Standardized Extract (HemoHIM) Ameliorated High Intensity Exercise Induced Fatigue in Mice

  • HemoHIM was used as a Korean traditional medicine for anti-inflammatory and antioxidant effects. However, there is no study on the effect of HemoHIM on fatigue. We examined the potential use of HemoHIM to determine whether it can induce anti-fatigue effects. Mice were administered with HemoHIM and VEH for 14 days. On the last day of treatment, mice were subjected to behavioral tests. Subsequently, their plasma and muscle were collected after the treadmill test to measure lactate, lactate dehydrogenase (LDH), ammonia, corticosterone, glycogen, and creatine kinase (CK). We found that HemoHIM moderately increased the running time (s) in the treadmill and mobility duration in the cold swimming tests. In addition, the VEH group showed a significant increase in lactate, LDH, and corticosterone levels in the plasma compared to the group that did not perform the test. However, this was moderately reduced in HemoHIM treatment. Moreover, the HemoHIM-treated group showed significant differences in LDH and glycogen levels, and showed significantly different CK levels in the muscle. HemoHIM is considered to be effective in improving fatigue, given the duration of cold swimming or running time on a treadmill. Also, HemoHIM treatment resulted in reduced concentrations of blood and muscle parameter analysis.

Short term effect of Allii tuberosi semen on muscular fatigue and muscular strength (단기간(短期間)의 구자 투여(投與)가 생쥐의 근피로(筋疲勞)와 근력(筋力)에 미치는 영향(影響))

  • Purpose: This study was conducted to investigate the muscle anti-fatigue effects of Allii tuberosi semen after short term administration. Methods: 4-6 weeks old ICR mice were used in this study and we administered the water soluble extracts of Allii tuberosi semen in the concentration of 1, 10 and 100mg/0.3ml to each 5 mice (ATS group) and same volume of normal saline was administered to 5 mice (control group) once a day for 30 or 60 days. After the administration we performed the swimming exercise test and the grip strength test. And we measured the concentration of glucose and the activity of lactate dehydrogenase in plasma regarded as biochemical indicator related with the muscular fatigue.. Results: In swimming exercise test. 10 and 100mg/0.3ml ATS groups showed increased time compared with relative control group after 30 days administration. But after 60 days adminstration all ATS groups showed increased results. In grip strength test, 100mg/0.3ml ATS group increased grip strength after 30days administration and 10mg/0.3ml ATS group showed increased result after 60days administration. All the results of plasma glucose and lactate dehydrogenase didn't showed statistically significant difference after 30 and 60 days administration. Conclusion: From this study we could find the anti-fatigue effect of Allii tuberosi semen in the muscle strength related test.

In the plasma concentration of activity level of lactate dehydrogenase, 1mg/0.3ml and 10mg/0.3ml ATS groups showed decreased concentration compared with relative control group after 90 days administration. But after 120 days adminstration 1mg/0.3ml ATS groups showed decreased results. Conclusion: From this study we could find the anti-fatigue effect of Allii tuberosi semen in the muscle strength related test.

Anti-Fatigue Properties of Cultivated Wild Ginseng Distilled Extract and Its Active Component Panaxydol in Rats

  • Objectives: Cultivated wild ginseng (cWG), called SanYangSanSam, has been used clinically in patients with chronic fatigue in Korea. Little is known about effects of the ginseng distilled (volatile) components produced during evaporizaiton. Recently, we first identified one major component from cWG distilled extract, panaxydol, by using mass spectrometry. However, functional properties of cWG distilled extract and panaxydol remains elusive. Therefore, the present study evaluated the effect of cWG distilled extract or panaxydol on exercise-induced fatigue in rats. Methods: Fatigue was induced by forced swimming and the immobility time was analyzed in male Sprague-Dawley rats. The animals received intraperitoneally either vehicle, cWG distilled extract, or panaxydol 10 min prior to beginning of the forced swimming test (FST) once daily for 5 days. After the FST on day 5, we also analyzed fatigue-related biochemical levels including blood urea nitrogen (BUN), lactate acid (LAC), and lactate dehydrogenase (LDH) in serum and levels of glycogen in liver and soleus muscle. Results: The forced swimming time in cWG distilled extract (0.6 mL/kg)-treated group was significantly longer than that of control group on day 4 and 5. Panaxydol (0.1 and 0.25 mg/kg)-treated groups showed significantly enhanced performance in the forced swimming, compared to control. In addition, a significant decrease in serum LDH level was found in panaxydol-treated group, while there were no alternations in levels of serum BUN and LAC and glycogen in liver or soleus muscle. Conclusion: The present study demonstrated cWG distilled extract and its active component panaxydol have a function of anti-fatigue.

Effects of Combined Preparation (DWP715) Containing Alaska pollack Extract, Maltol, Ascorbic Acid and Nicotinamide on Decreasing of Blood Alcohol Concentration, Anti- fatigue and Anti-oxidation (북어엑스 및 말톨 함유 복합 조성물(DWP715)의 혈중 알콜농도 저하, 항피로 및 항산화 효과)

  • Effect of combined preparation (DWP715) containing Alaska pollack extract, maltol, ascorbic acid and nicotinamide on decreasing of blood alcohol was evaluated in human blood. Treatment of DWP715 prior to administration of 25% alcohol (100 mL) decreased alcohol concentration in blood and showed significant difference after 2 hours. The pharmacokinetic parameters such as area under the concentration-time curve (AUC), $C_{max},\;T_{max}\;and\;T_{1/2}$ were also decreased and delayed when compared with control values. Effects of DWP715 on anti-fatigue and anti-oxidation activities were also studied in the restraint stress model using various parameters (GOT, GPT, LDH values and organ weights) on mild condition and examined through the content of lipid peroxide induced by 2% $CCl_4$ in mouse livers. While GPT level, thymus and adrenal weight were not influenced by DWP715 dosing, LDH, GOT level and spleen weight used as a parameter against fatigue and stress states were recovered almost to the nomal level. Furthermore, lipid peroxidation due to $CCl_4$ was significantly inhibited by DWP715 treatment. These results suggest that DWP715 seems to metabolize the blood alcohol rapidly and to restore the damaged liver and fatigue conditions which was caused by alcohol metabolism to normal condition.

Anti-fatigue effect of a beverage mixture containing red ginseng and Prunus mume fruit vinegar on high-intensity exercised rats (홍삼과 매실식초 혼합 음료의 고강도 운동을 실시한 흰쥐에서 항피로 효과)

  • New types of vinegar drinks are being processed and developed using various raw materials. In this study, a beverage containing a mixture of 0.25% red ginseng extract and 5% Prunus mume fruit vinegar (RPV) was evaluated for its anti-fatigue effect on high-intensity exercised rats. RPV administration markedly enhanced running endurance and significantly decreased fatigue-related serum biomarkers, such as inorganic phosphate, ammonia, and L-lactate, compared to the other groups. In addition, RPV administration increased glycogen contents in the liver and muscles and decreased creatine kinase activity in the serum and muscles. RPV administration also remarkedly increased the activity of lactate in the muscles. Furthermore, HPLC analysis revealed that main organic acids in RPV were acetic acid, malic acid, and citric acids. Overall, the results indicate that RPV improved fatigue recovery in exhausted rats, thus proving a promising material of functional food to attenuate fatigue.

Effects of Yeast Fermented Ganoderma Lucidum Extract on the Recovery of Exercise Fatigue in Forced Swimming Rats (효모를 이용한 발효영지 추출물이 흰쥐의 유영 운동 피로에 미치는 영향)

  • Objectives : Ganoderma Lucidum has been used in traditional oriental medicine to treat various conditions including chronic fatigue and cough. This experimental study was designed to investigate the effects of yeast-fermented Ganoderma Lucidum extract on the fatigue of rats elicited by forced swimming. Materials and methods : The rat subjects were divided into 4 groups: normal group, control group administered with 0.9% normal saline, experimental I(Exp I) group administered with yeast-fermented Ganoderma lucidum extract and experimental II(Exp II) group administered with Ganoderma lucidum extract for 4 weeks. All the serum samples collected by heart puncture were used to measure biochemical factors such as glucose, lactate, lactate dehydrogenase(LDH), creatine kinase(CK), inorganic phosphorus, ammonia, total cholesterol(TC), triglycerides(TG), high-density lipoprotein(HDL), low-density lipoprotein(LDL), aspartate transaminase(AST), alanine transaminase(ALT). Results : 1. Weight change and swimming time were not significantly different in the experimental groups as compared with control group. 2. Lactate was significantly decreased in the experimental groups as compared with control group. Inorganic phosphorus was also significantly decreased in the Exp I group as compared with control group(p<0.05). 3. Serum lipid concentration was not significantly different in the experimental groups as compared with control group. 4. AST and ALT were significantly decrease in the experimental groups as compared with control group. Conclusions : Yeast-fermented Ganoderma lucidum in forced swimming rats would appear to reduce fatigue-related factors, AST, ALT of serum and promote anti-fatigue effect.

Effect of Carbon Potential on the Carbide Formation and Pitting Fatigue Strength of Supercarburized Steel (고농도 침탄강의 탄화물 형성과 피팅 피로강도에 미치는 탄소 포텐셜의 영향)

  • In the present work, we investigated the effects of the carbon potential on the formation of carbide at the carburized surface and anti-pitting fatigue strength in the supercarburized steels. Two low carbon steels with different Cr concentrations were adopted and the repeated supercarburizing treatment carried out with the different carbon potential conditions. The microstructure and carbides at the supercarburized surface were observed by using optical microscope and scanning electron microscope. The microhardness test was performed and the hardness distribution and the effective case depth at the supercarburized surface were discussed. The roller pitting fatigue test was carried out and the fatigue strength was evaluated with different the carbon potential conditions. The microstructure of the fatigue specimen surface was observed by means of scanning electron microscope and scanning transmission electron microscope. Depending on the chemical composition of the steels and the carbon potential condition, the resistance of temper softening and pitting failure was influenced due to the carbide distribution and the formation of coarse network carbide. Thus, it was confirmed that the control of the carbide formation is a key factor to improve the anti-pitting fatigue strength in the supercarburized steels.

Study on the Anti-Fatigue and Immune-Enhancing Effects of Gami-Yukmi-Jihwang- Tang (가미육미지황탕의 항피노와 면역 증강 효과에 대한 연구)

  • The Korean formula medicine, Gami-Yukmi-Jihwang-Tang (GYJT) has been used for growing slowly, short of stature, incomplete development, fatigue, weak child, growing pain of child. However, it is still unclear how GYJT has an effect on experimental models. In the present study, the author investigated the immune-enhancing effect of GYJT. Forced swimming test (FST) was performed as a model of activity test in mice and measured blood urea nitrogen (BUN), aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactic dehydrogenase (LDH), glucose (Glc) and total protein (TP) in the serum. GYJT (1, 0.1 and 0.01 g/㎏) were orally administered to mice, once per day for 7 days using a feeding atraumatic needle. After 3 days, on FST, the immobility time was significantly decreased in the GYJT (0.01g/㎏/day)-fed group (120.75±5.71s) in comparison with the saline-fed group (153.80±10.74s). After 7 days, the immobility time was significantly decreased in the GYJT (0.1 and 0.01g/㎏/day)-fed group (125.67±5.36s and 107.67±3.71s) in comparison with the saline-fed group (167.67±12.99s). In addition, the contents of BUN and Glc in the blood serum were significantly decreased and the contents of AST, ALT and LDH were also decreased in the GYJT (1g/㎏/day)-fed group. However, the content of TP was not changed. The present results suggest that GYJT may be useful for the anti-fatigue and immune-enhancing agent. Also, the author investigated the effect of GYJT on the production of cytokines in human T-cell line, MOLT-4 cells. However, GYJT has not affected the production of IFN­γ, IL-2, IL-4. These results suggest that GYJT has immune-enhancing effect but does not affect T cell-mediated production of cytokines in the immune function improvement.

  • Recently, the steel parts used at the aerospace and automobile industries are required to be used light weight parts. Therefore, used material, steel have to be a high stress, which is an indispensable condition in this field. At the consideration of parts design, high hardness of the lightweight parts have an benefit of saving fuel and material. A high stress of metal has a point of difference according to the shape of design, external cyclic load and condition of vibration. A crack generates on the surface of metal or under yield stress by defect of inner metal defect or surface defect and slowly, this crack grow stable growth. Finally, rapidity failure phenomena is happen. Fatigue failure_phenomena, which happen in metal, bring on danger in human life and property therefor, anti-fatigue failure technology take an important part of current industries Currently, the shot peening is used for removing the defect from the surface of steel and improving the fatigue strength on surface. Therefore, this paper investigated the effect on frcature toughness using shot peening which is improve the resistance of crack growth and crack expansion rate by fatigue that make a compressive residual stress on surface.
