Synthesis of Oxazolidinone Phosphonate Derivatives, Part II

  • Several oxazolidinones, a new class of synthetic antibacterial agents, have shown biological activity against multidrug-resistant gram positive organisms such as staphylococci, streptococci, and enterococci. Previous results of our studies with benzoxazolidinone phosphonate derivatives have demonstrated very low antibacterial activity. In the course of our studies directed towards the discovery of noble antibacterial agents, we have synthesized several new derivatives of oxazolidinone phosphonates prepared efficiently from commercially available amino acids. These compounds are tested for in vitro antibacterial activity and one of the compounds showed promising results allowing us to pursue further studies.

  • 대전근교(大田近郊)의 목장(牧場)에서 자유중인 유우(乳牛)를 대상(對象)으로 CMT법(法)에 의하여 유방염(乳房炎)으로 판정(判定)된 양성우(陽性牛)의 유즙(乳汁)으로부터 분리(分離)한 Staphylococcus aureus를 공시(供試)하여 유방염치료(乳房淡治療)에 있어 보다 개선(改善)된 방법(方法)을 모색(模索)하기 위하여 시판(市販)되고 있는 복합처방(複合處方)된 항균제(抗菌劑)를 선택(選擇)하여 내성시험법(耐性試驗法)을 개발(開發)할 목적(目的)으로 실시(實施)한 결과(結果)를 다음과 같이 요약(要約)한다. 1. 치료용(治療用) 항균제(抗菌劑)를 계속(繼續) 반복(反復)하여 사용(使用)하게되면 차등(差等)있는 내성(耐性)을 나타내었다. 2. Rifamastene, Neo-cillin은 다같이 4회(回)까지 Abimasten-100, Neo-Masti, Special formula, Mastalone은 6회(回)까지 약(弱)한 내성(耐性)을 나타내었다. 3. Tardomyocel-L는 6회(回)까지 내성(耐性)이 거의 나타나지 않았다. 4. 내성시험(耐性試驗)을 거처 선택(選擇)된 약제(藥齊)를 3일간격(日間隔)으로 교체사용(交替使用)할 것을 권장한다.

  • The purpose of study was to develop new dental plaque disclosants which could replace erythrosine. Three food coloring agents(Red No.40, Blue No.1, and Mixed Green), erythrosine and fluorescein were tested for their color difference, antibacterial property, and biocompatibility. Color difference of Red No.40 was greater than that of erythrosine as concentration of solution increased. Color differences of Blue No.1 and Mixed Green were smaller than that of red dyes. Erythrosine showed obvious antibacterial property, but food coloring agents showed almost no antibacterial property. The taste and sensation of erythrosine was the worst, and the taste of Red No.40 and the sensation of Mixed Green were the most tolerable. Erythrosine stained dental plaque and oral soft tissue most deeply and long, and Blue No.1 was the next in the depth and longevity of stain.

가교제를 이용한 Sulfanilamide 중합체의 합성과 항균특성 (Characterization of Antibacterial activity and Synthesis of Sulfanilamide Polymer using Crosslinking Agent)

  • Drug delivery system(DDS) have been actively studied for the past twenty years. Dual action agents are unique chemical entities comprised of two different types of antibacterial compounds covalently linked together in a single molecule in such a way that both components are able to exert their bactericidal properties. In spite of the advent of the antibacterial agent the sulfa agents are the most widely used antibacterial agent today. In this study, new antibacterials derivative was synthesized using glutaraldehyde such as crosslinking agent for the purpose of dual-action as DDS study. Antibacterial activity of these new synthetic derivative between their structures and activities were examined by disc diffusion method. As a result, new synthetic derivative exhibited the broad antibacterial activities against Gram(+) and Gram(-) bacilli. Especially, the antibacterial effect of new synthetic derivative against Gram negative(Esherichia. coli) was much stronger than that against Gram positive.

천연물유래 여드름 치료제제의 항균활성 측정 (Antimicrobial Activities of the Anti-acne Compounds from Natural Sources)

  • The in vitro antibacterial activities of anti-acne agents prepared from the extracts of natural sources were investigated against several bacteria including antibiotic-susceptible and -resistant Propionibacterium acnes. SD-1 and SD-2 were prepared with different formulations and they showed strong antibacterial activities. The anti-acne agents completely inhibited the growth of the tested strains at the concentration of 0.5%. There was no difference in antibacterial activity between antibiotic-susceptible and -resistant P. acnes. The inhibitory activities of two agents showed time-dependent manner. In S. aureus, time-kill curve demonstrated 2.8- and 3.4-$log_{10}$-unit killing after 8 h with SD-1 and SD-2, respectively. In P. acnes, time-killing curve demonstrated 5.1- and 6.1-$log_{10}$-unit killing after 24 h with SD-1 and SD-2, respectively. SD-2 showed stronger antimicrobial activity than SD-1. From these results, we expect that SD-1 and SD-2 have strong antibacterial activities and have advantages for treating acne.

The Importance of Essential-Oils in the Green Synthesis of Silver Nanoparticles

  • The antibacterial activity of metallic nanoparticles (NPs), especially silver (Ag), has been investigated during the course of time in various chemical reactions for antibiotics free agents. Green synthesis of metallic NPs using either microorganisms or plant-extracts has appeared as a simple and replacement to chemical and physical methods. The synthesizing of these NPs through ecofriendly methods signifies an exceedingly applicable approach for offering economical, preferring scalability and possessing negligible ecological influences. Essential-oils are among the subordinate metabolites of plants and their antibacterial anti-inflammatory characteristics have been investigated widely and are commonly attained from the aromatic plants. The usage of essential-oils as reducing agents in biosynthesizing of Ag NPs bring together the interaction of a vital antibacterial agent that simplify the nucleation and growth process within the NPs formation. This review article is offering a progressive process of Ag NPs synthesis using essential oils along with proposing the most applicable formation mechanisms and their antibacterial activities.

화장품 성분들의 세균에 대한 항균 시너지 효과 연구 (A Study on Synergistic Antibacterial Activity of Cosmetic Ingredients)

  • 본 연구에서는 화장품에 널리 쓰이는 컨디셔닝제이면서 항균력을 가지는 성분들인 1,2-hexanediol, ethylhexylglycerin, glyceryl caprylate를 대상으로, 이들이 화장품 제형 내에서 다양한 제형 원료와 혼합될 때 그람 양성, 음성 세균에 대한 항균력이 어떻게 변화하는지 확인하고자 하였다. 검토할 제형 원료로는 제형 내에서 항균력을 향상시킬 것으로 예상되는 물질 7종(1,3-propanediol, 1,3-butanediol, glycerin, dipropylene glycol, niacinamide, EDTA-2Na, ethanol)을 선정하였으며, checkerboard assay를 이용하여 해당 물질들과 항균력을 지닌 3종 물질에 대한 항균 시너지 효과를 확인하였다. 시험 결과, 세균에 대해 눈에 띄는 항균 시너지 효과를 보인 물질은 ethanol과 EDTA-2Na였으며 특히 ethanol은 항균력을 지닌 컨디셔닝제 3종 모두와 세균에 대해 항균 시너지 효과를 나타냄을 확인할 수 있었다. Glyceryl caprylate는 시험한 물질 7종 모두와 그람 양성균인 S. aureus에 대해 항균 부가 및 상승 효과를 나타내었으며, 4종의 폴리올이 첨가된 제형에는 1,2-hexanediol, glyceryl caprylate 조합으로 방부처방을 하는 것이 그람 양성, 음성 세균을 모두 제어하기에 용이하다고 판단되었다. 본 연구는 화장품에서 자주 이용되는 항균력을 지닌 컨디셔닝제와 주요 제형 원료의 항균 시너지 효과를 확인함으로써 항균력을 지닌 컨디셔닝제의 제형 내 항균력을 예측하는 데 기여했다는 점에서 의의가 있다.

  • We synthesized peptide-resin conjugates (1 and 2) by immobilizing ${\beta}$-sheet antibacterial peptide and ${\alpha}$ helical antibacterial peptide on PEG-PS resin, respectively. Conjugate 1 showed considerable antibacterial activity in various conditions, whereas conjugate 2 did not exhibit antibacterial activity. The growths of various bacteria were inhibited by conjugate 1 even at lower concentrations than MIC. Conjugate 1 killed bacteria at MIC and had a potent synergistic effect with current antibacterial agents such as vancomycin and tetracycline, respectively. Overall results indicate that polymer surface modification using antibacterial ${\beta}$ sheet peptide is a powerful way to prevent microbial contamination on polymer surfaces.

  • A new series of substituted phenazone derivatives has been prepared through a series of reactions that are illustrated in Scheme I. The antibacterial and antineoplastic activities of the prepared compounds were evaluated. While none of the synthesized products showed marked antibacterial activity, all of them possessed a significant antitumor effect.

Synergistic Killing Effect of Synthetic Peptide P20 and Cefotaxime on Methicillin-Resistant Nosocomial Isolates of Staphylococcus aureus

  • The salt resistance of antibacterial activity and synergistic effect with clinically used antibiotic agents are critical factors in developing effective peptide antibiotic drugs. For this reason, we investigated the resistance of antibacterial activity to antagonism induced by NaCl and $MgCl_2$ and the synergistic effect of P20 with cefotaxime. P20 is a 20-residue synthetic peptide derived from a cecropin A (CA)-melittin(ME) hybrid peptide. In this study, P20 was found to have potent antibacterial activity against clinically isolated methicillin-resistant Staphylococcus aureus (MRSA) strains without hemolytic activity against human erythrocytes. The combination study revealed that P20 in combination with cefotaxime showed synergistic antibacterial activity in an energy-dependent manner. We also confirmed the synergism between P20 and cefotaxime by fluorescence-activated flow cytometric analysis by staining bacterial cells with propidium iodide (PI) and bis-(1,3-dibutylbarbituric acid) trimethine oxonol (BOX). This study suggests that P20 may be useful as a therapeutic antibiotic peptide with synergistic effect in combination with conventional antibiotic agents.
