Background and Objectives : The aim of this study was to investigate the anti-oxidant, anti-allergic and anti-inflammatory ability of the Taglisodog-eum(SHE) extract on the RAW 264.7 and EL4 cells Materials and Methods : Three types of experiments were implemented for this study: first, the experiment to study the anti-oxidant effect of SHE using Riboflavin; second, in vitro experiment to investigate the inhibition of Th 2 cell differentiation by SHE using EL4 cells (IL-4 mRNA expression); third, the suppression of $NF-{\kappa}B$ activation using RAW 264.7 cells (iNOS and COX-2 mRNA expression). Results : The anti-oxidant ability of SHE were dose-dependantly increased. From in vitro, the LPS-induced iNOS and COX-2 mRNA expression were dose-dependantly decreased in the RAW264.7 cells treated with SHE and the PMA-induced IL-4 mRNA expression were also dose-dependantly decreased in EL4 cells. $NF-{\kappa}B$ activation was suppressed, and iNOS & COX-2 production were inhibited by SHE Conclusion : The results suggest that SHE has dose-dependant anti-oxidant ability, and has anti-allergic and anti-inflammatory effects through the suppression of $NF-{\kappa}B$ activation and the inhibition of Th 2 cell differentiation.
Background and Objectives : The aim of this study was to investigate the anti-oxidant and anti-inflammatory effects of the Taglisodog-eum(TSE) extract on the RAW264.7 cell Methods : The RAW264.7 cell was cultured using Dulbecco's modified Eagle's medium(DMEM, USA), including the 10% fetal-bovine serum(FBS; Sigma, USA) in a $37^{\circ}C$, 5% CO2 incubator. Results : The anti-oxidant ability of TSE were dose-dependantly increased. The LPS-induced IKK, iNOS and COX-2 mRNA expression were dose-dependantly decreased in the RAW264.7 cells treated with TSE. $NF-kB$ activation was suppressed. Conclusion : The findings in this study show that TSE has anti-oxidant and anti-inflammatory effects, such as the inhibition of $NF-kB$ activity.
Objectives : This study was investigated to analyze the anti-oxidant activity of water and ethanol extracts from the Magnoliae Flos. Methods : The effects on electron donating ability (EDA), SOD-like activity and xanthine oxidase inhibition have been investigated the anti-oxidant activity for utilization as a natural material of cosmetics. Results : This study was investigated to analyze the anti-oxidant activity of water and ethanol extracts from the Magnoliae Flos. In the electron donating ability test, water and ethanol extract from Magnoliae Flos showed an effect of 75.6% and 83.5% at 1,000 ppm concentration, respectively. In the superoxide dismutase (SOD)-like activity, 1,000 ppm of water extract showed an effect of 45.2% and ethanol extract showed an effect of 64.2%. Xanthine oxidase showed a meaningful inhibitory effect of 68.8% in water extract and 23.2% at 1,000 ppm in ethanol extract. Conclusion : From these results, Magnoliae Flos showed the development possibility as a natural material of anti-oxidant cosmetics.
Many herbs have been used as therapeutics in Korean traditional medicine. In view of their clinical indications, anti-oxidant activity may contribute to their pharmacological effects. However, anti-oxidant information on these plants has not been available. In this study, seventy herbs which have been used in Korean traditional medicine were selected and screened for anti-oxidant activity using their water extracts. The anti-oxidant activity was assessed by their ability to inhibit three oxidation reactions; luminol/Fenton reagent, 2, 7-dichlorodihydrofluorescein(DCHF)/Fenton reagent and DCHF/peroxynitrite. In each assay, 70 herbs were divided into two groups; anti-oxidant group which inhibited the respective oxidation reaction and was majority(about 60 herbs), and pro-oxidant group which enhanced the oxidation reaction but was minority(more or less 10 herbs). When the herbs were listed in the order of their anti-oxidant strength, the orders obtained from each assay were found to be quite similar. The upper top rankers(more or less 10 herbs) in each assay showed strong activity compared to the others. The uppermost rankers in each assay were Rubus coreanus Miquel/Rubus schizostylus(覆盆子), Schisandra chinensis Baillon/Schizandra chinensis(五味子) and Terminalia chebula Retziusl Terminalia chebula(訶子). Of the pro-oxidant herbs, about 4-5 herbs were strongly pro-oxidant, which enhanced the control oxidation reactions to 150-300%. But the meaning of this observation is not known since few of them in one assay were also anti-oxidant in other assays. The results obtained in the present study may serve as information for understanding pharmacological effects of these herbs and developing new drugs from them.
Objective: This experimental study was performed to investigate the anti-inflammation and anti-oxidant effects of Sopungdojeok-tang(SPDJT) and Samulsopung-san(SMSPS) which were used to treat patient with atopic dermatitis Methods: Anti-inflammation and anti-oxidant effects of SPDJT and SMSPS were measured by the inhibitory ability of Nitric oxide(NO) production and the scavenging for 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical. Results: 1. SPDJT and SMSPS were not showed cell toxicity. 2. In inhibitory effects against NO production, all groups of SPDJT were showed anti-inflammation, but all groups of SMSPS were not showed anti-inflammation. There were statistical significances between $20{\mu}g/m\ell$ and 4, $10{\mu}g/m\ell$ in SPDJT groups. 3. In DPPH radical scavenging activity, all groups of SPDJT and all groups of SMSPS were showed anti-oxidant effects. There were statistical significances between $20{\mu}g/m\ell$ SPDJT group and 4, $10{\mu}g/m\ell$ SMSPS groups. Conclusion: These results suggest that the SPDJT groups are better than the SMSPS groups in antiinflammation and anti-oxidant effects.
We recently reported that a water extract of laurel or turmeric, 1,8-cineole enriched fractions, showed hypolipidemic activity in the zebrafish model. Therefore, the present study investigated the cineole's anti-oxidant and anti-inflammatory activities in lipoprotein metabolism in vitro and in vivo. Cineole had inhibitory effects on cupric ion-mediated oxidation of lipoproteins in general, while simultaneously enhancing ferric ion removal ability in high-density lipoprotein (HDL). Hypercholesterolemia was induced in zebrafish using cholesterol-feeding treatment, 4% cholesterol, for 3 weeks. After feeding with or without the addition of cineole, the results revealed that cineole possessed lipid-lowering and anti-inflammatory activities in hypercholesterolemic zebrafish. In addition, serum amyloid A and interleukin-6 levels were lowered and lipid accumulation was decreased in the liver. Conclusively, 1,8-cineole was found to have anti-oxidant activities in lipoprotein metabolism both in vitro and in vivo with simultaneous reduction of lipid accumulation in the liver of zebrafish.
Objective : Sanguisorbae Radix(SO). that belong to Rosaceae is widely distributed in Asia including Korea, Japan and China. It has been used as traditional medicine from old times as a treatment for anti-inflammatory drugs. This study was designed to investigate the effects of n-BuOH fractions of SO on anti-oxidant effect and anti-microbial effect on $Streptococcus$$mutans$ ($S.$$mutans$). Methods : The anti-microbial effect of $n$-BuOH fractions of SO. was assessed by the paper disk diffusion method and anti-oxidant effect was assessed by the DPPH radical scavenging effect, Superoxide anion radical scavenging effect and SOD like ability. Results : DPPH radical scavenging of the $n$-BuOH fraction from SO in 50 ug/mL was shown to be Fr-2 (89.9%), Fr-3 (88.7%), Fr-4 (76.3%), Fr-1 (59.4%), Fr-5 (56.2%). Superoxside anion radical scavenging activity of the n-BuOH fraction from SO. in 50 ug/mL for Fr-3 was shown to be 78%. SOD-like activity of n-BuOH fraction from the SO in 1000 ug/mL for Fr-3 were shown to be 76.1% respectively. The $n$-BuOH fraction from the SO had high anti-microbial effect on $S.$$mutans$. Conclusion : As a result, the $n$-BuOH fraction from SO. has good anti-microbial and anti-oxidant effects in a concentration-dependent manner.
나비류 추출물은 전통적으로 다양한 활성을 보유하여 의약용으로 사용되어졌다. 5종의 나비(산제비나비[Papilio maackii], 호랑나비[Papilio xuthus], 배추흰나비[Pieris rapae], 남방호랑나비[Eurema hecabe], 왕오색나비[Sasakia charonda])를 이용하여 물, dimethly sulfoxide (DMSO), ehtanol 및 methanol로 추출한 추출물로 항산화 활성 및 Cox-2 promoter assay를 수행하였다. 그 결과 산제비나비의 추출물이 전반적으로 높은 항산화활성을 나타내었으며, Cox-2 promoter assay에서는 호랑나비의 DMSO 분획이 가장 저해활성이 높은 것으로 나타났다. 활성이 높은 추출물을 대상으로 분리 및 정제를 통하여 유용한 식의약 소재로의 기초연구가 더 필요하다.

The aim of this study was to elucidate the difference in the amount of phytochemicals and anti-oxidant activities of the freeze-dried 5th instar 3rd day silkworm powder (FDSP) and 7 HongJams (HJs). Three primary colors of light of 8 silkworm powders revealed why each sample had unique color. The visual and absorption spectrum analyses of 80% methanol (MeOH) extracts of 8 silkworm powders revealed that white jade (WJ)-FDSP extracts showing strong forest-green colors had different absorption spectrum from 7 HJs. Among 7 HJs, the absorption spectrum of light green silkworm (LGS)-HJ extract was different from those of other HJ extracts. The amounts of total flavonoids and phenolic compounds of WJ-FDSP were 423.0±9.21% and 252.3±12.86%, respectively, more than those of WJ-HJ. Among 7 HJs, the amounts of total flavonoids and phenolic compounds in LGS-and red silkorm (RS)-HJ extracts were significantly more than those of WJ-HJ extract. Consistent with the amounts of phytochemicals, radical scavenging activity and reducing ability of WJ-FDSP extract were significantly higher that other HJ extracts. Among 7 silkworm powder extracts, LGS-HJ extract showed significantly higher radical scavenging activity and reducing ability than those of WJ-HJ extract. But, RS-HJ extract only showed significantly higher radical scavenging activity than that of WJ-HJ extract. Taken together, the amounts of phytochemicals in 8 silkworm powders contributed on their colors and anti-oxidant activities.
Objectives : The solvent extracts of Pruni persicae Flos were investigated for the activities of anti-oxidant and anti-inflammatory effects to apply as a functional ingredient for cosmetic products. Methods : In this study, the fractions of P. persicae Flos were extracted with 70.0% acetone and purified using Sephadex LH-20 column chromatography. As a result, eight fractions were isolated. We performed MTT assay, total polyphenol contents, DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay, astringent activity assay, hyaluronidase inhibition assay and the production of nitric oxide. Results : For anti-oxidant effects, the electron donating ability of fraction (Fr.) 2-5, Fr.-8 isolated from P. persicae Flos was above 90.0% at 100 ppm respectively. The superoxide dismutase (SOD) - like activity of Fr.-5 isolated from P. persicae Flos was 92.1% at 1,000 ppm. The xanthine oxidase inhibitory effect of Fr.-6 isolated from P. persicae Flos was about 83.3% at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 94.0% for Fr.-4 isolated from P. persicae Flos at 500 ppm. In the anti-inflammation effect, the Fr.-4 isolated from P. persicae Flos inhibited the generation of nitric oxide. Conclusions : All these findings suggested that the fractions of P. persicae Flos has a great potential as a cosmeceutical ingredient with a anti-oxidant and anti-inflammatory effects.
