The Effects of Lycium chinese Mill., Morus alba L. and Their Combination on the Asthmatic Murine Model (천식동물모델을 이용한 지골피(地骨皮), 상백피(桑白皮) 및 두 배합약물의 실험적 연구)

  • Objectives : To clarify the possible effect of Lycium chinese Mill (LC)., Morus alba L (MA)., and Lycium chinese Mill. +Morus alba L. (LC+MA), we have examined their influence on the development of pulmonary eosinophilic inflammation in the asthmatic murine model. Methods : Female Balb/c mice (5weeks) were immunized on two different days (21 days and 7 days before inhalational exposure) by intraperitonial injections of 0.2ml alum-precipitated Ag containing $100{\mu}g$ of OVA bound to 4 mg of aluminum hydroxide in PBS. Seven days after the second sensitization, mice were exposed to aerosolized ovalbumin for 30 minutes/day on 3 days/week for 8 weeks (at a flow rate of 250 L/min, 2.5% ovalbumin in normal saline) and, LC, MA, and LC+MA (500 mg/kg) were orally administered 3 times per a week for 8 weeks. Results : The suppressive effect of LC, MA, and LC+MA were demonstrated by the accumulation of eosinophills into airways, with the reduction of eosinophil, total lung leukocytes numbers. These were correlated with the marked reduction of IL-5, IL-13 and IL-4 levels in the BALF and serum. OVA-specific IgE levels were also decreased in serum and BAL from these mice. LC, MA, and LC+MA decreased eosinophil CCR3 expression and CD11b expression in lung cells. Conclusions : These results indicate that LC, MA, and LC+MA have high inhibitory effects on airway inflammation and hyper-responsiveness in the asthmatic murine model. The suppression of IL-5, IgE, eosinophil CCR3 expression and CD11b expression, and the increase of IFN-${\gamma}$ production in BALF seem to contribute to this effect. Hence, the results indicated that LC, MA, and LC+MA could act as a immuno-modulator which possesses anti-inflammatory and anti-asthmatic property by modulating the imbalance between Th1 and Th2 cytokines.

Anti-inflammatory Effects of Gamiyunjo-tang on Lipopolysaccharide Induced Inflammatory Responses in RAW 264.7 Cells (가미윤조탕(加味潤燥湯)이 LPS로 유도된 RAW 264.7 대식세포에서의 항염 효과 연구)

  • Objectives : Allergic disease has been well known as an IgE-dependent immunologic response. Recently, interest about the late inflammatory reaction has grown up as well as early allergic reaction characterized by IgE and mast cell. The purpose of this study was to find the anti-inflammatory effect of Gamiyunjo-tang(GMYJT) in allergic reaction. Methods : The experiment was performed using Raw 264.7 cells pretreated with GMYJT extracts. In this study, we observed the toxicity of cells by MTT analysis and measured the production of LPS-induced NO, $PGE_2$, IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ at a concentration of 50, 100, 200 and $400{\mu}g/ml$. Results : No toxicity of GMYJT (50, 100, 200, $400{\mu}g/ml$) on RAW 264.7 cells was found after 24 hours incubation. LPS-induced NO production was reduced after treatment with GMYJT (100, 200, $400{\mu}g/ml$)(P<0.05). $PGE_2$ was reduced after treatment with GMYJT (100, 200, $400{\mu}g/ml$)(P<0.05). IL-$1{\beta}$ did not decrease at any dose. IL-6 decreased at 200, $400{\mu}g/ml$(P<0.05). TNF-${\alpha}$ production decreased only at $400{\mu}g/ml$(P<0.05). Conclusions : These data suggest that GMYJT has anti-inflammatory effects in late allergic reaction.

  • Objective : This study was carried out for the purpose of proving the effect of anti-allergic efficacy on B cells and the mast cells of the BALB/C mouse by the abstraction from a Moutan cortex. Methods & Results : In order to know what the effect of an abstraction from Moutan cortex and about the expression of CD23 and IgE, IC-2 cell (mouse mast precursor cells that was dependent on IL-3), it was necessary to be activated. We then analyzed it from the flow of cytometry on the increase and the divorce of the B cells activated by anti-CD40. In order to know what the effect of it was on the organization of cytokine gene expression from the increase and divorce of the B cells and allergic acting by Moutan cortex, we found it necessary to examine the IC-2 cells and B cells. At the same time, as we examined the histamine release of IC-2 cells by ELISA method, we also examined the effect of Moutan cortex on the increase and divorce of the B cells by 3H-thymidine uptake method. We then analyzed the release of IL-4, IgE and histamine. Conclusions : As a results, Moutan Cortex promoted blood supply by extending the blood vessel of nasal mucous, which was contracted by the hypertrophied nasal mucous.

  • Gamipaidok-san(GPDS) is the Hyungbangpaidok-san prescription fortified with the additional ingredients known to be effective for dermatitis. So it has been used for atopic dermatitis in the clinic work actually. In this study, we investigated the effects of GPDS on in vitro and in vivo anti-allergic effect on RBL-2H3 rat basophilic leukemia Cells and on IgE-induced passive cutaneous anaphylaxis (PCA) in mice. The in vitro anti-inflammatory activity of GPDS in RAW 264.7 cells was investigated. GPDS potently inhibited $\beta$-hexosaminidase release from RBL-2H3 and the IgE-mediated PCA reaction in mice. GPDS inhibited LPS-induced NO and PGE2 production in a dose-dependent manner Furthermore, It also inhibited inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein expression in RAW 264.7 cells, and the activation of the transcription factor, NF-kB, in nuclear fraction. The antiallergic action of GPDS may originate from anti-inflammatory activities, and can improve the inflammation caused by allergies.

Anti-asthmatic activities of Cypress oil in a mouse model of allergic asthma (마우스 모델을 이용한 사이프러스 오일의 알러지성 천식 억제 효과)

  • This study was aimed to evaluate the effects of Cypress oil(CS) on anti-asthmatic activities in a mouse model of allergic asthma. Using an Ovalbumin-induced allergic asthma mouse model, 0.3% of CS was administered to experimental group using a nebulizer for 3 weeks on a basis of 3 times per week and 30min each time. The degree of airway hypersensitivity, the number of eosinophil in white blood cells, the number of immune cells and the change of cytokine in lung tissue were evaluated. The degree of airway hypersensitivity, the number of eosinophil, IL-5 and IL-13 levels in lung tissue, IgE in serum, the number of CCR3, CD3, CD4 cells were significantly decreased in experimental group treated with CS. These results suggested that CS may have a positive effects on Th2 cytokine and eosinophils which are major factors of asthma responses. Therefore CS might be of therapeutic value in treating asthma.

  • Acute disseminated encephalomyelitis (ADEM) and Guillain-$Barr{\acute{e}}$ syndrome (GBS) are both rare post-infectious neurological disorders. The co-existence of these conditions has often been reported despite of low incidence. We describe a 20-year-old male, who presented with acute flaccid paralysis and encephalopathy. The patient showed reversible MRI lesions suggesting ADEM. This case showed anti-GT1a IgG and anti-GM1 IgM antibodies positivity. We suggest that certain immunogenicity within central and peripheral nervous system may share a common autoimmune process during the disease course.

Specially, esosinophils and TNF-a in J+P combination group were significantly reduced in BALF and lung tissue.   • Experimental studies were done to research the clinical effects of Sopoongyangjetang on the Anti-allergic effect, Analgesic action and Anti-inflammatory action. The results obtained as follows ; 1. In effect of Sopoongyangjetang on vascular permeability responses to intradermal serotonin, it had tendency to decrease, but was none significant effect. 2. In effect of Sopoongyangjetang on vascular permeability responses to intradermal histamine, it revealed significant effect. 3. In the homologous PCA provoked by the IgE-like antibody against white egg albumin, Sopoonyangjetang showed significant effect. 4. In the delayed type hypersensitivity responses to picryl chloride, Sopoongyangjetang was proved significant effect. 5. In the delayed type hypersensitivity responses to SRBC, Sopoongyangjetang revealed significant effect. 6. In analgesic action by acetic acid method, Sopoongyangjetang was recognized significantly. 7. In Anti-inflammatory action by carrageenine method, Sopoongyangjetang showed significant effect.

Anti-atopic Activity of Sargassum micracanthum Ethanol Extracts (잔가시 모자반 에탄올 추출물의 항아토피 효과)

  • Atopic dermatitis (AD) is a common chronic inflammatory disease preceding the development of allergic disorders. The aim of this study was to evaluate the effect of Sargassum micracanthum ethanol extract (SMEE) on AD. AD was induced by spreading 2,4-dinitrochlorobenzen (DNCB) on the back sides of BALB/c mice. The efficacy of SMEE was tested by observing the skin clinical severity score, proliferations of Raw 264.7 cells and the secretion of cytokines and IgE. The secretion of IL-4, and IgE was significantly decreased by SMEE in a dose dependent manner, while IFN-${\gamma}$ was increased. In addition, SMEE alleviated the AD symptoms better when compared to the positive controls. In conclusion, these results suggest that SMEE has an inhibitory effect on AD, and may serve as a useful biomaterial for the development of cosmeceuticals.

A Study on the Immune Modulation of Chunghwatang(CHT) in Atopic Dermatitis Animal Models (아토피피부염 동물 병태 모델에서 청화탕(淸華湯)의 면역조절작용에 관한 연구)

  • Objectives : The aim of this study was to investigate the effect of Chunghwatang (CHT) for atopic dermatitis. Methods : CHT, a verified anti-oxidant and anti-inflammatory effect, was treated in atopic dermatitis animal model to investigate cytokine levels and immunoglobulin production. Results : Clinical skin index was 47.1%, suggesting significant efficacy of CHT in atopic dermatitis treatment. Serum IL-4, IL-6, IL-13, TNF-${\alpha}$ and histamine productions were significantly decreased to 52.3%, 61.8%, 68.0%, 37.4%, and 46.6%, respectively. The production of IL-5 was decreased to 33.3%. The increase of Immunoglobulin IgG1 production along with IgE through the interaction of IgE and IL-4 induced by IL-4 and IL-13 were measured as 20.7% and 23.4%, respectively. Conclusions : The results above, along with the in vitro test results, strongly supports the CHT samples as effective immunomodulator in AD treatments, suggesting its use in clinical practice and basic information for EBM.
