Teicoplanin 생산성이 우수한 Actinoplanes teichomyceticus ATCC31121 변이주 선별 및 배양학적 특성 (Screening and Characteristics of a Mutant of Actinoplanes teichomyceticus ATCC31121 Highly Producing Teicoplanin)

  • Teicoplanin은 Actinoplanes teichomyceticus가 생산하는 밴코마이신과 유사한 글리코펩티드계 항생제의 일종으로 메티실린 내성 황색포도상구균에 대해서도 효과가 우수한 치료제이다. 공시균주인 Actinoplanes teichomyceticus ATCC31121에 자외선을 조사하여 teicoplanin 생산성이 우수한 균주를 얻었다. 본 연구에서 우수 변이주 획득을 위한 UV 조사에 대한 균주의 치사곡선을 구하고 변이 유도 최적 조건을 확립하였다. 또한 한천 확산법을 사용하여 모균주와 변이주들의 teicoplanin 생산성 분포를 비교하였다. 변이주 가운데 가장 생산성이 높은 T1014-1를 최종적으로 획득하였으며 효소 활성도, 항생제 내성, 자가내성 및 teicoplanin 생산성을 조사하였으며 발효조에서 발효특성을 조사하였다.

Production and Biological Activity of Laidlomycin, Anti-MRSA/VRE Antibiotic from Streptomyces sp. CS684

  • Culture broth of a streptomycete isolate, Streptomyces sp. CS684 showed antibacterial activity on methicilin resistant Staphylococcus aureus (MRSA) and vancomycin resistant enterococci (VRE). Among purified substances from the organism, CSU-1, which is active against MRSA and VRE, is a $C_{37}H_{62}O_{12}Na\;(M^+,721.3875)$, and identified as laidlomycin. The anti-MRSA and anti-VRE activity of CSU-1 was stronger than oxacillin and vancomycin. Phylogenetic analysis showed that strain CS684 is very similar to Streptomyces ardus NRRL $2817^T$, whereas the ability of Streptomyces sp. CS684 to produce laidlomycin was shown to be unique.

Klebsiella pneumoniae 분리균의 생화학적 특성과 항생물질 내성유형 연구 (Biochemical Characteristics and Antibiotic Resistant Patterns of Klebsiella pneumoniae)

  • One hundred and thirty-six strains of Klebsiella pneumoniae were isolated from clinical specimen taken from pediatric patiants at 6 different hospitals and identified, characterized and investigated on the patterns of antibiotic resistance. The 136 strains showed the positive reactions in the 17 tests; Voges-Proskauer, ONPG, cirate utilization, KCN degradation, productions of lysine decarboxylase, acid and gas from glucose, utilizations of malonate, manitol, rhamnose, salicin, sucrose, raffinose, arabinose, lactose, sucrose, inositol and raffinose, but the strains showed the negative reactions in the 8 tests; production of $H_2S$, indole, arginine dehydrolase, ornithine decaraoxylase, phenylalanine deaminase, motility, methly red and gelatin liquefaction. 41 did not utilize dulcitol, and 32 did not utilize adonitol. The 36 out of them produced klebecin. The 136 strains were resistant to ampicillin, 2 to gentamicin, 14 to cephalothin, 16 to chloramphenicol, 8 to kanamycin, 13 to streptomycin, and 17 to tetracycline. Twenty strains were resistant to 2 kinds of antibiotics 5 strains to 3, 4 to 4 and 1 to 6 and 7.

Salmonellosis in swine: Clinical perspectives

  • Salmonella is one of the most important food-borne zoonotic pathogens, causing acute or chronic digestive diseases such as enteritis. The acute form of enteritis is common in young pigs of 2 - 4 months of age. The main symptoms include high fever ($41-42^{\circ}C$), loss of appetite, and increased mortality within 2 - 4 days of onset of the disease. It is often the cause of increasing mortality, decreasing growth rate and reducing feed efficiency of piglets. In the case of chronic enteritis in pigs, the main symptom is weight loss due to the continuing severe diarrhea. Salmonella enterica serovar Typhimurium and Salmonella enterica serovar Choleraesuis are typical pig adapted serotypes, which cause one of four major syndromes: enteric fever, enterocolitis/diarrhea, bacteremia and chronic asymptomatic carriage. These syndromes cause a huge economic burden to swine industry by reducing production. Therefore, it is necessary that swine industries should strive to decrease Salmonellosis in pigs in order to reduce economic losses. There are several measures, such as vaccination to prevent salmonellosis, that are implemented differently from country to country. For the treatment of Salmonella, ongoing antibiotic treatment is needed. However constant doses of antibiotics can be a problem because of antibiotic resistance. Therefore, the focus should be made more on prevention than treatment. In this review, we addressed the basic information about Salmonella, route of infection, clinical symptoms, and prevention of Salmonellosis.

Diagnosis of Clostridium difficile infection in patients with hospital-acquired diarrhea

  • Clostridium difficile infection (CDI) is a rapidly emerging infection that may have devastating consequences. Prompt and accurate diagnosis is crucial for management and control. The aim of this study was to determine the incidence of C. difficile associated diarrhea among hospitalized patients, and to compare different diagnostic laboratory methods for detection of toxin producing strains in clinical specimens. The study was conducted at a university hospital in Cairo during the period from May 2013 till June 2015. Subjects were under antibiotic therapy and presented with hospital-acquired diarrhea. Four hundred and sixty-five stool specimens were processed by different microbiological methods. C. difficile was recovered in culture in 51 of stool specimens. Of these, 86.3% to 98% were positive for toxin production by 2 different methods. This study showed that antibiotic intake is the major risk factor for development of hospital-acquired diarrhea. We evaluated different microbiological methods for diagnosis of C. difficile. We recommend the use of toxigenic culture as a gold standard for microbiological diagnosis of C. difficile.

Dual Inoculation of Native Rhizobium spp. and Arbuscular Mycorrhizal Fungi: An Impact Study for Enhancement of Pulse Production

  • Fifteen Rhizobium spp. from nodules of 6 common pulses collected from 6 districts of Assam were studied for their infectivity, intrinsic antibiotic resistance, nitrogenase activity and effect of dual inoculation with two native Arbuscular Mycorrhizal Fungi viz. Glomus mosseae(GM) and Gigaspora gilmarie(GG). Out of the 15 isolates 9 were found nodulation positive and 6 of them(AR1, BR8, BR12, AR10, UR10 & GR21) were subjected to intrinsic antibiotic sensitivity test of which AR1 showed resistance against all the 9 test antibiotics. Isolates AR1 and GR21 showed the highest(4.25 mole, $gm^{-1}hour^{-1}$) and the lowest(1.05 mole, $gm^{-1}hour^{-1}$) nitrogenase activity respectively. In Most Probable Number count, the maximum Rhizobium population $5.8{\times}10^5$, was found in both Blackgram and Greengram variety of pulses. The maximum dry weight of nodules(3.14 g), dry weight of shoot(10.08 g), nitrogen content(7.68 mg, $plant^{-1}$), chlorophyll content(1.89 mg, $g^{-1}$), phosphorus content of shoot(6.17 mg, $g^{-1}$) and yield(535.67 kg, $Ha^{-1}$) were found when AR1 dually inoculated with GM in Blackgram.

  • Enrofloxacin, a fluoroquinolone, is a broad-spectrum antibiotic widely used in veterinary medicine that inhibits the action of bacterial DNA gyrase, resulting in anti-bacterial effects. This study was performed to examine whether enrofloxacin has modulatory and anti-inflammatory activity on immune cells. A few studies have reported the anti-inflammatory effects of enrofloxacin. In this study, we used mouse spleen cells treated with lipopolysaccharide (LPS) and examined the effects of enrofloxacin. Several assays were performed in LPS-treated spleen cells after the enrofloxacin treatment. Enrofloxacin inhibited the metabolic activity and mitochondrial membrane potential of LPS-treated spleen cells significantly. On the other hand, enrofloxacin did not alter the proportion of the subsets in spleen cells, and did not induce cell death. The production of tumor necrosis factor-alpha in LPS-treated spleen cells was inhibited by enrofloxacin. Overall, enrofloxacin had modulatory activity in spleen cells treated with LPS. These data may broaden the use of enrofloxacin as an antibiotic with anti-inflammatory activity in veterinary clinics.

Antibiotic susceptibility of Clostridium perfringens type D isolated from feces of goats

  • Clostridium perfringens (C. perfringens) may cause diarrhea and enterotoxemia in adult and young livestock, leading to problems in the production and management of farms. Four hundred fecal samples were collected from 25 goat farms located in Gyeongsangbuk-do Province in the Republic of Korea. Sixteen C. perfringens strains were isolates from fecal samples, and the isolates were identified as type A (n=11) and type D (n=5). Additionally, ${\alpha}$- and ${\varepsilon}$-toxin genes were detected in 16 and 5 strains by PCR, respectively, and the enterotoxin gene was presented in 2 strains. The antibiotic susceptibility test was performed using the disk diffusion method and E-test method. In the disk diffusion method, ampicillin (n=16) and chloramphenicol (n=15) were highly susceptible to 16 C. perfringens isolates. In the E-test method, ampicillin, amoxicillin, amoxicillin/clavulanic acid and meropenem were susceptible to more than 14 of 16 C. perfringens isolates. This study indicates that administration of antibiotics such as ampicillin, amoxicillin/clavulanic acid and meropenem can prevent and treat C. perfringens infections in goats.

옻나무 추출액의 급여가 육계의 생산성, 맹장내 균총 및 유해가스 발생량에 미치는 영향 (Effects of Feeing Extracts from Rhus trees on Performance, Microflora Population and Noxious Gas Emission in the Cecum of Broiler)

  • The effect of feeding extracts from rhus trees on performance of broiler were performed to investigate development of natural antibiotic in process of broilers production. A total of 320 broiler chicks at 7 day of age were fed the commercial diet and water, water containing 0 ppm (control), 300 ppm (T1), 600 ppm (T2) and 1000 ppm (T3) of extracts from rims trees for 7 weeks. The body weight gain and feed conversion ratio were improvement by feeding extracts from rims trees at 7 to 21 days of age (P<0.05), but amount of feed intake and mortality were not different among the four group. When 21 to 35 days of age, body weight gain, amount of feed intake, feed conversion ratio and mortality were almost same results when 7 to 21days of age, but it was not significantly different. When 35 to 49 days of age, body weight gain in T1 and T3 group was significantly improvement than control group (P<0.05), also feed conversion ratio was improve by feeding extracts from rhus trees. There was a decrease in the microflora population of both E coli. and salmonella in the cecum contents and feaces in broilers by feeding extracts from rhus trees. Emission of ammonia and VFA gas from excreta were significantly decreased at 5 to 10 days of storaged period (P<0.05). The digestibility of dry matter, crude fat, and crude ash of feed were tend to increased in broiler feeding extracts from rims trees. Digestibility of crude protein of feed in broiler three treatmental groups was significantly improved (P<0.05) as compared with those in control group. These results indicated that the feeding extracts from rhus trees of 300 to 1000 ppm of broiler production were improvement in th6 body weight gain, feed conversion ratio, and mortality due to increasing nutrients digestibility from feed, decreasing of microflora population of both E coll. and salmonella in the cecum contents and feaces, decreasing emission of ammonia and VFA gas from excreta.

Pomyxin B의 장계세균항원(腸系細菌抗原)에 대(對)한 항체산생(抗體産生) 증강작용(增强作用)에 관(關)한 연구(硏究) (Studies on the Enhancing Effect of Polymyxin B on the Antibodies Response of Enterobacterial Antigens)

  • Various kinds of antibiotics are generally believed to have inhibitory effects on the antibody response. However, as polymyxin B which belongs to the cyclic polypeptide group of antibiotic was found to have some enhancing effects on the antibody response of rabbits to enterobacterial common antigen(CA) under specified conditions, experiments were carried out on this problem with the following results. 1. When mixture of polymyxin B and CA derived from Salmonella typhimurium(STM) was treated 30 minutes at $37^{\circ}C$ and injected three times into rabbits by intravenous route, the antibody response to CA was weaker than rabbits injected CA only. 2. Mixture of polymyxin B and CA showed a marked antibody production when injected into rabbits primed with small amounts of heat-extracted antigen of STM, while the injection of CA alone showed low titers of response. 3. Mixture of polymyxin B and heat-extracted CA-containing antigen of Escherichia coli 014 also showed a increased antibody production than CA alone in rabbits primed with antigen of STM. 4. The effect of polymyxin B appeared in different ways. This antibiotic did not enhance the CA antibody response in rabbits primed with small amounts of E. coli 0111 and 055, but enhance in rabbits primed with Shigella flexneri. 5. No enhancing effect on the antibody response was observed by polymyxin B in rabbits primed with CA. 6. No enhancing effect on the antibody response was also noted in rabbits primed with STM antigen in case polymyxin B and CA were administered simultaneously but in veins of different places. 7. Bacitracin did not enhance the CA antibody response in primed rabbits with STM antigen, but neomycin slightly enhance the response. 8. Lipopolysaccharide showed no priming effect on the CA antibody response, and no enhancement of the CA antibody response in rabbits printed with STM. 9. The priming effect of STM antigen against CA antibody response was very weak as compared with the effect of CA derived from STM antigen.
