Effects of Banhahubak-Tang Extract (BHTe) on Sociopsychological Stress (반하후박탕(半夏厚朴湯)의 사회·심리적 스트레스에 대한 실험적 연구(實驗的 硏究))

  • Objectives: This study is designed to investigate the effects of Banhahubak-Tang Extract (BHTe) on stress. Methos: BHTe was fed to ICR male mice ($20{\pm}2g$) orally with the dose of 100 mg/kg/day for five days. Mice were then exposed to sociopsychological stress by observing foot shock stressed mice for one hour for five days while restrained. Results: 1) The BHTe-administered group showed a tendency of decreasing of serum corticosterone secretion compared with the control group. 2) The BHTe-administered group showed an increase in noradrenalin secretions in the dorsal cortex of the brain, but it was not significant. 3) BHTe administration had no effect on the brain level of lipid peroxidation. 4) BHTe administration decreased the serum level of lipid peroxidation. 5) BHTe administration decreased the Cu,Zn-SOD in the brain. 6) BHTe administration had no effect on catalase activity in the brain. 7) BHTe administration had no effect on the brain level of GSH. 8) BHTe administration increased the serum level of DPPH free radical scavenging activity, but not significantly enough to make a comparison with BHT. 9) The elevated plus-maze test is designed to detect the effect of anxiolytic drugs. The BHTe group showed a significant increase in latency time. Conclusions: These results suggest that BHTe can effectively rid the subject of the effects of sociopsychological stress.

Comparison of Marker Components and Biological Activities of Gamiguibi-tang(Jiaweiguipi-tang) Decoction and Commercial Extract Granules (가미귀비탕 탕액과 시판제제의 성분 및 생리활성 비교)

  • Gamiguibi-tang (GGBT) is a traditional herbal medicine generally used to treat anemia, insomnia, anxiety, and nervousness. GGBT is being commercially produced in the form of extract granule and the quality control methods are specified in the Korean Herbal Pharmacopeia (KHP). However, there is no method to simultaneously analyze compound preparations. In this study, a HPLC method was developed and validated for the simultaneous determination of marker compounds in GGBT. And the contents of marker components and biological activities of the commercial GGBT extract granules (GGBT-2 and GGBT-3) were compared with those of the GGBT decoction (GGBT-1). We confirmed the robustness of simultaneous analytical method by monitoring the contents of the commercial GGBT products and carrying out validation. The marker components of GGBT were geniposide ($8.03{\sim}12.70{\mu}g/mL$), paeoniflorin ($2.79{\sim}4.25{\mu}g/mL$) and glycyrrhizic acid ($5.06{\sim}6.30{\mu}g/mL$). DPPH and ABTS radical scavenging activities were 47.34~63.17% and 21.52~33.61% in the GGBT products concentration of $1,000{\mu}g/mL$, respectively. The GGBT products significantly decreased NO, iNOS and COX-2 production in lipopolysaccharide (LPS)-induced RAW 264.7 macrophages in a concentration-dependent manner. The GGBT-2 had higher contents of marker components and biological activities than GGBT-1 and GGBT-3. The research suggest that be used in developing quality control methods for enhancing the quality of herbal medicines.

Development of Elderly Diet Using Inhibitory Plant Against Aging Process - Optimization for preparation conditions of Barley gruel with Green Tea - (노화억제 식물을 이용한 노인식 개발 - 녹차를 이용한 보리죽의 제조조건의 최적화 -)

  • Green Tea consists of 15-30% catechins (a type of polyphenol), which act as super antioxidants, inhibitory action against aging process. Antioxidants fight radical-free oxygen, an agent which can begin the process of cancer by damaging essential body chemicals and harming DNA. This study was peformed to develop the elderly diet using Green Tea as an inhibitory action against aging process. Nokchaborijook (Baley gruel added rice powder and Green Tea) was manufactured by various levels of Green Tea(1, 4, 7%) and the grinding period(15, 30, 45 sec). The optimum levels of added Green Tea and grinding periods on Nokchaborijook were determined with the results of sensory evaluation by response surface methodology and analysis of composition. The Anti-oxidant Vitamin A, C, E and Flavonoid were increased with increased levels of Green Tea and grinding periods be decreased. As the levels of Green Tea and grinding periods were increased, the green color and penetrated force became stronger. Among the sensory attributes, Bitterness, Green Tea Flavor and Hashness were greater depending on increasing Green Tea. Jujube flavor was greater relying on decreasing Green Tea. Greenness and spreadability of particles were increased as both the amounts of Green Tea and the grinding period were increased. This result was used to determine the optimum conditions of adding levels of Green Tea and grinding periods. The optimum conditions of Nokchaborijook was established as adding of 5.8% Green Tea grinded for 15 seconds.

Efficacy of Uniconazole as a Phytoprotectant Against $SO_2$ Injury in Snap Bean (강남콩에 대한 $SO_2$ 피해경감제로서 uniconazole의 효과에 관한 연구)

  • This study was conducted to determine the efficacy of using uniconazole,[(E)-1-(4-chlorophenyl)-4,4-dimethyl-2-(1,2,4-triazole-1-yl)-1-penten-3-ol)] as a phytoprotectant against $SO_2$ injury in snap been (Phaseolus vulgaris L. 'Strike'). Thirteen days prior to $SO_2$ fumigation, plants were given a 100 ml soil drench of uniconazole solution at concentrations of 0.02, 0.10, 0.25 and 0.50 mg/pot. All four uniconazole concentrations were significantly effective in providing protection against $SO_2$ exposure(3 h at 1.5 ppm), but uniconazole treatment above 0.02 mg/pot severely reduced stem elongation, leaf enlargement, flowering date and pod number and weight. Uniconazole treatment had little or no effect on stomatal conductance but reduced transpiration rate on a whole plant basis by nearly 40%. This may reflect an alteration in canopy structure by reducing stem elongation and leaf enlargement. Although uniconazole did not increase the activities of superoxide dismutase(SOD) and peroxidase(POD) in non-$SO_2$-fumigated plants, it significantly increased those enzyme activities in $SO_2$-fumigated plants. Chlorophyll concentration on the basis of unit area was increased 50-60% by uniconazole. However, the difference was not detected on the basis of dry weight. $SO_2$ increased variable chlorophyll fluorescence (Fv) 48% after 1.5 h of exposure in non-uniconazole treated plants but decreased Fv in the plants after 3 h of exposure. By appliing uniconazole, it was possible to maintain high Fv values in the latter group of plants. These results suggest that the phytoprotective effects of uniconazole are related to its growth-retarding properties as an anti-gibberellin as well as the increase of activites of free radical scavengers such as SOD and POD.

Physiological Activities of Cudrania tricuspidata Extracts (Part I) (꾸지뽕나무 추출물의 생리 활성(제1보))

  • Cudrania tricuspidata has been used for a long time as a traditional herb medicine in Korea nad China. This paper has shown the experimental results about the physiological activities of water-, ethanol-, ethyl acetate-soluble fractions from ethanolic extracts of leaves, stems and roots of Cudrania tricuspidata. The effects of these fractions on the growth of various cells have exhibited that the ethyl acetate fractions from leaves, stems and roots inhibited significantly the growths of macrophage(RAW 264.7 cell), melanoma cell(B16-F10 cell), fibroblast cell(CCD-986sk cell), and lung carcinoma cell(A549 cell). The water and ethanol fractions of leaves and ethanol fraction of stems demonstrated better antioxidant activities scavenging radicals than other fractions when compared with the concentrations of different fractions for scavenging free radical DPPH (di(phenyl)-(2,4,6-trinitrophenyl) iminoazanium).

Protective Effect of Oenanthe javanica Extract on Acetaminophen-induced Hepatotoxicity in Rats (Acetaminophen으로 유도한 쥐의 간 독성에 대한 미나리(Oenanthe javanica) 추출액의 간 보호 작용)

  • The hepatoprotection by the methanol extract of Oenanthe javanica DC (water dropwort) (OJME) was investigated in Sprague Dawley rats with inducing liver damage by acetaminophen. After OJME administration for 1 week, the increase of hepatic lipid peroxide level by acetaminophen-induced hepatotoxicity was significantly reduced. In case of phase I microsomal enzyme systems including cytochrome P-450, aminopyrine N-demethylase and aniline hydroxylase, any significant differences between in control and in OJME-pretreated group was observed after acetaminophen treatment. However, the pretreatment of OJME maintained the hepatic glutathione level and the activity of liver cytosolic glutathione S-transferase, which was significantly decreased by the acetaminophen intoxication. Among the glutathione-generating system, glutathione reductase was more responsible for its biosynthesis rather than ${\gamma}-glutamylcystein$ synthetase. OJME itself showed the strong inhibition activity on DPPH radical generation. In conclusion, OJME administration maintains the liver glutathione pool and hepatic glutathione S-transferase activity, in addition with its high anti-oxidative capability, to show hepatoprotective effect from acetaminophen intoxication.

Wound Healing Effect of Curcumin Gel for Transdermal Delivery (경피전달을 위한 커큐민 젤의 창상치유효과)

  • The aim of this study was to investigate the effect of trasdermal delivery of the curcumin gel on healing of the rats' dorsum wounds. Carbopol 934 and propylene glycol were used to prepare gels containing 1% curcumin. Curcumin gel was evaluated for various properties such as antioxidant, cell viability, anti-inflammatory, in vivo wound healing. The free radical scavenging activity using 1,1-diphenyl-2-picryl hydrazyl (DPPH) was 50% at 12.5 ppm concentration. Inhibition of nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells by curcumin gel. In the curcumin gel-treated group, the re-epithelialization in wounds was significantly increased compared to the control group throughout the experimental period. These results suggested that curcumin may be helpful for the promotion of wound healing.

Physiological and Pharmacological Activites of Nutraceutical Tea by Leaves and Flowers of Domestic Camellia(Camellia japonica)

  • This project was conducted to development several camellia tea mixed herb teas having any physiological effects. Leaves of tea tree contain many compounds, such as polysaccharides, volatile oils, vitamins, minerals, purines, alkaloids(eg. caffeine) and polyphenols(catechins and flavonoids). Although all three tea types(green, oolonr and black) have antibacterial and free radical capturing(antioxidizing) activities, the efficacy decreases substantially the darker the variety of tea is. This is due to lower contents of anti-oxidizing polyphenols remaining in the leaves. Unlike tea tree(Camellia sinensis), the biochemical features and effects of camellia(Camellia japonica) are not well known. Fresh mature leaf of sasanqua camellia(C. sasanqua), roasted young leaf tea(C. japonica) and fresh mature leaf and bark of camellia had high antibacterial activity against P. vulgaris and B. subtilis. In antifungal activity bioassay, young leaf roasted teas of camellia and sasanqua camellia had high activity against C. albicans and T. beigelil. Plant extracts from Camelia japonica had higher inhibitory activity against fungi than against bacteria. In cytotoxic effect against human acute myelogenous leukaemia cell extracts including fresh leaf(200$\mu\textrm{g}$/m1), bark(230$\mu\textrm{g}$/ml) and flower tea (320$\mu\textrm{g}$/m1)inhibited growth of AML cells.(중략)

Stem bark of Maackia amurensis Extract according to extraction Solvent (추출용매에 따른 다릅나무 수피 추출물의 항산화 활성)

  • Objectives : The objective of this research was to investigate the antioxidant activities of stem bark of Maackia amurensis extract.Methods : Stem bark of Maackia amurensis extract were prepared using 70% methanol. Methanol extracts were fractionated to hexane, chloroform, ethyl acetate, butyl alcohol, water fractions and investigated. The antioxidant activities of fractions was evaluated by four different assays as total polyphenol contents, total flavonoid contents, DPPH(1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity and ABTS(2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) scavenging ability.Results : The yield of methanolic extracts from stem bark of Maackia amurensis was 10.16%, whereas those of its solvent fractions (hexane, chloroform, ethyl acetate, butyl alcohol, and water) were 5.45, 11.39, 13.88, 26.07, and 40.80%, respectively. The total polyphenol contents and electron donating ability of 70% methanol extracts from stem bark of Maackia amurensis were 15.44 mg/g and 194.15 μg/mL of its IC50, respectively. The 70% methanol extracts showed the highest antioxidant activity. The total polyphenol content and total flavonoid content of chloroform fractions were higher in each of 201.98 mg/g and 13.55 mg/g. The chloroform fraction showed the lowest levels of DPPH(IC50, 183.95 μg/mL) and ABTS scavenging activity(IC50, 10.0 μg/mL). The antioxidant activity was detected in methanol extract, chloroform fractions.Conclusions : These results indicate that 70% methanol extract and its fractions of stem bark of Maackia amurensis, especially chloroform fraction, have the properties of anti-oxidant suggesting stem bark of Maackia amurensis may be a candidate for natural and functional materials.

Structural, Electrochemical, DNA Binding and Cleavage Properties of Nickel(II) Complex [Ni(H2biim)2(H2O)2]2+ of 2,2'-Biimidazole

  • A nickel(II) complex $[Ni(H_2biim)_2(H_2O)_2](ClO_4)_2{\cdot}H_2O$ (1) of biimidazole ligand has been synthesized and characterized (Where $H_2biim$ = 2,2'-biimidazole). The single crystal X-ray diffraction of the complex shows a dimeric structure with six coordinated psudo-octahedral geometry. The cyclic voltammograms of complex exhibited one quasireversible reduction wave ($E_{pc}=-0.61V$) and an irreversible oxidation wave ($E_{pa}=1.28V$) in DMF solution. The interaction of the complex with Calf-Thymus DNA (CT-DNA) has been investigated by absorption and fluorescence spectroscopy. The complex is an avid DNA binder with a binding constant value of $1.03{\times}10^5M^{-1}$. The results suggest that the nickel(II) complex bind to CT-DNA via intercalative mode and can quench the fluorescence intensity of EB bind to CT-DNA with $K_{app}$ value of $3.2{\times}10^5M^{-1}$. The complex also shown efficient oxidative cleavage of supercoiled pBR322 DNA in the presence of hydrogen peroxide as oxidizing agent. The DNA cleavage by complex in presence of quenchers; viz. DMSO, KI, $NaN_3$ and EDTA reveals that hydroxyl radical or singlet oxygen mechanism is involved. The complex showed invitro antimicrobial activity against four bacteria and two fungi. The antimicrobial activity was nearer to that of standard drugs and greater than that of the free ligand.
