Isolation of Antifungal Substances by Bacillus amyloliquefaciens IUB158-03 and Antagonistic Activity against Pathogenic Fungi (Bacillus amyloliquefaciens IUB158-03이 생산하는 항진균물질의 분리와 항균활성)

  • For the control of pathogenic microorganisms, Bacillus spp. were isolated from diseased pepper fruits in Korea. Among them, Bacillus sp. IUB158-03 showed high inhibitory effect on mycelial growth and spore germination of C. gloeosporioides and Botrytis cinerea. The strain was identified as B. amyloliquefaciens IUB158-03 based on its physiological, biochemical characteristics and Microlog analysis. The highest level of antifungal substances by B. amyloliquefaciens IUB158-03 were obtained when the bacterium was cultured in medium containing 2% soluble starch, 3% yeast extract, 0.5% tryptone, 0.5% $NH_4H_2PO_4$, and 1% NaCl (pH 6.0) at $25^{\circ}C$ for 72 hrs. The antifungal substances were purified by butanol extraction, silica gel column chromatography, preparative thin layer chromatography, and high performance liquid chromatography. The purified antifungal substance was confirmed $R_f$ 0.27 by TLC. This substance exhibited antifungal activity against Fusarium solani, Rhizoctonia solani, Botrytis cineria, Alternata alternaria of plant pathogenic fungi and Trichophyton mentagrophytes, Epidermophyton floccosum, Cryptococcus neoformans of human pathogenic fungi.

Role of Dopamine on Motility of Duodenal bulb in rabbits (토끼 십이지장구의 운동성에 미치는 dopamine의 영향)

  • The present study was undertaken to see an interaction of dopamine and cholecystokinin on spontaneous contractility of the small intestine including the duodenal bulb. A possible neural mechanism of the interaction was alto examined. The spontaneous isometric contractility of a segment of the duodenal bulb, duodenum, jejunum and ileum obtained from the rabbit anesthetized with ether was recorded in a chamber filled with Krebs-Ringer's solution. The solution was constantly kept at $37^{\circ}C$ and aerated with $O_2$ containing 5% $CO_2$. After 20 min from beginning of the contraction, dopamine $(10^{-4}M)$, CCK-8($10^{-8}M$), domperidone($10^{-5}M$) and tetrodotoxin ($10^{-6}M$) were administered into the chamber The following results were obtained by analyzing changes in the contractility of the intestinal segments. 1) Dopamine inhibited the spontaneous contractility of the duodenal bulb, duodenum, jejunum and ileum. The inhibitory action of dopamine on all parts of the small intestine except the ileum was reduced by tetrodotoxin. 2) Domperidone knwon to be a specific peripheral dopamine receptor antagonist blocked the inhibitory action of dopamine on all parts of the small intestine. The antagonistic action of domperidone on all parts of the small intestine except the ileum was completely abolished by tetrodotoxin. 3) CCK-8 reduced the inhibitory action of dopamine on all parts of the small intestine. The effect of CCK-8 on the dopamine action was diminished by tetrodotoxin. These results suggest that dopamine inhibits the spontaneous contractility of the small intestine including the duodenal bulb and CCK-8 reduces the inhibitory action of dopamine through the enteric nervous system.

Inhibition of Hydrogen Sulfide-induced Angiogenesis and Inflammation in Vascular Endothelial Cells: Potential Mechanisms of Gastric Cancer Prevention by Korean Red Ginseng

  • Previously, we reported that Helicobacter pylori-associated gastritis and gastric cancer are closely associated with increased levels of hydrogen sulfide ($H_2S$) and that Korean red ginseng significantly reduced the severity of H. pylori-associated gastric diseases by attenuating $H_2S$ generation. Because the incubation of endothelial cells with $H_2S$ has been known to enhance their angiogenic activities, we hypothesized that the amelioration of $H_2S$-induced gastric inflammation or angiogenesis in human umbilical vascular endothelial cells (HUVECs) might explain the preventive effect of Korean red ginseng on H. pylori-associated carcinogenesis. The expression of inflammatory mediators, angiogenic growth factors, and angiogenic activities in the absence or presence of Korean red ginseng extracts (KRGE) were evaluated in HUVECs stimulated with the $H_2S$ generator sodium hydrogen sulfide (NaHS). KRGE efficiently decreased the expression of cystathionine ${\beta}$-synthase and cystathionine ${\gamma}$-lyase, enzymes that are essential for $H_2S$ synthesis. Concomitantly, a significant decrease in the expression of inflammatory mediators, including cyclooxygenase-2 and inducible nitric oxide synthase, and several angiogenic factors, including interleukin (IL)-8, hypoxia inducible factor-1a, vascular endothelial growth factor, IL-6, and matrix metalloproteinases, was observed; all of these factors are normally induced after NaHS. An in vitro angiogenesis assay demonstrated that NaHS significantly increased tube formation in endothelial cells, whereas KRGE pretreatment significantly attenuated tube formation. NaHS activated p38 and Akt, increasing the expression of angiogenic factors and the proliferation of HUVECs, whereas KRGE effectively abrogated this $H_2S$-activated angiogenesis and the increase in inflammatory mediators in vascular endothelial cells. In conclusion, KRGE was able to mitigate $H_2S$-induced angiogenesis, implying that antagonistic action against $H_2S$-induced angiogenesis may be the mechanism underlying the gastric cancer preventive effects of KRGE in H. pylori infection.

  • The toxicities of binary mixtures of the four insecticides acephate, demeton-S-methyl, cypermethrin and pirimicarb to the two strains of green peach aphid (Myzus persicae Sulz.) resistant to acephate and demeton-S-methyl were investigated and compared to the toxicities of their individual insecticides. The synergistic action of the insecticide mixtures to the insects were greatly varied with the kind of insecticide combinations, their mixture ratios, and the origin of resistance by an insecticide. The maximum synergistic action of acephate for the acephate resistant strain was obtained at 1:1 mixed with demeton-S-methyl. However, there were some antagonistic effects in all acephate mixtures with pirimicarb and cypermethrin. With the strain of demeton-S-methyl resistance, acephate, cypermethrin, and pirimicarb were synergized at the given mixture ratios by demeton-S-methyl. The maximum synergistic effect was observed at 2:3 with acephate, 1:1 with cypermethrin and 3:2 with pirimicarb.

  • Antagonistic microorganisms from rhizosphere soil were isolated, identified, and applied successfully as the biocontrol agents of damping-off caused by Rhizoctonia spp. Rhizosphere antagonists isolated from rhizosphere soil were identified as Trichoderma viride, T. harzianum, T. hamatum, T. polysporum, Gliocladium sp., Pseudomonas fluorescence, P. stutzeri, P. cepacia, Enterobacter sp., Serratia sp. and Erwinia herbicola. Of these, the most promising ones in vitro were T. virdie, T. harzianum, Gliocladium sp., Serratia sp., P. stutzeri, and P. cepacia. These above six antagonists were efficient in reducing disease incidence to $40{\sim}70%$ when the reselected rhizosphere antagonists preparations were applied to the soil at $10^6$ propagules per gram. Among six antagonists, T. viride was the most promising biocontrol agents against R. solani isolates in soil. The suppressive effect was more evident in steam-sterilized soil than in non-sterilized field soil.

  • Chrysanthemum indicum L. (Asteraceae) is a traditional herbal medicine that has been used for the treatment of inflammation, hypertension, and respiratory diseases due to its strong antagonistic activity against inflammatory cytokines. The effects of Chrysanthemum indicum L. Extract (CIE) for increasing cell growth, alkaline phosphatase (ALP) activity, and collagen content were totally inhibited, suggesting that the effect of CIE might be partly involved with estrogen activity. Furthermore, the protective effects of CIE on the response of osteoblasts to oxidative stress were evaluated. Osteoblastic MC3T3-E1 cells were incubated with hydrogen peroxide and/or CIE, and markers of osteoblast function and oxidative damage were examined. CIE significantly increased cell survival, ALP activity, and calcium deposition, and decreased the production of Reactive Oxygen Species (ROS) and Tumor Necrosis Factor-${\alpha}$ (TNF-${\alpha}$) in osteoblasts. Taken together, these results indicate that the enhancement of osteoblast function by CIE may prevent osteoporosis and inflammatory bone diseases.

  • Bacillus subtilis KMU-13 was isolated from the Lillehammer forest soils at Norway and shown a strong antifungal activity on cucumber scab, Cladosporium cucumerinum KACC 40576. B. subtilis KMU-13 produced a maximum level of antifungal substance under incubation aerobically at $30^{\circ}C$, 180 rpm for 48 hours in LB broth containing 0.5% maltose and 0.5% bactopeptone and initial pH adjusted to 6.0. Butanol extract of cultured broth was confirmed inhibitory zone by plate assay and Rf 0.64 value substance by thin layer chromatography (TLC) represented high antifungal activity against C. cucumerinum KACC 40576 and also shown fungal growth inhibitory activity against Botytis cinerea KACC 40573, C. gloeosporioides KACC 40804, D. byoniae KACC 40669, F. oxysporum KACC 40037, F. oxysporum KACC 40052, F. oxysporum f. sp. radicis-lycopersici KACC 40537, F. oxysporum KACC 40902, M. cannonballus KACC 40940, P. cambivora KACC 40160, R. soiani AG-1 KACC 40101, R. solani AG-4 KACC 40142, and S. scleotiorum KACC by agar diffusion method.

Synbiotic Potential of Yoghurt Manufactured with Probiotic Lactic Acid Bacteria Isolated from Mustard Leaf Kimchi and Prebiotic Fructooligosaccharide (갓김치로부터 분리한 Probiotic 유산균과 Prebiotic Fructooligosaccharide로 제조한 요구르트의 Synbiotic 가능성)

  • In the present work, the influence of prebiotic fructooligosaccharide (FOS) on adhesion to Caco-2 cells, viability, acid and bile tolerance, antibacterial, antioxidant, enzymatic, and metabolic activities of the probiotic starters Lactobacillus acidophilus GK20 and Lactobacillus paracasei GK74, has been explored. Experiments were conducted with fermented yoghurt over a period of 7 days at $4^{\circ}C$. When compared to control fermentations without prebiotic, the addition of FOS was seen to significantly (p<0.05) increase the viable cell counts of the probiotics, overall viscosity, and concurrently reduce the pH of the fermented yoghurts. Both Escherichia coli ATCC 11229 and Salmonella enteritidis ATCC 13076 were inhibited by the probiotics' antibacterial activities, while the synbiotic yoghurt containing mixed probiotics and FOS was noted to highly improve antagonistic action. When fermented with mixed starters, the addition of FOS (1.0%) resulted in the highest proteolytic ($1.06{\pm}0.06$ unit) and ${\beta}$-galactosidase activities ($20.14{\pm}0.31$ unit). However, FOS did not affect acid and bile tolerance, adhesion to Caco-2 cells or the antioxidant activity of the probiotics, although both L. acidophilus GK20 and L. paracasei GK74 had functionality as probiotic strains. Hence, a significant synbiotic effect was observed in fermented yoghurt after 7 days of storage at $4^{\circ}C$, and as a result, such synbiotic yoghurt can be said to possess synergistic actions which improve the gastrointestinal environment and promote of health.

Suppression of Meloidogyne arenaria by different treatments of Pasteuria penetrans (Pasteuria penetrans의 처리방법에 따른 땅콩뿌리혹선충 (Meloidogyne arenaria) 방제효과)

  • This study was investigated to compare the suppression of Meloidogyne arenaria by different treatments of Pasteuria penetrans which is known for biological control agent against Meloidogyne spp.. In order to select proper number of P. penetrans showing good suppression effect, P. penetrans were mixed with M, arenaria for attachment using three different concentration such as $3{\times}10^4$, $3{\times}10^5$ and $3{\times}10^6$ endospores/5 g medium, followed by treating them onto the roots of tomato. After 14 weeks incubation, P. penetrans at $3{\times}10^6$ endospores showed highest activity against the formation of gall caused by M, arenaria. At a dose of $3{\times}10^5$ endospores/5 g medium, P. penetrans was treated into soil either mixing with soil or spray onto soil surface for comparing of suppressive efficacy. When the antagonistic bacterium was treated by the former method, it suppressed more effectively Using P. penetrans at $3{\times}10^6$ endospores and mixing with soil method, suppression was compared among P. penetrans, $PASTORIA^{(R)}$(Japan) and $Fosthiazate^{(R)}$(Korea). P. penetrans was more potent than $PASTORIA^{(R)}$(Japan) and as similar as $Fosthiazate^{(R)}$(Korea). Therefore, these results suggested that P. penetrans can be used for controling of M. arenaria as biological control agent. Furthermore, thess results can be provided to develop environmentally-friendly nematicide.

  • The objective of this study was to achieve biological control of green mold disease in Pyogo mushrooms using antagonistic microorganisms. Bacillus subtilis BSM320 cells inhibited mycelial growth by 48-60% against three Trichodermaisolates including T. hazianumisolated from the substrates of Lentinula edodes, showing their antifungal activity.The bacteria were cultured to a high density of 4.2 × 109±113.7 cfu/mlin aqueous extract of composted spent mushroom substrates of L. edodes containing 1% glucose and showed a higher growth rate than that observed when using the commercial medium, Luria-Bertani broth. The bacterial culture showed a 75% protective effect without damaging the mushroom fruiting bodies. These results suggest that B. subtilis BSM320culture is suitable for biological control of green mold disease during mushroom cultivation.
