Study on The Anti-HSV (Herpes Simplex Virus) Activity of Korean Traditional Prescriptions (Herb complexes) (한약 탕제분획의 항 Herpes simplex virus 활성에 관한 연구)

  • Herpes simplex viruses(HSV) are one of the most common infectious virus of man. Though chemotherapies and antibiotics against HSV have been developed in many countries, but anti-HSV agents were not satisfactory to mankind by their toxic reaction and side effects. In order to search for anti-HSV agents from Korean traditional prescriptions, we extended the number of specimens. Both methanol extract and boilling water extract of the Korean traditional prescriptions were screened to detect anti-HSV activities by MTT assay. Korean traditional prescriptions showing anti-HSV activities as methanl extracts were Paekyopsan, Chesupwilyungtang, Yongdamsagantang, and prescription 11. Four methanol extracts showing anti-HSV activities were freationated by hexane and their efficacies were tested. Hexane freationations of Paekyopsan, Chesupwilyungtang, and prescription 11 showed in anti-HSV activities both haxane and methanol fractionation.

Prevalence of Anti-bovine Leukemia Virus Antibodies in Dairy and Korean Native Cattle (우리 나라 젖소 및 한우의 Bovine Leukemia Virus 항체 분포조사)

  • As a baseline study for the establishment of bovine leukemia virus(BLV)-free herd in Korea, the prevalence of anti-BLV antibody was determined in the present study. Sera from Korean native cows of 8 provinces and from dairy cattle of 9 provinces were subjected to enzyme-linked immunosorbent assay. Anti-BLV antibodies were positive in two (0.14%) of 1,413 Korean native cows. In contrast. 54.2% of 2,415 dairy cows were positive for anti-BLV antibodies, and their seropositive herd rate was 86.8%. And no differences were found in the sero-positive rates with age. The results indicate that the BLV infection rate has been increased continuously in Korea and that the establishment of BLV-free herd is imminent.

  • It is easy to access to the infinity information and programs, but it gives rise to the side effect. There are many side effects(ex. Hacking, Cracking, expose the personal information, etc). Nowadays, the computer virus raise the serious problems. The making program called Vaccine is work out a count measure. The Anti-Virus programs install on the client side computer and upgrade by downloading on the server's signature, the latest date, the program bound both of them is shown, but these programs have the defect that they have no remote control and no signature update because user's unconcern, This paper reported the research of existing virus infecting technology and the development of Web based Anti-Virus Scanner using the remote control on the internet server. Through this paper, I want to set up the counter measure for new virus easily, and to make more fast the vaccine for virus.

Detection of PED virus by the immunoelectron microscopy and immunogold conjugate immunoelectron microscopy (면역전자현미경(免疫電子顯微鏡)(IEM) 기법(技法) 및 immunogold conjugate 면역전자현미경(免疫電子顯微鏡)(IGC-IEM) 기법(技法)을 이용(利用)한 돼지 분변내 PED 바이러스의 검출(檢出))

  • Both immunoelectron microscopy(IEM) and immunogold conjugate immunoelectron microscopy (IGC-IEM) techniques were developed for the detection of porcine epidemic diarrhea virus(PEDV) from the feces. Fecal samples were incubated sequentially with anti-PEDV monoclonal antibody(MoAb) and immunogold conjugated goat anti-mouse IgG+IgM. Then negatively stained, mounted on the formvar carbon-coated copper EM grids and observed by the transmission electron microscope. By the direct electron microscopy(DEM), coronavirus particles were observed from 17 cases of total 33 fecal samples of grower pigs and sows. The virons of coronavirus were moderately pleomorphic but mostly spherical, with a diameter ranged from 90 to 190nm. PED virus particles were identified from 15 cases of 17 DEM positive samples by the IEM and IGC-IEM techniques. Aggregates of PED virus coated with specific antibody were seen in fecal samples incubated with homologous anti-PED virus MoAb but not in control samples incubated with anti-TGE virus MoAb. Following incubation with immunogold-conjugated secondary antibody, the gold granules were usually distributed around and among the virus particles and soluble and viral particle-associated antigen. So, IEM and IGC-IEM techniques were proved a rapid and sensitive methods for detection and identification of PED virus from fecal and intestinal contents.

  • To evaluate anti-influenza virus activity of 113 specimens of Korean traditional medicine both water and methanol extracts were examined using haemagglutination inhibition test. The water extract from Citrus junos was found to inhibit influenza virus A/Taiwan/l/86(H1N1). The survival rates of virus were determined by in situ cellular enzyme-linked immunosorbent assay. The water extract of Citrus junos was fractionated by chromatographic separating using Amberlite XAD-4, 40% MeOH and 60% MeOH layer had antiviral activity. The half inhibition concentration $(IC_{50})$ of 40% MeOH layer on survival of influenza virus was $MIC>361.5{\mu}g/ml$ and $IC_{50}$ value of fr. 40-4 fractionated from 40% MeOH layer was $677.19{\mu}g/ml$. These results suggested that the fractions of Citus junos have potent anti-influenza A virus activity.

Vitamin C Is an Essential Factor on the Anti-viral Immune Responses through the Production of Interferon-${\alpha}/{\beta}$ at the Initial Stage of Influenza A Virus (H3N2) Infection

  • L-ascorbic acid (vitamin C) is one of the well-known antiviral agents, especially to influenza virus. Since the in vivo antiviral effect is still controversial, we investigated whether vitamin C could regulate influenza virus infection in vivo by using Gulo (-/-) mice, which cannot synthesize vitamin C like humans. First, we found that vitamin C-insufficient Gulo (-/-) mice expired within 1 week after intranasal inoculation of influenza virus (H3N2/Hongkong). Viral titers in the lung of vitamin C-insufficient Gulo (-/-) mice were definitely increased but production of anti-viral cytokine, interferon (IFN)-${\alpha}/{\beta}$, was decreased. On the contrary, the infiltration of inflammatory cells into the lung and production of pro-inflammatory cytokines, tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-${\alpha}/{\beta}$, were increased in the lung. Taken together, vitamin C shows in vivo antiviral immune responses at the early time of infection, especially against influenza virus, through increased production of IFN-${\alpha}/{\beta}$.

Seroprevalence of Hepatitis A and B Virus Antibody of Employees among Three Companies with Different Health Policy (사업장 보건관리에 따른 A형, B형 간염 항체 양성률)

  • Objectives: We checked hepatitis A virus antibody(anti-HAV IgG) and hepatitis B virus antibody(HBsAb) in three large companies. The result could be a guideline to establish proper health policy for Hepatitis A and B virus preventive plan in company. Methods: We investigated the seroprevalence of anti-HAV IgG and HBsAb, and demographic characteristics of employee volunteer in three companies in southern area of Korea. Company was divided three according to health policy for hepatitis A and B. Results: The seroprevalence rate of anti-HAV IgG in company A, B, and C was 53.6%, 25.8%, and 17.7%(P<0.001), respectively. The seroprevalence rate of HBsAb in workplace A, B, and C was 79.7%, 82.4%, and 70.9%(P<0.001), respectively. Anti-HAV IgG showed more considerable difference among the companies. Conclusions: The results confirmed that low rate of IgG anti-HAV and HBsAb, particularly in the company that had low level of hepatitis education and vaccination program. This study was important for establishing hepatitis education policies, to prevent and control outbreaks in companies.

The Combined Anti-apoptotic Effect from Tamiflu and Pinoresinol of Forsythia fructus Extract Against Influenza Virus Infection (연교 추출물 Pinoresinol와 Tamiflu의 병용효과로부터 Influenza Virus 감염에 의한 세포사멸 억제효과)

  • The fruit body of Forsythiae Fructus (Oleaceae), a common Korean medical herb, is widely used in the treatment of cold and inflammation. In order to elucidate the action mechanism and the active principles from the plant against anti-influenza virus, the influenza virus hemagglutinin (HA) and neuraminidase (NA) gene RT-PCR and Viral Screening & Identification (VSI) assay were conducted, and the activity against viral replication was also investigated. Consequently, one active constituent, namely pinoresinol showed the in vitro antiviral principle using a cytopathic effect (CPE) reduction method, indicating pinoresinol possessed anti-influenza viral activity. Furthermore, combination of pinoresinol and Tamiflu exhibited higher activities than Tamiflu alone against influenza virus (H3N2) infection. The results suggested that combination of pinoresinol with Tamiflu could be a better candidate for an ant-H3N2 viral agent in the treatment of the influenza.

Antiviral Activity of Korean Traditional Prescriptions against Influenza Virus Type A (한약 처방 (복합체)의 Influenza Virus Type A에 대한 항바이러스 활성 효과)

  • In order to search for anti-influenza virus type A agents from Korean traditional prescriptions (herb complexes), we selected 63 traditional prescriptions, based on a review of the Korean traditional medicine books. Both methanol extracts and boiling-water extracts were tested, by means of the Haemagglutination Inhibition Test (HIT). Three of the 63 methanol extracts: CM-22, CM-26, CM-48 (see explanation of nomenclature below), showed efficacy against influenza virus type ACM-22 showed anti-influenza virus type A activity at the range of $313{\mu}g/ml$ to $9.75{\mu}g/ml$, CM-26 showed antiviral activity at the range of $156{\mu}l/ml$ to $4.87{\mu}g/ml$, CM-48 showed anti-influenza virus type A activity at the range of $625{\mu}g/ml$ to $19.5{\mu}g/ml$, respectively. Three of the water extracts: CW-14, CW-34, CW-61 were active. CW-14 showed anti-influenza virus type A activity at the range of l0mg/ml to $78{\mu}g/ml$, CW-34 showed antiviral activity at the range of 10mg/ml to $625{\mu}g/ml$ and CW-61 showed anti-influenza virus type A activity at the range of l0mg/ml to $313{\mu}g/ml$, respectively. In order to determine cytotoxicity of each extracts, chicken red blood cells were incubated with the various concentration of extracts of Korean traditional prescriptions. CW-14, CW-34 and CW-61 did not show cytotoxic effect against red blood cells whereas CM-22, CM-26 and CM-48 showed cytotoxic effect against red blood cells at the range of l0mg/ml to $625{\mu}g/ml$, 10mg/ml to $313{\mu}g/ml$ and 10mg/ml to $313{\mu}g/ml$, respectively. These results indicated that Korean traditional pres criptions may be inhibit either attachment of virus to cell surface receptor or penetration of the virus into cell during the initial stage of infection.
