MCF-7 유방암 세포에서 영릉향 추출물에 의한 Apoptosis 유도에 관한 연구 (The Extract from Lysimachia foenum-graecum Induces Apoptosis in MCF-7 Breast Cancer Cells)

  • The extract from Lysimachia foenum-graecum (LFE) has been known to possess various instructive characters including anti-oxidant, anti-obesity, fungicidal activities. However, the accurate mechanism of those effects of LFE is not well known. In that respect, we evaluated the apoptotic effect and anti-cancer efficacy of extracts of LFE in MCF-7 breast cancer cells. In this study, we hypothesized that LFE may exert cancer cell apoptosis through regulating p53 and mitochondria-mediated apoptotic proteins. And this substance can generate ROS to cause free radical-induced apoptosis. Accordingly, the generation of ROS by LFE triggers the activation of p53 which are accompanied by pro-apoptotic protein activation and suppression of pro-survival proteins. We determined with MTT assay, flow cytometry for detection of intracellular ROS and Annexin V-PI staining, Western blotting. Consequently, our researches demonstrated that the treatment of LFE to breast cancer cells resulted in an activation of p53, Puma, Bax, cleaved-PARP and an inhibition of Bcl-2 expressions.

  • 개똥쑥 추출물(AAE)은 암에 효과적인 약초로 알려져 있다. Apoptosis는 프로그램화된 세포사멸로 미토콘드리아는 세포사멸 조절에 중요한 역할을 한다. 이 연구는 A549 폐암세포에서 Bcl-2 하위조절과 미토콘드리아 경로를 통한 AAE의 p53 비의존적인 세포사멸을 보여주고 있다. AAE는 p-Akt, cox-2, p53 그리고 미토콘드리아 조절 단백질을 통해 암세포의 사멸을 촉진한다. p-Akt/cox-2 단백질은 세포 증식과 생존에 중요한 역할을 한다고 알려져 있다. Bax, Bak, Bim과 같이 세포사멸을 촉진하는 Bcl-2 단백질은 미토콘드리아 외막의 투과성을 조절한다. AAE의 처리는 p-Akt, p-Mdm2, cox-2 그리고 anti-apoptotic 단백질과 같이 세포사멸을 억제하는 단백질들의 발현을 감소시키는 반면에 암 억제자인 p53과 pro-apoptotic 단백질들을 증가시킨다. Bax/Bak의 활성화는 caspase를 활성화시키기 위해 cytochrome c를 미토콘드리아에서 세포질로 방출하도록 한다. Caspase-3는 apoptosis 과정과 관련된 주요 effector caspase이다. Caspase-3는 일반적으로 pro-enzyme형태로 세포질에 존재한다. Apoptosis의 개시단계에서 caspase-3는 proteolytic cleavage에 의해 활성화되고 활성화된 caspase-3는 PARP를 분해한다. Apoptosis와 관련된 단백질들의 신호전달 사이의 상관관계를 알기 위해 Pifithrin-α (p53 inhibitor)와 Celecoxib (cox-2 inhibitor)을 처리했다. 이러한 결과를 통해 A549 폐암 세포에 AAE를 처리하였을 때 p53-independent 경로를 통해 apoptosis가 유도된다는 것을 확인하였다.

Apoptotic effect of physcion isolated from marine fungus Microsporum sp. in PC3 human prostate cancer cells

  • Background: Apoptosis is a process of programmed cell death, and apoptosis defect results in serious diseases such as cancer. Apoptosis induction is one of the key mechanisms of anti-cancer agents. This study was aimed to find anti-prostate cancer compounds from marine-derived fungus Microsporum sp. Results: We found that physcion isolated from the fermentation broth extract of the marine fungus Microsporum sp. strain MFS-YL decreases the cell proliferation of PC3 human prostate cancer cells. Physcion induced cell apoptosis as determined by Annexin V/propidium iodide double staining. Physcion downregulated the anti-apopotoic proteins such as Ras, Bcl-xL, and Bcl-2, whereas upregulated the pro-apoptotic Bax. Physcion also activated caspase-3, caspase-8, and caspase-9. Conclusion: These results suggest that physcion from Microsporum sp. inhibits the proliferation of PC3 human prostate cancer cells via the pathway leading to apoptotic cell death. Physcion may be a potential candidate in the field of anticancer drug discovery against human prostate cancer.

Association of a Methanol Extract of Rheum undulatum L. Mediated Cell Death in AGS Cells with an Intrinsic Apoptotic Pathway

  • Objectives: Rheum undulatum L. has traditionally been used for the treatment of many diseases in Asia. However, its anti-proliferative activity in cancer has still not been studied. In the present study, we investigated the anti-cancer effects of methanol extract of Rheum undulatum L. (MERL) on human adenocarcinoma gastric cell lines (AGS). Methods: To investigate the anti-cancer effect of MERL on AGS cells, we treated the AGS cells with varying concentrations of MERL and performed 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assays. Cell cycle analyses, measurements of the mitochondrial membrane potential (MMP), caspase activity assays and Western blots were conducted to determine whether AGS cell death occurred by apoptosis. Results: Treatment with MERL significantly inhibited growth of AGS cells in a concentration dependent manner. MERL treatment in AGS cells leaded to increased accumulation of apoptotic sub G1 phase cells in a concentration dependent manner. In control cultures, 5.38% of the cells were in the sub G1 phase. In MERL treated cells, however, this percentage was significantly increased (9.95% at $70{\mu}g/mL$, 15.94% at $140{\mu}g/mL$, 26.56% at $210{\mu}g/mL$ and 38.08% at $280{\mu}g/mL$). MERL treatment induced the decreased expression of pro-caspase-8 and -9 in a concentration dependent manner, whereas the expression of the active form of caspase-3 was increased. A subsequent Western blot analysis revealed increased cleaved levels of poly (ADP-ribose) polymerase (PARP) protein. Also, treatment with MERL increased the activities of caspase-3 and -9 compared with the control. MERL treatment increased the levels of the pro-apoptotic truncated Bid (tBid) and Bcl2 Antagonist X (Bax) proteins and decreased the levels of the anti-apoptotic B-cell lymphoma 2 (Bcl-2) protein, whose is the stabilization of mitochondria. However, inhibitions of p38, extracellular signal regulated kinases (ERKs) and C-Jun N-terminal kinases (JNK) by MERL treatment did not affect cell death. Conclusion: These results suggest that MERL mediated cell death is associated with an intrinsic apoptotic pathway in AGS cells.

Anti-apoptotic Effects of Red Ginseng on Oxidative Stress Induced by Hydrogen Peroxide in SK-N-SH Cells

  • Ginseng (Panax ginseng C.A. Meyer) has been shown to have anti-stress effects in animal studies. However, most studies have only managed to detect altered levels of biomarkers or enzymes in blood or tissue, and the actual molecular mechanisms by which ginseng exerts these effects remain unknown. In this study, the anti-oxidative effect of Korean red ginseng (KRG) was examined in human SK-N-SH neuroblastoma cells. Incubation of SK-N-SH cells with the oxidative stressor hydrogen peroxide resulted in significant induction of cell death. In contrast, pre-treatment of cells with KRG decreased cell death significantly. To elucidate underlying mechanisms by which KRG inhibited cell death, the expression of apoptosis-related proteins was examined by Western blot analysis. KRG pre-treatment decreased the expression of the pro-apoptotic gene caspase-3, whereas it increased expression of the anti-apoptotic gene Bcl-2. Consistent with this, immunoblot analysis showed that pre-treatment of the SK-N-SH cells with KRG inhibited expression of the pro-inflammatory gene cyclooxygenase 2 (COX-2). RT-PCR analysis revealed that the repression of COX-2 expression by KRG pre-treatment occurred at the mRNA level. Taken together, our data indicate that KRG can protect against oxidative stress-induced neuronal cell death by repressing genes that mediate apoptosis and inflammation.

참나무 목초액에 의한 전립선암세포의 apoptosis 유발기전에 관한 연구 (Up-regulation of Bax and Down-regulation of Bcl-2 in Oak Smoke Flavoring(Holyessing)-induced Apoptosis of Human Prostate Carcinoma Cells)

  • We investigated the effects of Oak smoke flavoring (OSF, Holyessing) on the growth of DU145 and PC-3 human prostate carcinoma cells. OSF treatment resulted in a concentration-dependent growth inhibition in both DU145 and PC3 cell lines. The anti-proliferative effect of OSF treatment was associated with the induction of apoptotic cell death which was confirmed by morphological change such as membrane shrinking, rounding up and chromatin condensation in DU145 and PC-3 cells. DNA flow cytometry analysis confirmed that OSF treatment increased population of apoptotic sub-G1 phase. Furthermore, we observed an increase of pro-apoptotic protein Bax expression and a decrease of anti-apoptotic protein Bcl-2 by OSF treatment in a dose-dependent manner. OSF also induced a proteolytic cleavage of specific target proteins such as poly(ADP-ribose) polymerase (PARP) and β-catenin proteins. The present results indicated that OSF-induced inhibition of human prostate carcinoma cell proliferation is associated with the induction of apoptosis.

Inhibitory Effect of Bee Venom Toxin on Lung Cancer NCI H460 Cells Growth Through Induction of Apoptosis via Death Receptor Expressions

  • Objectives : I investigated whether bee venom inhibit cell growth through enhancement of death receptor expressions in the human lung cancer cells, NCI-H460. Methods : Bee venom(1-5 ${\mu}g/ml$) inhibited the growth of NCI-H460 lung cancer cells by the induction of apoptotic cell death in a dose dependent manner. Results : Consistent with apoptotic cell death, expression of TNF-R1, TNF-R2, FAS, death receptors(DR) 3, 4, 5 and 6 was increased in the cells. Expression of DR downstream pro-apoptotic proteins including Caspase-8, -3, -9 was upregulated and Bax was concomitantly overwhelmed the expression of Bcl-2. NF-kB were inhibited by treatment with bee venom in NCI-H460 cells through TNF response change led by TNF-R1 and TNF-R2. Conclusions : These results suggest that bee venom should exert anti-tumor effect through induction of apoptotic cell death in NCI-H460 human lung cancer cells via enhancement of death receptor expression, and that bee venom could be a promising agent for preventing and treating lung cancer.

Fungal Taxol Extracted from Cladosporium oxysporum Induces Apoptosis in T47D Human Breast Cancer Cell Line

  • Purpose: The present study concerns molecular mechanisms involved in induction of apoptosis by a fungal taxol extracted from the fungus Cladosporium oxysporum in T47D human breast cancer cells. Materials and Methods: Apoptosis-induced by the fungal taxol was assessed by MTT assay, nuclear staining, DNA fragmentation, flow cytometry and pro- as well as anti-apoptotic protein expression by Western blotting. Results: Our results showed inhibition of T47D cell proliferation with an $IC_{50}$ value of $2.5{\mu}M/ml$ after 24 h incubation. It was suggested that the extract may exert its anti-proliferative effect on human breast cancer cell line by suppressing growth, arresting through the cell cycle, increase in DNA fragmentation as well as down-regulation of the expression of NF-${\kappa}B$, Bcl-2 and Bcl-XL and up-regulation of pro-apoptotic proteins like Bax, cyt-C and caspase-3. Conclusions: We propose that the fungal taxol contributes to growth inhibition in the human breast cancer cell through apoptosis induction via a mitochondrial mediated pathway, with possible potential as an anticancer therapeutic agent.

Impact of imatinib administration on the mouse ovarian follicle count and levels of intra-ovarian proteins related to follicular quality

  • Objective: The impact of imatinib, a tyrosine kinase inhibitor, on ovarian follicles and several proteins related to follicular function and apoptosis was investigated in mice. Methods: Saline, cyclophosphamide (Cp; 50 or 75 mg/kg), or imatinib (7.5 or 15 mg/kg) was injected once intraperitoneally into female B6D2F1 mice (18 mice in each group). In multiple ovarian sections, the number of various types of follicles and the proportion of good-quality (G1) follicles were counted. The levels of six proteins (anti-Müllerian hormone [AMH], BCL-xL, BAX, acid sphingomyelinase [A-SMase], caspase-3, and α-smooth muscle actin [α-SMA]) within the whole ovaries were quantified using Western blots. Results: Compared to the saline group, a significant reduction of the primordial follicle count was observed in the group treated with imatinib 7.5 and 15 mg/kg, as well as in the group treated with Cp 75 mg/kg. Administration of Cp significantly decreased the proportion of G1 primordial follicles, but administration of imatinib did not. No differences in the AMH, anti-apoptotic BCLX-L, pro-apoptotic BAX, and A-SMase levels in the ovarian tissues were observed among the five groups. However, caspase-3 and α-SMA levels were significantly higher in the imatinib and Cp groups than in the saline group. Conclusion: The administration of imatinib to mice significantly reduced the primordial follicle count and increased the protein levels of caspase-3 and α-SMA. Our findings suggest that imatinib potentially exerts ovarian toxicity via apoptotic processes, similarly to Cp.

매실(Prunus mume) 메탄올 분획물의 처리에 따른 인체 전립선암세포 LNCaP의 apoptosis 유도 효과 (Induction of Apoptosis by Treatment of Human Prostate Cancer LNCaP Cells with Methanol Fractions from Prunus mume)

  • 본 연구에서는 매실 메탄올 추출물(maesil methanol fraction, MMF)을 제조하여 인체 전립선암세포 LNCaP, RC-58T 및 PC-3에 대한 증식억제 효과를 확인하였다. 인체 전립선암세포인 PC-3 및 RC-58T와 비교해보았을 때, LNCaP은 MMF의 처리에 따른 증식억제 효과에 가장 민감했다. LNCaP의 형태학적 관찰과 apoptotic body 형성을 관찰해보았으며, MMF의 처리로 인한 형태의 변화, 핵 손상 및 응축을 확인했다. MMF의 처리로 인한 인체 전립선암세포 LNCaP에서 성장억제 효과가 내인성 apoptosis 경로와 관련 있는지 확인한 결과, pro-apoptotic 단백질인 Bax, caspase-3, caspase-9, PARP의 발현이 증가하였고, anti-apoptotic 단백질인 Bcl-2의 발현이 감소하는 것을 확인했다. MMF와 AIF inhibitor인 N-phenylmalemide (N-PM)의 병용처리군에 비해 MMF 단독처리군의 증식억제 효과가 유의적으로 나타났으며 AIF 및 Endo G의 발현 증가를 통해 외인성 apoptosis 경로에 영향을 미치는 것을 확인했다. 또한 PI3K inhibitor인 LY294002와 MMF의 병용처리군에 비해 MMF 단독처리군의 증식억제 효과가 유의적으로 나타났으며 PI3K, p-Akt, p-mTOR의 발현 감소를 통해 PI3K/Akt/mTOR 신호경로에 영향을 미치는 것을 확인했다. 결론적으로 인체 전립선암세포 LNCaP에서 MMF의 증식억제 효과는 천연물 유래 기능성 식품의 소재로써의 가능성을 보여준다.
