Synergistic Antibacterial Effect and Antibacterial Action Mode of Chitosan-Ferulic Acid Conjugate against Methicillin-Resistant Staphylococcus aureus

  • We evaluated the synergistic antibacterial effect in combination with the chitosan-ferulic acid conjugate (CFA) and β-lactam antibiotics, such as ampicillin, penicillin, and oxacillin, against methicillin-resistant Staphylococcus aureus (MRSA) using fractional inhibitory concentration (FIC) indices. CFA clearly reversed the antibacterial activity of ampicillin, penicillin, and oxacillin against MRSA in the combination mode. Among these antibiotics, the combination of oxacillin-CFA resulted in a ΣFICmin range of 0.250 and ΣFICmax of 0.563, suggesting that the oxacillin-CFA combination resulted in an antibacterial synergy effect against MRSA. In addition, we determined that CFA inhibited the mRNA expression of gene mecA and the production of PBP2a, which is a key determinant for β-lactam antibiotic resistance, in a dose-dependent manner. Thus, the results obtained in this study supported the idea on the antibacterial action mechanism that oxacillin will restore the antibacterial activity against MRSA through the suppression of PBP2a production by CFA.

부안산 참굴(Crassostrea gigas) 추출물로부터 피부 상재균에 대한 새로운 항균 펩타이드, cgCAFLP의 탐색 및 정제 (Screening and Purification of a Novel Antibacterial Peptide, cgCAFLP, Against Skin Pathogens from the Extract of the Pacific Oyster Crassostrea gigas from Buan in Korea)

  • This study was performed to screen the antimicrobial activities of the extract from the Pacific oyster Crassostrea gigas against skin pathogens and to purify the relevant antibacterial peptide. The acidified extract showed potent antibacterial activities against gram-positive and gram-negative bacteria but showed no activity against Candida albicans and no significant cell toxicity. Among acne-causing pathogens, the acidified extract showed potent antibacterial activity only against Staphylococcus aureus, and its antibacterial activity was completely abolished by treatment with trypsin or chymotrypsin, and was inhibited by salt treatment. The acidified extract showed strong DNA-binding ability but did not show bacterial membrane permeabilizing ability. Based on antimicrobial activity screening and cytotoxic effects, a novel antibacterial peptide was purified from the acidified gill extract using solid-phase extraction, cation-exchange, and reversed-phase HPLC. The resulting peptide had a molecular weight of 4800.8 Da and showed partial sequence homology with the carbonic anhydrase 4 (CA4) protein in the hard-shelled mussel. Overall, we purified a novel antibacterial peptide, named cgCAFLP, which is related to carbonic anhydrase 4 (CA4) protein, against skin pathogens. Our results suggest that the Pacific oyster extract could be used as an additive to control some acne-related skin pathogens (S. aureus).

Evaluation of an Appropriate Replacement Cycle for Copper Antibacterial Film to Prevent Secondary Infection

  • The use of copper antibacterial films as an effective infection prevention method is increasing owing to its ability to reduce the risk of pathogen transmission. In this study, we evaluated the bacterial contamination of the antibacterial copper membrane attached to a door handle at a university over time. Six mounting locations with high floating population were selected. In three sites, the door handles with the antibacterial film were exposed, while the remaining three were not attached with the antibacterial films. On days 7 and 14, isolated bacterial strains were inoculated in BHI broth and agar, respectively. Colony-forming units (CFU) were determined after incubation. Strain identification was performed using bacterial 16s rRNA PCR and sequencing. Results showed that the bacterial population on day 14 significantly increased from 6 × 109 CFU/mL (day 7) to 2 × 1010 CFU/mL. Furthermore, strain distribution was not different between the on and off the copper antibacterial film groups. In conclusion, although copper has an antibacterial activity, microbial contamination may occur with prolonged use.

해양 유해세균에 대한 감태 추출물의 항균특성 (Antibacterial Property of Ecklonia cava Extract against Marine Bacterial Pathogens)

  • 사람과 어류에게 감염증을 유발하는 Edwardsiella sp., Streptococcus sp. 및 Vibrio sp. 의 해양 유해세균에 대한 해조류 추출물의 항균 활성을 조사하였다. 연구에 사용된 4종의 식용 해조류 중에서 감태 MeOH 추출물이 본 연구에서 사용 된 6종의 모든 해양 유해세균에 대해 넓은 범위의 항균 활성을 나타내었다. 감태의 MeOH 추출물의 유기용매 분획 추출물들 중에서, EtOAc 분획 추출물이 가장 높은 항균활성을 나타내었으며 6종의 해양 유해세균에 대하여 $128{\mu}g/mL$에서 $256{\mu}g/mL$의 MIC 값을 나타내었다. 또한, HPLC 분석에 의해 감태 EtOAc 분획 추출물에 phlorotannin 화합물인 dieckol이 다량 존재하고 있는 것이 확인 되었다. 결론적으로, 감태 추출물의 phlorotannin 화합물이 여러 유해세균에 대한 강한 항균활성을 나타내는 것처럼, 해양 유해세균에 대해서도 강한 항균활성을 나타내는 것으로 판단된다.

  • To study photocatalytic mechanism of metal doped $Ti0_2$, we investigated photodecomposition effect, photocurrent effect and antibacterial effect. When aluminium content was 2 wt %, photodecomposition effect was better than the others. Silver doped thin films had high photocurrent efficiency and antibacterial effect. This reactions were caused by dissolved oxygen in solution and oxygen adsorbed on surface of thin films.

동충하초, 쑥 및 솔잎 추출물의 항균작용 (Antibacterial Activities of Cordyceps spp., Mugwort and Pine Needle Extracts)

  • 동충하초(눈꽃 동충하초, 번데기 동충하초), 쑥 및 솔잎 추출물의 항균작용을 조사하기 위하여 동충하초의 자실체와 균사체, 쑥 및 솔잎을 물과 70% 에탄올로 추출하였다. 항균실험에는 3종류의 그람양성균(Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus)과 3종류의 그람음성(Escherichia coli, Shigella sonnei, Salmonella typhimurium) 식중독 및 병원성 세균을 사용하였다. 동충하초의 추출수율은 물과 에탄올 모두 자실체의 수율(39∼58%)이 균사체(9∼24%)에 비하여 2.4∼4.4배 높았으며, 쑥과 솔잎 추출물의 수율은 9%미만이었다. 눈꽃 동충하초의 에탄올추출물(JME)은 1%농도에서 L. monowtogenes에 대하여 항균활성을 나타내었으며 번데기 동충하초의 에탄올추출물(MFE)은 3%농도에서 S. aureus에 대하여 항균활성을 나타내었다. 쑥의 에탄올추출물(MGE)은 1% 농도에서 L. monocygenes와 S. aureus에 대하여 항균활성을 나타내었으나 눈꽃 동충하초, 번데기 동충하초 및 쑥의 물추출물은 시험균주에 대하여 항균활성을 나타내지 않았다. 솔잎 물추출물은 E. coli를 제외한 모든 균주에 대하여, 솔잎 에탄올추출물은 실험에 사용한 모든 균주에 대하여 항균활성을 나타내어 솔잎추출물들은 가장 폭넓은 항균스펙트럼을 나타내었다. 액체배지내에서 솔잎추출물의 세균증식 억제작용은 에탄올추출물이 물추출물에 비하여 큰 편이었다.

Antibacterial Activity of Clove Oil against Foodborne Pathogenic Bacteria and Sensory Attributes in Clove Oil-Enriched Dairy Products: A Preliminary Study

  • This study was conducted to evaluate the antibacterial activity against Cronobacter sakazakii and Salmonella enteritidis as well as the sensory attributes of milk products supplemented with various concentrations (control, 0.5%, 1.0%, 1.5%, and 2.0%) of clove oil. In this study, clove oil was shown to have strong antibacterial activities. In addition, all the samples were assessed by ten researchers trained in five sensory attributes, namely, taste, flavor, color, texture, and overall acceptability. Compared to the control, 5% clove oil supplemented was the best in market milk, while in yogurt and kefir, 1.0% supplementation was the best. In terms of sensory attributes, the low score of color and flavor of market milk, yogurt, and kefir is attributed to the characteristics of the supplemented clove oil. Consequently, this study presents the possibility of producing bio-functional milk products supplemented with clove oil, and for controlling the growth of foodborne pathogenic bacteria in milk products using clove oil.

쑥 추출물을 이용한 향균 및 김서림 방지에 관한 연구 (Study on Antibacterial and Anti-Fogging using mugwort extract)

  • 코로나 19 이후 향균에 대한 인식이 많아 달라졌다. 외출 후의 손세정은 필수사항이 되었다. 또한 마스크 착용에 따라 안경에 김서림이 많이 발생한다. 이 문제점은 지금까지의 일상생활에 많은 불편함을 야기하고, 향균과 김서림 방지에 대한 우리들의 인식을 바꾸게 하였다. 이에 본 연구에서는 쑥 추출물을 이용하여 향균 및 김서림 방지에 효과가 있는지 연구를 하였다. 쑥 추출물을 얻기 위해 쑥의 건조과정, 알코올 추출과정, 동결건조과정을 거쳤다. 쑥 추출물과 3:7로 혼합된 징크옥사이드, 천연제올라이트 혼합물을 사용하여 향균성 테스트를 실시하였으며, 쑥 추출물과 기타 혼합물을 사용하여 김서림 방지 테스트를 실시하였다. 그 결과 향균성 테스트에서 시험균주로 사용된 E. coli와 K. pneumoniae의 경우 갯쑥추출물 1% 첨가배지부터 세균이 배양되지 않아 대표적인 그람음성균에 대한 항균력이 확인되었다. 김서림 방지 테스트에서 쑥 추출물 10wt% 혼합물이 고온부와 저온부 모두 매우 양호한 결과를 나타냈다. 우리는 쑥 추출물이 향균과 김서림 방지에 효과가 있어 코로나 19를 극복하는데 많은 도움을 줄 것으로 기대한다.

연교김패전(連翹金貝煎)의 Escherichia coli에 대한시험관내 항균력 평가 (In Vitro Antibacterial Effects of Yeonkyokeumpae-jeon against Escherichia coli)

  • Objectives: The object of this study was to observe the in vitro antibacterial effects of Yeonkyokeumpae-jeon (YKKPJ) have been used for treating various gynecological diseases including mastitis in Korea, and individual six kinds of herbal composition aqueous extracts - Forsythiae Fructus (FF), Millettiae Caulis (MC), Lonicerae Flos (LF), Fritillaria Thunbergii Bulb (FT), Taraxci Herba (TH) and Prunellae Spica (PS) against E. coli. Methods: Antibacterial activities against E. coli of YKKPJ, FF, MC, LF, FT, TH and PS aqueous extracts were detected using standard agar microdilution methods. In addition, the effects on the bacterial growth curve were also monitored at MIC and $MIC{\times}2$ levels. The effects on the intracellular killing and bacterial invasion of individual test materials were also observed using Raw 264.7 and MCF-7. The results were compared with ciprofloxacin, a second generation of quinolone antibiotics in the present study. Results: MIC of YKKPJ, FF, MC, LF, FT, TH, PS aqueous extracts against E. coli were detected as $0.039{\pm}0.013mg/ml$, $0.064{\pm}0.033mg/ml$, $0.108{\pm}0.053mg/ml$, $0.078{\pm}0.027mg/ml$, $16.250{\pm}8.385mg/ml$, $15.625{\pm}9.375mg/ml$, $0.254{\pm}0.131mg/ml$, repectively. YKKPJ, FF, MC, LF, FT, TH, PS aqueous extracts showed antibacterial effects against to E. coli, except for FT and TH, which were showed negligible antibacterial effects, respectively. In addition, ciprofloxacin with YKKPJ, FF, MC, LF and PS aqueous extracts also showed marked dosage-dependent inhibition of bacterial growth, and favorable inhibitory effects on the both bacterial invasion and intracellular killing assays using MCF-7 and Raw 264.7 cells were detected in this experiment. Conclusions: The results obtained in this study suggest that traditional polyherbal formula YKKPJ aqueous extracts showed more favorable antibacterial activities as compared to individual six kinds of herbal composition aqueous extracts. The antibacterial effects of YKKPJ against E. coli considered as results of complicated synergic effects of their six kinds of herbal components rather than simple antibacterial effects of single herbal components. It means, YKKPJ aqueous extracts may show potent anti-infectious effects against E. coil for mastitis.

경옥고(瓊玉膏)가 결핵균(結核菌)에 미치는 영향(影響)에 관(關)한 실험적(實驗的) 연구(硏究) (Experimental Studies of The Effects of Kyungok-go against Mycobacteria tuberculosis)

  • Objective : In order to know the antibacterial effects of Kyungok-go against Mycobacteria tuberculosis, Methods : In this study, I inverstigated these effects in terms of combination of other antibiotics with and without Kyungok-go on several different media conditions. Results: On Ogawa medium, Kyungok-go of the $10{\mu}/ml$ concentration showed the anti-Mycobacteria tuberculosis activity against antibiotic drug-sensitive strain. On Middle-blue medium, Kyungok-go of the $10{\mu}/ml$ concentration showed the anti-Mycobacteria tuberculosis activity against antibiotic drug-sensitive strain. Kyungok-go showed the anti mycobacteria tuberculosis activity with the meaningful result above a certain concentration. The resistance against M, tuberculosis as the concentration of Kyungok-go was decreased significantlly on the high concentration($500{\mu}/ml$) When rifampicin and Kyungok-go were used together, the resistance was decreased with the statistical significance as to the persistant antibacterial effect against M. tuberculosis, When ciprofloxacin and Kyungok-go were used together, the resistance was decreased with the statistical significance as to the persistant antibacterial effect against M. tuberculosis, The combination of treatment, Kyungok-go with both rifampicin and ciprofloxacin, showed much better antibacterial effect against M, tuberculosis than antibiotics alone. Conclusions : This study shows that Kyungok-go has antibacterial effect against M. tuberculosis and in the combination of treatment, Kyungok-go with antibiotics, showed much better antibacterial effect against M. tuberculosis than antibiotics alone,.
