Lipopolysaccharide로 유발한 급성 간손상 마우스 모델에서 와송과 발효 와송의 항산화 조절과 염증 예방 효과 비교 연구 (Orostachydis Herba and Fermented Orostachydis Herba Enhances Anti-Inflammatory and Anti-oxidant Effect against Lipopolysaccharide-Induced Acute Liver Injury in Mice)

  • Objectives : This study aimed to evaluate the protective effect of Orostachydis Herba (OH) and Fermented OH (OHF) against the acute liver injury by lipopolysaccharide (LPS). Methods : OHF by 4 lactic bacteria such as (Lactobacillus hilgardii (OHF1), Leuconostoc mesenteroides (OHF2), Pediococcus acidilactici (OHF3), Saccharomyces cerevisiae (OHF4)) were prepared. Samples were selected to OHF0, OHF2, OHF3 based on UPLC analysis, DPPH, ABTS radical scavenging activities. To evaluate the protective effect of OHF on liver injury mice, ICR mice were divided into 5 groups: Normal mice (Nor), LPS (20 mg/kg) treated mice (Veh), administrated OHF0, OHF2 OHF3 200 mg/kg body weight during 8 days before LPS injection. Serum and liver were collected 24 hours after LPS injection. Results : The activity was high in order of OHF0 and OHF3 in DPPH and ABTS radical scavenging activities. The quercetin contents for bioactive ingredient of OH was 5.39, kaempferol contents was 9.94 by UPLC analysis. The LPS-treated vehicle group significantly increased liver weight, and aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in serum. In contrast, administrated OHF3 group decreased liver weight, AST, ALT. In addition, OHF3 groups reduced the elevated levels of reactive oxygen species (ROS) in serum and tissues. Moreover, AP-1, iNOS and COX-2 were significantly decreased in OHF2 and OHF3. But $NF-{\kappa}B$ p65 and $TNF-{\alpha}$ only showed a significant reduction in OHF3. Conclusions : Therefore, these results suggest that fermented Orostachydis Herba might be protective effect on liver injury through anti-oxidant effect.

차가버섯 물 추출물의 cisplatin에 의해 유도된 HEI-OC1세포 손상에 대한 항산화효과 (Anti-oxidant Effects of the Water Extracts from the Inonotus Obliquus against Cisplatin- Induced Damage in HEI-OC1 Cells)

  • The medicinal mushroom Inonotus obliquus is a traditional and widely used multi-functional fungus. In this study, we have investigated whether Inonotus obliquus (Chaga mushroom) extracts exerts anti-oxidant effects on cisplatin-induced cytotoxicity in auditory cell line, HEI-OC1 cells. First of all, Chaga extracts has no harmful effects on viability of HEI-OC1 cells in the dose range of $65{\sim}125{\mu}g/m{\ell}$. Moreover, it shows cyto-protective effects on the cells treated with cisplatin-induced cytotoxicity in HEI-OC1 cells and the damage of hair cells arrays of the rat primary organ of Corti explants in the presence of cisplatin. Pretreatment with Chaga extracts inhibited the cell death, reactive oxygen species generation (ROS), lipid peroxidation induced by cisplatin. These effects were associated with the induction of antioxidant enzyme by Chaga extracts. We further investigated the effects of Chaga extracts on expression of antioxidant enzymes such as Cu, Zn superoxide dismutase (SOD 1) and Mn SOD (SOD 2) by RT-PCR. In addition, Chaga extracts shows SOD activity and SOD protein expression in cisplatin treated group induced similar to control group. Taken together, these results indicate that Chaga extracts can prevent cisplatin-induced cytotoxicity by radical-scavenging activity (SOD activity) in HEI-OC1 cells. It might be an effective as antioxidant and further studies on the chemo-preventive mechanisms of Inonotus obliquus are needed.

Production of Polyphenols and Flavonoids and Anti-Oxidant Effects of Lactic Acid Bacteria of Fermented Deer Antler Extract

  • The deer antler has been used as a major drug in oriental medicine for a long time. Recently, the demand for easy-to-take health functional foods is increasing due to economic development and changes in diet. As part of research on the development of functional materials for antlers, lactic acid fermentation of antler extract was performed. It was intended to develop a functional material with enhanced total polyphenol and flavonoid content and enhanced antioxidant activity. Lactic acid bacteria fermentation was performed by adding 4 types of lactic acid bacteria starter products, B. longum, Lb. Plantarum, Lb. acidophilus and mixture of 8 types of lactic acid bacteria to the antler water extract substrate, respectively. During the fermentation of lactic acid bacteria, the number of proliferation, total polyphenol and total flavonoid content, DPPH radical scavenging and antioxidant activity were quantified and evaluated. As a result of adding these four types of lactic acid bacteria to the antler water extract substrate, the number of lactic acid bacteria measured was 2.04~5.00×107. Meanwhile, a protease (Baciullus amyloliquefaciens culture: Maxazyme NNP DS) was added to the antler extract to decompose the peptide bonds of the contained proteins. Then, these four types of lactic acid bacteria were added and the number of lactic acid bacteria increased to 2.84×107 ~ 2.21×108 as the result of culture. The total polyphenol contents were 4.82~6.26 ㎍/mL in the lactic acid bacteria fermentation extracts, and after the reaction of protease enzyme and lactic fermentation, increased to 14.27~20.58 ㎍/mL. The total flavonoid contents were 1.52~2.21 ㎍/ml in the lactic acid bacteria fermentation extracts, and after the protease reaction and fermentation, increased to 5.59 ~ 8.11 mg/mL. DPPH radical scavenging activities of lactic acid bacteria fermentation extracts was 17.03~22.75%, but after the protease reaction and fermentation, remarkably increased to 32.82~42.90%.

사과락(絲瓜絡)의 항산화효과 및 3T3-L1분화 시 Cytokine류에 미치는 영향 (Experimental Study on Antioxidative Effect of Luffae Fructus Retinervus and Their Effects on Cytokines to 3T3-L1 Cell Lines)

  • In recent year, We are concerned in anti-aging, disease-protection, long-life, many method are used in solving this problem. Recently, We heard that Luffae Fructus Retinervus(LFR) has effect of anti-aging, disease-protection, long-life. So I let made a experiment for this result. The purpose of this study is to; 1) the anti-oxidant effect of Luffae Fructus Retinervus(LFR) used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity, Superoxide anion radical scavenging activity, 2) cultivation 3T3-L 1 Preadipocytes and Protein chip used for ProteoPlexTM 16-Well Murine Cytokine Array Kit. We measured level of DPPH radical scavenging activity. And we experienced that the ability of DPPH radical's elimination was increased by rising concentration of LFR. When the concentration of LFR was 5 mg/ml, the ability of DPPH radical's elimination was Maximum. We measured level of Nitric oxide(NO) radical scavenging activity. And we founded that the ability of NO radical's elimination was significant when concentration of LFR was from 1.25 mg/ml to 2.5 mg/ml. We measured level of Superoxide anion radical scavenging activity. And we founded that the ability of Superoxide anion radical's elimination was maximum when concentration of LFR was 0.3125 mg. When we inspected Antioxidative Effects with BSA, we experienced that ability of defense was increased by rising concentration of LFR. We known the immunity of LFR about 3T3-L1 Preadipocytes and gained the increase of Cytokines(IL-2, IL-4, GM-CSF) without IL-12p70, $INF-{\gamma}$, $TNF-{\alpha}$ So I guess that Luffae Fructus Retinervus(LFR) has effects of anti-aging, disease-protection, long-life, etc.

아사이베리 분말을 첨가한 머핀의 품질특성 및 레시피 최적화 연구 (A Study on Quality Characteristics and Optimized recipe of Muffin with added Acai Berry powder)

  • In this study Muffins adding each 0, 1, 3, 5 7% of Acai Berry powder, which has very high anti-oxidant constituent, have been made, and its quality characteristics have been surveyed. From the volume and specific volume measurement, it has been decreased for MA1, 1% of Acai Berry powder added muffin, but the tendency thereafter has shown increasing (p<0.001). From texture characteristics gumminess and chewiness of MA0, reference group, was the highest by 885.86 g/ cm and 6645.71 g, and has shown decreasing tendency as per the adding rate of Acai Berry powder increased (0.001). The electronics scavenging activity of MA7, 7% added muffin, was the highest by 68.91%, and the ascending order was MA1 < Reference Group < MA3 < MA5 < MA7. From the preference test of Acai Berry powder added muffin, MA3, 3% added muffin, was the most preferable one from color, taste as well as total preference. As the added volume of Acai Berry powder is 5% or more, the typical taste and flavor of Acai Berry gives influence to the taste and flavor of muffin, so the preference level has been decreased. Thus the added volume of Acai Berry powder 5% or more is not recommendable.

Amelioration of $Cd^{++}$ Toxicity by $Ca^{++}$ on Germination, Growth and Changes in Anti-Oxidant and Nitrogen Assimilation Enzymes in Mungbean(Vigna mungo) Seedlings

  • The present study describes the ameliorating effect of $Ca^{++}\;on\;Cd^{++}$ toxicity on the germination, early growth of mungbean seedlings, nitrogen assimilation enzyme. s-nitrate reductase (NR), nitrite reductase (NIR), anti-oxidant enzymes (POD, CAT and SOD) and on the accumulation of hydrogen peroxide and sulphydryls. $Cd^{++}$ inhibited seed germination and root and shoot length of seedlings. While NR activity was down- regulated, the activities of NIR, POD and SOD were up- regulated with $Cd^{++}$ treatment. $Cd^{++}$ treatment also increased the accumulation of sulphydryls and peroxides, which is reflective of increased thiol rich proteins and oxidative stress. $Ca^{++}$ reversed the toxic effects of $Cd^{++}$ on germination and on early growth of seedlings as well as on the enzyme activities, which were in turn differentially inhibited with a combined treatment with calcium specific chelator EGTA. The results indicate that the external application of $Ca^{++}$ may increase the tolerance capacity of plants to environmental pollutants by both up and down regulating metabolic activities. 益氣化瘀湯加味方이 고지방 식이로 유발된 생쥐의 항고지혈 및 항산화에 미치는 효과 (Anti-hyperlipidemic and Anti-oxidant effects of Kgihwaetang-gamibang in High Cholesterol Diet-induced Mice Model)

  • This study was conducted to investigate the anti-hyperlipidemic and anti-oxidative activities of Ikgihwaetang-gamibang (IK). We observed the effect of IK on lipid factor, inflammation factor, body weight and organ weight, and liver after administering IK to hyperlipidemia mice triggered by high fat diet. Additionally, we used Raw 264.7 cells to investigate the changes in anti-oxidative activity and the effects of anti-inflammation. Administration of IK resulted in reduction of total cholesterol, LDL cholesterol, TG, and glucose in blood serum while a clear increase in HDL cholesterol. The production of IL-1β and IL-6, and ROS and NO also decreased. The results showed that IK has anti-hyperlipidemic and anti-oxidative effects, and suggested its potential use in prevention and treatment of hyperlipidemia and its associated diseases.

블랙 커런트의 화학성분 및 항산화 활성 (Chemical Components and Anti-oxidant Activities of Black Currant)

  • 블랙 커런트를 건강기능성 식품 소재 및 가공품을 개발하기 위한 기초자료로 활용하기 위하여 화학성분 및 항산화 활성을 조사하였다. 블랙 커런트 착즙액의 pH는 3.36, 가용성고형분은 $15.11^{\circ}Brix$ 및 총산도는 1.65%였으며, L값 18.20, a값 5.13 및 b값 1.08으로 나타났다. 일반성분은 수분 77.64%, 가용성 무질소물 17.41%, 조섬유 3.08%, 조단백 1.28%, 조회분 0.31% 및 조지방 0.28% 순이었다. 주요 무기성분은 K, P 및 Ca으로 그 함량은 각각 177.36 mg/100 g, 54.74 mg/100 g 및 26.45 mg/100 g이었다. 블랙 커런트의 주요 유리당은 glucose(7.71%) 및 fructose(5.88%)였으며, 가장 많이 함유되어 있는 아미노산은 glutamic acid(105.73 mg/100 g)이었고, 가장 적게 함유되어 있는 아미노산으로는 cystine(5.29 mg/100 g)이었다. 비타민 C 함량과 총페놀 함량은 각각 112.19 mg/100 g 및 34.48 mg/GAE g이었다. 블랙 커런트의 농도 10 및 1.25 mg/mL 일때 각각 99.48%와 89.03%의 ABTS 및 DPPH 라디칼 소거활성을 보였으며, 또한 환원력과 FRAP은 농도의존적인 항산화 활성을 보였다. 따라서 블랙 커런트는 항산화제와 같은 기능성 식품소재로 활용가능성이 매우 높을 것으로 생각된다.

소나무과 일부 수종의 에탄을 추출물에 대한 생물학적 평가 (Evaluation of Biological Activities on the Extractives of Pinaceae)

  • 소나무과는 우리 생활과 친숙한 수종이지만 천연물로써 그 평가는 전래되는 사용범위에 국한되어 있다. 본 연구에서는 소나무과 수종 Pinus koraienis S et Z. Pinus banksiana LAMBERT. Pinus rigida MILL. Pinus deniflora S. et Z.의 에탄을 추출물에 대하여 피부 노화방지에 대한 화장품 소재로써의 평가를 수행하였다. 추출물은 각 수종의 식물 부위를 껌질, 잎, 심재 등의 부위별 제조되었으며 각 부위에 따른 추출물은 생물학적 평가에 항산화 효과(DPPH, superoxide radical 소거실험), elastase 활성 억제 효과 및 세포 생존율에 과한 MTT assay를 실시하였다. 항산화 효과는 수종에 차이 없이 껍질 >잎>심재 순으로 활성이 나타났으며 껍질과 잎은 $10{\mu}g/mL$ 농도에서 L-ascorbic acid나 $\alpha$-tocopherol과 유사한 항산화 효과를 나타내었다. Human keratinocyte cell line인 HaCaT 세포주를 사용하여 추출물을 10, In $100{\mu}g/mL$ 농도로 처리하여 세포 생존율을 실험하였다 껍질부분의 추출물은 농도에 상관없이 세포 증식효과를 나타내었으며 잎과 심재 부분의 추출물 $10{\mu}g/mL$ 농도에서 세포 증식 효과가 보이나 $100{\mu}g/mL$ 농도에서는 세포 증식이 저하되었다. 또한 세포기저층을 형성하는 분자 중 elastin을 분해하는 elastase 활성억제를 측정하여 사용된 식물 부위 중 껍질에서 elastase 활성 억제효과를 확인하였다. 따라서 소나무과 수종의 활용에 있어 껍질 부분의 활용을 기대할 수 있으며 일반적 조림용 이외 목재로 사용되는 경우 산림폐기물을 이용한 부가가치를 극대화하는 기능성화장품 소재로써의 사용이 가능할 것이다.

RAW 264.7 세포에서 Desmodium heterocarpon 추출물의 항산화 및 항염증 활성 (Anti-oxidative and Anti-inflammatory Activities of Desmodium heterocarpon Extract in RAW 264.7 Cells)

  • Desmodium heterocarpon은 콩과에 속하는 덩굴식물로 주로 한국, 일본 등의 아시아 국가에 분포되어 있다. 본 연구에서는 식물에 존재하는 신규 기능성 소재 개발의 일환으로 Desmodium heterocarpon 에탄올 추출물(DHEE)의 항산화 및 항염증 생리활성을 DPPH 라디칼 소거능, ROS 소거능, NO 생성 억제능 및 관련 단백질 발현을 통해 분석하였다. 먼저 DHEE의 항산화능을 DPPH 라디칼 소거능을 통해 분석한 결과 높은 소거활성을 보여 DHEE가 매우 강한 항산화능을 보유함을 확인하였다. 또한 RAW 264.7 세포주에서 H2O2에 의해 유도된 ROS에 대한 DHEE의 소거능을 분석한 결과 농도의존적인 강한 ROS 소거능을 보였다. 뿐만 아니라 대표적인 항산화 효소 중 하나로 항산화능 보유 천연물에 의해 발현이 유도되는 HO-1 및 그 전사 인자인 Nrf2의 단백질 발현이 DHEE의 처리에 의해 유의적으로 증가됨을 보였다. 한편 DHEE가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 농도 의존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이러한 결과를 통해 DHEE의 높은 항산화능과 항염증 활성을 확인하였으며 향후 잠재적인 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다. 추후 계속적인 연구를 통해 활성 물질의 규명이 필요할 것으로 보인다.
