The plasma membrane and microsomes, isolated from the cells treated with hematoporphyrm derivative (HpD) for 1 and 24 h, accumulated the aggregated porphyrin. The quantity of aggregated porphyrin was same in the plasma membrane and microsomes after isolating them from cells treated with HpD for 1 h whereas the microsomes accumulated higher quantity of aggregated porphyrin when cells were treated with HpD for 24 h. Photodynamic action of aggregated porphyrin on plasma membrane and microsomes was investigated using lipid specific fluorescent probes: 1,6-diphenyl-1,3,5-hexatrine (DPH) and 1-(4-trimethylammonium), 6-diphenyl-1,3,5-hexatrine(TMA-DPH). The time dependent anisotropy of these probes in the membranes was measured and the decay of anisotropy was analyzed using wobbling in cone model. Upon irradiation both the plasma membrane and the microsomes showed an increase in the limiting anis~)tropy and order parameter and a decrease in the cone angle of the lipid probes. The increase in the limiting anisotropy was pronounced in membranes isolated from the cells treated with HpD for 24 h. Photoinduced change in the limiting anisotropy was dependent on the duration of incubation of cells with HpD before isolating the membranes. In both the membranes. the membrane core was affected more as compared to the outer leaflet. In addition to the structural changes, a decrease in Na$^+$-K$^+$-ATPase and NADPH cyt c reductase activity was also observed upon irradiation of HpD treated cells. Inhibition in NADPH cyt c reductase was more when cells were treated with HpD for 24 h, however, Na$^+$-K$^+$-ATPase activity did not depend on the duration of the treatment of cells with HpD before irradiation. Our results suggest that the extent of photoinduced perturbations in the membranes varies as a function of duration of the treatment of cells with HpD and the membrane core is more susceptible to the photodynamic action of aggregated porphyrin.
Conformational heterogeneity of directly linked multiporphyrin arrays with larger molecular length retards their utilities in practical applications such as two-photon absorption and molecular photonic wire. In this regard, here we adopted a way to overcome the conformational heterogeneity through hydrogen bonding by selective binding of meso aryl substituents of porphyrins (host) with urea (guest) to form helical structure. Using steady-state and time-resolved spectroscopy, we observed the enhanced fluorescence quantum yield by ~1.8 to 2.4 times, enhanced anisotropy values and the disappearance of fast fluorescence decay component in the host-guest helical forms. In addition, the enhanced nonlinear optical responses of helical arrays infer the extended inter-porphyrin electronic coupling due to a significant change in dihedral angle between the neighboring porphyrin moieties. The current host-guest strategy will provide a guideline to improve the structural homogeneity of the photonic wire.
Photoresponsive side chain polymers containing aminonitro azobenzene were synthesized for studying optically induced birefringence. Four different copolymers were prepared using methacrylate, a-methylstyrene, and itaconate monomer. Two copolymers are totally amorphous and the other two are liquid crystalline in nature. Trans-to-cis photoisomerization was observed under the exposure of UV light with UV-VIS absorption spectroscopy. Reorientation of polar azobenzene molecules induced optical anisotropy under a linearly polarized light at 532 nm. The dynamic parameters of optically induced birefringence let us compare the effect of polymeric structure on the rate of growth and decay of the birefringence. Besides the effect of glass transition temperature on the dynamics of photoinduced birefringence, we focused our interests on the geometrical hindrance of polar azobenzene molecules and cooperative motion of environmental mesogenic molecules in the vicinity of polar azobenzene moiety.
Molecular dynamics simulation studies for thermotropic liquid crystalline systems conposed of rodlike molecules with 6 Lennard-Jones interaction sites wre performed in NPT ensemble. Within the range of temperature studied, the system exhibited isotropic and smectic phase. For the characterization of the smectic phase, we examined the structure of the liquid crystalline phase via the radial distribution function, its longitudinal and transverse components to the director, and other orientational correlation function, its longitudinal and transverse components to the director, and other orientational correlation functions. In the smectic A phase, our results showed a large anisotropy in translational motion (i.e.,$D_⊥ >> D_∥$), and the decay of the collective orientational correlation function of rank two became slower than that of the single particle orientational correlation function of rank one. Comments on the spontaneous growth of orientational order directly from the isotropic phase are given.
We extended the measurable time scale of DNA dynamics to microsecond using $[Ru(phen)_2(dppz)]^{2+}$ (phen = 1,10-phenanthroline, dppz = dipyrido[3,2-a:2',3'-c]phenazine) (RuPD), which displays a mean lifetime near 500 ns. To evaluate the usefulness of this luminophore (RuPD) for probing nucleic acid dynamics, its intensity and anisotropy decays when intercalated into supercoiled and linear pBluescript (pBS) II SK(+) phagemids were examined using frequency-domain fluorometry with a blue light-emitting diode (LED) as the modulated light source. The mean lifetime for the supercoiled phagemids (< $\tau$ > = 489.7 ns) was somewhat shorter than that for the linear phagemids (< $\tau$ > = 506.4 ns), suggesting a more efficient shielding from water by the linear phagemids. The anisotropy decay data also showed somewhat shorter slow rotational correlation times for supercoiled phagemids (997.2 ns) than for the linear phagemids (1175.6 ns). The slow and fast rotational correlation times appear to be consistent with the bending and torsional motions of the phagemids, respectively. These results indicate that RuPD can have applications in studies of both bending and torsional dynamics of nucleic acids.

$[Ru(bpy)_2(dppz)]^2+$ (bpy=2,2'-bipyfidine, dppz=dipyrido[3,2-a:2',3'-c]phenazine) (RuBD), a long-lifetime metal-ligand complex, displays favorable photophysical properties. These include long lifetime, polarized emission, but no significant fluorescence from the complex that is not bound to DNA. To show the usefulness of this luminophore (RuBD) for probing the bending and torsional dynamics of nucleic acids, its intensity and anisotropy decays when intercalated into supercoiled and relaxed pTZ18U plasmids were examined using frequency-domain fluorometry with a blue light-emitting diode (LED) as the modulated light source. The mean lifetimes for the supercoiled plasmids (< $\tau$ >=148 ns) were somewhat shorter than those for the relaxed plasmids (< $\tau$ >=160 ns). This suggests that the relaxed plasmids were shielded more efficiently from water. The anisotropy decay data also showed somewhat shorter slow rotational correlation times for supercoiled plasmids (288 ns) than for the relaxed plasmids (355 ns). The presence of two rotational correlation times suggests that RuBD reveals both the bending and torsional motions of the plasmids. These results indicate that RuBD can be useful for studying both the bending and torsional dynamics of mucleic acids.
We have studied electro-optic characteristics as a function of splay elastic constants ($K_{11}$) in the fringe-field switching (FFS) mode using the LC with positive dielectric anisotropy. When $K_{11}$ is increased from 7.7pN to 11.7pN, a maximum transmittance is slightly increased and rising time become a little bit fast. However, operating voltage and threshold voltage is independent. In opposition to rising time, decay time is not affected by $K_{11}$. We already know that $K_{11}$ affects tilt angle of liquid crystals. Therefore, on the occasion of high $K_{11}$, liquid crystals are mainly affected by twist deformation because the higher $K_{11}$, the less tilt angle. In the FFS device, high $K_{11}$ is favorable to reduce tilt angle in on state and thus improve rising response time.
We have studied a field sequential liquid crystal display (FSLCD) with electrically controlled birefringence (ECB). The ECB mode exhibiting fast response time, high transmittance, low operating voltage and adequate viewing angle. The positive liquid crystal (LC) is better than negative LC on dielectric anisotropy, birefringence and rotational viscosity. Because $K_{11}$ value out of the elastic coefficient of LC is also larger than $K_{22}$ value, decay time of VA-ECB is advantage. However, the transmittance & response time reduced with decreasing the cell gap. This drawback can be overcome by using LC with high ${\Delta}n$ but VA-ECB is occured loss of light efficiency because the $\gamma$ value increased by high ${\Delta}n$ of LC. Consequently, the HA-ECB mode is one of strongest candidate for FSLCD application.

In this paper, a complement to the Hoek-Brown criterion is proposed in order to derive the strength of anisotropic rock from strength of the corresponding truly intact rock. The complement is a decay function, which unlike other modifications or suggestions made in the past, is multiplied to the function of the original Hoek-Brown failure criterion for intact rock. This results in a combined and extended form of the criterion which describes the strength of anisotropic rock as a varying fraction of the corresponding truly intact rock strength. Statistical procedures and in particular regression analyses were conducted into data obtained in experiments conducted in the current research program and those collected from the literature in order to define the Hoek-Brown's criterion complement. The complement function was best described by a simple polynomial including only three constants to be empirically evaluated. Further investigations also showed that these constants can be related to the other readily available parameters of rock material which further facilitate determining the constants. A great and prime advantage of the proposed complement is that it is mathematically simple including the least possible number of empirical constants which are easily estimated with minimum experimental effort. Moreover, proposed concept does not suggests any change to the original Hoek-Brown criterion itself or its constants and serves whenever anisotropy does exist in the rock. This further implies on the possibility of using any other failure criterion for intact rock in conjunction with the compliment to reach the strength of anisotropic rock.
