Isolation and Properties of Amino Acid Antimetabolite from Streptomyces sp. YS-943 (Streptomyces sp. YS-943균주가 생산하는 아미노산 대사길항물질의 정제와 성상)

  • A Streptomyces strain YS-943, which produced amino acid antimetabolite, was isolated from soil. During the course of screening for new amino acid antimetabolites from the culture broth of Actinomycetes, we found that the strain produced a substance active against Gram-positive bacteria and its activity was reversed by L-methionine and L-histidine on the synthetic minimal agar medium in the culture broth.The morphological and cultural characteristics serve to identify the producing organism strain YS-943 as the genus Streptomyces. Fermentation was carried out in the synthetic medium at 28$\CIRC$C for 48 hours. The fermentation yield reached about 12 mg per liter of the broth. The YS-943 substance was obtained as white powder, mp 194$\CIRC$C and has the molecular formular of C$_{4}$H$_{8}$N$_{2}$O$_{4}$. Its structure was determined to be o-carbamyl-D-serine by spectroscopic data. It is active against some Gram-positive bacteria and reversed by L-methionine and L-histidine.

Relationship Between Antimetabolite Toxicity and Pharmacogenetics in Turkish Cancer Patients

  • Introduction: Antimetabolites may cause severe toxicity and even toxic death in cancer patients. Our aim was to evaluate the relationship between antimetabolite toxicity and pharmacogenetics in patients with severe clinical toxicity or alanine transaminase (ALT) elevation after fluorouracil (5FU), capecitabine or methotrexate administration. Patients and Methods: Cancer patients with severe antimetabolite toxicity were evaluated for methylenetetrahydrofolate reductase (MTHFR) gene C667T, thymidilate synthase (TS) gene 5´UTR variable number of tandem repeats (VNTR), dihydroprymidine dehydrogenase (DPYD) gene IVS14+1G/A, Xeroderma pigmentosum (XPD) gene Lys751Gln and X-ray repair cross-complementing group 1 (XRCC1) gene Arg399Gln polymorphisms. Results: Eighteen patients were enrolled, with a male/female ratio of 0.8. They had osteosarcoma in methotrexate group (n=7), gastrointestinal malignancies in 5FU group (n=9) and breast cancer in the capecitabine group (n=2). Mucositis and dermatitis occurred in all groups, together with ALT elevation in the methotrexate group and 2 toxic deaths were encountered. DPYD, TS, MTHFR, XPD and XRCC1 gene polymorphism rare allele frequencies were observed to be higher than in the general population. Conclusion: Pharmacogenetics might contribute to tailored therapy.

방선균이 생산하는 아미노산 대사길항물질, YS-460의 분리 정제 및 특성

  • An amino acid antimetabolite named YS-460 was isolated from the culture filtrate of a newly isolated Actinomycetes identified as Streptomyces sp. Fermentation was carried out in the synthetic medium at 30$\circ$C for 5 days. Purification was done by ion exchange resin, active carbon, silica gel column chromatography and obtained 38 mg of pure active substance per liter of the broth. YS-460, an amino acid like substance, has the molecular formula of C$_{7}$H$_{11}$NO$_{3}$- Its structure determined to be furanomycin by spectral analysis. It is active against some bacteria on a chemically defined medium and reversed competitively by L-isoleucine and non-competitively by L-leucine and L-valine.

  • The effects of an antimetabolite, 6-aminonicotinamide (6-AU) on the levels of glucose, glycogen, catechoamines and mucleotides in mice brain were investigated. The level of glucose in the blood starts increasing from 3 h after administration of 6-AU while those in the brain tissue start increasing from 9 h after administration of 6-AN. The concentration of brain glvcogen remained unchanged at all time points except 11h. The level of epinephrine in the brain was found to reach maximum value at initial 3 h following 6-AU administration, after urhich it started dec$\ulcorner$easing si역서cantle. The Brvel of brain norepinephrine remained virtually unchanged before 24 h time point at which it starts decreasing significantly. ATP, CTP, UMP and UTP levels were significantly reduced but AMP and CMP levels urere not affected.

  • A Streptomyces strain No. 182-27, which produced amino acid antimetabolite, was isolated from soil. During the course of screening for new amino acid antimetabolites from the culture broths of Actinomycetes, we found that the strain produced a substance active against Gram-positive bacteria and its activity was reversed by L-Ieucine on the synthetic minimal agar medium in the culture broth. The morphological and cultural characteristics serve to identify the producing organism strain 182-27 as the Streptomyces, although the species of this strain should be resolved in further studies. Fermentation was carried out in the synthetic medium at $28^{\circ}C$ for 78 hours. The fermentation yield reached about 2 mg per liter of the broth. Purification was done by ion exchange resin, active carbon, silica gel column chromatography and obtained 20 mg of pure active substance from the 20 $\ell$ culture broth. The 182-27 substance was obtained as white powder, mp 18SoC. From the physicochemical characteristics of the substance, it was amino acid like substance but unknown about its chemical structure. It is active against some Gram-positive bacteria and reversed by L-Ieucine.

  • The effects of antimetabolite 6-aminonicotinamide on levels of soluble proteins, free amino acids and protease activity in various tissues of quail have been in vestigated. The levels of soluble proteins m liver, heart and pectoral muscle were markedly lowered and the specific activity of protease in kidney and pectoral muscle was markedly increased. The concentrations of aspartic acid / asparagine, alanine, valine, methionine, isoleucine, leucine and lysine in the liver were markedly increased. En the kidney the concentrations of aspartic acid I asparagine, arginine, threonine, alanine, proline and lysine were markedly increased but those of glutamic acid I glutamine were decreased. The concentrations of glutamic acid / glutamine and serine in the heart were reduced but those in glycine and methionine were increased. In the pectoral muscle the concentration of arginine was decreased but the concentration of alanine and threonine was increased. The overall results suggest that antimetabolite 6-aminonicotinamide may act to enhance concentrations of amino acids related to the generation of energy and to depress the biosynthesis of some specific amino acids.

  • The Synthesis of E, E, E-12-fluorofarnesol and E, Z-6-fluorofarnesol which are key intermediates for the study of biosynthesis of some sesquiterpenes, is described. E, E-Farnesyl acetate is treated with selenium dioxide to give E, E, E-12-hydroxy farnesyl acetate, whih is transformed by DAST into E, E, E-12-hydroxy farnesyl acetate, which is transformed by DAST into E, E, E,-12-fluorofarnesylacetate. The latter compound is hydrolyzed to E, E, E,-12-fluorofarnesol. The reformatsky reaction of 6-methyl-5-hepten-2-one with ethyl bromofluoroacetate affords ethyl 2-fluoro-3-hydroxy-3, 7 dimethyl-6-octanoate. This ester is acetylated and eliminated to give ethyl (Z)-2-fluoro-3, 7-dimethylocta-2, 6-dienoate, which is transformed to allyl bromide via allylic alcohol. The allyl bromide is treated with dianion of methyl acetate to give-keto ester. The $\beta$-keto ester is converted to diethyl phosphoryloxy compound. The conjugate addition of lithium dimethylcuprate to the latter compound gives fluoro ester, which is treated with DIBAL to afford E, Z-6-fluorofarnesol.

  • To investigate the cytotoxicity and genotoxicity of the DNA alkylating agnet, mitomycin C and the antimetabolite, 5-Fluorouracil (5-FU) in cultured rat fibroblasts, the colorimetric assay of netural red (NR) for cytotoxicity and for genotoxicity, sister chromatid exchange (SCE) assay and the measurement of the rate of DNA synthesis were performed in cells cultured in media containing various concentrations of mitomycin C and 5-FU. The uptake ability of neutral red decreased does-dependently. NR90 and NR50 values of mitomycin C were 1.49 nM and 6.87mM and 5-FU were 38.4mM AND 284.4Mm respectively.

Influence of Pretreatment with Immunosuppressive Drugs on Viral Proliferation

  • Immunosuppressive drugs are used to make the body less likely to reject transplanted organs or to treat autoimmune diseases. In this study, five immunosuppressive drugs including two glucocorticoids (dexamethasone and prednisolone), one calcineurin inhibitor (cyclosporin A), one non-steroid anti-inflammatory drug (aspirin), and one antimetabolite (methotrexate) were tested for their effects on viral proliferation using feline foamy virus (FFV). The five drugs had different cytotoxic effects on the Crandell-Ress feline kidney (CRFK) cells, the natural host cell of FFV. Dexamethasone-pretreated CRFK cells were susceptible to FFV infection, but pretreatment with prednisolone, cyclosporin A, aspirin, and methotrexate showed obvious inhibitory effects on FFV proliferation, by reducing viral production to 29.8-83.8% of that of an untreated control. These results were supported by western blot, which detected viral Gag structural protein in the infected cell lysate. As our results showed a correlation between immunosuppressive drugs and susceptibility to viral infections, it is proposed that immune-compromised individuals who are using immune-suppressive drugs may be especially vulnerable to viral infection originated from pets.

Ultrastructural Changes of the Spinal Cord after Treatment with 6-Aminonicotinamide (6-Aminonicotinamide 투여 후 햄스터 척수의 미세구조 변화)

  • The effects of antimetabolite, 6-aminonicotinamide (6-AN), on ultrastrudural changes in the spinal cord of golden hamster were investigated. Intraperitoneal administration of 6-AN (10 mg/kg body weight) every two days gave rise to a marked reduction of about $30\sim40%$ in body weight after $26\sim28$ days ($13\sim14th$ injection). In the lesions of the spinal cord, neuroglial cells such as astrocytes and oligodendrocytes were severely damaged, but neurons and blood vessels were not affected by 6-AN. The myelin sheath was also affected by 6-AN. Vacuoles observed in the lesions were produced by the swelling and degenerating changes of neuropils and neuroglial cells. Numerous swollen mitochondria and cisterns of rough endoplasmic reticulum were observed in the watery cytoplasm of damaged neuroglial cells, but intermediate filaments were well preserved. Especially in the damaged astrocytes, the outer nuclear membrane were partially swollen and formed a halfmoonlike structure. It is suggested that as well as the multivesicular bodies protruding from the swollen dendrites, the conjugation of adjacent vacuoles also participated in the formation of large vacuoles.
