왕고들빼기 메탄올 추출물의 항산화성 (Antioxidant Activity of Methanol Extracts from $Lactuca$ $indica$)

  • 본 연구에서는 왕고들빼기의 비타민 C, 총 폴리페놀, 총 플라보노이드 함량과 메탄올 추출물의 항산화력을 측정하였다. 왕고들빼기 잎과 뿌리의 비타민 C 함량은 각각 24.14 mg/100 g, 0.38 mg/100 g으로 나타났다. 왕고들빼기 잎의 총 폴리페놀 함량과 총 플라보노이드 함량은 각각 42.08 mg/g, 23.09 mg/g로 나타났으며, 왕고들빼기 뿌리의 총 폴리페놀 함량과 총 플라보노이드 함량은 각각 7.66 mg/g, 0.77 mg/g로 나타났다. DPPH radical 소거능은 왕고들빼기 잎의 경우 추출물 농도가 300 ${\mu}g$/mL 일 때 약 78%로 높은 항산화 능력을 보였으며, 뿌리에 비해 높은 활성을 나타냈다. ABTS radical 소거능은 추출물 농도가 5 mg/mL 일 경우 잎과 뿌리 각각 92.3%, 10.21%로 나타났다. Reducing power(환원력)를 측정한 결과 잎 추출물 농도가 1.6 mg/mL 일 때 1에 가까운 흡광도 값을 보였고, $Fe^{2+}$-chelating 효과는 추출물 농도가 14 mg/mL 일 때 잎과 뿌리 각각 97.17%, 34.26%의 활성을 나타냈다. 왕고들빼기의 항산화 활성의 $IC_{50}$을 측정한 결과 잎의 경우 DPPH radical 소거능, ABTS radical 소거능, $Fe^{2+}$-chelating 효과의 $IC_{50}$은 각각 0.19 mg/mL, 2.7 mg/mL, 6.27 mg/mL로 나타났다. 왕고들빼기 뿌리의 경우 잎에 비해 그 값이 높게 나타났다. 이상의 결과에서 왕고들빼기의 잎은 뿌리에 비해 비타민 C, 폴리페놀, 플라보노이드 함량이 높고 우수한 항산화 활성을 보여 기능성 식품 및 기능성 식품재료로써의 이용가능성이 매우 높은것으로 확인되었다.

부자탕 추출물이 골관절염 동물 모델에 미치는 영향 (Effects of Buja-tang Extract on Osteoarthritic Animal Model)

  • Objectives The present study was designed to find out the therapeutic effects and possible underlying mechanism of Buja-tang, a herbal complex formula on experimental monosodium iodoacetate (MIA)-induced osteoarthritis. Methods Osteoarthritis models were created via intra-joint injection of MIA (50 μL with 80 mg/mL) in rats. Rats were divided into five groups and each group consisted of seven. Normal group was not injected MIA and did a normal diet. Control group injected MIA and received distilled water. Indo injected MIA and oral administration of 5 mg/kg of indomethacin. BJTL injected MIA and oral administration of 100 mg/kg of Buja-tang. BJTH injected MIA and oral administration of 200 mg/kg of Buja-tang. We analyzed weight-bearing ability of hind paws, oxidative stress related factor, antioxidant protein, inflammatory protein, inflammatory messenger and cytokine in joint tissue. Pathological observation of knee cartilage tissue structures was also performed with hematoxylin & eosin and safranin-O chromosomes. Results Weight-bearing ability of hind paws showed a tendency to reduce pain. The incidence of nicotinamide adenine dinucleotide phosphate oxidase and p22phox in articular tissue was significantly reduced, and the incidence of nuclear factor-erythroid 2-related factor 2 and heme oxygenase-1 and superoxide dismutases was significantly increased. The incidence of phosphorylated inhibitor of κBα, nuclear factor-kappa B p65, inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis factor alpha, interleukin (IL)-6, and IL-1β decreased significantly. In pathological observation, cartilage tissue damaged by MIAs in biopsy has significantly recovered from Buja-tang administration. Conclusions Buja-tang has anti-inflammation, antioxidation and pain relief effects. So this is thought to inhibit the progress of osteoarthritis in rat caused by the MIA.

케나프 장대 품종의 부위별 에탄올 추출물의 항혈전 및 항산화 활성 (Evaluation of Antithrombosis and Antioxidant Activities of the Ethanol Extract of Different Parts of Hibiscus cannabinus L. cv. 'Jangdae')

  • 세계 4대 섬유작물중의 하나로 알려진 케나프(Hibiscus cannabinus L.)는 탄소 저감용 및 친환경 생활소재용으로 각광받고 있으며, 2013년 국내 채종 가능한 '장대' 품종이 확립되면서 전국적으로 재배되고 있다. 그러나, 케나프의 부위별 생리활성, 특히 '장대'의 부위별 생리활성 평가는 미미한 실정이다. 본 연구에서는 케나프 '장대'의 종실(seed), 종자 꼬투리(seedpod), 잎(leaf), 줄기(stem), 뿌리(root) 에탄올 추출물을 조제하여 이의 항혈전 및 항산화 활성을 평가하였다. 그 결과 종실 추출물에서 강력한 혈액응고인자 저해활성을, 종자 꼬투리 추출물에서 프로트롬빈 저해 활성을, 줄기 추출물에서 트롬빈 저해활성을 확인하였다. 또한 종자 꼬투리 추출물에서는 항혈전 활성과 연관된 DPPH 음이온 소거능, ABTS 양이온 소거능, 환원력 및 nitrite 소거능이 여타의 부위 추출물보다 유의적으로 강력한 활성을 나타냄을 확인하였다. 본 연구결과는 국내 케나프 친환경 섬유산업의 부산물로 얻어지는 종자 꼬투리, 뿌리 및 줄기를 이용한 고부가가치 생물소재 개발이 가능함을 제시하고 있다.

들깨박 추출물의 항산화 및 항노화 효과 (Antioxidant and Anti-aging Activities of Ethanol Extracts from Defatted Perilla frutescens)

  • 목적: 본 연구는 들깨 부산물의 이용가치를 높이는 방법으로 들깨박 에탄올 추출물의 항산화 작용, collagenase와 elastase 활성의 저해작용을 통하여 항노화 활성이 있는지를 연구하였다. 그 외에 기능성 화장품 소재로서의 이용가능성을 연구도하였다. 방법: 들깨박 추출물의 항산화 기능을 연구하기 위하여 DPPH 자유기의 소거능력을 분석하여 electron spin resonance (ESR)를 광도계로 측정하였다. 결과: 들깨박의 에탄올 추출물의 diphenyl-β-picrylhydrazyl (DPPH) 자유기 소거의 항산화 기능은 추출물 농도 10 ㎍/ml에서는 약 68%, 20 ㎍/ml에서는 85%, 40-100 ㎍/ml에서는 90% 이상의 자유기의 소거 활성을 보여서 항산화능이 있음을 나타냈다. collagenase의 저해활성은 에탄올 추출물 농도 20 mg/ml에서는 4.1%, 40 mg/ml에서는 12%, 100 mg/ml에서는 26.7%로 증가하였다. 저해활성이 농도에 비례하여 증가하였다. elastase의 저해활성은 에탄올 추출물 농도 20 mg/ml에서는 3.8%, 40 mg/ml에서는 15%, 80 mg/ml에서는 28%, 100 mg/ml에서는 35.7%로 증가하였다. 결론: 이상의 연구결과에서 들깨박의 에탄올 추출물이 항산화효과, collagenase와 elastase의 활성을 저해하여 피부의 노화와 주름의 개선효과를 나타낼 수 있다고 본다.

Cordycepin Enhanced Therapeutic Potential of Gemcitabine against Cholangiocarcinoma via Downregulating Cancer Stem-Like Properties

  • Cordycepin, a valuable bioactive component isolated from Cordyceps militaris, has been reported to possess anti-cancer potential and the property to enhance the effects of chemotherapeutic agents in various types of cancers. However, the ability of cordycepin to chemosensitize cholangiocarcinoma (CCA) cells to gemcitabine has not yet been evaluated. The current study was performed to evaluate the above, and the mechanisms associated with it. The study analyzed the effects of cordycepin in combination with gemcitabine on the cancer stem-like properties of the CCA SNU478 cell line, including its anti-apoptotic, migratory, and antioxidant effects. In addition, the combination of cordycepin and gemcitabine was evaluated in the CCA xenograft model. The cordycepin treatment significantly decreased SNU478 cell viability and, in combination with gemcitabine, additively reduced cell viability. The cordycepin and gemcitabine co-treatment significantly increased the Annexin V+ population and downregulated B-cell lymphoma 2 (Bcl-2) expression, suggesting that the decreased cell viability in the cordycepin+gemcitabine group may result from an increase in apoptotic death. In addition, the cordycepin and gemcitabine co-treatment significantly reduced the migratory ability of SNU478 cells in the wound healing and trans-well migration assays. It was observed that the cordycepin and gemcitabine cotreatment reduced the CD44highCD133high population in SNU478 cells and the expression level of sex determining region Y-box 2 (Sox-2), indicating the downregulation of the cancer stem-like population. Cordycepin also enhanced oxidative damage mediated by gemcitabine in MitoSOX staining associated with the upregulated Kelch like ECH Associated Protein 1 (Keap1)/nuclear factor erythroid 2-related factor 2 (Nrf2) expression ratio. In the SNU478 xenograft model, co-administration of cordycepin and gemcitabine additively delayed tumor growth. These results indicate that cordycepin potentiates the chemotherapeutic property of gemcitabine against CCA, which results from the downregulation of its cancer-stem-like properties. Hence, the combination therapy of cordycepin and gemcitabine may be a promising therapeutic strategy in the treatment of CCA.

병풀(Centella asiatica) 및 아시아티코사이드는 미토콘드리아 호흡 및 TLR4 경로를 통해 H2O2 유도 세포염증 조절 (Centella asiatica and Asiaticoside Regulate H2O2-induced Cellular Inflammation via Mitochondrial Respiration and the TLR4 Pathway)

  • 이 연구는 인간진피섬유아(HDF)세포에서 병풀 및 아시아티코사이드가 H2O2 유래 세포주기 정지기, 미토콘드리아 활성 및 염증성 사이토카인에 미치는 영향을 조사하였다. 병풀 80% 메탄올 추출물, 에틸아세티이트 분획물 및 병풀의 대표물질인 아시아티코사이드를 사용하였다. 병풀 추추물, 에틸아세테이트 분획 및 아시아티코사이드로 처리한 세포는 낮은 수준의 TNF-α 및 IL-6을 분비하였고, 아시아티코사이드의 항산화 효과는 병풀 추출물 및 에틸아세테이트 분획물보다 높았다. 아시아티코사이드 처리는 미토콘드리아의 막포텐셜을 증가시키고, 미토콘드리아를 정상으로 되돌렸다. 스트레스 유도 후 에틸아세테이트 분획물 및 아시아티코사이드에 의해 미토콘드리아 산소 소비율이 증가하였고, TLR4-MyD88-TRAF6-p65 경로가 재감소하였다. 이러한 결과는 병풀 추출물, 에틸 아세테이트 분획 및 아시아티코사이드가 HDF 세포의 미토콘드리아 활성을 조절할 뿐 아니라 항산화 및 항염증에 효과 있음을 시사한다.

슈퍼자미의 이화학적 특성 및 항산화 활성 (Physicochemical Properties and Antioxidant Activity of Superjami)

  • This study attempted to investigate the characteristics of Superjami, which had a high C3G (cyandin-3-glucoside) content by comparing it with common rice (Ilpum) and black rice (Heougjinju, Suwon 415) for their components and physio-chemical characteristics. There were no significant differences in the water content, however there were significant differences in crude protein and crude fat in the order of Ilpum < Superjami < Heougjinju. As for the morphological characteristics of grains, it had been learned that Superjami was heavier and longer than the common rice, moreover Superjami had bigger seeds. As for the hardness of grains, the longer the time of steeping, the more decreased their hardness was at large. In the case of color, they showed significant differences in all the L, a, and b values of grains and flours. As a result of the experiment of pigment elution after steeping of the rice flours at $20^{\circ}C$ and $40^{\circ}C$, the longer the time steeping, the more increased the pigment elution became. The values are bigger with Heougjinju and Superjami than with Ilpum. As for the elution pH after rice steeping, the longer the time of steeping, the lower pH all the assays tended to have. As a result of the analysis of the total polyphenol contents of Ilpum, Heougjinju, and Superjami, it had turned out that the total polyphenol contents of Heougjinju and Superjami are 1.2 times as high as Ilpum, a common rice, and that Superjami is significantly higher than Heougjinju. As a result of the experiment of DPPH radical scavenging ability, there are significant differences among the assays in the order of Superjami > Heougjinju > Ilpum. Futher, it has turned out that Superjami has a higher DPPH radical scavenging ability than Heougjinju. Consequently, it can be stated that Superjami has a strong anti-oxidative ability. Thus, we should more precisely grasp the cooking characteristics of Superjami, which is in the state of brown rice, via comparing it with a common brown rice, and also provide opportunities to apply Superjamii to more foods by studying its starch characteristics in addition to its grain and flour properties.

혈당저하를 위해 처방된 천연물 복합제의 항산화 활성 및 3T3-L1 세포의 분화에 미치는 영향 (Anti-oxidative activity of the herb mixture prescribed to induce blood glucose level and effect on the differentiation of 3T3-L1 fibroblast)

  • 혈당저하를 위한 천연물복합제를 한의학 처방 및 문헌 등을 기초로 하여 구성하고, 기능성 식품 소재로서의 이용가능성을 조사하기 위하여 추출물의 이화학적 특성을 조사하였다. 천연물복합제 5가지 추출물의 수율은 $19.52{\sim}29.79%$의 범위로 나타났으며, 총페놀성 화합물 함량은 $349.24{\sim}1752.21\;mg%$, 총 플라보노이드 함량은 $163.06{\sim}1,118.47\;mg%$의 범위로 조사되었다. 특히 2번, 3번 및 5번 복합제에서 1,000 mg% 이상의 높은 함량을 나타내었으며, 전자공여능 또한 이들 복합제에서 높은 활성을 보여주었다. 아질산염 소거능은 2번 및 5번 천연물복합제가 pH 1.2에서 70% 이상의 높은 활성을 보여주었으며, pH가 증가할 수록 감소하였다. 인슐린성 물질의 함유정도를 조사하기 위하여 섬유아세포 3T3-L1의 지방세포로의 분화정도를 측정한 결과, 천연물복합제 5가지 추출물 중 5, 1, 4, 3, 2번의 순으로 높은 값을 나타내었다. 한편 이들 천연물복합제의 기능성 식품으로 이용 가능성을 검증하기 위한 추가적인 생리활성 및 효능 연구를 진행할 필요성이 있다고 판단된다.

Inhibition of MMP-2 and MMP-9 Activities by Limonium tetragonum Extract

  • Matrix metalloproteinases (MMPs) are crucial extracellular matrices degrading enzymes that take important roles in metastasis of cancer progression as well as other significant conditions such as oxidative stress and hepatic fibrosis. Natural products are on the rise for their potential to provide remarkable health benefits. In this context, halophytes have been of interest in the nutraceutical field with reported instances of isolation of bioactive compounds. In this study, Limonium tetragonum, an edible halophyte, was studied for its ability to inhibit MMP-2 and -9 using HT1080 fibrosarcoma cells. Results showed that L. tetragonum extract was able to inhibit the enzymatic activity and mRNA expression of MMP-2 and -9 according to gelatin zymography and RT-PCR assays, respectively, but it was not able to significantly change the MMP pathway related factors such as tissue inhibitors of metalloproteinases. Also, Mitogen-activated protein kinases pathway-related protein levels and their phosphorylation were assayed. While the phosphorylated p38 levels were decreased, extracellular signal-regulated kinase and c-Jun N-terminal kinase were not affected by L. tetragonum treatment. In conclusion, it was suggested that L. tetragonum contains substances acting as MMP inhibitors on enzymatic activity rather than intracellular pathway intervention, which could be useful for further utilization of L. tetragonum as a source for anti-MMP agents.

Effect of Dietary Grape Pomace Fermented by Saccharomyces boulardii on the Growth Performance, Nutrient Digestibility and Meat Quality in Finishing Pigs

  • Fifty-six [(Duroc${\times}$Yorkshire)${\times}$Landrace] pigs with an average initial BW of 19.3${\pm}$0.17 kg were used in this 15-wk growth experiment to investigate the effects of grape pomace fermented by Saccharomyces boulardii on pig growth performance, nutrient digestibility and quality attributes of pork. Pigs were allotted to 2 dietary treatments (7 replications) based on their initial BW in a randomized complete block design. The experimental treatments were: i) control (CON; basal diet), ii) FGPP (CON+30 g/kg fermented grape pomace product). Dietary FGPP improved (p<0.05) average daily gain (ADG), coefficient apparent total tract digestibility (CATTD) of dry matter (DM) and nitrogen (N) during 35-70 d of the experiment. Similarly, pigs fed the FGPP supplemented diet had a higher N digestibility (p<0.05) in the finisher phase (day 71-105). Dietary FGPP increased (p<0.05) the marbling score, the redness ($a^*$) and yellowness ($b^*$) values, as well as the anti-oxidative ability (lower TBARS). The inclusion of FGPP reduced palmitic acid (C:16:0), stearic acid (C:18:0), arachidic acid (C:20:0) and SFA levels (p<0.05) in subcutaneous fat. An increased (p<0.05) linoleic acid (C18:2n6), total PUFA and PUFA/SFA ratio were observed in the FGPP group. Dietary FGPP supplementation decreased the arachidic acid (C:20:0) level in longissimus muscle (LM). In conclusion, dietary inclusion of FGPP at the level of 30 g/kg improved the growth performance, nutrients digestibility and altered the fatty acid pattern in the subcutaneous fat as well as some attributes of pork meat.
