STRAINS OF FOOT-AND-MOUTH DISEASE VIRUS IN DIFFERENT DISTRICTS OF BANGLADESH

  • An investigation was carried out to find out the strains of foot-and-mouth disease (FMD) virus in 24 districts of Bangladesh. A total of 505 FMD virus samples were collected from June, 1989 to June, 1991 and tested by complement fixation test (CFT). Of these, 276 (54.7%) were found positive for different strains of FMD virus and the rest 45.3% were either negative or anticomplementary. Strains identified were O, C, Asia-1 and sub-strains $A_5$ and $A_{22}$. Strain O was found to be most prevalent(39.8%) followed by Asia-1 (5.7%), C (5.3%), $A_5$ (3.4%) and $A_{22}$ (0.4%). Prevalence of sub-strain $A_5$ was reported for the first time in Bangladesh. District-wise typing of FMD virus has been done which would be helpful for appropriate vaccination programme in different districts of Bangladesh for control of the malady.

Serological Study on Q Fever by Detection of Complement Fixation Antibodies to Coxiella burnetii in Dairy Cattle in Kyuuggi Province, Korea (Coxiella burnetii 보체결합항체 검사에 의한 경기지역 젖소에 있어서의 Q열에 대한 혈청학적 조사연구)

  • Regarding to Q fever which is one of the most important zoonoses in Food (Milk) Hygiene and in Environmental Public Health, a sero-epidemiological study was carried out to detect the complement fixation antibodies to Coxiella burnetii Nine Mile strain phase II antigen among the milking cows in Kyunggi Province. The results obtained were summarized as follows: 1. The overall prevalence of the CF antibodies to Q fever among 989 milking cows of 75 dairy farms in Kyunggi Province was revealed as high as 58.7% by the farms and 27.8% by the individual cows with higher prevalence in Kyunggi Central and Kyunggi East regions than any other regions in the Province. 2. Anticomplementary reactions were appeared as 7.5% (74/989) and it ranged from 1.0% to 16.0% according to the regions investigated. 3. In the titration of the positively reacted sera, the figures of 16.7%, 37.5%, 29.8%, 9.5%, 2.9% and 3.6% at the serum dilutions of 1: 10, 1: 20, 1: 40, 1: 80, 1: 160 and higher than 1: 160, respectively. 4. It was recognized that the relatively higher cumulated frequency distribution of the CF antibodies was shown in the sera collected from the regions with higher prevalence of Q fever. 5. There was a high correlation between the prevalence of Q fever CF antibodies and the age which is usually equivalent to one year older than the calving history of the milking cows.

Pharmacological Activities of the Polysaccharide extracted from Phellinus linteus (상황버섯으로부터 분리한 고분자 다당류의 약리활성)

  • To examine the pharmacological activity of the polysaccharide extracted from phellinus linteus, we obtained the polysaccharide treated by the alkali and hot water, molecular weights of hot water was 10kD and 225 kD. Anticomplementary activity were highly observed in hot water fraction than alkali fraction. The tumor inhibition ratio of the polysaccharide extracted by hot water and alkali of Phellinus linteus against sarcoma 180 were 72.5% and 67.6%, respectively. These results suggest that protein bound polysaccharides extracted by a hot water and alkali of Phellinus linteus were similar to the pharmacological activities and chemical properties.

  • The highest antitumor activity was observed in water soluble AS fraction of the Ganoderma lucidum IY 009. AS fraction did not show any cytotoxicity on sarcoma 180 cell but stimulated antibody production, opsonization of macrophage in ICR mouse and superoxide ion production from isolated macrophage. AS fraction activated complement C3 in human serum, and their antitumor activity was inhibited by EDTA, a chelator of cation related complementary activation. AS fraction exerted om prolong of life span and ingibition of tumor growth in the leukemia P388 or L1210 transplanted inbreed mouse,k BDF1 but krestin did not. AS fraction did not show any serious and lethal effects through oral administration on ICR mouse, and LD$_{50}$ of those was above 2,230 mg/kg.

  • Exo-polysaccharide (BWS) obtained from submerged cultivation of Ganoderma lucidum mycelium was fractionated. Antitumor activity of their fractions was investigated in comparison with the mycelial polysaccharide fractions. Eight kinds (BWS-DN, BWS-DA, BWS-DN-GI, BWS- DA-GI, MWS-DN, MWS-DA, MWS-DN-GI and MWS-DA-GI) of polysaccharide fractions were obtained by DEAE-cellulose ion exchange chromatography and Sepharose CL-4B gel chromatography from BWS and MWS, which were isolated from culture fiuid and mycelial cell, respectively. The anticomplementary activities (ITCH$_{50}$%) of the exo-polysaccharide fractions showing 15% to 30% were lower than those of mycelial polysaccharide fractions showing 15% to 70%. The acidic fractions of BWS-DA and BWS-DA-GI fractionated from BWS, showed the highest activity of 30%. In the MTT assay, BWS-DN and MWS against mouse leukemia L1210 exhibited high inhibition ratio of 86 and 89%, respectively at the concentration of 600 $\mu$g/ml. High inhibition ratio of 50% (IC$_{50}$) was achieved for BWS, BWS-DA and MWS-DA fractions against human colon adenocarcinoma COLO-205 and for BWS-DA, BWS-DN and MWS-DN fractions against human leukemia HL-60 at the concentra- tion of 300 $\mu$g/ml among the six polysaccharide fractions, respectively.

Biological Activities of Solvent Fractions from Methanolic Extract of Crataegi fructus (산사(Crataegifructus) 메탄올 추출물로부터 용매분획된 분획물의 생리활성)

  • The purpose of this study is to determine the possibility of using Crataegi fructus as natural health food source. The research was conducted to determine the biofunctional activities of Crataegi fructus extract. Methanolic extract from Crataegi fructus was partitioned by using organic solvents, including n-hexane, ethyl acetate, n-butanol, and water. Ethyl acetate soluble fraction was shown to have the strongest antioxidant activity ($RC_{50}=4.39{\mu}g/m{\ell}$) among the fractions. In the antimicrobial activity assays, ethyl acetate soluble fraction was effective in bacterial inhibition, against the cases of Escherichia coli and Klebsiella pneumonia, with minimum inhibitory concentrations in $125{\mu}g/m{\ell}$. In the anticomplementary activity assays, water soluble fraction was the most effective exhibiting 18.4% inhibitory activity.

Chemical Properties and Physiological Aacitivities of Stromata of Cordyceps militaris

  • The chemical properties and physiological activities of the freeze-dried stromata of Cordyceps militaris were examined. A proximate analysis exhibited that the stromata consisted mainly of crude carbohydrate (74.3%), crude protein (11.5%), and moisture content (8.7%), with a low content of crude ash (4.%) and fat (1.3%). The carbohydrate was mostly composed of glucose (88.6%). A large quantity of essential fatty acids, including linolenic acid (33.8%) was also observed. An analysis of the component amino acid showed a relatively high ratio of the essential amino acids, lysine (101.2 mg/g), methionine (62.7 mg/g), and acidic amino acids of glutamic aicd (57.5 mg/g) and aspartic acid (43.9 mg/g), whereas a low of tyrosine content (4.7 mg/g). An examination of the cordycepin content indicated tht the stromata and mycelium-embedded media (silkworm pupae) contained 0.2% and 0.5%, respectively. From the examination of the physiological activities, based on methanol extract (M), ethylacetate extract (EA), and hot-water extract (HW) fractions of the stromata, the hot-water extract (HW) fraction showed the most potent intestinal immune modulating activity, anticoagulant activity, and anticomplementary activity, whereas the ethylacetate extract (EA) fraction exhihited a radical scavenging activity. Therefore, the results from the present study indicate that the stromata of C. militaris contain various healthy chemical ingredients, and that especially boiled water extract of whole body would apprear to provide beneficial physiological activities.

Chemical Properties and Physiological Activities of Synnemata of Beauveria bassiana

  • Chemical properties and physiological activities of the freeze-dried synnemata of Beauveria bassiana were examined. A proximate analysis showed that the synnemata consisted mainly of carbohydrate (49.86%), protein 11.36%), and a moisture content of 30.64%. It contained a low amount of crude ash (4.76%) and fat (3.38%). The carbohydrate was composed mainly of mannose (52.3%), galactose (31.5%), glucose (13.2%), and rhamnose (3%). Trace amounts of arabinose, xylose, and fructose were present. Major amino acids In the synnemata were glutamic acid, glycine, aspartic acid, arginine, threonine, alanine, valine, leucine, lysine, and aspartic acid with the amounts of 30.42, 25.22, 17.17, 15.12, 12.65, 15.23, 12.47, 11.47, 14.24, and 17.17 mg/g, respectively. Among extracts from the synnemata, the hot-water extract showed 67% of anticomplementary activity compared to that of the positive control, followed by ethyl acetate extract (17%) and methanol extract (15%). The hot-water extract also had anticoagulant activity with 55 sec of coagulating time and this fraction exhibited the most potent Intestinal immune system modulating activity. The methanol extract showed the highest inhibitory activity (25%) on the 12-O-tetradecanoyl phorbol-13-acetate-induced superoxide ($O_2^-$) generation, followed by hot-water extract (18%) and ethyl acetate extract (10%). The data in the present study indicate that the extract of Beauveria bassiana synnemata contains some healthful chemical ingredients and it could provide beneficial physiological activities. These features of the synnemata should be of interest to the food industry as well as other industrial fields.

  • Flos Sophora japonica L. (Leguminosae), commonly called scholar tree, is a well-known traditional medicine used for the treatment of bleeding and as an antihemorrhagic agent. This research was conducted to determine biofunctional activities of Flos Sophora japonica extract. In anticomplementary activity assays, water soluble fraction was the most effective exhibiting 21% inhibitory activity.

Pharmaceutical Substances of Platycodon grandiflorus (jacquin) A. De Candolle (도라지의 약리성분)

  • Platycodon root contains abundant pharmaceutical substances and is widely used as a food and a medicinal herb. The three-year-old and the 24-year-old Platycodon roots cultivated in a local farm were analysed and compared with their pharmaceutical substances to identify the major components. Reverse column HPLC analysis of ethanol extracts from the 24-year-old roots showed some distintive peaks after the retention time of 50 min which were absent in the extracts from the three-year-old roots. This indicates that several pharmaceutical substances are present in the older roots. There were no differences in sterol composition and saponin content between the 24-year and three-year-old roots. The 15-year-old and the 23-year-old Platycodon roots, the 6-year-old Ginseng root, and the three-year-old Condonopsis root all did not show significant anticomplement activity.
