 보라색을 띄며 식탁에서 널리 애용되고 있는 자주색 대표식품인 적채의 항균 및 암세포 성장저지 및 QR 유도 효과를 일으키는 생리활성 물질의 존재와 기전 규명에 유익한 자료가 될 것으로 사료된다.
