MRSA에 대한 울금 추출 및 분획물의 항균활성과 항생제 증강 효과 (Antibacterial Activity and Enhancing Antibiotic Effect of Extract and Fractions from Curcuma longa against MRSA Strain)

  • Curcumin content of butanol fraction from C. longa was found to be 22.4942% of the highest content. However, in DPPH radical scavenging ability and antibacterial activity against methicillin resistance Staphylococcus aureus(MRSA, CCARM3696), ethylacetate fraction contained 2.5791% of curcumin was exhibited highest activity. In comparison of enhancing antibiotic(ampicillin) effect against MRSA, ethanol extract contained 1.7838% of curcumin showed more strong activity. This indicates that the ethanol extract and some fractions from C. longa can have antibacterial activity and enhancing antibiotic effect possibly without curcumin. Appropriate use of antimicrobial agent was important point prior to the development of new antibiotics. And in that sence, extract and fractions of C. longa were worth using as synergist of antibiotics and natural antimicrobial agent.

  • Background: The objective of this study were to determine the antibacterial activity and antibiotic activity-enhancing effect of 70% ethanol extract of the root of Rumex japonicus Houtt. and its fractions when used in combination with gentamicin against aerobic skin flora. Methods and Results: The antibacterial activity and antibiotic (gentamicin) activity enhancing effect against aerobic skin flora were determined using the disc diffusion assay. Chloroform fraction (CF) and ethyl acetate fraction (EF) showed higher activities against Staphylococcus aureus and Staphylococcus epidermidis than those shown by other fractions. Regarding the antibiotic (gentamicin) activity-enhancing effect against aerobic skin flora, the n-hexane fraction (HF) and CF showed strong activity. The combination of HF and CF with gentamicin was evaluated using the broth dilution assay to determine the inhibitory effect on the growth of aerobic skin flora. The combination of CF with gentamicin exhibited the highest inhibitory effect on the growth of S. aureus and S. epdermidis. MTT assay performed to determine the viability of L929 cells revealed that EF treatment resulted in viability of 33.96 - 116.76% at the tested concentration. The combination of 70% ethanol extract and its other fractions with gentamicin showed low cell toxicity. Conclusions: Appropriate use of antimicrobial agents is important prior to the development of new antibiotics. The 70% ethanol extract of the root of R. japonicus Houtt. and its fractions showed significant synergism with gentamicin when used in combination against S. aureus and S. epdermidis. Thus, R. japonicus Houtt. could be used as a functional materials in antimicrobial-related fields.

Coicis Semen Reduces Staphylococcus aureus Persister Cell Formation by Increasing Membrane Permeability

  • Unlike resistant cells, persister cells resist antibiotics due to a decreased cellular metabolic rate and can transition back to normal susceptible cells when the antibiotic is removed. These persister cells contribute to the chronic symptoms of infectious diseases and promote the emergence of resistant strains with continuous antibiotic exposure. Therefore, eliminating persister cells represents a promising approach to significantly enhance antibiotic efficacy. Here, we found that Coicis Semen extract reduced Staphylococcus aureus persister cells at a concentration of 0.5 g/L. Linoleic acid and oleic acid, the major components of Coicis Semen extract, exhibited a comparable reduction in persister cells when combined with three antibiotics: ciprofloxacin, oxacillin, and tobramycin. Conversely, these effects were nullified in the presence of the surfactant Tween 80 (1%), suggesting that the hydrophobic characteristics of linoleic acid and oleic acids play a pivotal role in reducing the number of S. aureus persister cells. Considering the concentration-dependent effects of linoleic acid and oleic acid, the persister-reducing activity of Coicis Semen extract was primarily attributed to these fatty acids. Moreover, Coicis Semen extract, linoleic acid, and oleic acid increased the cell membrane permeability of S. aureus. Interestingly, this effect was counteracted by 1% Tween 80, indicating a close association between the reduction of persister cells and the increase in cell membrane permeability. The identified compounds could thus be used to eliminate persister cells, thereby enhancing therapeutic efficacy and shortening treatment duration. When used in conjunction with antibiotics, they may also mitigate chronic symptoms and significantly reduce the emergence of antibiotic-resistant bacteria.

FOX hunting system을 이용한 배추 기능유전자 탐색 (Systematic approaches to identify functional genes using the FOX-hunting system in Chinese cabbage)

  • Full-length cDNAs are essential for the correct annotation of genomic sequences and for the functional analysis of genes and their products. To elucidate the functions of a large population of Chinese cabbage (Brassica rapa) genes and to search efficiently for agriculturally useful genes, we have been taking advantage of the full-length cDNA Over-eXpresser (FOX) gene hunting system. With oligo dT column it purify the each mRNA from the flower organs, leaf and stem tissue. And about 120,000 cDNAs from the library were transformed into $\lambda$-pFLCIII-F vector. Of which 115,000 cDNAs from the library were transformed into T-DNA binary vector, pBigs for transformation study. We used normalized full-length cDNA and introduced each cDNA into Arabidopsis by in planta transformation. Full-length Chinese cabbage cDNAs were expressed independently under the CaMV 35S promoter in Arabidopsis. Selfed seeds were harvested from transgenic Arabidopsis. We had selected 2,500 transgenic plants by hygromycin antibiotic tolerant test, and obtained a number of transgenic mutants. Each transgenic Arabidopsis was investigated in morphological changes, fertility and leaf colour. As a result, 285 possible morphological mutants were identified. Introduced cDNA was isolated by PCR amplification of the genomic DNA from the transgenic mutants. Sequencing result and BLAST analysis showed that most of the introduced cDNA were complete cDNAs and functional genes. Also, we examined the effect of Bromelain on enhancing resistance to soft rot in transgenic Chinese cabbage 'Osome'. The bromelain gene identified from FOX hunting system was transformed into Chinese cabbage using Agrobacterium methods. Transformants were screened by PCR, then RT-PCR and real time PCR were performed to analyze gene expression of cysteine protease in the T1 and T2 generations. The anti-bacterial activity of bromelain was tested in Chinese cabbages infected with soft rot bacteria. The results showed that the over-expressed bromelain gene from pineapple conferred enhanced resistance to soft rot in Chinese cabbage.

무김치에서 분리한 Leuconostoc 속의 특성과 면역증강 효과 (Characteristics of Leuconostoc spp. isolated from radish kimchi and its immune enhancement effect)

  • 본 연구는 무김치에서 우점균인 Leuconostoc속의 균주 특성을 살펴보고 유용한 균주를 발굴하여 소재화하기 위해 수행되었는데, 이들 Leuconostoc속 4균주의 효소학적 특성, 안전성 평가, 및 면역증강 효과는 양성대조구 LGG와 함께 비교·분석하였다. 효소활성 평가에서 모든 시험균주들은 벤조피렌과 같은 발암성 전구물질 생성과 연관된 β-glucuronidase 효소활성을 갖고 있지 않았고, β-1,4 당쇄결합을 유리시키는 β-glucosidase 효소활성을 나타내어 식품, 세제, 화학 등 여러 산업 분야로의 활용성을 기대할 수 있는 특징을 보유하고 있었다. 또한, 시험균주들에 대한 항생제 내성 평가결과는 8종 항생제들에 대해 MIC 값이 EFSA 기준치 이하로 감수성을 나타내었다. 용혈성 현상 실험 결과, 4종의 시험균주와 양성대조구로 사용된 LGG균은 모두 용혈능이 없는 감마 용혈 현상을 나타내었고, 병원성 미생물 4종에 대한 항균활성 결과에서도 Leu. mesenteroides K2-4 균주가 B. cereus와 Sta. aureus에 대하여 각각 19±0과 20±0 mm의 억제환을 나타내었는데, 이는 양성대조구인 LGG균보다 높은 항균활성을 보였다. 대식세포 RAW cell에 시험균주들을 처리하였을 때 NF-kB/AP-1 전사인자의 활성도는 모든 유산균주가 처리 농도가 낮을수록 농도 의존적으로 증가하였다. 이들 세포들에 의해 염증성매개물질 중 TNF-α와 IL-6의 생성량 역시 시험균주의 처리 농도가 낮을수록 높은 값을 보였는데, 특히 Leu. mesenteroides K2-4 균주는 모든 항목에서 다른 균주들에 비해 뚜렷한 차이를 보였다. 또한, mRNA 수준에서 TNF-α와 IL-6 발현량 역시 통계적 유의수준(p<0.05)으로 높은 활성을 저농도에서 처리하였을 때 유의적(p<0.05)으로 높은 값을 나타내었다. 따라서 이들 균주들이 분비한 사이토카인들은 T세포와 B세포를 자극하여 체내 면역체계를 활성화시켜 면역증강에 기여할 수 있을 것으로 사료된다. 종합적으로 본 연구에서 사용한 모든 Leuconostoc 균주들은 효소활성과 안전성이 확인되었는데, 그 중 Leu. mesenteroides K2-4균주가 높은 면역활성능을 보여, 향후 면역 기능 개선을 위한 건강기능식품 개발을 위한 후보소재로서 유용하게 이용될 수 있을 것으로 기대된다.
