Identification of Antifungal Substances of Lactobacillus sakei subsp. ALI033 and Antifungal Activity against Penicillium brevicompactum Strain FI02

  • This study was performed to investigate the antifungal substances and the antifungal activity against fungi of lactic acid bacteria (LAB) isolated from kimchi. LAB from kimchi in Imsil showed antifungal activity against Penicillium brevicompactum strain FI02. LAB LI031 was identified as Lactobacillus sakei subsp. Antifungal substances contained in L. sakei subsp. ALI033 culture media were unstable at high pH levels. Both, the control and proteinase K and protease treated samples showed clear zones, suggesting that the antifungal substances produced by ALI033 were non-protein substances unaffected by protesases. Both, the control and catalase showed clear zones, suggesting that the antifungal metabolite was not $H_2O_2$. The molecular weights of the antifungal substances were ${\leq}3,000Da$. The organic acid content of crude antifungal substances produced by L. sakei subsp. ALI033 showed high concentrations of lactic acid (502.47 mg/100 g). Therefore, these results suggest that antifungal substance produced by L. sakei subsp. ALI033 is most likely due to its ability in producing organic acid.

Isolation of Bacterial Strain Antagonistic to Pyricularia oryzae and Its Mode of Antifungal Action

  • An antagonistic bacterium PM-1 which strongly inhyibits the growth of Pyricularia oryzae was isolated and identified as paenibacillus macerans. The antifungal substances of the strain PM-1 showed the broad antifungal spectra against P.oryzae races. Relating to the localization test, it was found that the antifungal substances existed not only in the cytoplasm but also in the culture supernatant, and importantly the antifungal activity of the latter was stronger than that of the former. The extracellular antifungal substances were extremely heat-stable up to $121^{\circ}C$ for 15 min. The substances were optimally produced at $20^{\circ}C$ and pH 10.0 in a potato dextrose broth. The culture filtrate of the strain PM-1 caused a partial swelling of the mycelia of P.oryzae, and it prevents the normal growth of the fungus as well. This result suggested that the antifungal substances secreted by the strain PM-1 potentially inhibited the germination of P.oryzae.

Isolation and Numerical Identification of Streptomyces humidus strain S5-55 Antagonistic to Plant Pathogenic Fungi

  • To search for the antifungal substances, various actino-mycete isolates were obtained from various soils of Korea using plate dilution method on the humic acid vitamin agar plates. In the screening procedures using a dual culture method, 32 actionomycete isolates were selected, which showed the inhibitory activity against mycelial growth of plant pathogenic fungi Altirnaria mali, Colletotrichum gloeosporides, Fusarium oxysporum f.sp. cucumerinum, Magnaporthe grisea, Phytophthora capsici, and Rhizoctonia solani. Bioassay of the crude extracts from culture filtrates and mycelial mets revealed that 12 antagonistic actionomycetes produced highly active antifungal substances. Actinomycete strain S5-55 which showed the substantial antifungal activity against the tested fungi was selected for production of the antifungal substances. Based on the cytochemical and morphological characteristics, strain S5-55 was identified as a Streptomyces species. The results of the numerical identification using the TAXON program confirmed that Streptomyces strain S5-55 was identical with Streptomyces humidus including in TAXON major cluster 19. The production of antifungal substance was most favorable when S. humidus strain S5-55 was cultivated for 10 dats on soluble starch broth supplemented with $K_2$HPO$_4$. The antifungal substances active against the plant pathogenic fungi P. capsici and M. grisea were partially purified using $\textrm{C}_{18}$ reversed-phase column chromatography.

천연 항균물질을 이용한 황토모르타르의 항곰팡이 성능 평가 (Antifungal Performance of Hwangtoh Mortars with Natural Antifungal Substances)

  • 본 연구는 다양한 천연물계 항균물질로부터 추출된 추출액(extract)들의 자체 항곰팡이 성능을 평가하고, 이를 적용한 항균모르타르의 항곰팡이 성능과 그 지속성능, 그리고 항균성능 등에 대해 종합적으로 고찰함으로써 효율적인 건축물 내 항균대책 및 친환경 항균 건자재(모르타르 포함) 개발을 위한 기초적 자료를 제공하고자 수행하였다. 시험결과, 마조람 추출액 및 이를 적용한 항균모르타르의 항곰팡이 성능은 현재 널리 사용되고 있는 유기계 화학 항곰팡이제 및 이를 적용한 모르타르와 동등수준의 높은 항곰팡이 성능을 나타내었다. 특히 마조람 추출액 5% 혼입 모르타르는 장기 항곰팡이 성능뿐 아니라 대장균, 녹농균 및 황색포도상구균에 대한 항균성능에서도 뛰어나 그 활용가능성이 크게 기대되었으며, 향후 이에 대한 보다 상세한 검토가 요구된다.

  • 쇠비름즙액(汁液)(원액(原液))은 사과 부란병균(病菌)(Valsa mali), 배 검은무늬병균(病菌)(Alternaria kikuchiana), 벼 도열병균(病菌)(Pyricularia oryzae)등 몇가지 식물병원균(植物病原菌)에 대한 항균작용(抗菌作用)을 지니고 있다. 이와 같은 항균작용(抗菌作用)을 나타내는 물질(物質)은 메타놀에 잘 용해(溶解)되며 지질(脂質)에 속하는 물질(物質)로 추정(推定)된다. 그리고 쇠비름즙액(汁液)에 함유(含有)되어 있는 항균물질(抗菌物質)은 균채신장억제(菌采伸長抑制) 뿐만 아니라 포자발아억제(胞子發芽抑制) 효과(效果)를 보여주고 있다.

토양길항세균 Bacillus sp. KL-3의 대사산물을 이용한 벼도열병균 Pyricularia oryzae의 생물학적방제

  • Biocontrol of plant pathogens provides an alternative means of reducing the incidence of plant diseases without the negative aspects of chemical pesticides. Nowdays, as the resistant fungi about the chemical fungicides have revealed and the concern of environment has increased, the biological control of phytopathogenic fungi by the antagonistic microorganisms is very much indispensable. For the selection of strong antagonistic bacterium for biological control agent of rice leafblast and cucumber gray mold rot, the antifungal strain KL-3 strain was selected among 120 strains isolated from the rhizosphere soils. And the strain was identified to be a species of Bacillus subtilis or closely related strain. In several biochemical and in vitro antibiosis tests, antifungal substances of Bacillus sp. KL-3 were presumed heat stable, micromolecular antibiotic substances. In vivo test and vinyl house field test, the antifungal substances of Bacillus sp. KL-3 represented excellent biocontrol ability aganist Alternaria mali, Phyricularia oryzae, and Alternaria kikuchiana as well as broad spectrum of other fungi. In particular, Bacillus sp. KL-3 strain showed more predominant activity than some chemical fungicides against fungi shown to resist chemcal fungicides.

Chemical Constituents of the Fruiting Bodies of Clitocybe nebularis and Their Antifungal Activity

  • During a continuing search for antimicrobial substances from Korean native wild mushroom extracts, we found that the methanolic extract of the fruiting body of Clitocybe nebularis exhibited mild antifungal activity against pathogenic fungi. Therefore we evaluated the antifungal substances and other chemical components of the fruiting body of Clitocybe nebularis, which led to the isolation of nebularine, phenylacetic acid, purine, uridine, adenine, uracil, benzoic acid, and mannitol. Nebularine showed mild antifungal activity against Magnaphorthe grisea and Trichophyton mentagrophytes, and phenylacetic acid potently inhibited the growth of Pythium ultium and displayed moderate antifungal activity against Magnaphorthe grisea, Botrytis cinerea, and Trichophyton mentagrophytes. The other isolated compounds showed no antimicrobial activity.

Microscopic Examination of the Suppressive Action of Antifungal Substances from Pseudomonas aeruginosa on Asexual Sporulation of Fungi

  • Two fractions with unusual antifungal activity that suppress asexual sporulation of several fungi were obtained from culture filtrate of Pseudomonas aeruginosa and were partially purified through the repeated silicagel flash column chromatographies. The sporulation-suppressive actions of these fractions in Aspergillus nidulans, Rhizopus stolonifer, and Coprinus cinereus, were analyzed by light and electron microscopes. The germination ability of the spores produced in the presence of these fractions were also checked to determine the persistent effects of these antifungal substances on the next generation. Light microscopic observation of developing sporangia of R. stolonifer grown in the presence of both fractions revealed that the significant number of sporangia failed to reach maturity, and frequently, uncontrolled growths of hyphae and rhizoids from the sporangiophores were found. In A. nidulans addition of these fractions appeared to cause different classes of morphological abnormality in conidia development, which included aborted formation of conidiogenous cells from the apex of conidiophores and enhanced hyphal growths either at the tip or middle of the conidiophores. Germination abilities of spores obtained from the cultures grown in the presence of antifungal fractions were 40∼60% in Aspergillus, 50∼80% in Coprinus (thallic spores), and 30∼40% in Rhizopus compared to those of normal spores.

Bacillus amyloliquefaciens IUB158-03이 생산하는 항진균물질의 생화학적 특성 및 독성 (Toxicity and Characteristics of Antifungal Substances Produced by Bacillus amyloliquefaciens IUB158-03)

  • B. amyloliquefaciens IUB158-03균주에서 정제된 항진균물질은 극성인 용매에 잘 용해되고, pH 6.0~10.0와 $-70{\sim}121^{\circ}C$에서와 같이 넓은 범위의 온도 및 pH에서 안정성을 보였다. 항진균물질의 FAB-MS, UV 흡수 스펙트럼, 아미노산 조성 등을 분석한 결과 분자량은 1,042 이었고, TLC를 이용하여 분석한 결과 ninhydrin solution에서 보라색으로 발색되었다. UV 스펙트럼은 220 nm, 277 nm에서 ${\lambda}max$를 보였으며, $Asn_3$, $Gln_2$, $Ser_1$ $Gly_1$, $Tyr_1$의 아미노산 조성을 갖는 것으로 나타났다. 그리고 $^1H$-NMR spectrum, $^1H$-COSY, HMQC 을 분석한 결과 iturin A계에 속하는 물질로 확인되었다. NIH3T3 섬유아세포에 대해 항진균물질이 세포독성을 나타내지 않는 것은 물론 마우스에 항진균물질을 경구투여하여 장기 내의 변화와 백혈구 수, 생체내의 생리적인 기능면에서 정상 마우스와 차이를 보이지 않았으므로 생체독성이 없는 것으로 나타났다. 따라서 본 연구를 통하여 B. amyloliquefaciens IUB158-03에서 분리된 항진균물질이 앞으로 고추탄저병의 생물적 방제제로 이용될 수 있는 잠재성을 갖고 있는 것으로 사료된다.

Purification and Antifungal Activities of an Antibiotic Produced by Gliocladium virens G1 Against Plant Pathogens

  • This study was undertaken to separate and identify antifungla substances produced by Gilocladium virens G1, a biocontrol agent used for the control of plant diseases caused by Rhizoctonea solani. The culture of G. virens G1 effectively inhibited the growth of R. solani, Colletotrichum gloeosporioides, and Phytophthora capsici, but less that of Fusarium oxysporum. The n-hexane extract of the G. virens culture, which was used for the purification of responsible substances, strongly inhibited R. solani and C. gloeosporioides, but not P. capsici, although the n-butanol extract was effective on all of the pathogens tested. An antifungal substance was purified using the n-hexane extract by Silica gel column chromatography and HPLC. The substance was examined for purity by HPLC and for nature by UV spectrometry, which differed from known antibiotic compounds such as gliotoxin, viridin and gliovirin. The antifungal substance was very liphophilic based on its solvent-solubility and Rf values on TLC, and more inhibitory to C. gloeosporioides than other fungal pathogens tested.
