The trend towards ageing populations has been observed over many years in Europe and the US but has accelerated significantly in developed countries in Asia including Japan and South Korea. In the latter country the elderly population (65+) has increased 5-fold between 1960 and 2000 and this group will comprise 40% of the population by 2050. This creates a new socio-economic group with specific demands and considerable spending power. As ageing occurs a range of changes occur in the body that can be moderated by adjustments in nutrition. A significant body of evidence points to changes in the balance of glutathione synthesis and utilisation as people age. Glutathione is tile most important natural anti-oxidant of the body and the amounts present can become limited by available cysteine in the diet. A cysteine-enriched peptide product, Cysteine Peption$^{TM}$ has been developed by DMV International for dietary supplement and food applications. A qualitative consumer trial has indicated benefits including improved sleep and more energy. Animal and clinical trial will be described that provide indications on bioavailability and possible mechanisms of action of Cysteine Peption$^{TM}$ with particular focus on the ageing population.

Bitter melon (Momordica charantia L.) has various biological functions, including anti-diabetic, anti-cancer, anti-inflammatory, anti-viral, and antioxidant activities. However, the development of foods using bitter melon (BM) is unexplored due to its bitter taste. In this study, BM pickle was prepared, and changes in quality characteristics and antioxidant activity during ageing were determined. After 4 weeks of ageing, pH levels, salinities, soluble solids, and reducing sugars of BM pickle decreased, whereas acidities increased. In addition, total viable, lactic acid bacteria, and yeast viable cell numbers greatly increased until 1 week, after which they slightly decreased. Levels of soluble phenolics increased during ageing, antioxidant activity increased accordingly. Crude protein, K, and P contents of pickles were lower than in raw material of BM, whereas crude fat and Na contents increased. Corn silk extracts were added to BM pickle during ageing in order to improve the bitter taste and flavor. The best sensory quality was obtained by adding 0.25% corn silk extracts.
Anti-aging is one of the key words to have characterized Korean society, and Korean adults in their 40s and 50s are very interested in this word. In this study, their cognitive ages and body images which are expected to have been influenced by anti-ageing were determined and correlation between them was examined. A survey was conducted and 368 of 500 questionnaires were used in data analysis. Data were analyzed by descriptives, factor analysis, reliability analysis, correlation analysis, and multiple response analysis through SPSS 12.0. The results are as follows. First, the cognitive ages of Korean adults in their 40s and 50s showed to be 37.5 years old which are 9.1 years old younger than their average actual ages. Second, their attitudinal body images appeared to be composed of appearance orientation and appearance evaluation, and appearance orientation represented to be a little higher than appearance evaluation. In addition, it showed that the correlation between cognitive ages and the attitudinal body images was low and negative. Third, people in their 40s and 50s revealed to think good health and impression as ideal body images. It was found that they regarded a face as the most important body part for ideal body images and were sensitive of their weight. Forth, correlation between cognitive ages and variables of ideal body images was identified to be significant in 4 variables which are slender, shapely and slim figure and good body proportions. In conclusion, it was shown that Korean adults in their 40s and 50s recognized themselves to be 9.1 years old younger and, the younger they perceive themselves, the younger body images they want. Also, it could be confirmed that anti-ageing is a megatrend in Korean society.

Previously, we reported LVH peptiede derived from Aurelia aurita as cosmecetuticals with anti-winkle activity. In this study, we synthesized pal-LVH using palmitic acid to enhance skin permeability of LVH and examined the effects as cosmeceuticals of pal-LVH. To evaluate these effects, we performed cell toxicity, wound healing, and patch test for skin irritation with LVH and pal-LVH and compared these results for their effects. As a result. pal-LVH was not showed in cytotoxicity and allergenic effect like as LVH. Besides, pal-LVH had almost same excellent anti-ageing properties in high concentration and anti-winkle effect in low concentrationwas as LVH. These results suggested synthesis of palimitic acid and LVH did not affect any functions as cosmeceuticals with increasing skin permeability. Therefore, pal-LVH can be adaptable as new cosmecetuticals with anti-winkle and anti-ageing materials and applied in the development of medicine through various convergence study.
Living cells are continuously subject to all sorts of stress such as ultraviolet rays on skin cells. Tests made in various laboratories show that when young fibroblasts (Le. at the beginning of their proliferate life) were repeatedly put under stress at subletal doses, they acquired a phenotype similar to Senescence Induced Prematurely by Stress (SIPS). The work presented hereafter was made on a new model of senescence induced prematurely by stress from ultraviolet Brays (UVB). The human fibroblast model was put under repeated UVB stress, causing SIPS. Several ageing biomarkers were used in order to characterise the cells that underwent stress:. an increase in the proportion of positive cells with senescence associated $\beta$-galactosidase activity (SA $\beta$-gal) measured by a specific coloration,. the proportion in the different morphological stages that fibroblasts undergo during culture visualised by microscopic observation,. the expression of genes known for overexpressing during senescence, particularly fibronectin and apolipoprotein J, measured by Real Time-PCR,. the common deletion of 4,977 bp in mitochondrial DNA, evaluated by nested PCR. Studying the variation of these 4 biomarkers, we have evaluated the protective effect of a Laminaria digitata extract (LDE) that can be used as a natural active ingredient for anti-ageing cosmetics.

Ageing has been known to be deeply related to oxidative activities. Furthermore, inflammation is a response initiated by conditions such as infection and injury. It has been known that complex connections occur between oxidative stress and the inflammatory response, however underlying mechanism needs to be investigated. Chinese resource plants have been widely used as functional food and medicine for a long time, however it is not studied well how many of these resource plants work. We first decided testing anti-oxidant and anti-inflammatory activities with 95% ethanol extracts of the chinese resource plants, Boehmeria pilosinscula (Blume) Hassk., Carlemannia tetragona Hook f. and Clausena emarginata C.C.Huang. To measure anti-oxidant activity, we performed DPPH assay in Raw264.7 cells with 95% ethanol extracts. Clausena extracts showed significantly higher anti-oxidant activity than those of Boehmeria and Carlemannia. We then performed Nitric Oxide assay to measure the inflammation suppression levels with 95% ethanol extracts of these plants. Clausena extracts showed significantly higher suppression of nitric oxide production than those of Boehmeria and Carlemannia. This indicates that inflammation levels are significantly reduced by Clausena. After measuring anti-oxidant and anti-inflammatory activities, we then performed MTT assay to measure the cell survival rate of Raw264.7 cells treated with these extracts. Boehmeria showed much more cell survival rate than Carlemannia and Clausena. Taken together, this result suggests that Clausena extracts have more anti-oxidant and antiinflammatory activities than Boehmeria and Carlemannia, while Boehmeria extracts have more cell survival rate than Carlemannia and Clausena.
With the progress of chemical researches in ginseng studies, efforts to elucidate the pharmacological actions of ginseng have been greatly increased. The majority of ginseng reaserches in the past has been performed with crude extracts from ginseng roots to verify scientifically the empirical application of ginseng in men and animals recently. Ginseng reaserches have been done mostly with pure ginsenosides and there has been a shift in focus to the various biochemical pathways. It was demonstvated that ginseng had diverse effects by modulating the second-messenger system, such as cyclic nucleotides. calcium The demonstration in 1987 of the formation of nitric oxide(NO, endothelium-derlled rectating factor) by an enzyme in vascular endothelial cells opened up a new area of biological reaserches of ginseng. It was shown that vascular relaxations induced by glnsenosides are mediated by release of nitric oxide from endothelial cells. According to the literature search from'hledline". There have been 737 original and review articles during the last 30 years. In these review articles, an attempt has been made to summalize some results from some of these published papers. Ginseng has a wide range of phal.macologtcal and therapeutical actions. It acts on the centralral nervous system and cardiovascular system, promotes immune function and metabolism. Possesses anti-stress. Anti-cancer and anti-ageing activities, and so on.o on.
Bee venom (Apis mellifera L. BV) has been used as a cosmetic ingredient for anti-ageing, anti-inflammatory and antibacterial functions. The aim of this study was to evaluate the acute toxicity after a single dermal administration of BV, BV was administered to 2 groups of Sprague-Dawley (SD) male and female rats (5 animals/group) at doses of 0 and 1,500 mg/kg body weight (BW). Mortality, clinical signs, body weight changes and gross findings were continually monitored for 15 days following the single dose. There were no unscheduled deaths in any groups during the study period. No BV related clinical signs and body weight changes were observed in any groups during the study period. There were no abnormal gross findings at necropsy on day 15 after the treatment. On the basis of the above results, it was concluded that there were no treatment-related effect on mortality, clinical signs, body weight changes and gross findings in SD rats treated with a single dermal dose of BV at dose of 1,500 mg/kg BW. Therefore, the approximate lethal dose of BV was considered to be over 1,500 mg/kg/day for both sexes of rats. BV may provide a developmental basis for a cosmetic ingredient or external application for topical uses.

The Raf-1 kinase inhibitory protein (RKIP) can regulate multiple key signaling pathways. Specifically, RKIP binds to Raf-1 kinase and inhibits the Ras-Raf-1-MEK1/2- ERK1/2 pathway. Additionally, Raf-1 has been shown to translocate to mitochondria and thereby protect cells from stress-mediated apoptosis. Recently, HBx was found to stimulate the mitochondrial translocation of Raf-1, contributing to the anti-apoptotic effect. We found that RKIP was downregulated during HBx-mediated hepatocarcinogenesis. In this study, we show that RKIP bound to Raf-1 and consequently inhibited the translocation of Raf-1 into mitochondria. This promoted the apoptosis of cells treated with apoptotic stimulus. Thus, the downregulation of RKIP increased the level of free Raf-1 and thereby elevated the mitochondrial translocation of Raf-1 during HBx-mediated hepatocarcinogenesis. The elevated Raf-1 mitochondrial translocation induced the increased anti-apoptotic effect and subsequently promoted HBx-mediated hepatocarcinogenesis.
