Antitumor Activities of Red Ginseng Acidic Polysaccharide(RGAP) as an Immunomodulator

  • A red ginseng acidic polysaccharide(RGAP) with immunomodulating antitumor activities was isolated from Korean red ginseng, The molecular weight of RGAP was estimated to be 12-450 kDa by gel filtration chromatography, RGAP was found to increase survival rate and to inhibit of tumor growth significantly in a dose dependent manner in mice transplanted with tumor cells. RGAP significantly promoted nitric oxide(NO) production from peritoneal macrophages bothin vivo and in vitro. Western blot analysis exhibited a newly synthesized inducible nitric oxide synthase(iNOS) protein band in the RGAP treated group. It seems likely that immunomodulating antitumor activities of RGAP are mainly mediated by NO production of macrophage. RGAP was further purified by ultrafiltration and anion exchange chromatography on DEAE-sepharose, followed by gel filtration on Sephacryl S-300 to give an active fraction(GFP) with stronger NO production in murine macrophages. GFP increased survival rate ten times compared to RGAP in male ICR mice transplanted with sarcoma 180 and also showed more potent tumoricidal activities of natural killer cells than RGAP. Sugar $composition(mol\%)$ of GFP was found to be arabinose:rhamnose:xylose:galacturonic acid:mannose:galactose:glucose(10:9:1:25:8:20:27) by GC/MS. The results suggest that clinical trials of RGAP in immunotherapy against cancer are highly feasible.

Hydrogen Production from Water Electrolysis Driven by High Membrane Voltage of Reverse Electrodialysis

  • The voltage produced from the salinity gradient in reverse electrodialysis (RED) increases proportionally with the number of cell pairs of alternating cation and anion exchange membranes. Large-scale RED systems consisting of hundreds of cell pairs exhibit high voltage of more than 10 V, which is sufficient to utilize water electrolysis as the electrode reaction even though there is no specific strategy for minimizing the overpotential of water electrolysis. Moreover, hydrogen gas can be simultaneously obtained as surplus energy from the electrochemical reduction of water at the cathode if the RED system is equipped with proper venting and collecting facilities. Therefore, RED-driven water electrolysis system can be a promising solution not only for sustainable electric power but also for eco-friendly hydrogen production with high purity without $CO_2$ emission. The RED system in this study includes a high membrane voltage from more than 50 cells, neutral-pH water as the electrolyte, and an artificial NaCl solution as the feed water, which are more universal, economical, and eco-friendly conditions than previous studies on RED with hydrogen production. We measure the amount of hydrogen produced at maximum power of the RED system using a batch-type electrode chamber with a gas bag and evaluate the interrelation between the electric power and hydrogen energy with varied cell pairs. A hydrogen production rate of $1.1{\times}10^{-4}mol\;cm^{-2}h^{-1}$ is obtained, which is larger than previously reported values for RED system with simultaneous hydrogen production.

Development of indirect ELISA for the detection of canine adenovirus type 2 antibodies in dog sera

  • Background: Canine adenovirus type 2 (CAV-2) induces infectious laryngotracheitis in members of the family Canidae, including dogs. To date, no ELISA kits specific for CAV-2 antibody have been commercialized for dogs in Korea. Objectives: We aimed to develop new indirect enzyme-linked immunosorbent assay (I-ELISA) to perform rapid, accurate serological surveys of CAV-2 in dog serum samples. Methods: In total, 165 serum samples were collected from dogs residing in Chungbuk and Gyeongbuk provinces between 2016 and 2018. The Korean CAV-2, named the APQA1701-40P strain, was propagated in Madin-Darby canine kidney cells and purified in an anion-exchange chromatography column for use as an antigen for I-ELISA. The virus-neutralizing antibody titers of CAV-2 in the dog sera were measured by virus neutralization (VN) test. Results: We compared the results obtained between the VN and new I-ELISA tests. The sensitivity, specificity, and accuracy of new I-ELISA were 98.6%, 86.4% and 97.0% compared with VN test, respectively. New I-ELISA was significantly correlated with VN (r = 0.91). Conclusions: These results indicate that new I-ELISA is useful for sero-surveillance of CAV-2 in dog serum.

Direct determination of gadolinium in urania-gadolinia nuclear fuels by inductively coupled plasma atomic emission spectrometry (유도결합플라스마 원자방출분광법을 이용한 UO2-Gd2O3 핵연료 중 가돌리늄 분석)

  • The urania-gadolinia fuels were dissolved with nitric acid. The analytical conditions of ICP-AES for the direct determinations of gadolinium in the uranium matrices without separation process were investigated. Based on the effect of uranium on gadolinium intensity, the best wavelength for gadolinium was 336.223 nm. The relative deviation of two methods, direct and indirect measurements with anion exchange chromatography, was less than 5 %. Therefore it was possible for this procedure directly to measure 5~10 wt.% of gadolinium in urania-gadolinia fuels without separation by ICP-AES.

The Antifungal Efficacy of Extracts Derived from Kimchi Filtrates

  • Secondary metabolites in the culture filtrates of lactic acid bacteria offer varied chiral moieties, making them a valuable resource for drug design scaffolding. Our previous methodology included using a combination of anion exchange resins, Amberlite IRA-67 and Purolite A420S, to purify significant quantities of Lactobacillus plantarum LBP-K10 peptidyl compounds. However, current experimental evidence regarding the impact of native culture extracts and/or filtrates on pathogenic fungi in vivo/in vitro is insufficient. This study analyzed the antifungal properties of two different probiotic cultures: the CH2Cl2-extracted filtrate of Chinese cabbage kimchi (CH2Cl2-extracted CCKWLB and CH2Cl2-extracted CCKWOLB) and the non-extracted filtrate of Chinese cabbage kimchi (non-extracted CCKWLB and non-extracted CCKWOLB). The samples were divided into two groups: one group was inoculated with probiotics while the other group remained non-inoculated. Filtrates from both experimental groups were utilized for antifungal assays. The treatments employing CCKWLB, with an initial inoculation of Lb. plantarum LBP-K10 as a starter, demonstrated significant antifungal activity under various experimental conditions. Our study offers new perspectives on the antifungal properties of CH2Cl2-extracted kimchi filtrates, which are naturally produced by lactobacilli. The efficacy of antifungal compounds is supported by substantial evidence demonstrating their efficient uptake by cells and the antifungal properties exerted by metabolites.

Protein Patterns on a Vaginal Mucus during Spontaneous and Estrus Synchronization using CIDR in Korean Native Cattle (Hanwoo)

  • The aim of the present recent study was to compare the protein patterns in the vaginal mucus of Hanwoo cattles during spontaneous and CIDR induced-estrus. Ten cattles, who had been observed in estrus, received no treatment and served as the group of cattles with normal spontaneous estrus. Thirteen cattles in the CIDR received an CIDR insert on day 14 were removed and cattles were injected GnRH on day 15. Vaginal mucus samples were collected from all cattles at the same time the single AI in cattles with spontaneous estrus and the AI in cattles with induced estrus. Spontaneous and CIDR-induced estrus vaginal mucus samples were analyzed on two different array surfaces: cation-exchange (CM10), anion-exchange (Q10). In addition, using the NaCl solution by which the proteins combined after washing are 0.5, 1 and 2 M, it was fractionated and a protein was collected successively. The results are summarized as follows: 1) Ionic surfaces chemistries (Q10 and CM10) gave the best results in terms of detectable protein peaks, with more than 100 protein peaks in the two fractions and under each condition. 2) Protein mass spectrometer using 11 different proteins in protein identification of 7 were able to determine the protein. List of identified proteins as follows; Ribosome-binding protein 1, GRIP 1-associated protein 1, Katanin p60 ATPase-containing subunit A-like 1, Protein FAM44A, DUF729 domain-containing protein 1, Prolactin precursor, Dihydrofolate erductase. Conclusively, on the basis of this study, protein expression in the vaginal mucus could be used as an indicator for time of estrus manifestation in order to increase conception rates by applying AI at an optional time.

Effect of pH on PAH Transport in Brush Border Basolateral Membrane Vesicles of Rabbit Proximal Tubule (가토 신장 근위세뇨관의 Brush Border 및 Basolateral Membrane Vesicle에서 PAH 이동에 미치는 pH의 영향)

  • The effect of pH on the rate of PAH uptake was studied in rabbit renal basolateral membrane vesicles (BLMV) and brush border membrane vesicles (BBMV). In the absence of Na in incubation medium, a decrease in external $pH(pH_0)$ led to an increase in probenecid-sensitive PAH uptake by BLMV. In the presence of Na, the probenecid-sensitive PAH uptake was unaltered when the $pH_0$ decreased from 8.0 to 6.0 but further decrease in $pH_0$ to 5.5 increased significantly the uptake. The probenecid-sensitive PAH uptake was not affected by an alteration in pH per se in the absence of a pH gradient with or without the presence of Na. However, the presence of Na stimulated the probenecid-sensitive PAH uptake in all pH ranges tested over that measured in the absence of Na. A similar pattern of pH dependence on the PAH uptake was observed in BBMV but the presence of Na did not alter the probenecid-sensitive PAH uptake in the presence and absence of a pH gradient. Kinetic analysis for BLMV showed that Na or pH gradient increased Vmax of the probenecid-sensitive PAH uptake without a change in Km value. These results suggest that PAH is transported by $OH^-/PAH$ exchange process in the luminal membrane, but the pH dependence in the BLMV is not unequivocally consistent with an anion exchange process. The PAH transport is dependent on Na in BLMV but not in BBMV.

  • Copolymerization of the 4-vinylpyridine with vinylacetate and divinylbenzene initiated by azobis-isobutyronitrile was carried out in DMF in presence $BaCl_2$ at $98^{\circ}C$. Ion exchange res in, poly 4-vinylpyridine-vinylsulfonic acid-divinylbenzene was prepared by sulfonation of 4-vinylpyridine-vinylacetatp-divinylbenzene with concentrated sulfuric acid. The compositions of each synthetic resin were identified by means of ir adsorption spectroscopy. Anion and cation capacities of 4-vinylpyridine-vinylsulfonic acid-divinylbenzene ion exchanger were 2.5meq/g and 4.8meq/g, respectively. Adsorption of Cd(II) and Cu(II) ions have showed larger quantity in alkalie media. A study also was made of the influence of alcohol on the distribution coefficient of Cd(II) and Cu(II) ions between the synthetic ion exchanger, and solution containing hydrochloric acid, various alcohols and water.   • This study was conducted to efficiently separate the Ig from Korean native cattle colostrum and to utilize them as an immunogen for the production of antibodies aginst rabbit. The results obtained were as follows : 1. About 84% of Ig G could be separated from Korean native cattle colostrum by·gel filtration using Superose 12 column on HPLC. The separation profile of Korean native cattle colostral immunoglobulin was similar that of Holstein colostral Ig. 2. Separation of Korean native cattle colostral Ig by anion exchange chromatography using Mono Q column on HPLC was poor resolution chromatographic pattern. 3. Hi-Trap Protein G column showed better results than the Protein A Sepharose CL-4B column in the Ig G binding capacity from Korean native cattle colostral Ig. 4. Protein G Sepharose Fast Flow system resulted in higher Ig g binding capacity as the industrial size scale-up approach. 5. Sufficient titer reaction of antibody to Korean native cattle colostral Ig G was confirmed by ELISA.

Peptide Profiling and Selection of Specific-Expressed Peptides in Hypoglycemic Sorghum Seed using SELDI-TOF MS (SELDI-TOF MS를 활용한 혈당강하 수수 종자의 펩타이드 프로파일링 및 특이 발현 펩타이드 선발)

  • Sorghum seed is traditionally used as secondary food sources in addition to rice in Korea. While the hypoglycemia regulating phytochemicals have been found in sorghum seed, peptides related with hypoglycemia never been studied before. To obtain the peptide characteristics and the specifically high-expressed peptides in hypoglycemic sorghum seed, peptide profiles of seven hypoglycemic and five non-hypoglycemic sorghum lines bred in RDA were determined using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF MS). The twelve sorghum lines exhibited 104 peptides on CM10 protein chip array (weak cation exchange) and 95 peptides on Q10 (weak cation exchange) in the molecular mass range from 2,000 to 20,000 Da. Heat map via supervised hierarchical clustering of the significantly different peptides (p < 0.01) in peak intensity among the 12 lines effectively revealed the specifically upregulated peptides in each line and distinguished between 7 hypoglycemic and 5 non-hypoglycemic lines. Through the comparison with hypoglycemic and non-hypoglycemic lines, 10 peptides including 2231.6, 2845.4, 2907.9, 3063.5, 3132.6, 3520.8, 4078.8, 5066.2, 5296.5, 5375.5 Da were specifically high-expressed in hypoglycemic lines at p < 0.00001. This study characterized seed peptides of 12 sorghums and found ten peptides highly expressed for hypoglycemic sorghum lines, which could be used as peptide biomarkers for identification of hypoglycemic sorghum.
