Effects of Horse oil on the DNCB-induced Contact Hypersensitivity in Balb/c Mice (마유(馬油)가 DNCB로 유발된 알레르기성 접촉성피부염에 미치는 영향)

  • Objectives : Horse oil (HO) has been used long time as the folk medicine of many Asian countries such as Korea, Mongol, China, India and Japan. HO has been used for anti-bacterial, anti-inflammatory, and anti-pruritic purposes in skin. However, it is still largely unknown whether HO modulates the skin condition. In this study, we attempted to evaluate the anti-inflammatory effect of HO on the 1 % of 2, 4-dinitro-1-chlorobenzene (DNCB)-induced contact hypersensitivity in Balb/c mice. Methods : To find the anti-inflammatory effect of HO, contact hypersensitivity, a local inflammatory response of skin, was induced on the back of Balb/c mice by sensitization and repeated application by 1% DNCB and HO treated 2 weeks on the 1% of DNCB-treated Balb/c mice. Excised mice skins were stained with hematoxylin and eosin and serum IgE level was measured by mouse IgE ELISA kit. Results : In this study, we found that HO reduced erythema by 1% of DNCB treated Balb/c mice. Also, HO recovered histopathological features such as the thickening of epidermis, hyperkeratosis and the infiltration of inflammatory cells in 1% of DNCB treated Balb/c mice. In addition, HO reduced IgE level on the serum obtained from blood of 1% of DNCB-treated Balb/c mice. Conclusion : Taken together, these results showed that HO could be used as a pharmaceutical material with anti-inflammatory effects by reducing of erythema, IgE level and recovering of histopathological features skin on DNCB-induced contact hypersensitivity in Balb/c mice model.

Effects of Piperis Longi Fructus on Regulatory T Cells Number, IgE, Histamine Production in Asthma Model Mice and Th1/Th2 Cytokine Balance in vitro (천식 모델 생쥐에서 필발이 CD25+T 세포수, IgE, Histamine 생성량과 in vitro에서 Th1/Th2 Cytokine Balance에 미치는 영향)

  • Objectives : It has been recently shown that Piperis Longi Fructus (PLF) is involved in the reduction of eosinophil recruitment and production of Th2 cytokines in vivo. However, the main therapeutic mechanisms of PLF remains a matter of considerable debate. To investigate the therapeutic mechanisms of PLF, we examined the influence of PLF on regulatory T cells number, IgE, histamine production in vivo and Th1/Th2 cytokine balance in vitro. Methods : All mice were immunized on two different days (21 days and 7 days before inhalational exposure) by i.p. injections of 0.2 $m\ell$ alum-precipitated Ag containing 100 ${\mu}g$ of OVA bound to 4 mg of aluminum hydroxide in PBS. Seven days after the second sensitization, mice were exposed to aerosolized ovalbumin for 30 min/day on 3 days/week for 12 weeks(at a flow rate of 250 L/min, 2.5% ovalbumin in normal saline) and PLF (150 mg/kg) were orally administered 3 times a week for 8 weeks. Splenocytes from C57BL/6 mice at 8 weeks of age were stimulated with anti-CD3 (1 mg/ml) plus anti-CD28 (1 mg/ml) antibody for 48hrs. IL-4 and IFN-$\gamma$ in the culture supernatants were measured by ELISA Results : The suppressive effects of PLF on asthma model were demonstrated by the increase the number of regulatory T cells and by reducing IgE, histamine production in vivo and modulation of Th1/Th2 cytokine balance. Conclusions : These results indicate that PLF has a deep inhibitory effects on asthma model mice by increase the number of regulatory T cells, and by reducing IgE, histamine production.

  • In a bioassay-guided search for anti-allergic compounds from higher plants of Korea, poly-methoxyflavones, 3',4',5,6,7,8-hexamethoxyflavone (I), 5-hydroxy-3',4',6,7,8-pentamethoxyflavone (II) and 3',4',5,7,8-pentamethoxyflavone (III) have been isolated from the immature peels of Citrus unshiu. Structures of these compounds were elucidated on the basis of spectroscopic techniques. Compounds I and II inhibited dose-dependent histamine release from the rat peritoneal mast cells activated by compound 48/80 or anti-DNP IgE.

Five cases of ocular toxocariasis confirmed by serology

  • We report 5 cases of ocular toxocariasis in Korean adults complaining of visual impairment along with floating or bubbling sensation. Fundoscopic examination revealed a retinal detachment along with exudate in 4 cases. They all showed typical reaction by ELISA and immunoblot against Toxocra excretory-secretory antigen. One case showed high level of anti-Toxocara IgE antibodies (34.000 Toxocara units/L) as well as increased level of serum total IgE antibodies and the specific IgE antibodies for 3 inhalant antigens, suggesting that high level of anti-Toxocara IgE antibodies was associated with an atopic status. Clinical manifestations were improved after the sequential use of steroids then mebendazole. We also suggest that ocular toxocariasis should be thoroughly investigated even when an evocative uniocular inflammatory lesion is encountered in peripheral retina without a systematic disease.

Extract of Malus sieboldii Suppresses IgE-mediated Mast Cell Activation through Inhibition of Syk Kinase (아그배나무(Malus sieboldii)의 IgE 매개성 알레르기 반응 억제 효과 및 기전)

  • Malus sieboldii is a dicotyledonous plant that grows widely in Jeju Island and Ganghwa Island in Korea. Malus sieboldii has been known as a detoxifying and antioxidant plant, but study on allergic diseases is not known. In this study, we investigated the effect of Malus sieboldii extract (MSE) on the activation of mast cells, which is well known to be a critical causative cell to induce allergic diseases. As a result of our experiments, MSE inhibited the degranulation and inflammatory cytokine secretion from mast cells by antigen stimulation. As the mechanism of MSE in mast cells, it inhibited the activation of Syk kinase, a essential signaling protein activated by antigen, and further inhibited activation of $PLC{\gamma}$ and MAP kinase(P38, ERK1/2, and JNK). Furthermore, in vivo animal studies showed that MSE significantly inhibited IgE-mediated passive cutaneous anaphylaxis and passive systemic anaphylaxis in a dose-dependent manner. Taken together, the results of this study showed for the first time that MSE inhibited IgE-mediated allergic responses by suppressing Syk kinase in mast cells. Therefore, it could be considered that MSE is worth developing as an anti-allergic material.

  • The effect of aqueous extract of Terminalia chebula fruit (Combretaceae) (TCAE) by anal administration on mast cell-dependent immediate-type anaphylactic reactions was investigated. TCAE (0.005 to 1 g/kg) inhibited systemic anaphylaxis induced by compound 48/80 in mice. When TCAE was pretreated at the same concentrations with systemic anaphylaxis, the plasma histamine levels were reduced in a dose-dependent manner. TCAE (0.1 and 1 g/kg) also significantly inhibited local anaphylaxis activated by anti-DNP IgE. TCAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-DNP IgE. Moreover, TCAE (0.01 and 0.1 mg/ml) had a significant inhibitory effect on anti-DNP IgE-mediated tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$ production from RPMC. These results provide evidence that anal therapy of TCAE may be beneficial in the treatment of systemic and local mast cell-dependent anaphylaxis.

Intracellular Signaling Pathways for Type II IgE Receptor (CD23) Induction by Interleukin - 4 and Anti - CD40 Antibody

  • Since the role of CD40 on the interleukin-4(IL-4) -induced B cell activation has been strongly implicated in the agumentation of IgE production and response, we have investigated the intracelluar signaling pathways utilized by IL-4 and CD40 for type II IgE receptor (CD23) expression. IL-4 and anti-CD40 antibody treatment of human B cells, independently caused a rapid induction of CD23 gene activation within 2 h. There was a noticeable synergism between the action of the two agents inducing CD23 expression: the addition of anti-CD40 to the IL-4-treated culture significantly agumented the IL-4-induced CD23 on both mRNA and surface protein levels, and the inclusion of IL-4 in the anti-CD40-treated cells caused a further increase of CD23 expression far above the maximal level induced by anti-CD40. Protein tyrosine kinase (PTK) inhibitors effectively suppressed the both IL-4- and anti -CD40-induced CD23 expression. whereas protein kinase C (PKC) inhibitors had no effects. Electrophoretic mobility shift assays (EMSA) have shown that IL-4 and anti-CD40 induce the activation of NF-IL-4 and $NF-_{K}B$, respectively, binding to the CD23 promoter, both in a PKC-independent and PTK-dependent manner. These data suggest that the synergistic activation of CD23 gene expression by IL-4 and anti-CD40 is mediated by co-operative action of distinct nuclear factors. each of which is rapidly activated via PKC-independent and PTK-dependent process.

  • We studied the effects of the powders of Spirulina platensis (SPP) on anaphylactic reactions. SPP inhibited systemic anaphylaxis induced by compound 48/80 in rats. SPP also inhibited local anaphylaxis activated by anti-dinitrophenyl(DNP) IgE. Moreover, SPP dose-dependently inhibited histamine release in rat peritoneal mast cells activated by compound 48/80 or anti DNP-IgE. These results suggest that SPP may contain compounds with actions that inhibit mast cell degranulation in the rat.

  • We studied the effects of sulfasalazine(SSZ) on anaphylaxis. SSZ was found to exhibit a inhibitory activity on the compound 48/80-induced anaphylaxis. SSZ also inhibited local a naphylaxis activated by anti-dinitrophenyl(DNP) IgE. Moreover, SSZ dose-dependently inhibited histamine ralease in rat peritoneal mast cells activated by compound 48/80 or anti DNP IgE. We investigated the effects of SSZ on cAMP of rat peritoneal mast cell. The level of cAMP in rat peritoneal mast cells, when SSZ was added, transiently and significantly increased approximately 16-fold compared with that of basal cells. These results suggest that the antianaphylactic action of SSZ may be associated with an increase in the intracellular cAMP content of the mast cells as the result of an inhibition of the cAMP phosphodiesterase.

Detection of Escherichia coli O157:H7 Using Combined Procedure of Immunomagnetic Separation and Test Strip Liposome Immunoassay

  • A model system for the immnunochemical detection of Escherichia coli O157:H7 using a combined immunomagnetic separation (IMS) and test-strip liposome immunoassay (LIA) procedure was developed. Immunomagnetic beads coated with anti-E. coli O157 IgG antibodies were used to separate the E. coli O157 (including the H7 serotype) from culture. Immunoliposomes, whose surface was conjugated to goat anti-E. coli O157:H7 IgG and which encapsulated the marker dye, sulforhodamine B, were used as a detection label. The test strip, onto which antibodies to goat IgG were immobilized, was the immunosensor capturing immunoliposomes that did not bind to E. coli O157:H7 on the immunomagnetic bead-E. coli O157:H7 complexes. In experiments, pure cell culture suspensions of $10^5 E.$ coli O157:H7 organisms per ml produced a measurable signal inhibition, whereas a weak yet detectable signal inhibition occurred with $10^3CFU/ml$. The inhibition signals increased, when the incubation time for IMS was extended to 90 min and higher IgG-tag density (0.4mol%) was used on the liposomes. With 0.2 and 0.4mol% IgG-tagged liposomes, the IMS-LIA procedure showed more improved signal inhibitions than those of a direct (no IMS) LIA. The combined assay, which measures the instantaneous signal from immunoliposomes, can be completed within 90 min, making it significantly faster than conventional plating methods and enzyme-linked immunosorbent assay (ELISA). Accordingly, it is quite feasible to use the combined immunoassay format of IMS and dye-loaded immunoliposomes for the detection of E. coli O157:H7.
