Effect of Fat Contents on Thermal Resistance, Antibiotic Sensitivity, and Caco-2 Cell Invasion of Listeria monocytogenes

  • This study evaluates the effects of fat contents on the thermal resistance, antibiotic sensitivity, and Caco-2 cell invasion of Listeria monocytogenes. Ten strain mixture of L. monocytogenes in milk (0, 1, and 4% fat) and pork sausage patties (10, 20, and 30% fat) were exposed to $63^{\circ}C$. To evaluate effects of fat on the antibiotic sensitivity of L. monocytogenes, the L. monocytogenes strains NCCP10811 (most antibiotic resistant to streptomycin) and NCCP10943 (most antibiotic sensitive to streptomycin) were exposed to different fat contents in milk and pork sausage patties, and L. monocytogenes from the foods were used for antibiotic sensitivity assays. The most invasive L. monocytogenes strains (NCCP10943) was exposed to different fat contents in milk or pork sausage patties, and L. monocytogenes from the foods were used for the Caco-2 cell invasion assays. The reductions of L. monocytogenes populations were not generally influenced by fat contents. The L. monocytogenes subjected to milk fat had increased sensitivities (p<0.05) due to some antibiotics. In addition, Caco-2 cell invasion efficiency of L. monocytogenes NCCP10943 increased (p<0.05) as fat contents increased. These results indicated that higher fat contents may be related to L. monocytogenes invasions and heat resistances in pork sausage patties, but the relationship between fat and antibiotic sensitivity varied according to antibiotics, strains, and fat contents.

청소년의 올바른 항생제 사용을 위한 웹 기반 교육프로그램 개발 및 평가 (Development and Evaluation of a Web-based Education Program on Appropriate Antibiotic Use in Korean Adolescents)

  • Purpose: This study was done to develop a web-based education program on appropriate antibiotic use and test the effects of the program on knowledge and attitudes towards antibiotic use in Korean adolescents. Methods: The web-based education program was developed through an extensive literature review and professional advisory meetings including technical assistance from a web-based education programmer and content experts. A convenience sample of 851 students from middle and high schools participated in the assessment of effects of the program. Knowledge and attitudes of the students towards antibiotic use and satisfaction with the web-based education program were measured. Descriptive statistics and paired t-test were used to analyze the data. Results: There were significant improvements in knowledge and attitudes towards antibiotic use following self-learning via the web-based education program in both middle and high school students. High school students demonstrated higher scores in knowledge and attitudes than middle school students. Conclusion: The results of this study indicate that this web-based education program on appropriate antibiotic use is a convenient and effective medium for self-learning in adolescents. Therefore the web-based program should be put into wide use as an effective and convenient teaching method for health education in secondary schools.

  • A vast increase of antibiotics usage in concentrated animal feeding operations (CAFOs) over the last few decades has led to an environmental risk due to the presence of antibiotic residuals in different environmental compartments. Especially in Korea, the use of antibiotics in CAFOs is much greater than in other developed countries. One of the primary adverse impacts of antibiotic residuals in the environment is that they readily produce antibiotic resistant bacteria (ARB), which exert detrimental effects on the ecosystem as well as human health. In this article, the impacts of veterinary antibiotic residuals with regard to their quantification and management, and desirable remediation technologies have been widely reviewed. This review article concluded that the continuous monitoring should be required to ensure the safety of antibiotic residuals in the surrounding environments. Furthermore, the management guidelines of antibiotic residuals need to be developed in the future.

항생제 처방 질 관리를 위한 항생제 처방 지침의 개발 (Development of antibiotic prescription guidelines for antibiotic prescription quality management)

  • 이 연구의 목적은 치과 진료실에서 항생제 남용에 대처하기 위해 항생제 투여의 필요성을 결정하는 과정을 소개하고 적절한 항생제의 선택과 사용에 대한 정보를 공유하고, 지속적인 모니터링을 통해 항생제 처방의 적절성을 증가시키는 것이다. 본 병원의 가이드라인은 최신의 항생제 처방에 대한 연구 결과에 따랐으며, 향후에도 지속적인 항생제 처방 가이드 라인에 대한 보완이 필요할 것이다. 본 가이드라인에 의하면, 병력 검사에 의한 페니실린 알레르기 조사와 피부에서의 페니실린 알레르기 검사를 통해 아목시실린을 일차 선택적 항생제로 처방하였다. 증상의 경과에 대한 재평가 과정을 거친 후 아목시실린에 의한 약물 효과가 없다면 보다 광범위한 항생제로 대체하였고, 증상이 호전되지 않으면 환자를 상급병원으로 전원하도록 설계하였다. 사과나무치과병원 직원은 정기적으로 항생제 처방에 대해 모니터링을 하였고 이 결과에 대해 주기적으로 교육을 받도록 하였다. 현재의 가이드라인은 지속적으로 보완되어야 하며, 항생제 남용의 제어에 긍정적인 결과를 가질 것으로 생각되며, 전반적 치과 진료의 흐름에도 기여할 것으로 보인다.

사상자 추출물의 자돈 병원성 대장균 억제 효과 (The effects of Torilis fructus extracts against enteropathogenic Escherichia coli in Piglets)

  • The antibiotic chemotherapy produces sometimes side effects and fails to eliminate bacterial infection. The occurrence of strains resistant to antibiotics would be expected to increase, and it is nowadays important to search for non-antibiotic substances. We are aimed to evaluate the effects of Torilis fructus extracts against enteropathogenic Escherichia coli (E. coli) in Piglets. The piglets were divided with three groups; Negative control group, E. coli-infected positive control group, and the Torilis fructus extracts treated group with E. coli infection. During the study period, we compared clinical signs, weight increase rate, fecal scores, gross findings between the treated group and non treated group. After necropsy, necropsy findings and histopathological findings were conducted with the comparison between the groups. As the results of this study, the Torilis fructus extracts additive showed the effects on the suppression of E. coli-induced lesions. On the basis of this study results, our data suggest that the Torilis fructus extracts additive have the antimicrobial effects. The Torilis fructus extracts additive could be used as the alternative material for antimicrobial feed additives.

Structure and Antibiotic Activity of Fragment Peptides of Antifungal Protein Isolated From Aspergillus giganteus

  • In order to determine the functional region of the antifungal protein (AFP) isolated from Aspergillus giganteus responsible for growth inhibitory activity and the promotion of phospholipid vesicle aggregation, overlapping peptides covering the complete sequence of AFP were synthesized. The antibiotic activity against bacterial, fungal, and tumor cells, and the vesicle-aggregation activity of the synthetic peptides were investigated. The AFP functional sequence responsible for antibiotic and vesicle-aggregation activity was determined to be located within the region between AFP residues 19 to 32. AFP (19-32) exhibited an a-helical conformation in a cell membrane-like environment. AFP (19-32) displayed potent antibiotic activity against bacterial, fungal, and tumor cells without peptide toxicity as indicated by hemolysis. Accordingly, AFP (19-32) could be used as a good model for the design of effective antibiotic agents with powerful antibiotic activity yet without any cytotoxic effects against the host organism.

어성초 추출물이 에멀젼에 미치는 영향에 관한 연구 (Study on the Effects of Houttuynia Cordata Extracts on Emulsions)

  • Objectives : Since cosmetics have been one of the help of life, unlike medicine, natural products have been used for cosmetics, generally giving the image of safety and relief compared to synthetic products. Among them, Houttuynia cordata has been known as a useful herbal medicine with antibiotic, anti-inflammatory and anti-oxidative activities. In this study, we were to use Houttuynia cordata extract on formulation of cosmetic emulsions. Methods : The effects of Houttuynia cordata extract on emulsion stability and viscoelastic properties of emulsion were measured using turbiscan and rheometer. And we assessed the anti-oxidative and antibiotic activities of Houttuynia cordata extract. Results : 1. The results of this investigation for emulsion stability showed that the stability emulsion containing Houttuynia cordata extract was decreased depending on concentration of Houttuynia cordata extract. 2. Super oxide dismutase activity was strongly dependents on concentration of Houttuynia cordata extract. 3. Houttuynia cordata extract showed good anti-microbial activity against Staphylococcus aureus. Conclusions : Houttuynia cordata extract can be effectively used in cosmetic emulsions when the relation between natural product extracts and formulation of cosmetics is elucidated.

감초의 메티실린 내성 황색포도구균에 대한 생육억제 효과 (Antibacterial Activity of Glycyrrhizae Radix against Methicillin-resistant Staphylococcus)

  • Objectives : Methicillin-resistant Staphylococcus aureus (MRSA) CCARM 3251 and S. aureusKCTC 1928 have been known to be resistant to many kinds of antibiotics. The extract of Glycyrrhizae Radix showed antibacterial activity against MRSA and antibiotics-resistant S. aureus. Methods : We examined the effects of the water-soluble extract and the methanol-soluble extract of Glycyrrhizae Radix on MRSA and antibiotic-resistant S. aureus. The methanolic extract was further fractionated with organic solvents such as hexane, chloroform, and ethyl acetate in that order. Results and Conclusions : The methanol-soluble extract of Glycyrrhizae Radix showed relatively high antibacterial activity against MRSA and antibiotic-resistant S. aureus. However, the water-soluble extract of Glycyrrhizae Radix showed no antibacterial activity against MRSA and antibiotic-resistant S. aureus. Among the fractions tested, the chloroform fraction showed the highest antibacterial activity against MRSA and antibiotic-resistant S. aureus. The methanol-soluble extract of Glycyrrhizae Radix minimal inhibitory concentrations (MICs) against MRSA and antibiotics-resistant S. aureus were $5{\;}mg/m{\ell}$ in both. The methanol-soluble extract of Glycyrrhizae Radix was separated using thin-layer chromatography and detected with UV -detector. Further study should be carried out to identify which effects cell growth inhibition of MRSA and antibiotics-resistant S. aureus.

항생제, 생균제 및 효모제 첨가가 육계의 성장과 육질에 미치는 영향 (Effects of Supplemention of Antibiotic, Probiotic and Yeast Culture of Performance and Meat Quality in Broiler Chicks)

  • The present study was conducted to investigate the effects of dietary supplementions of 0.1% probiotic and 0.1% yeast culture on the growth performance and meat quality of broiler chicks. A total of 160 Arbor Acre broiler chicks were randomly allotted to 16 pens ; four pens per treatment and 10 birds per pen. Feeding trial lasted for 6 weeks. The results obtained are summerized as follows : The body weight gain and feed intake was slightly higher in group of broiler chick supplemented antibiotic than those of other treatment groups, but were not significantly different among treatments. The feed conversion were slightly lower in groups of broiler chick supplemented antibiotic and probiotic than those of other treatment groups, but were not significantly different among treatments. The pH of thigh musle was slightly higher in group of broiler chick supplemented probiotic than those of other treatment groups. Crude protein, crude fat and crude ash of thigh muscle were lower in groups of broiler chick supplemented probiltic and yeast culture than those of other treatment groups, but were not significantly different among treatments. The saturated fatty acid content of the thigh muscle was tende to be higher in group of broiler chick supplemented yeast culture, but the unsaturated fatty acid content was tende to be lower in group of broiler chick supplemented yeast culture than those of other treatment groups.

Activities of Essential Oils from Perilla frutescens var. acuta against Antibiotic-Susceptible and -Resistant Vibrio and Salmonella Species

  • We determined the inhibitory activity of the essential oil fraction obtained by steam distillation from the fresh and dried leaves of Perilla frutescens var. acuta against some pathogenic Salmonella and Vibrio spp. The activities of compounds isolated from the essential oils, apiol and myristicin, were also tested and the results were compared with those of the essential oil fraction. The Perilla essential oil fraction and its main components showed significant inhibition against antibiotic-susceptive and antibiotic-resistant strains of the tested Salmonella and Vibrio strains. Synergistic or additive effects were identified by combing the oils with ampicillin by checkerboard-titer tests. We conclude that essential oils from P. frutescens can be useful in the treatment of Salmonella and Vibrio infections and as safe additives to food materials for the prevention of contamination of food by these bacteria. This is especially important because of the rapid increase in antibiotic-resistant strains, which could cause severe symptoms in humans.
