비기환이 신생혈관형성 억제에 미치는 효과 (Effects of Bikihuan (BKH) on anti-angiogenesis)

  • Objective: To evaluate the effects of Bikihaun (BKH) on angiogenesis. Method: We examined the anti-angiogenic effect of BKH in invasion assay model. We performed proliferation assay, migration assay, tube formation assay and Chicken Chorioallantoic Membrane (CAM) assay. Results: In proliferation assay, at lower dose under 125 ${\mu}g/m{\ell}$ anti-angiogenesis effect of the group treated BKH made no difference with the control group. But, at the dose of 250 ${\mu}g/m{\ell}$ or more anti-angiogenesis effect of the group treated BKH showed more effective as compared to the control group. In migration assay, BKH did not affect migration of vascular endothelial cell. In tube formation assay, at lower dose under 100 ${\mu}g/m{\ell}$ showed mild effect of anti-tube formation. But, at the dose of 1000 ${\mu}g/m{\ell}$ showed more effective anti-tube formation. In CAM assay, BKH showed anti-angiogenesis effect at the dose of 10 ${\mu}g/m{\ell}$. Conclusion: BKH has antiangiogenetic properties in vitro.

Influence of Tyrosol on Cell Growth Inhibition of KB Human Oral Cancer Cells

  • Tyrosol, a phenylethanoid and a derivative of phenethyl alcohol, possesses various biological properties, such as anti-oxidative and cardioprotective activity. Olive oil is the principal source of tyrosol in the human diet. However, so far the anti-cancer activity of tyrosol has not yet been well defined. This study therefore undertakes to examine the cytotoxic activity and the mechanism of cell death exhibited by tyrosol in KB human oral cancer cells. Treatment of KB cells with tyrosol induced the cell growth inhibition in a concentration- and a time-dependent manner. Furthermore, the treatment of tyrosol induced nuclear condensation and fragmentation of KB cells. Tyrosol also promoted proteolytic cleavage of procaspase-3, -7, -8 and -9, increasing the amounts of cleaved caspase-3, -7, -8 and -9. In addition, tyrosol increased the levels of cleaved PARP in KB cells. These results suggest that tyrosol induces the suppression of cell growth and cell apoptosis in KB human oral cancer cells, and is therefore a potential candidate for anti-cancer drug discovery.

  • Phytomedicines are believed to have benefits over conventional drugs and are regaining interest in current research. Moringa oleifera is a multi-purpose herbal plant used as human food and an alternative for medicinal purposes worldwide. It has been identified by researchers as a plant with numerous health benefits including nutritional and medicinal advantages. Moringa oleifera contains essential amino acids, carotenoids in leaves, and components with nutraceutical properties, supporting the idea of using this plant as a nutritional supplement or constituent in food preparation. Some nutritional evaluation has been carried out in leaves and stems. An important factor that accounts for the medicinal uses of Moringa oleifera is its very wide range of vital antioxidants, antibiotics and nutrients including vitamins and minerals. Almost all parts from Moringa can be used ad a source for nutrition with other useful values. This mini-review elaborates on details of its health benefits.

Endlicheria anomala 메탄올 추출물에 의한 인체 폐암세포주와 간암세포주의 자가사멸 유도 (Induction of Apoptosis by Methanol Extract of Endlicheria anomala in Human Lung and Liver Cancer Cells)

  • 본 연구에서는 인체 폐암세포주인 A549 세포와 간암세포주인 HepG2 세포를 사용하여 Endlicheria anomala 메탄올 추출물(Methanol extract of E. anomala, MEEA)의 항암 활성 및 그 분자적 기전에 관하여 분석하였다. 먼저 MEEA가 인체 폐암세포와 간암세포의 증식에 미치는 영향을 분석한 결과, 암세포의 증식을 억제하는 효과가 탁월하였다. 그 후 MEEA에 의한 세포 증식이 억제되는 원인을 분석하기 위하여 Flow cytometry analysis, AnnexinV & 7-AAD 이중 염색을 수행한 결과, 두 세포에서 모두 농도의존적으로 apoptosis 유발군인 SubG1기의 세포 분포가 증가하였고, early apoptosis에서 late apoptosis로 전환되는 공통적인 현상을 확인할 수 있었다. 또한 apoptosis의 유발로 일어날 수 있는 세포의 형태 변화를 관찰하기 위한 DAPI 염색과 DNA fragmentation을 통하여 MEEA 처리에 의한 A549의 염색질 응축, 사멸체 형성 및 DNA의 끌림 현상을 관찰할 수 있었으며, 이와 관련된 분자적 기전 분석을 위한 Western blot을 추가로 수행하여 caspase, PARP, pro-, anti-apoptotic 단백질의 발현을 관찰하였다. 이상의 결과로 MEEA는 인체 폐암세포와 간암세포에서 p53의 발현 증가와 Bcl-2 family의 변화를 유도하며, caspase-3와 관련된 경로를 통하여 농도의존적으로 apoptosis를 유발시킨다는 것을 증명하였다. 이는 MEEA가 항암 활성을 보유하고 있고, 인체 폐암세포와 간암세포 사멸의 기전 연구를 위한 중요한 자료가 될 수 있음을 시사한다.

Anti-cancer and Anti-inflammatory Properties of Korean Citrus Fruits (Citrus aurantium L.)

  • Purpose. Citrus aurantium L.(familyRutaceae), alsoknownasbitter orange, have been used as traditional herbal medicine in many Asian countries since ancient times. Hence, the purpose of the study was to briefly discuss the new findings about anti-inflammatory and anti-cancer activities of Citrus aurantium L in-vitro. Methods. The articles for this study were collected from pubmed and scopus electronic resources. Results. Citrus aurantium L contains an abundant Flavonoids, including hesperidin, naringin and nobiletin. These Flavonoids has reported to have various medicinal benefits that include antioxidant, antimicrobial, anti-inflammatory, anticancer, anti-diabetic activities, and also used to treat cardiovascular diseases. Conclusion. Based on the above evidence, we propose that Flavonoids from Korea Citrus aurantium L would be a therapeutic potential for cancer treatment and pharmacological benefit for inhibiting the inflammatory effect.

Anti-cancer and -Metastatic Effects of Lactobacillus Rhamnosus GG Extract on Human Malignant Melanoma Cells, A375P and A375SM

  • Human malignant melanoma is an aggressive skin cancer which has been rising at a greater rate than any other cancers. Although various new therapeutic methods have been developed in previous studies, this disease has properties of high proliferation and metastasis rate which remain obstacles that have lead to a poor prognosis in patients. It has been reported that a specific Lactobacillus extract has anti-cancer and -metastasis effect in vitro and in vivo. However, previous research has not specified precisely what effect the Lactobacillus rhamnosus GG (LGG) extract has had on human malignant melanomas. In this study, we showed that the LGG extract has anti-cancer and -metastasis effects on the human malignant melanoma cell lines, A375P and A375SM. At first, it was found that, while the LGG extract affects human neonatal dermal fibroblasts slightly, it induced the dose-dependent anti-cancer effect on A375P and A375SM by a WST-1 proliferation assay. As a result of a real-time PCR analysis, the expression patterns of several genes related to cell cycle, proliferation, and apoptosis were modulating in a manner that inhibited the growth of both malignant melanoma cell lines after the treatment of the LGG extract. Furthermore, genes related to the epithelial-mesenchymal transition were down-regulated, and migration rates were also decreased significantly by the LGG extract. Our study showed that the LGG extract could be used as a potential therapeutic source.

Sulfasalazine Induces Autophagic Cell Death in Oral Cancer Cells via Akt and ERK Pathways

  • Sulfasalazine (SSZ) is an anti-inflammatory drug that has been used to treat inflammatory bowel disease and rheumatoid arthritis for decades. Recently, some reports have suggested that SSZ also has anti-cancer properties against human tumors. However, little is known about the effects of SSZ on oral cancer. The aim of this study was to investigate the anti-cancer effects of SSZ in oral squamous cell carcinoma (OSCC) cells and to elucidate the mechanisms involved. The authors investigated the anti-proliferative effect of SSZ using the MTT method in HSC-4 cells (an OSCC cell line). Cell cycle analysis, acidic vesicular organelle (AVO) staining, monodansylcadaverine (MDC) staining and Western blotting were also conducted to investigate the cytotoxic mechanism of SSZ. SSZ significantly inhibited the proliferation of HSC-4 cells in a dose-dependent manner. In addition, SSZ induced autophagic cell death, increased microtubule-associated protein 1 light chain (MAP1-LC; also known as LC) 3-II levels, as well as induced punctate AVO and MDC staining, resulted in autophagic cell death. Furthermore, these observations were accompanied by the inhibition of the Akt pathway and the activation of ERK pathway. These results suggest that SSZ promotes autophagic cell death via Akt and ERK pathways and has chemotherapeutic potential for the treatment of oral cancer.

Anti Tumoral Properties of Punica granatum (Pomegranate) Seed Extract in Different Human Cancer Cells

  • Background: Punica granatum (PG) has been demonstrated to possess antitumor effects on various types of cancer cells. In this study, we determined antiproliferative properties of a seed extract of PG (PSE) from Iran in different human cancer cells. Materials and Methods: A methanolic extract of pomegranate seeds was prepared. Total phenolic content (TPC) and total flavonoid content (TFC) were assessed by colorimetric assays. Antioxidant activity was determined with reference to DPPH radical scavenging activity. The cytotoxicity of different doses of PSE (0, 5, 20, 100, 250, 500, $1000{\mu}g/ml$) was evaluated by MTT assays with A549 (lung non small cell carcinoma), MCF-7 (breast adenocarcinoma), SKOV3 (ovarian cancer cells), and PC-3 (prostate adenocarcinoma) cells. Results: Significant (P<0.01) or very significant (P<0.0001) differences were observed in comparison to negative controls at all tested doses ($5-1000{\mu}g/ml$). In all studied cancer cells, PSE reduced the cell viability to values below 23%, even at the lowest doses. In all cases, IC50 was determined at doses below $5{\mu}g/ml$. In this regard, SKOV3 ovarian cancer cells were the most responsive to antiproliferative effects of PSE with a maximum mean growth inhibition of 86.8% vs. 82.8%, 81.4% and 80.0% in MCF-7, PC-3 and A549 cells, respectively. Conclusions: Low doses of PSE exert potent antiproliferative effects on different human cancer cells SKOV3 ovarian cancer cells as most and A549 cells ar least responsive regarding cytotoxic effects. However, the mechanisms of action need to be addressed.
