Characterization of Antibacterial Activity and Synergistic Effect of Cationic Antibacterial Peptide-resin Conjugates

  • We synthesized peptide-resin conjugates (1 and 2) by immobilizing ${\beta}$-sheet antibacterial peptide and ${\alpha}$ helical antibacterial peptide on PEG-PS resin, respectively. Conjugate 1 showed considerable antibacterial activity in various conditions, whereas conjugate 2 did not exhibit antibacterial activity. The growths of various bacteria were inhibited by conjugate 1 even at lower concentrations than MIC. Conjugate 1 killed bacteria at MIC and had a potent synergistic effect with current antibacterial agents such as vancomycin and tetracycline, respectively. Overall results indicate that polymer surface modification using antibacterial ${\beta}$ sheet peptide is a powerful way to prevent microbial contamination on polymer surfaces.

  • In the preceding paper, we reported the synthessis and biological properties of cephalosporins having quinoliniumthiomethyls at the C-3 position and demonstrated that quinoliniumthiomethylcephalosporins exhibited an extended antibacterial spectrum including antipseudomonal activity, especially strong antiGram-positive activity. With the aim of elaboration to optimize the antibacterial potency of the quaternary ammonium cephalosporins, we have studied the modification of substituents in the pyrrolidine ring of cefepime 1 which is characterized by its potent activity.

  • The syntheses of mercaptotetrazoles and cephalothin analogs are described. Their in vitro potency was established. The compounds exhibited high antibacterial activity against Gram-positive bacteria and moderate activity against Gram-negative bacteria.

렌넷 커드 내 히스타민 생성에 관한 프로바이오틱 유산균이 생산한 항균 물질의 영향 (Effect of antibacterial substances produced by probiotic lactic acid bacteria on histamine formation in rennet curd)

  • 본 연구에서는 히스타민을 생성하는 유산균과 이에 항균 활성을 나타내는 프로바이오틱 유산균을 혼합 접종하여 제조한 렌넷 커드 내 항균물질과 히스타민 생산에 영향을 미치는 인자를 조사하였다. 프로바이오틱 유산균인 Lactobacillus plantarum FIL20과 Lactobacillus sakei PIL52는 히스타민 생성균인 Lactobacillus brevis LAS129, Enterococcus faecium SBP12 및 Enterococcus faecalis SBP58에 대해 강력한 항균 물질을 생산하였고, 프로바이오틱균이 생산한 유산과 박테리오신의 항균 활성은 농도의존적이었다. 렌넷 커드 제조를 위해 접종한 프로바이오틱 유산균의 균수가 많을수록 항균 물질 생성량이 증가되어 히스타민 생성균에 대한 항균 활성도 높아졌다. 프로바이오틱 유산균과 히스타민 생성균은 NaCl 10% 첨가에 의해 증식이 억제됨으로써 렌넷 커드 내에 항균 물질과 히스타민의 함량은 대조구보다 유의하게 낮았다(P < 0.05). 한편, 프로바이오틱 유산균과 히스타민 생성균을 혼합하여 제조한 렌넷 커드를 $25^{\circ}C$에서 5일간 저장 했을 때 히스타민의 함량은 유의하게 증가되지 않았으나, $20^{\circ}C$에서 20일간 저장한 경우에는 프로바이오틱 유산균이 생산한 항균 물질의 활성이 감소됨에 따라 렌넷 커드 내에 히스타민의 함량이 유의하게 증가되었다(P < 0.05).

  • 건강한 닭의 맹장에서 분리한 Lactobacillus sakei LAB 2와 LAB 8의 생균제로 이용을 알아보기 위해 기본적인 특성과 면역활성을 시험하였다. LAB 2와 LAB 8은 높은 내산성과 내담즙성을 가지고 있으며, 1차 대사산물인 유기산에 기인한 항균활성을 보였다. Salmonella 종을 병원성균으로 이용하여 최대 항균력을 나타내는 시간을 시험한 결과 48시간으로 나타났으며, 생성된 유기산 중 젖산의 생산량은 다른 Lactobacillus 균주보다 높게 조사되었다. In vitro 모델로 알아본 면역증진효과는 면역관련 세포의 증식과 사이토카인의 생산량 증가로 확인하였다. 이에 Lactobacillus sakei LAB 2와 LAB 8은 유용한 생균제로 개발할 수 있는 균으로 생각된다.

  • 3-[(1-Methyl-1H-tetrazol-5-y)thiomethyl] is one of the most important side chains in cephaloporins. This side chain which occures in numerous antibacterial agents, suc as cefamandole, latamoxef, cefoperazone, and cefmenoxime, contributes remarkable potency and broad spectrum. For the continuous work of study on the development of broad spectrum cephalosporins, we become interested in the effect of structural modification of the substituent at the N-1 position of mercaptoterazole toward biological activity because our recent work had demonstraated that the arylsubstituted mercaptotetrazoles (Fig. 1) had favorable effect on the activity against gram-positive bacteria. The main focus of this report was to investigate the relationship between activity and functional groups in the mercaptotetrazole.

  • Syntheses of cefotaxime analogs with halosubstituted phenyltetrazolthiomethyl at the $C_{3^-}$position are described. Their in vitro potency was established. The compounds exhibited a broad antibacterial spectrum. Some of these compounds showed activity against Gram-positive bacteria superior to the parent cefotaxime. Against Gram-negative bacteria, these compounds are less effective than cefotaxime.

Functional Characteristics of Kombucha Fermented with Lactic Acid Bacteria, Yeast, and Acetic Acid Bacteria Derived from Korea Traditional Foods

  • In this study, to determine the importance of lactic acid bacteria (LAB) in Kombucha fermentation, biological functions, such as organic acid production and anti-inflammatory and antibacterial activities, of Kombucha, with or without LAB inoculation, were evaluated. Lactobacillus paracasei DK215, Saccharomyces cerevisiae C3, and Acetobacter pasteurianus P2 were selected as the inoculants. Organic acids were measured every 3 days from the end of fermentation using HPLC; the organic acid content of LAB-inoculated Kombucha was relatively high. Samples with or without LAB inoculation showed high antibacterial activity against Escherichia coli. The MTT assay results indicated no significant difference in concentration difference and cell death. In the NO production test, compared with the uninoculated Kombucha sample, the LAB-inoculated Kombucha sample exhibited a value similar to that of the group without LPS treatment. The levels of cytokine (IL-1α, IL-6, TNF-α) production were significantly lower than those of the LPS(+) group, indicating the anti-inflammatory activity potential of the Kombucha sample. This improvement in the biological function of the LAB-inoculated Kombucha further verifies the value of LAB in the fermented food and beverage industry.
