Effects of β-Glucan Supplementation on Lymphocyte Proliferation, Macrophage Chemotaxis and Specific Immune Responses in Broilers

  • Immunomodulatory feed additives might offer alternatives to antimicrobial growth promoters in poultry production. This experiment was carried out to test the effect of $\beta$-glucan supplementation on the growth performance and immune response in broilers. Total of 160 day-old broilers were randomly assigned to 4 treatment groups fed corn-soybean diets containing 0, 0.012, 0.025 or 0.05% of $\beta$-glucan supplement in a 6 week feeding experiment. Growth performance, antibody titer against New Castle vaccine, lymphocyte blastogensis, and peritoneal macrophage chemotaxis activity of broilers were evaluated. Results showed that there were no significant differences in weight gain and feed efficiency among the treatments, and no differences in antibody titer was observed. Supplementation of $\beta$-glucan did not elevate the lymphocyte blastogensis among treatments, following stimulation with different mitogens. However, supplementation with 0.025 and 0.05% $\beta$-glucan enhanced the macrophage chemotaxis activity of broilers. These results suggest that $\beta$-glucan may enhance some cell-mediated immune responses of chickens by modulate macrophages ability.

An epidemiological study on the malaria for residents in Kangwha-Gun(1999) (경기 강화지역 주민에 대한 말라리아의 역학적 특성에 관한 연구(1999년))

  • Epidemic malaria is prevalent in the demilitarized zone and the northern area of Kyonggi province in Korea. Malaria has broken out in Gangwha-Gun since 1997 due to geographical condition. The results obtained from an epidemiological surveillance for the malaria that happened in 1999 were as follows ; Anopheles sinensis collected in UV-light traps in the barn in Wolgogri and Dangsanri of Songhaemyon showed the distribution of 27,156 of 49,280 figures, corresponding to 55.1%, between July and August. The numbers of patients with malaria were 48(32.4%) in July, 36(24.3%) in August, and 38(25.7%) in September with a prevalence with the ages of the twenties to the seventies. Of a total 154 patients, the male were 86(58.1%) and the female were 68(41.8%). The annual parasite index(API) were 6.44, 5.89 and 4.45 in Songhaemyon, Yangsamyon and Kyodongmyon, respectively. A high infection rate was found Particularly in the areas within 2km from the Han river. By the immunofluorescence assays(IFA), 26 cases were positive with an antibody titers of $\geq$ 1 : 32, of 220 residents in Songhaemyon and 336 ones with a potential case and contacted with the patients. By the blood test, there was no significance in alkaline phosphatase, alanine aminotransferase, total cholesterol, glucose, hemoglobin, hematocrit, white blood cell, red blood cell, and platelet between residents with an antibody titers of $\geq$ 1 : 32 and < 1 : 32 in Songhaemyon.

  • Porcine respiratory coronavirus (PRCV) is antigenically related to transmissible gastroenteritis virus (TGEV). Differential serological diagnosis between PRCV and TGEV infection is not possible with the classical sero-neutralization test. Infection with PRCV or TGEV induces antibodies which neutralize both viruses to the same titer. However, the enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR) can differentiate between PRCV and TGEV infection. This study was carried out to investigate the prevalence of PRCV infection of swine in Gyeongnam province. A total of 391 serum samples from 37 herds in Gyeongnam were examined for antibody to PRCV using blocking ELISA. All serum samples were collected from 130- to 150-day-old pigs between August and December 2006. By ELISA, 182 out of 391 sera tested (46.5%) and 29 out of 37 sample herds (78.4%) were positive against PRCV. Our data suggested that seropositive herds for PRCV are distributed diffusely throughout Gyeongnam. The PCR methods were established to diagnose PRCV spike protein (S) gene. PCR were conducted to identify the PRCV genome against 150 pigs in PRCV antibody positive herds.

Production and characterization of monoclonal antibodies against porcine transmissible gastroenteritis virus (돼지 전염성 위장염 바이러스에 대한 단크론항체 생산 및 특성)

  • Eight monoclonal antibodies(MAbs) against the transmissible gastroenteritis virus (TGEV) were produced and characterized. Four of the MAbs were produced against a reference TGEV, Purdue strain(P115) and the others were produced against the Korean vaccine virus, Pyungtaek strain. Only one MAb(5C8) produced against P115 had neutralizing activity and was found to be E2 protein-specific. The other seven MAbs(4E2, 4G8, 5H6, 1F8, 2C6, 5H5, and 3A6) had specificity of nucleocapsid protein and no neutralizing activity. All MAbs reacted with different strains of TGEV, but none of the MAbs was reactive with porcine enteropathogenic viruses such as rotavirus, epidemic diarrhea virus and enterovirus by fluorescence antibody(FA) test.

  • This study was performed to verify the stability and efficacy of Streptococcus iniae formalin killed cells on storage at refrigerator temperature in olive flounder, Paralichthys olivaceus. The vaccines preserved for 6, 12 and 15months showed high stability of potency. The antibody titers and protection efficacy to challenge test were significantly higher in booster immunized groups than prime immunized groups during storage. Especially, above 60% of relative percent survival obtained at low antibody revel in prime immunized groups indicates that innate and non-specific immune system might act against the S. iniae challenged.

A Serological Study on Toxoplasma gondii Infection Among People in South of Tehran, Iran

  • Although Toxoplasma gondii infection generally shows subclinical courses in adults, it may have severe pathologic effects on the fetus. For a survey of toxoplasmosis in Shahr-e-Rey area, south of Tehran, Iran, we conducted a serological study on 1,187 general populations using indirect fluorescent antibody (IFA) test. In this cross sectional study, cases were selected randomly by a local primary health care (PHC) system. Blood samples were collected in microcapillary tubes and sera were examined by IFA after centrifugation. It has been shown that 68.3% of cases in all age groups had IFA titer of 1:10 and higher, which we consider as positive cases, and the rate for 1:400 or over was 30.2%. There was meaningful correlation between anti-Toxoplasma antibody titers and age, sex, and occupation of subjected people (P < 0.05%). The incidence was higher in females and highest in the children age group (10-14 years old). The present study demonstrates high prevalence of Toxoplasma infection among the people in south of Tehran, Iran.

  • To establish a statistically reliable sampling strategy for serological surveillance of classical swinefever (CSF) in Korea, antibody test data from CSF surveillance conducted during year 2005 were analyzed.The most appropriate sampling method was determined to be stratified multi-stage random sampling strategy,in which the primary sampling unit is a pig farm and the secondary are the pigs by the strata of breedersand finishers in the selected farm. The optimum sample size was 5 to 19 including 1 to 2 breeders accordingto the number of pigs in the farm. The optimum sampling strategy demonstrated in this study was veryFindings of our study provide practical guidelines for surveillance of herd immunity level to CSF in Korea.

Study on Infection of Theileria Sergenti in Neonatal Calves (신생 송아지에 있어서 Theileria sergenti의 감염에 관한 연구)

  • The rate of 58 neonatal calves in infection of Theileria sergenti was investigated in random samples on the farms located in Kyunggi, Chonbuk districts of Korea. 1. The criteria used in veryfying infection with T. sergenti included the detection of parasites by giemsa stain and acridine orange stain in the blood smear slides. 2. Further evidence of current or previous exposure to T. sergenti was based on demonstration of T. sergenti specific antibody and antigen by the western immunoblot and the directed immunofluorescent antibody test in the peripherial blood of the calves. 3. The prevalence rates were 35%, 50% in Kyunggi, Chonbuk provinces respectively and the overall prevalence in all the farms was 43.2% by means of acridine orange stain. 4. The parasites that were observed in the peripherial blood of calves was showen surely by the western immunoblot to the characteristic 34KD antigen among the proteins of T. sergenti (Korean isolate). 5. And the antigen of the neonatal calves reacted at the very highest titer(1 : 2, 560) 6. These data highlight the significances of T. sergenti in the neonatal calf disease in Korea.

  • Generally known psittacosis or ornithosis is a disease of birds caused by the bacterium Chlamydia psittaci. Humans are accidential hosts and are most commonly infected from avian sources. It raises hepatitis or neurosis. As major outer membrane protein (MOMP) of Chlamydia psittaci has been known to play a role in the avoidance of host immune defenses, research on developing a Chlamydia vaccine has focused on the MOMP. In this study, the gene encoding the major outer membrane protein (MOMP) of the Chlamydia psittaci strain 6BC was cloned and expressed in Escherichia coli strain M-15. The recombinant DNA was cloned by fusion prokaryotic expression vector pQE30-GFPII. Expression of the recombinant protein was performed in E. coli and was induced by IPTG. The size of expressed recombinant protein is 74.220 kDa (MOMP, 43.260 kDa; GFP expression region, 30 kDa; $6{\times}His$ tag, 960Da). This protein was purified by using his-tagging-inclusion body. Recombinant protein was reconfirmed through ELISA test and western blot with antibody against pQE30-GFPII. It will be useful antibody development.

Juvenile onset acquired myasthenia gravis in a Shih-tzu dog

  • A 7-month-old female Shih-tzu dog was presented with intermittent trembling, dyspnea, generalized muscle weakness, and unconsciousness after exercise. No remarkable findings were shown in the complete blood counts and the radiographic examination. On serum biochemical profiles, alkaline phosphatase and creatine phosphokinase were mildly elevated. Based on history takings, physical examination, and neurological findings, presumptive diagnosis was made as a myasthenia gravis (MG). Clinical signs of this patient were dramatically improved after administration of neostigmine. The result of acetylcholine receptor antibody test in serum was 0.89 nmol/L and the histopathology of muscle were normal. Clinical sign of the patient evaluated in this study is stabilized with long-term administration of pyridostigmine at this time. This case report here describes clinical and clinicopathological findings of a juvenile onset acquired MG in a Shih-tzu dog.
