Many skin diseases are caused by microbial infections. Representative pathogenic fungus and bacterium that cause skin diseases are Candida albicans and Staphylococcus aureus, respectively. Malassezia pachydermatis is a fungus that causes animal skin diseases. In this study, we propose a method for removing pathogenic microorganisms from the skin using relatively safe edible herbal extracts. Herbal extracts were screened for skin health through the removal of pathogenic microorganisms, and combinations for effective utilization of the screened extracts were identified. In this study, among methanol extracts of 240 edible plants, C. albicans, S. aureus, and M. pachydermatis were killed by extracts of 10 plants: Acori Gramineri Rhizoma, Angelicae Tenuissimae Radix, Cinnamomi Cortex, Cinnamomi Ramulus, Impatientis Semen, Magnoliae Cortex, Moutan Cortex Radicis, Phellodendri Cortex, Scutellariae Radix, and Syzygii Flos. By evaluating the synergistic antifungal activities against C. albicans using all 45 possible combinations of these 10 extracts, five new synergistic antifungal combinations, Acori Gramineri Rhizoma with Magnoliae Cortex extracts, Acori Gramineri Rhizoma with Phellodendri Cortex extracts, Angelicae Tenuissimae Radix with Magnoliae Cortex extracts, Magnoliae Cortex with Phellodendri Cortex extracts, and Phellodendri Cortex with Syzygii Flos extracts, were identified. By utilizing the selected extracts and five combinations with synergistic antifungal effects, this work provides materials and methods to develop new and safe methods for treating candidiasis using natural products.

The present study investigated effects of antifungal and carboxylesterase inoculant on rumen fermentation with different rumen pH. Corn silage was treated without inoculant (CON) and with a mixed Lactobacillus brevis 5M2 and L. buchneri 6M1 (MIX). Rumen fluid was collected from two cannulated Hanwoo heifers before morning feeding (high rumen pH at 6.70) and 3 h after feeding (low rumen pH at 6.20). Dried corn silage was incubated in the rumen buffer (rumen fluid + anaerobic culture medium at 1:2 ratio) for 48 h at 39℃. Eight replications for each treatment were used along with two blanks. Both in a high and a low rumen pH, MIX silages presented higher (p<0.05) the immediately degradable fraction, the potentially degradable fraction, total degradable fraction, and total volatile fatty acid (VFA) than those of CON silages. Incubated corn silages in a low rumen pH presented lower (p<0.05) total degradable fraction, ammonia-N, total VFA (p=0.061), and other VFA profiles except acetate and propionate, than those in a high rumen pH. The present study concluded that application of antifungal and carboxylesterase inoculant on corn silage could improve degradation kinetics and fermentation indices in the rumen with high and low pH conditions.
Essential oils (EOs) have been shown to be plant-extracted antimicrobial agents. However, there are limited studies investigating the efficacy of EOs against pathogens. Among them, tea tree oil (TTO) is extracted from Melaleuca alternifolia, which is also used as an antifungal agent. In this study, the effect of TTO was investigated on the suppression of melon powdery mildew caused by Podosphaera xanthii and tomato leaf mold disease caused by Passalora fulva. Both powdery mildew and leaf mold diseases were significantly suppressed by a spray of TTO. Eighty percent of powdery mildew and 81% of leaf mold disease of the control value were suppressed by 0.5% TTO liquid, when sprayed 3 times every 7 days on the melon and tomato leaves. Inhibition of mycelial growth was also greatly affected by different concentrations of TTO against four different fungal pathogens. Ninety-eight percent of Pseudocercospora fuligena, 97% of P. fulva, 95% of Botrytis cinerea, and 94% of Phytophthora infestans mycelial growth were inhibited by 0.2% to 1.0% of TTO contained in plate media, respectively. However, phytotoxicity in plants by the TTO treatments was revealed when melon and tomato leaves were sprayed with a 1% and 2% concentration of TTO, respectively. Therefore, our findings show that TTO has high antifungal effects against various plant pathogens that occur during crop cultivation. We also suggest that when applying TTO to plant leaves, it is necessary to establish an accurate treatment concentration for different crops.
Antifungal activities of crude extractum of Nanshancha Seed Cake (NSC), to inactivate postharvest pathogens were investigated. Highest inhibitory rate was found against C. musae, C. gloeosporioides and C. papaya P.Henn, which was much stronger than that by tea saponin. Compared to tea saponin, effects of NSC extractum was relatively weak and similar on C. gloeosporioides Penzig and P. italicum. In an in vivo study, best controlling effects by NSC extractum was found with banana anthracnose disease development, which showed no inhibitory effects by tea saponin. NSC extractum controlled in vitro C. musae growth through directly inhibiting germination rate and germ tube elongation, and causing distortation, rupture and indentation of C. musae mycelium. In banana fruit subject to C. musae inoculation, higher PAL, POD, GLU and CHT activity was observed in banana fruit treated with crude NSC extractum than that of water control fruits. Current study proved the best controlling effects of crude NSC extractum in C. musae in vitro and in vivo development, which through direct inhibition of C. musae growth and increasing defense system of the banana fruit.
The antifungal activity of polygodial, which was isolated from Polygonum hydropiper used as a hot spice in Japan, was investigated through measuring its MICs and against food-contaminants including 4 strains of Saccharomyces cerevisiae, 3 strains of Zygosaccharomyces species, and 5 strains of Aspergillus species. All yeast-like fungi exhibited strong antifungal susceptivilities with MICs and MFCs below 3.13${\mu}g$/ml. Strains of Aspergillu species showed moderate with MIC of 25-50${\mu}g$/ml and MFC of 50-100${\mu}g$/ml. However these strains to sorbic acid, as already known, revealed very weak with MICs of 200-800${\mu}g$/ml and MFCs above 1600${\mu}g$/ml. In addition, effects of various pHs and temperatures on the antifungal activity of polygodial were tested against S. cerevisiae. At $4^{\circ}C$, the fungicidal activity of polygodial gradually increased with incubation time and reached the maximum(MFC of 3. 13${\mu}g$/ml) in 5 hours. At temperatures in the range of 30-$45^{\circ}C$, the activity of polygodial increased in proportion to temperature rise and in particular at $45^{\circ}C$ was possible with the concentration of 0.1${\mu}g$/ml. mOn the other hand, medium pH was also identified to affect the antifungal activity of polygodial. Namely, compared to neutral (pH7), acidic(pH 3) and basic(pH 9) medium synergized its activity (MIC and MFC) to 8- and 2- fold, respectively.
To define the optimum conditions for the mass production of four antifungal Pseudomonas spp. isolated from soil, we have investigated culture conditions and effects of various nutrient sources on the bacterial growth and evaluated antagonistic activity against Rhizoctonia solani and Sclerotinia homoeocarpa, plant pathogens. The optimum temperature and pH for the growth of these isolates were determined as pH 7.0 and $20^{\circ}$ or $25^{\circ}C$, respectively. Sucrose, tryptone, and $K_2HPO_4$ generally were more adequate for better growth as carbon, nitrogen and mineral source, respectively. The nutrient sources were also found to be very effective for high antifungal activities against R. solani and S. homoeocarpa. It was elucidated that YUD-F group (P. mandelii and P. fluorescens), which inhabit regions at relatively low temperature, had more broad spectrum and higher antifungal activity than YUD-O group (P. trivialis and P. jessenii) generally against R. solani and S. homoeocarpa. It is thought that the differences of the average temperature in the various habitats of Pseudomonas spp. influence the optimal growth temperature and antifungal activity. Especially, Pseudomonas spp. of YUD-O group showed the better antifungal activity against dollar spot caused by S. homoeocarpa, but showed relatively weaker antifungal activity against brown patch caused by R. solani.

This study was carried out in order to investigate the potential of using plant oils derived from Leptospermum petersonii Bailey and Syzygium aromaticum L. Merr. Et Perry as natural antifungal agents. The antifungal effects of essential oils at concentrations of 0.05, 0.1, 0.15, and 0.2 mg/ml on the dermatophytes Microsporum canis (KCTC 6591), Trichophyton mentagrophytes (KCTC 6077), Trichophyton rubrum (KCCM 60443), Epidermophyton floccosum (KCCM 11667), and Microsporum gypseum were evaluated using the agar diffusion method. The major constituents of the active fraction against the dermatophytes were identified by gas chromatography-mass spectrometry and high-performance liquid chromatography analysis. The antifungal activities of S. aromaticum oil (clove oil) against the dermatophytes tested were highest at a concentration of 0.2mg/ml, with an effectiveness of more than 60%. Hyphal growth was completely inhibited in T. mentagrophytes, T. rubrum, and M. gypseum by treatment with clove oil at a concentration of 0.2 mg/ml. Eugenol was the most effective antifungal constituent of clove oil against the dermatophytes T. mentagrophytes and M. canis. Morphological changes in the hyphae of T. mentagrophytes, such as damage to the cell wall and cell membrane and the expansion of the endoplasmic reticulum, after treatment with 0.11 mg/ml eugenol were observed by transmission electron microscopy (TEM). At a concentration of 0.2 mg/ml, L. petersonii oil (LPO) was more than 90% effective against all of the dermatophytes tested, with the exception of T. rubrum. Geranial was determined to be the most active antifungal constituent of L. petersonii oil. Taken together, the results of this study demonstrate that clove and tea tree oils exhibited significant antifungal activities against the dermatophytes tested in this study.
The antifungal activities of the essential oil from Agastache rugosa and its main component, estragole, combined with ketoconazole, one of the azole antibiotics commonly used to treat infections caused by Trichophyton species, were evaluated in this study. The combined effects were measured by the checkerboard microtiter and the disk diffusion tests, against T. erinacei, T. mentagrophytes, T. rubrum, T. schoenleinii and T. soudanense. Susceptibility of the five Trichophyton species to the oil alone, or ketoconazole alone, differed distinctly. The fractional inhibitory concentration indices (FICI) of ketoconazole combined with estragole or A. rugosa essential oil, against the tested Trichophyton species, were between 0.05 and 0.27, indicating synergistic effects. These drug combinations exhibited the most significant synergism against T. mentagrophytes, with FICIs of 0.05 and 0.09 for estragole and the essential oil fraction from A. rugosa, respectively. Isobolograms based on the data from checkerboard titer tests also indicated significant synergism between ketoconazole and the Agastache oil fraction or estragole, against the Trichophyton species evaluated. Trichophyton susceptibility to ketoconazole was significantly improved by combination with the Agastache rugosa oil fraction or its main component, estragole.
The present study aimed to assess the antifungal properties of the essential oil fraction from Ligusticum chuanxiong (Umbelliferae) and its components against five clinically important Aspergillus species. The essential oil fraction was extracted from the underground parts of the plant by steam distillation, and its main components, namely, Z-ligustilide, butylidene phthalide, and p-cresol were isolated by column chromatography. The antifungal activities of the essential oils were evaluated by the broth dilution method. Both the total essential oil fraction of L. chuanxiong and its components showed significant anti-Aspergillus activity against all five tested strains with MICs between 62.5 and 250 ${\mu}g$/ml, respectively. In a checkerboard microtiter assay, the combination of antibiotics, itraconazole with the essential oil fraction of L. chuanxiong or its main components exhibited synergistic or additive, and in some cases indifferent, effects against the tested Aspergillus species, resulting in FICIs (fractional inhibiting concentration indices) ranging from 0.12 to 2, while the combination of antibiotics, amphothericin B with L. chuanxiong essential oils mostly showed antagonistic effects.

The objective of this study was to determine inhibitory activities of four volatile plant essential oils (cinnamon oil, fennel oil, origanum oil and thyme oil) on in vitro growth of Fusarium oxysporum f. sp. fragariae causing Fusarium wilt of strawberry plants. Results showed that these essential oils inhibited in vitro conidial germination and mycelial growth of F. oxysporum f. sp. fragariae in a dose-dependent manner. Cinnamon oil was found to be most effective one in suppressing conidial germination while fennel oil, origanum oil and thyme oil showed moderate inhibition of conidial germination at similar levels. Cinnamon oil, origanum oil and thyme oil showed moderate antifungal activities against mycelial growth at similar levels while fennel oil had relatively lower antifungal activity against mycelial growth. Antifungal effects of these four plant essential oils in different combinations on in vitro fungal growth were also evaluated. These essential oils demonstrated synergistic antifungal activities against conidial germination and mycelial growth of F. oxysporum f. sp. fragariae in vitro. Simultaneous application of origanum oil and thyme oil enhanced their antimicrobial activities against conidial germination and fungal mycelial growth. These results underpin that volatile plant essential oils could be used in eco-friendly integrated disease management of Fusarium wilt in strawberry fields.
