Objective : In 2001, the rate of obesity in Korea reached 30.6%. There are many therapeutic ways to reduce body weight, such as low and very low calorie diet, exercise therapy, behavior modification therapy, etc. However, in many cases the patients feel stress under obesity treatment because of starvation. This study was aimed to evaluate the anti-starvation stress effect of Coicis Semen on mice. Methods : First, the mice were divided into 6 groups : Normal (group with no starvation), Control (administrated normal saline 6 times before starting 36 hours starvation), and Samples A, B, C, and D (administrated 0.25, 0.5, 1.0, 3.0 g/kg Coicis Semen respectively 6 times before starting 36 hours starvation). Then the plasma corticosterone level and rectal temperature were measured. The norepinephrine, dopamine, DOPAC (dihydroxy-phenylacetic acid), 5-HT (5-hydroxytryptamine) and 5- HIAA (5-hydroxy-indole-acetic acid) in the hypothalamus were measured by the HPLC method. Result : I. The rectal temperature in Sample group D showed a significant difference (P<0.05) compared with the Control group. 2. The DOPAC in Sample groups A, C and D showed the significant difference (P<0.05) compared with the Control group. Conclusion : It might be recognized that Coicis Semen has an anti-starvation stress effect.
This study aimed to evaluate the anti-stress effects of soyosan and chyengansoyosan on the contents cold swimming stress.The experimental animals were forced cold swimming stress for 3 minutes, and administered 4㎎/10g of soyosan and chyengansoyosan extract for 7 days before stress. The monoamines contents were measured by HPLC method in various part of mouse brain. The following results were obtained :1. In frontal cortex, the contenrs of norepinephrine ans dopamine increased significantly in the group administered chyrngansoyosan compared with in the group administered non chyengansoyosan and soyosan(control group) of mice stressed by cold swiming.2. In hypothalamus, the content of norepinephrine increased with statistical significance in the group administered soyosan compared with control group of mice stressed by cold swimming. 3. The content of dopamine increased with statistical significance on the group administered chyenfansoyosan and compared with control group of mice stressed by cold swimming in hypothlaamus and hippocampus.4. The content of serotonin increased with statistical significance in the group administered soyosan and compared with control group of mice stressed by cold swimming in hippocampus and corpus striatum.5. In corpus striatum and hippocampus, the content of norepineohrine decreased with statistical significance in the group administered soyosan and chyengandoyosan compared with control group of mice stressed by cold swimming.Judgding from the above results, this sthdy reaches a conclusion that soyosan or chyengansoyosas has significant effects in reducing stress.
The effects of the ethanol extracts from Gynostemma pentaphyllum (GP extracts) on body weights, grip strengths, endurances and catecholamine levels after electric footshock (EF) stress in mice and rats were investigated. The animals were treated with GP extracts (50 mg/kg/day, p.o.) for 21 days before exposure to EF (duration and interval 10 sec for 3 min, 2 mA) once a day. The increases in body weights were delayed by 13.1% of the control levels by EF-induced stress in mice, which were recovered to 24.1% of the control levels in GP extract-treated groups. The grip strengths were significantly decreased by EF stress in mice and the EF-stressed groups treated with GP extracts increased grip strengths to 115.2% compared to control levels. The endurance times by forced swimming, which reduced significantly by EF stress, were also maintained similar to control levels by GP extracts in rats. In addition, the levels of norepinephrine and epinephrine in serum and brain, and dopamine in brain were significantly increased to 17.5-95.0% of the control levels after exposure of EF stress in mice. However, EF stressinduced increases in norepinephrine and epinephrine in serum were reduced to 17.1-17.3% of the control levels by treatments of GP extracts, and those in dopamine, norepinephrine and epinephrine in brain were also reduced to 5.0-19.5%. These results suggest that GP extracts showed the protective effects on EF stress-induced physiological functions and can be developed as the promising anti-stress agents.
The effects of herbal ethanol extract from Gynostemma pentaphyllum (GP extract) on chronic stress-induced anxiety in mice were investigated. The animals were treated with GP extracts (50 and 100 mg/kg/day, p.o.) for 21 days before exposure to electric footshock (EF; duration and interval 10 sec for 3 min, 2 mA) for chronic stress once a day. The ambulatory locomotor activity was reduced by chronic EF stress and it was recovered by 12.9-15.1% in GP extract-treated groups. The grip strength was also significantly decreased by chronic EF stress, however, the EF-stressed groups treated with GP extract increased grip strength from 13.9% to 56.8% compared to EF-stressed groups. In addition, the serum levels of corticosterone were significantly elevated by chronic EF stress to 197% of the control levels, which was reduced to 73.1% by treatment with GP extract (100 mg/kg). In contrast, the brain levels of dopamine and serotonin were reduced to 67.6% and 63.1% by chronic EF stress, which was recovered to 90% of the control levels by treatment with GP extract. These results indicate that GP extract shows the ameliorating effects on chronic EF stress-induced anxiety in mice and it can be developed as the promising anti-anxiety agent.
Objective : The study was aimed to evaluate the anti-stress effect of KYT on the cold-stressed Mice. Methods: The experimental animals were stressed in cold room $(4{\pm}0.5^{\circ}C)$ for one hour in a day during two weeks, and administred 131mg/100g KYT extract for two weeks before stress. For two weeks, while the experiment was taking place, we conducted a research about the change of weight and content of norepinephrine, epinephrine, dopamine, cortisol and GOT, GPT in the mice serum. Results : KYT extract significantly inhibited the decrease of body weight induced by stress, compared with the control group. KYT extract had no significant decreasing effect on the change of content of the serum GOT but had significant decreasing effect on the change of content of the serum GPT compared with the control group. KYT extract had significant decreasing effect on the change of content of the serum cortisol compared with the control group. KYT extract had decreasing effect on the change of content of the serum dopamine compared with the control group. KYT extract had no decreasing effect on the change of content of the serum norepinephrine compared with the control group. KYT extract had significant decreasing effect on the change of content of the serum epinephrine compared with the control group. Conclusions : According to the above results, it is concluded that KYT will be useful as a preventive and remedy against stress disease.
This study aimed to evaluate the anti-stress effect of Yangsimtang and Yangsimtang+Siyup on the rats in immobilization stress.The experimental animals were immobilized in the stress box(5${\times}$5${\times}$20cm) for 12 hours in a day suring 3 days, and administered 1g/100g of Yangsimtang and Yangsimtang+Siyup and Siyup extract for 12 days before stress. The norepinephrine, epinephrine, dopamine and serotonin contents were measured by HPLC method in various part of rat brain.The following results were observed.1. In frontal cortex the norepinephrine content of control group was 561.${\pm}$24.46 ng/g brain tissue, that of saple 1 group was 430.8$\pm$41.2 ng/g brain tissue, and that of sample 2 group was 417.2$\pm$38.5 ng/g brain tissue. The differences was statistically significant.2. In corpus striatum, the norepinephrine content of control group was 561.3$\pm$27.3 ng/g brain tissue, and that of sample 1 group was 422.1$\pm$21.2 ng/g brain tissue, the dopamine content of control group was 1205.1$\pm$75.9 ng/g brain tissue, that of sample 2 group was 685.6$\pm$41.5 ng/g brain tissue. The differences was statistically significant.3. In hypothalamus, the norepinephrine content of control group was 1165.1$\pm$162.6 ng/g brain tissue, that of sample 2 group was 947.2$\pm$35.7 ng/g brain tissue. The differences was statistically significant.4. In hippocampus, the norepinephrine content of control group was 931.6$\pm$82.2 ng/g brain tissue, that of sample 1 group was 652.1$\pm$47.5 ng/g brain tissue, and that of sample 2 group was 627.4$\pm$31.2 ng/g brain tissue, the dopamine contrnt of control group was 315.4$\pm$28.4 ng/g brain tissue, that of sample 2 group was 208.5$\pm$23.7 ng/g brain tissue. The differences were statistically significant.Base on the above results, it may be concluded that Yangsimtang and Yangsimtang+Siyup are effective to reduce stress.

As an effort to direct differentiation of human embryonic stem (hES, MB03) cells to dopamine-producing neuronal cells, Nurr1 was transfected using conventional transfection protocol into MB03 and examined the expression of tyrosine hydroylase (TH) after differentiation induced by retinoic acid (RA) and ascorbic acid (AA). Experimentally, cells were transfected with linearized Nurr1 cDNA in pcDNA3.1 (+)-hygovernight followed by selection in medium containing hygromycin-B (150 $\mu$/ml). Expression of Nurr1 mRNA was confirmed by RT-PCR and protein by immunocytochemistry in the drug resistant clones. In order to study the effect of Nurr1 protein on the differentiation pattern of ES cells, one of the positive clones (MBNr24) was allowed to form embryoid body (EB) for 2 days and were induced to differentiate for another 4 days using RA (1 $\mu M$) and AA (50 mM) (2-/4+ protocol) followed by selection in N2 medium for 10 or 20 days. After 10 days in N2 medium, cells immunoreactive to anti-GFAP, anti-TH, or anti-NF200 antibodies were 38.8%, 11%, and 20.5%, respectively. After 20 days in N2 medium, cells expressing GFAP, TH, or NF200 were 28%, 15% and 44.8%, respectively but approximately 9% of MB03 expressed TH protein when the cells were induced to differentiate using a similar prorocol, These results suggest that ectopic expression of Nurr1 enhances generation of TH+ cells as well as neuronal cells when hES cells were differentiated by 2-/4+ protocol.
In order to, study the anti-stress effect of $Tae{\breve{u}}min$$Ch{\breve{o}}ngsimy{\breve{o}}njat^{\prime}ang$, several measures of stress, including catecholmine changes, body weight changes, and gastric ulcers were used.the following result were observed: 1. Plasma epinephrine contents of stress by food in rats, in the controlled group were $219.3{\pm}18.4pg/ml$, but in the experimental group the value was decreased to $169.0{\pm}16.6pg/ml$. 2. Plasma norepinephrine content of stress by food in rats in the controlled group was $303.0{\pm}18.9pg/ml$ but in the experimental group the value was decreased to $243.6{\pm}17.6pg/ml$. 3. Plasma dopamine contents of stress by food in rats in the controlled group was $16.9{\pm}1.9pg/ml$ but in the experimental group the value was derceased to $12.8{\pm}1.5pg/ml$, but it was nonsignificant. 4. Effects of body weight of stress by food in the Shay rats in controlled group was $175.9{\pm}3.0g$, but in the experimental grou the value was increased to $187.7{\pm}4.8g$. 5. Gastric ulcers of stress by food in Shay Rats, in the experimental group, the glade was decreased more than in the control group. 6. It is considered that the anti-stress effect of Taeumin Chongsimyonjat' depends on its concentration rate. Based on the results above, it may be concluded that Taeumin chongsimyonjat' aug is effective to prevent the harmful effects of stress.

Various kinds of related parameters on hypertension such as anti-oxygen effect, ACE, weight of body, hwart and kidney, blood pressure, heartbeat rates, contents of aldosterone, catecholamine, change rates, of plasma constituents, uric acid, BUN, creatinine were determined to verify the effects on hypertension by Kamisamul-tang (KSMT). And the results are concluded as follows. KSMT did not show any cytotoxicity at the range of concentration (1-250 ${\mu}g/m{\ell}$) on the human fibroblast cell (hFCs). KSMT decreased the production of reactive oxygen species (ROS) and DPPH generation depending on the concentration. KSMT significantly inhibited angiotensin converting enzyme(ACE) activity depending on the concentration compared with control. KSMT maintained body weight of body, heat and kidney nearly normal group in hypertensive rat induced by DOCA-salt. KSMT significantly blood pressure and heart beat rate compared with control in hypertensive rat induced by DOCA-salt. KSMT significantly decreased aldosterone, dopamine, norepineph- rine, epinephrine compared with control in hypertensive rat induced by DOCA-salt. KSMT significantly decreased the level of potassium and cloride compared with control wheareas increased that of calcium significantly in hypertensive rat induced by DOCA-salt. KSMT significantly decreased the level of uric acid and BUN compared with control in hypertensive rat induced by DOCA-salt. It is verified experimentally tat Kamisamultang(KMST) which has been used broadly as a clinical therapeutics in oriental medicine is effective for anti-hypertension mechanism. And it could be applied to develope the reliable prescriptions for anti-hypertension in the future.

Purpose: Edible insect extracts have been used as an alternative source for medicinal supplements due to their significant antioxidative and anti-inflammatory activity. Recent studies have reported that anti-microbial peptides from insects have neuroprotective effects on dopamine toxins. The purpose of this study was to investigate the protective functions of mealworm (Tenebrio molitor) extract (MWE) on N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced cellular toxicity in SH-SY5Y neuroblastoma cells. Methods: Cellular toxicity induced by the MPTP toxin and the impact of MWE on cell survival were analyzed using MTT assays. DAPI staining was performed to observe apoptotic phenomena caused by MPTP. Changes in caspase-3 activity and protein expression were observed using enzyme activity assays and western blot assays, respectively. Results: MWE exerted significant antioxidant activity, which was measured by both DPPH and ABTS radical assays, with a dose-dependent relationship. Furthermore, MWE resulted in cellular proliferation in SHSY5Y cells in a dose-dependent manner. Furthermore, MWE pretreatment significantly inhibited MPTP-induced cytotoxicity, with a dose-dependent relationship. The morphological characteristics of apoptosis and increased reactive oxygen species induced by MPTP were also significantly reduced by MWE pretreatment. Conclusion: MWE treatment significantly attenuated MPTP-induced changes in the levels of proteins associated with apoptosis, such as caspase-3 and PARP. These findings suggest that MWE exerts neuroprotective effects on human neuroblastoma SH-SY5Y cells subject to MPTP-induced dopaminergic neurodegeneration.
