Anti-aging and Anti-inflammatory Activities of the Extracts of Calamagrostis arundinacea (Calamagrostis arundinacea (실새풀) 추출물의 항노화 및 항염증 활성)

  • The anti-aging and anti-inflammatory activities of hot-water (Ca-HW) and 70% ethanol (Ca-E70) whole-plant Calamagrostis arundinacea extracts, as well as their bioactive potentials, were investigated using cell-free and cell-mediated experimental systems. Use of the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical decolorization assay to evaluate the antioxidant activity of the Ca-HW and Ca-E70 extracts revealed DPPH radical scavenging activities of 27% and 48%, respectively. Neither extract caused significant cytotoxicity, and both showed cell proliferation and promotion effects using RAW 264.7, B16F10, and CCD986sk cells. B16F10 melanoma cells showed higher melanin synthesis when treated with 100 mg/ml Ca-HW or Ca-E70 than with arbutin, indicating a stronger inhibitory effect of arbutin on melanin synthesis. Ca-HW and Ca-E70 increased pro-collagen biosynthesis in the human fibroblast CCD986-SK cell line by 24.69% and 12.55%, respectively. Analysis of the anti-inflammatory effects of different concentrations of Ca-HW and Ca-E70 in RAW264.7 cells revealed that Ca-E70 appeared to inhibit the lipopolysaccharide-induced production of nitric oxide and IL-6, a proinflammatory cytokine; therefore, Ca-E70 showed an anti-inflammatory effect. These results suggested that C. arundinacea extracts could have skin anti-aging and anti-inflammatory properties.

  • Various kinds of positive effects of green tea extracts had been studied for long time which included anti-inflammation, anti-aging, and cardiometabolic effects. Although topical steroid and non-steroidal calcineurin inhibitors may control clinical symptoms of allergic contact dermatitis, some of patients also present allergic reaction to these topical agents. Therefore, we have tried green tea extracts for managing this skin disorder with expectation of anti-inflammatory effect without potential side effects including skin irritation and toxic responses. The toxicity test of green tea extract also did not show any sign of irritation in the skin throughout the test period. Moderate severity of allergic contact dermatitis presented satisfactory clinical outcome at second week follow-up which was final visit of outpatient. This result mean that green tea extract has a positive effect for managing allergic contact dermatitis but its potency and efficacy seem to be so not strong enough to control moderate severity allergy skin lesion. In this pilot study, we were able to conclude that green tea cell extracts might be applied for potential anti-inflammatory soaking without skin toxicity.

The Influence of Long-term Aloe Supplement on Anti-oxidative Defenses and Cholesterol Content in Brain and Kidney of Aged Rats

  • The present study was investigated the anti-oxidative effects of aloe vera ingestion on brain and kidney in aged rats by monitoring several oxidative-related parameters. Male specific pathogen-free Fischer 344 rats were randomly divided into four groups of five rat each: Group A was fed test chow without aloe supplementation; Group B was fed a diet containing a 1% freeze-dried aloe filet; Group C was fed a diet containing a 1% charcoal-processed, freeze-dried aloe filet; and Group D was fed a diet containing a charcoal-processed, freeze-dried, whole leaf aloe in drinking water. Analyses of tissues were done at 4 months and 16 months of age. Results showed that a long-term intake of aloe, regardless of the preparation used, enhanced antioxidant defenses against lipid peroxidation, as indicated by reduced phosphatidylcholine hydroperoxide levels in both brain and kidney. The additional benefit of aloe intake on the anti-oxidative action was evidenced by enhanced superoxide dismutase and catalase activity in all aloe-ingested groups. Another beneficial effect of aloe shown in this study, although not an anti-oxidative parameter, was its cholesterol-lowering effect as detected in brain and kidney with significant decreases at age16 months of aloe-fed rats. Based on these findings, we conclude that a long-term dietary aloe supplementation modulated the anti-oxidative defense systems and cholesterol level.

Anti-inflammation effect of extract from Zostera marina using UVB-induced damage on keratinocytes (잘피 추출물의 UVB로 손상을 유도한 각질형성세포에 대한 항염 효능)

  • Objectives : In order to confirm whether extracts of different parts of Zostera marina (ZM), a marine flowering plant, can be used as cosmetic ingredients, this study evaluated their cytotoxicity and cytoprotective effects against ultraviolet B (UVB). Inflammatory responses induced by UV stimuli are also associated with the aging of the skin.Methods : We investigated the effects of ZM extracts on cells through the water soluble tetrazolium salt-1(WST-1) assay for cell viability. In order to investigate the anti-inflammatory effects, we evaluated the suppression of Cyclooxygenase-2 (COX-2) expression by ZM extracts in HaCaT cells with UVB-induced damages, and also evaluated the production of Prostaglandin E2 (PGE2) in RAW 264.7 cells with LPS-induced damages.Results : High cell viabilities above 90% were observed in all types of ZM extracts, except for whole ZM extract at 0.5 mg/ml; in keratinocytes with UVB-induced damages, the cell viabilities were above 80% when treated with all types of ZM extracts. We confirmed their anti-inflammatory effects by investigating the suppression of inflammatory mediators. In keratinocytes with UVB-induced damages, COX-2 expression decreased in the experimental group treated with ZM extract. Similarly, in RAW 264.7 cells where inflammation was induced with LPS, the biosynthesis of PGE2 was inhibited.Conclusion : These results suggest that ethanol extracts from Zostera marina may have value as the potential anti-inflammatory medicinal plant. Also based on the abovementioned results, ZM extract protects skin cells from UV-induced damages, and thus can be used in topically applied products for skin protection.

Anti-Oxidant Effect of Hyeongbangjiwhang-tang Decoction in Kidney, Bladder and Spleen Cell of SD Rats (형방지황탕(荊防地黃湯) 전탕액이 노화쥐의 신장, 비장, 방광 세포의 항산화능에 미치는 영향)

  • 1. Objectives The purpose of this study is to investigate the anti-aging and anti-oxidant effects of Hyeongbangjiwhang-tang decoction(HGD) in SD rats. 2. Methods This experiment was used the tissue of kidney, bladder and spleen cells of 6, 52 and 68 weeks old SD rats. Each age group was again divided into three groups. One group, as normal group, was not-treated cells, another group, as control group, was saline-treated cells, and the last group, as experimental group, was HG-treated cells. After culture for 48 hours, each groups measured the level of SOD, GSH, MDA and NO in the tissue of kidney, bladder and spleen cells. 3. Results and Conclusions The activity of SOD were significantly increased in kidney cell of 68 w-HGD group, bladder cell of 52 w-HGD group and in spleen cell of 52,68 w-HGD group compared with those of the control groups. The level of GSH were significantly increased in bladder cell of 52w-HGD group and in spleen cell of 68w-HGD group compared with those of the control groups. The level of MDA were significantly decreased in bladder cell of 68 w-HGD group and in spleen cell of 68w-HGD group compared with those of the control groups. The level of NO were significantly decreased in kidney cell of 68 w-HGD group, bladder cell of 52, 68w-HGD groups, and in spleen cell of 68w-HGD group compared with those of the control groups. These results suggest that HGD(Hyeongbangjiwhang-tang) has anti-oxidant effects in aged rats.

  • This study was designed to investigate the effects of pine (Pinus densiflora Sieb et Zucc) needle extract (PNE) on membrane fluidity and oxidative stress in liver membranes of Sprague-Dawley (SD) rats as a study on investigation of anti-aging effect and determination of chemical structures of PNE through the animal experiments. Male SD rats were fed basic diets (control group) and experimental diets (0.5% and 1.0%-PNE group) for 6 weeks. Administrations of 0.5% and 1.0%-PNE resulted in a marked decreases (15∼25% and 23∼26%, respectively) in cholesterol accumulations of liver mitochondria and microsomes compared with control group. Membrane fluidities were significantly increased (15∼25%) in liver microsomes of 0.5% and 1.0%-PNE groups compared with control group. Formations of basal and induced oxygen radicals (BOR and IOR) in liver mitochondria were significantly inhibited (11∼12% and 10∼15%, respectively) by administrations of 0.5% and 1.0%-PNE compared with control group. Lipid peroxide (LPO) levels were remarkbly decreased about 20% in liver mitochondria and microsomes of 0.5% and 1.0%-PNE groups compared with control group. Oxidized protein levels calculated with carbonyl group were significantly decreased about 15% in liver mitochondria of 1.0%-PNE group compared with control group. These results suggest that PNE may play a effective role in a attenuating a oxidative stress and increasing a membrane fluidity.

  • These studies were designed to investigate the effects of pine (Pinus densiflora Sieb et Zucc.) needle extract (PNE) on oxygen radicals and their scavenger enzymes in liver membranes of Sprague-Dawley (SD) rats as a study on investigation of anti-aging effect and determination of chemical structures of PNE through the animal experiments. Male SD rats were fed basic diets (control group) and experimental diets (0.5% and 1.0%-PNE group) for 6 weeks. There were no significant differences in hydroxyl radical (·OH) formations of liver mitochondria and microsomes in 0.5%-PNE group, while ·OH formations were significantly decreased (10% and 18%, respectively) in liver mitochondria and microsomes of 1.0%-PNE group compared with control group. Microsomal hydrogen peroxides and cytosolic superoxide radicals were remarkably decreased (20% and 20∼25%, respectively) in 0.5% and 1.0%-PNE groups compared with control group. Mn-SOD activities in mitochondria were significantly increased about 10% in 1.0%-PNE group, while Mn-SOD activities in mocrosomes were remarkably increased (16∼20%) in 0.5% and 1.0%-PNE groups compared with control group. There were no significant differences in Cu, Zn-SOD activities of liver cytosol in 0.5% and 1.0%-PNE groups, while glutathione peroxidase (GSHPx) and catalase (CAT) activities were significantly decreased (28∼30% and 15∼30%, respectively) in liver cytosols of 0.5% and 1.0%-PNE groups compared with control group. These results suggest that these PNE may play a effective role in a attenuating a oxygen radical formations and increasing a scavenger enzyme activities.

Effect of Melothria heterophylla Extract on Expression of Matrix Metalloproteinase-1 in Human Skin Fibroblasts (백렴 추출물의 Matrix Metalloproteinase-1 발현 저해 효과)

  • Matrix metalloproteinases (MMPs) are known to play an important role in photoaging by mediating the degradation of extracellular matrix proteins. In this study, to develop a n ew anti-aging agent, we investigated the antioxidant activity and the inhibitory effect of Melothria heterophylla extract on expression of MMP-1 in UVA-irradiated human dermal fibroblasts and MMP-1 activity. The M.heterophylla extract was found to scavenge radicals and reactive oxygen species (ROS) with the $SC_{50}$ values of $13{\mu}g/ml$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and $20{\mu}g/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. UVA-induced MMP-1 expression was reduced about 80% by $100{\mu}g/ml$ of the M.heterophylla extract but MMP-1 Mrna expression was not inhibited. Therefore, we conclude that the M.heterophylla extract significantly inhibits MMP-1 expression at the protein level. Also, the M.heterophylla extract inhibited MMP-1 activity in a dose dependent manner. From these results, we suggest that the M.heterophylla extract can be used as a new anti-aging agent by antioxidant activity, regulation of UVA-induced MMP-1 production, and inhibition of MMP-1 activity.

Effect of laser-assisted multi-point zona thinning on development and hatching of cleavage embryos in mice

  • Objective: This study aimed to examine the effect of laser-assisted zona thinning (LAZT) at one or four-points on the blastocyst formation and hatching process in mice with respect to female age. Methods: Eight-cell or morula embryos collected from superovulated C57BL female mice with different ages (6-11 and 28-31 weeks) were treated with LAZT at one-point (LAZT1) or four-points (LAZT4). The zona pellucida was thinned to more than 70% of its initial thickness by making two holes of $15-20{\mu}m$. Results: In the young mice, LAZT resulted in a significant increase in early hatching and hatching rates compared to the control group (p<0.05). However, in the old mice, LAZT significantly increased blastocyst formation as well as early hatching and hatching compared to the controls (p<0.05). These effects were more remarkable in LAZT4 than in LAZT1 and in aged mice than in young ones. Conclusion: These results show that multi-point LAZT leads to a significant improvement of blastocyst formation and hatching in mice compared to controls.

Antimicrobial Activity on Respiration Diseases Inducing Bacteria and Antioxidant Activity of Water Extracts from Wild Edible Vegetables (산채류 추출물의 항산화 활성 및 호흡기 질환을 유발하는 세균에 대한 항균활성)

  • To investigate the antimicrobial activity on bacteria causing a respiration disease and antioxidant effects of water extracts from 12 kinds of wild edible vegetables, we extracted the water extracts for 72 h in $7^{\circ}C$ using distilled water as solvent. The water extracts except Ixeris dentate and Allium monanthum had high concentrations of phenol compounds and flavonoids. Liguraia fischeri specially had the highest level on total phenol compounds and flavonoids with 205 ${\mu}g/mL$ and 98. 86 ${\mu}g/mL$, respectively. The each 0.05% extracts of Sedum sarmentosum and Liguraia fischeri had high effect on the DPPH radical scavenging activity among wild edible vegetables and the most extracts promoted antioxidant activity with increasing concentration of extract. The catalase activity of Erysimum aurantiacum and Aralia elata showed more than 150 units per g of fresh tissue. The effect of antimicrobial activity on water extracts showed characteristic activity. Only Staphylococcus aureus KCTC 1928 and Corynebacterium diptheriae KCTC 3075 were inhibited cell growth on the other hands, the remainder of bacteria was not inhibited cell growth. Nevertheless, the extracts of wild edible vegetables had specific concentration as MIC for antimicrobial activity respectively. In case of the extract of Aster scaber, Erysimum aurantiacum, and Allium monanthum had over 30% antimicrobial activity on the bacteria causing a respiration disease. In results, the wild edible vegetables include high concentrations of total phenol compounds and flavonoids that give a good antioxidant activity and antimicrobial activity. Therefore the wild edible vegetables are functional food for anti-aging and physiological activation.
